Dehydrating stresses trigger the accumulation of abscisic acid(ABA),a key plant stress-signaling hormone that activates Snf1-Related Kinases(SnRK2s)to mount adaptive responses.However,the regulatory circuits that terminate the SnRK2s signal relay after acclimation or post-stress conditions remain to be defined.Here,we show that the desensitization of the ABA signal is achieved by the regulation of OST1(SnRK2.6)protein stability via the E3-ubiquitin ligase HOS15.Upon ABA signal,HOS15-induced degradation of OST1 is inhibited and stabilized OST1 promotes the stress response.When the ABA signal terminates,protein phosphatases ABI1/2 promote rapid degradation of OST1 via HOS15.Notably,we found that even in the presence of ABA,OST1 levels are also depleted within hours of ABA signal onset.The unexpected dynamics of OST1 abundance are then resolved by systematic mathematical modeling,demonstrating a desensitizing feedback loop by which OST1-induced upregulation of ABI1/2 leads to the degradation of OST1.This model illustrates the complex rheostat dynamics underlying the ABA-induced stress response and desensitization.

This research is focused on the analysis of the sequence stratigraphic units of F3 Block,within a wave-dominated delta of Plio–Pleistocene age.Three wells of F3 block and a 3D seismic data,are utilized in this research.The conventional techniques of 3D seismic interpretation were utilized to mark the 11 surfaces on the seismic section.Integration of seismic sequence stratigraphic interpretation,using well logs,and subsequent 3D geostatistical modeling,using seismic data,aided to evaluate the shallow hydrocarbon traps.The resulting models were obtained using System Tract and Facies models,which were generated by using sequential stimulation method and their variograms made by spherical method,moreover,these models are validated via histograms.The CDF curve generated from upscaling of well logs using geometric method,shows a good relation with less percentage of errors(1 to 2 for Facies and 3 to 4 for System Tract models)between upscaled and raw data that complements the resulted models.These approaches help us to delineate the best possible reservoir,lateral extent of system tracts(LST and/or HST)in the respective surface,and distribution of sand and shale in the delta.The clinoform break points alteration observed on seismic sections,also validates the sequence stratigraphic interpretation.The GR log-based Facies model and sequence stratigraphy-based System Tract model of SU-04-2 showed the reservoir characteristics,presence of sand bodies and majorly LST,respectively,mainly adjacent to the main fault of the studied area.Moreover,on the seismic section,SU-04-2 exhibits the presence of gas pockets at the same location that also complements the generated Facies and System Tract models.The generated models can be utilized for any similar kind of study and for the further research in the F3 block reservoir characterization.

Object recognition and tracking are two of the most dynamic research sub-areas that belong to the field of Computer Vision.Computer vision is one of the most active research fields that lies at the intersection of deep learning and machine vision.This paper presents an efficient ensemble algorithm for the recognition and tracking of fixed shapemoving objects while accommodating the shift and scale invariances that the object may encounter.The first part uses the Maximum Average Correlation Height(MACH)filter for object recognition and determines the bounding box coordinates.In case the correlation based MACH filter fails,the algorithms switches to a much reliable but computationally complex feature based object recognition technique i.e.,affine scale invariant feature transform(ASIFT).ASIFT is used to accommodate object shift and scale object variations.ASIFT extracts certain features from the object of interest,providing invariance in up to six affine parameters,namely translation(two parameters),zoom,rotation and two camera axis orientations.However,in this paper,only the shift and scale invariances are used.The second part of the algorithm demonstrates the use of particle filters based Approximate Proximal Gradient(APG)technique to periodically update the coordinates of the object encapsulated in the bounding box.At the end,a comparison of the proposed algorithm with other stateof-the-art tracking algorithms has been presented,which demonstrates the effectiveness of the proposed algorithm with respect to the minimization of tracking errors.

With the rapid progress of deep convolutional neural networks,several applications of crowd counting have been proposed and explored in the literature.In congested scene monitoring,a variety of crowd density estimating approaches has been developed.The understanding of highly congested scenes for crowd counting during Muslim gatherings of Hajj and Umrah is a challenging task,as a large number of individuals stand nearby and,it is hard for detection techniques to recognize them,as the crowd can vary from low density to high density.To deal with such highly congested scenes,we have proposed the Congested Scene Crowd Counting Network(CSCC-Net)using VGG-16 as a core network with its first ten layers due to its strong and robust transfer learning rate.A hole dilated convolutional neural network is used at the back end to widen the relevant field to extract a large range of information from the image without losing its original resolution.The dilated convolution neural network is mainly chosen to expand the kernel size without changing other parameters.Moreover,several loss functions have been applied to strengthen the evaluation accuracy of the model.Finally,the entire experiments have been evaluated using prominent data sets namely,ShanghaiTech parts A,B,UCF_CC_50,and UCF_QNRF.Our model has achieved remarkable results i.e.,68.0 and 9.0 MAE on ShanghaiTech parts A,B,199.1 MAE on UCF_CC_50,and 99.8 on UCF_QNRF data sets respectively.

