Absent MicroRNAs in Different Tissues of Patients with Acquired Cardiomyopathy

MicroRNAs （miRNAs） can be found in a wide range of tissues and body fluids, and their specific signatures can be used to determine diseases or predict clinical courses. The miRNA profiles in biological samples （tissue, serum, peripheral blood mononuelear cells or other body fluids） differ significantly even in the same patient and therefore have their own specificity for the presented condition. Complex profiles of deregulated miRNAs are of high interest, whereas the importance of non-expressed miRNAs was ignored. Since miRNAs regulate gene expression rather negatively, absent miRNAs could indicate genes with unaltered expression that therefore are normally expressed in specific compartments or under specific disease situations. For the first time, non-detectable miRNAs in different tissues and body fluids from patients with different diseases （cardiomyopathies, Alzheimer＇s disease, bladder cancer, and ocular cancer） were analyzed and compared in this study, miRNA expression data were generated by microarray or TaqMan PCR-based platforms. Lists of absent miRNAs of primarily cardiac patients （myocardium, blood cells, and serum） were clustered and analyzed for potentially involved pathways using two prediction platforms, i.e., miRNA enrichment analysis and annotation tool （miEAA） and DIANA miRPath. Extensive search in biomedical publication databases for the relevance of non-expressed miRNAs in predicted pathways revealed no evidence for their involvement in heart-related pathways as indicated by software tools, confirming proposed approach.

Assessment of two faecal sludge treatment plants in urban areas:Case study in Beijing

Every day,human beings produce excreta all over the world,and the sludge that accumulates in waste disposal systems is referred to as the‘faecal sludge(FS)’.FS can cause serious environmental pollution in urban areas if it cannot be disposed of properly.A complete FS management system must include onsite sanitation technologies,FS collection and transport,a treatment plant,and resource recovery or disposal of the treatment *** on the treatment and reuse/disposal step of a FS complete service chain,this research presents two cases of FS treatment in *** Case 1,FS biogas plant adopts anaerobic digestion(AD)to treat FS,and the digestate can be used as biofertilizer in the surrounding *** Case 2,several technologies including solid-liquid separation,dewatering,pyrolysis,AD and co-composting are integrated to find innovative solutions for FS treatment.A comprehensive assessment including the aspects of technology,economy and environment is conducted for further SWOT(Strengths,Weaknesses,Opportunities and Threats)***,critical strategies are developed,which include(1)selecting project site for optimized transportation,maximum waste reuse,minimum environmental impact and convenient final effluent disposal;(2)planning technical options at a feasible study stage,considering resource recovery,secondary pollution prevention and fire protection;(3)exploring market channels for by-products sale to increase profitability;(4)guaranteeing engineering quality and service life for the purpose of sustainable operation;(5)minimizing health risks to persons exposed to the untreated FS;and(6)providing necessary training for hygiene *** cases in Beijing can provide valuable lessons for urban areas in developing countries and the strategies can provide a reference for stakeholders and decision-makers who intend to develop FS treatment projects.

Defensin expression in chronic pouchitis in patients with ulcerative colitis or familial adenomatous polyposis coli

AIM. Pouchitis develops in ileoanal pouches in up to 50% of patients with ulcerative colitis during the first 10 years after pouch surgery while being rare in patients after proctocolectomy for familial adenomatous polyposis coil （FAP） syndrome. Defensins are major components of the innate immune system and play a significant role in gastrointestinal microbial homeostasis. Pouch defensin and cytokine expression were correlated with states of pouch inflammation to study their role in pouchitis.METHODS： Patients with ulcerative colitis and FAP syndrome were stratified into groups with pouches after surgery, pouches without or with pouchitis. Biopsies from terminal ileum from a healthy intestine or from normal terminal ileum of patients with ulcerative colitis served as controls, mRNA from pouches and controls was analysed for defensin and cytokine expression.RESULTS： Expression of defensins was increased in all pouches immediately after surgery, compared to ileum of controls. Initially, pouches in ulcerative colitis revealed higher defensin expression than FAP pouches. Defensin expression declined in both patient groups and increased again slightly in pouchitis in patients with ulcerative colitis. FAP pouches without pouchitis had strong expression of β-defensin hBD-1, while all other defensins remained at low levels. Cytokine expression in ulcerative colitis pouches was high, while FAP pouches showed moderately elevated cytokines only after surgery.CONCLUSION： Development of pouchitis correlates with decreased defensin expression in ulcerative colitis in addition to high expression of cytokines. The low incidence of pouchitis in FAP pouches correlates with increased expression of hBD-1 β- defensin in association with low cytokine levels.

