Electrical potentials up to 800 mv can be observed between different metallic dental restorations. These potentials produce fields in the mouth that may interfere with microbial communities. The present study focuses on the impact of different electric field strengths （EFS） on the growth of Staphylococcus aureus （ATCC 25923） and Escherichia coil （ATCC 25922） in vitro. Cultures of S. aureus and E. coil in fluid and gel medium were exposed to different EFS. Effects were determined by calculation of viable counts and measurement of inhibition zones. In gel medium, anodic inhibition zones for S. aureuswere larger than those for E. coliat all field strength levels. In fluid medium, the maximum decrease in the viable count of S. aureus cells was at 10 v.m-1. Field-treated S. aureus cells presented ruptured cell walls and disintegrated cytoplasm. Conclusively, S. aureus is more sensitive to increasing electric field strength than E. coll.

Association of rare SPINK1 gene mutation with another base substitution in chronic pancreatitis patients

AIM: To verify and expand the known spectrum of serine protease inhibitor Kazal type 1 (SPINK1) gene mutations in chronic pancreatitis. METHODS: DNA extracted from 172 chronic pancreatitis patients was assayed for SPINK1 gene mutations by PCR and DNA sequencing. A control cohort of 90 unrelated healthy individuals was analysed by the same methods for presence of common populational polymorphisms, and frequency of five-loci haplotypes was calculated. Linkages of gene aberrations in single SPINK1 gene copies were analysed by long-distance PCR followed by allele-specifi c PCR and DNA sequencing. RESULTS: The most frequent SPINK1 gene mutation N34S was found at a frequency of 6%. Furthermore, we detected the heterozygous intervening sequence (IvS) 3 + 2 T > C mutated gene in 2 German patients and 1 Macedonian chronic pancreatitis patient. In all three SPINK1 gene copies an additional rare base substitution was found: 5’untranslated region (UTR)-215 G > A. Poly-morphism analysis revealed that all three affected genes carried the same fi ve-loci haplotype. DNA sequencing of another chronic pancreatitis-related gene PRSS1 (cationic trypsinogen) did not reveal any mutations in these 3 ***: We found in 3 (2%) of 172 chronic pancreatitis patients an IvS3 + 2 T > C SPINK1 gene mutation and a base substitution 5’UTR-215 G > A inthe same gene copy. Most probably the 5’UTR-215 G >A represents a rare polymorphism and not a mutationas previously concluded. Haplotype analysis suggests acommon origin of the IvS3 + 2 T > C mutation in thesepatients.

Exposure to disinfection by-products（DBP） such as trihalomethanes（THM） in swimming pools has been linked to adverse health effects in humans, but their biological mechanisms are unclear. We evaluated short-term changes in blood gene expression of adult recreational swimmers after swimming in a chlorinated pool. volunteers swam 40 min in an indoor chlorinated pool. Blood samples were drawn and four THM（chloroform,bromodichloromethane, dibromochloromethane and bromoform） were measured in exhaled breath before and after swimming. Intensity of physical activity was measured as metabolic equivalents（METs）. Gene expression in whole blood m RNA was evaluated using Illumina Human HT-12v3 Expression-Bead Chip. Linear mixed models were used to evaluate the relationship between gene expression changes and THM exposure. Thirty-seven before-after pairs were analyzed. The median increase from baseline to after swimming were： 0.7 to 2.3 for MET, and 1.4 to 7.1 μg/m^3 for exhaled total THM（sum of the four THM）.Exhaled THM increased on average 0.94 μg/m^3 per 1 MET. While 1643 probes were differentially expressed post-exposure. Of them, 189 were also associated with exhaled levels of individual/total THM or MET after False Discovery Rate. The observed associations with the exhaled THM were low to moderate（Log-fold change range：-0.17 to 0.15）. In conclusion, we identified short-term gene expression changes associated with swimming in a pool that were minor in magnitude and their biological meaning was unspecific. The high collinearity between exhaled THM levels and intensity of physical activity precluded mutually adjusted models with both covariates. These exploratory results should be validated in future studies.

We describe the footprint and input catalog for bright nights in the LAMOST Pilot Survey, which began in October 2011. Targets are selected from two stripes in the north and south Galactic Cap regions, centered at δ = 29°, with 10° width in declination, covering right ascensions of 135° to 290° and -30° to 30° re- spectively. We selected spectroscopic targets from a combination of the SDSS and 2MASS point source catalogs. The catalog of stars defining the field centers （as re- quired by the Shack-Hartmann wavefront sensor at the center of the LAMOST field） consists of all v 〈 8m stars from the Hipparcos catalog. We employ a statistical selection algorithm that assigns priorities to targets based on their positions in mul- tidimensional color/magnitude space. This scheme overemphasizes rare objects and de-emphasizes more populated regions of magnitude and color phase space, while en- suring a smooth, well-understood selection function. A demonstration of plate design is presented based on the Shack-Hartmann star catalog and an input catalog that was generated by our target selection routines.