合成了一系列含磁性反离子的非对称双疏水链长的阳离子表面活性剂,其中三氯一溴铁合十六烷基戊基二甲基铵(C_(16)C_5DMA^+[FeCl_3Br]-~)与偶氮羧酸钠盐(AzoNa_4)在酸性条件水溶液中形成磁性囊泡凝胶。运用cryo-透射电镜(TEM)、冷冻蚀刻TEM(FF-TEM)、流变仪、傅里叶变换红外光谱(FT-IR)和超导量子干涉(SQUID)等表征技术对囊泡凝胶进行了结构和性质研究,结果发现:凝胶含有曲率多变的融合性的双层囊泡,这些双分子层结构模构了自然界中各种物象的结构轮廓,展现了不可预测的多变曲率和良好柔性。聚集体双分子层膜内由长短不对称烷基链采取交错相扣的双分子层排列模式,这种构建模式结构稳定,短烷基链可游离出囊泡双分子层并伸向外部水相介质。两个相邻囊泡间的短链在疏水相互作用下形成非共价的囊泡"补丁",疏水的囊泡"补丁"克服相邻囊泡之间的斥力而融合。磁性反离子[FeCl_3Br]^-不仅赋予囊泡磁性,且在囊泡的形成过程中调控烷基链的组装。这种多形态融合性囊泡为揭示膜曲率的调节机制和构建人工细胞提供实验数据和理论参考。

DNA origami is a promising technology for its reproducibility,flexibility,scalability and *** the several potential applications,DNA origami has been proposed as a tool for drug delivery and as a contrast agent,since a conformational change upon specific target interaction may be used to release a drug or produce a physical signal,***,its conformation should be robust with respect to the properties of the medium in which either the recognition or the read-out take place,such as pressure,viscosity and any other unspecific interaction other than the desired target *** we report on the read-out robustness of a tetragonal DNA-origami/gold-nanoparticle hybrid structure able to change its configuration,which is transduced in a change of its plasmonic properties,upon interaction with a specific DNA *** investigated its response when analyzed in three different media:aqueous solution,solid support and viscous *** show that,once a conformational variation is produced,it remains unaffected by the subsequent physical interactions with the environment.

Storage roots of cassava plants crops are one of the main providers of starch in many South American,African,and Asian *** varieties with high yields is crucial for growing and *** requires a better understanding of the dynamics of storage root formation,which is usually done by repeated manual evaluation of root types,diameters,and their distribution in excavated *** introduce a newly developed method that is capable to analyze the distribution of root diameters automatically,even if root systems display strong variations in root widths and clustering in high *** application study was conducted with cassava roots imaged in a video acquisition *** root diameter distribution was quantified automatically using an iterative ridge detection approach,which can cope with a wide span of root diameters and *** approach was validated with virtual root models of known geometries and then tested with a time-series of excavated root *** on the retrieved diameter classes,we show plausibly that the dynamics of root type formation can be monitored qualitatively and *** conclude that this new method reliably determines important phenotypic traits from storage root crop *** method is fast and robustly analyses complex root systems and thereby applicable in high-throughput phenotyping and future breeding.

Gonadal damage and options for fertility preservation in female and male cancer survivors

It is estimated that in 2010, 1 in every 250 adults will be a childhood cancer survivor. Today, oncological surgery, radiotherapy and chemotherapy achieve relatively high rates of remission and long-term survival, yet are often detrimental to fertility. Quality of life is increasingly important to long-term survivors of cancer, and one of the major quality-of-life issues is the ability to produce and raise normal children. Developments in the near future in the emerging field of fertility preservation in cancer survivors promise to be very exciting. This article reviews the published literature, discusses the effects of cancer treatment on fertility and presents the options available today thanks to advances in assisted-reproduction technology for maintaining fertility in male and female patients undergoing this type of treatment. The various diagnostic methods of assessing the fertility potential and the efficacy of in vitro fertilization （IVF） after cancer treatment are also presented.

Malnutrition is described as a state of insufficient intake of energy,protein and other nutrients leading to changes in body composition(weight loss,reduced fat-free mass)as well as adverse functional and clinical outcomes.[1]Depending on the assessments and definition used,the prevalence in older patients ranges between 12%in communitydwelling adults to 60%of patients in geriatric care facilities.[2–5]Older populations are at high risk of nutritional deficiencies because of risk factors such as multimorbidity,polypharmacy,cognitive and physical decline,poor appetite,depressive syndromes and socioeconomic changes.[4,6].

Mesmerism, Sexuality, and Medicine：＂Karezza＂ and the Sexual Reform Movement

Within the scope of certain social and religious movements, mesmerism had a considerable impact on the Americanway of life during the last decades of 19th century. A famous example is the Christian Science of Mary Baker EddyBut also less known groups adopted mesmeric ideas and practices. The paper focuses on the concept of＂Karezza＂.It combined a specific sexual practice with religious ideas of divine love, birth control, social reform, women＇semancipation and health education. It was created by Alice Bunker Stockham （1833-1912）, an obstetrician andgynecologist from Chicago. Among other authorities, the writer Leo Tolstoy and medical psychologist HavelockEllis appreciated her approach. She was a pioneer of the sexual and marriage reform promoting practical advice foreveryday life. As a doctor, she stressed especially the disastrous consequences of a brutal sexual life destroyingbody and mind （not only of the women）. As an antidote, she propagated the ＂Karezza love＂ avoiding ejaculation ada punctual orgasm （climax） during intercourse. The leading idea was the imagery of a mesmeric ＂fluidum＂ unitingindividuals spiritually. Bunker＇s publications were translated into German. They corresponded to the Lebensreformmovement, but never became popular. Also the early sexology about 1900 ignored Bunker＇s concept as well as thesexual medicine later on. It is worthwhile to reconsider it within the context of the history of medicine, culture, andanthropology.