We outline the design of the dark nights portion of the LAMOST Pilot Survey, which began observations in 2011 October. In particular, we focus on Milky Way stellar candidates that are targeted for the LEGUE （LAMOST Experiment for Galactic Understanding and Exploration） survey. We discuss the regions of sky in which spectroscopic candidates were selected, and the motivations for selecting each of these sky areas. Some limitations due to the unique design of the telescope are discussed, including the requirement that a bright （v 〈 8） star be placed at the center of each plate for wavefront sensing and active optics corrections. The target selection categories and scientific goals motivating them are briefly discussed, followed by a detailed overview of how these selection functions were implemented. We illustrate the difference between the overall input catalog - Sloan Digital Sky Survey （SDSS） photometry - and the final targets selected for LAMOST observations.

The weather at the Xinglong Observing Station, where the Guo Shou Jing Telescope （GSJT） is located, is strongly affected by the monsoon climate in north- east China. The LAMOST survey strategy is constrained by these weather patterns. We present statistics on observing hours from 2004 to 2007, and the sky brightness, seeing, and sky transparency from 1995 to 2011 at the site. We investigate effects of the site conditions on the survey plan. Operable hours each month show a strong cor- relation with season： on average there are eight operable hours per night available in December, but only one-two hours in July and August. The seeing and the sky trans- parency also vary with season. Although the seeing is worse in windy winters, and the atmospheric extinction is worse in the spring and summer, the site is adequate for the proposed scientific program of the LAMOST survey. With a Monte Carlo simulation using historical data on the site condition, we find that the available observation hours constrain the survey footprint from 22h to 16h in right ascension; the sky brightness allows LAMOST to obtain a limiting magnitude of v = 19.5 mag with S/N= 10.

背景在类癌性心脏病患者的心脏手术中，常因类癌危象、心血管功能障碍和失血导致血流动力学不稳定。在使用奥曲肽的同时，合用血管加压药物的安全性以及抑肽酶的益处尚不明确。方法我们回顾了从1985年到2003年的18年中，连续100例类癌性心脏病患者行心脏手术的病例，对奥曲肽给药期间合用血管加压药物和抑肽酶的影响以及患者的死亡率进行了单因素分析。由于抑肽酶可以暂时性降低死亡率，因此行双因素分析以鉴别其他与死亡率相关的因素。结果分别用奥曲肽（胛=89）和（或）血管加压药（门=93）处理类癌症状和低血压。血管加压药并没有随着奥曲肽用量的增加而增加。手术中需要使用肾上腺素的患者死亡率较高，但其手术前的纽约心脏协会分级就较差，尿中5．羟哼J哚乙酸水平以及对输血的需求都较高。抑肽酶（，z=54）可降低患者对输血的需求，并增加奥曲肽的用量，但对死亡率无影响。死亡率方面，从1985年到1994年的28％，下降至1995年到2003年的6％，平均总死亡率为13％。死亡率与大量输血和长时间心肺转流有关。结论血管加压药物可以与奥曲肽合用于类癌患者。死亡率的增加可能与肾上腺素的使用有关，这主要是由选择性差异导致的，而不是药物的原发不良反应。类癌患者的存活率随着时间推移而增加是由多因素造成的，与使用抑肽酶无关，说明在应用奥曲肽的情况下，在这类患者中进一步抑制激肽释放酶——激肽系统对预后并无益处。

Bacteroides fragilis Supernatant Extracts Enriched in Phenylacetic Acid Induce a Cytotoxic Effect in Mammalian Cells

Bacteroides species are nearly half of the fecal flora community and some are host symbionts crucial to host nutrition and systemic immunity. Among Bacteroides species B. fragilis strains are considered to be the opportunistic ones, being the most isolated anaerobic bacteria in clinical samples. Cell-free supernatants of 65 B. fragilis strains were assayed and they were capable of inducing vacuolating phenotype on vero cells lineage. The supernatant of the Bacteroides fragilis ATCC 23745 strain was elicited to have the strongest vacuolating effect on vero cells monolayers and peritoneal macrophages. Some drastic cell alterations were observed, such as a general disorganization of cytoplasm and chromatin condensation, evidencing cell death. By transmission electron microscopy it was confirmed that the vacuoles observed were, in fact, swollen mitochondria. An immunocytochemical assay, TUNEL, was used to confirm this hypothesis and showed that vero cells and peritoneal macrophages were dying by apoptotic process after exposition of B. fragilis cell-free supernatant. Physical analysis of the apoptotic factor has revealed properties similar to short-chain fatty acids. After gas chromatography and mass spectrometry analysis, phenylacetic acid (PA) was characterized as the major compound present in the most purified active fraction. We believe that the PA is responsible for the pro-apoptotic effect elicited by the supernatant of B. fragilis cultures.