Objective:To delineate the comparative immunomodulatory roles of H1R-H4R in antibody generation profile in rabbit model.Methods:The cohort comprised of eight groups containing 18(9 male and 9 female) rabbits in each group.GroupⅠremained non-immunized and received only vehicle(sterile distilled water,1 mL/kg×b.i.d.) intramuscularly.GroupⅡreceived vehicle (1 mL/kg×b.i.d.) while GroupsⅢ-Ⅶ(drugs-treated) received subcutaneous histamine (100μg/kg×b.i.d.),and intramuscular H1R-antagonist(pheniramine,10 mg/kg×b.i.d.), H2R-antagonist(ranitidine,10 mg/kg×b.i.d.),H3R-antagonist(iodophenpropit,1μg/kg×b.i.d.) and H4R-antagonist(JNJ 7777120,10μg/kg×b.i.d.),and GroupⅧDMSO(1 mL/kg×b.i.d.),respectively for 10 days(starting from day 1).They were subsequendy immunized with intravenous injection of sheep red blood cells(SRBC) at day 3.The estimation of serum Igs,IgM and IgG were done by ELISA,and observed at day 0(pre-immunization),and 7,14,21,28 and 58(post-immunization).Results:It was shown that histamine and HRs-antagonists could influence a detectable antibody response to SRBC as early as day 7-post-immunization(post-Ⅰ), which lasted until day 58 post-Ⅰ.The results were found statistically significant(P<0.05,). Conclusions:This study suggests that histamine receptors play important roles in modulation of antibody generation in which H1R,H2R and H4R have immunosuppressive roles and conversely, H3R playes an immune enhancing role.The findings of this study may have clinical significance and provide the baseline information for future study.

Biodiesel utilization has been rapidly growing worldwide as the prime alternative to petrodiesel due to a global rise in diesel fuel demand along with hazardous emissions during its thermochemical conversion.Although,several debatable issues including feedstock availability and price,fuel and food competition,changes in land use and greenhouse gas emission have been raised by using edible as well as inedible feedstocks for the production of biodiesel.However,non-crop feedstocks could be a promising alternative.In this article,waste cooking oils have been recommended as a suitable option for biodiesel production bearing in mind the current national situation.The important factors such as the quantity of waste cooking oil produced,crude oil and vegetable oil import expenses,high-speed diesel imports,waste management issues and environmental hazards are considered.Moreover,process simulation and operating cost evaluation of an acid catalyzed biodiesel production unit are also conducted.The simulation results show that the production cost of waste cooking oil-based biodiesel is about 0.66 USD·L-1.We believe that the present overview would open new pathways and ideas for the development of biofuels from waste to energy approach in Pakistan.

High HbA1c is associated with higher risk of ischaemic stroke in Pakistani population without diabetes

Context:The role of glycated haemoglobin(HbA1c)in the prediction of ischaemic stroke in individuals without diabetes is underestimated.Aims:We performed a study to analyse the role of HbA1c in the risk prediction of ischaemic stroke in Pakistani population without diabetes.We further studied the difference between HbA1c values of individuals with diabetes and without diabetes with stroke.Settings and design:Single centre,case–control.Materials and methods:In phase I,a total of 233 patients without diabetes with ischaemic stroke and 245 as controls were enrolled.Association of HbA1c levels,lipid profiles and blood pressure recordings with ischaemic stroke was analysed.In phase II,comparison was done between diabetics and nondiabetics with stroke.Statistical analysis:Comparison of the mean variables was performed with Student’s t-tests.Logistic regression analysis with ischaemic stroke as the dependent variable was performed for phase I.Results:In phase I,the ischaemic stroke group had significantly higher HbA1c levels(5.9±2.9%vs 5.5±1.6%)compared with controls(p0.05).Conclusions:Higher HbA1c indicated a significantly increased risk for ischaemic stroke.An HbA1c value above 5.6%(prediabetic range)predicted future risk of stroke and efforts to maintain glucose level within the normal range(≤5.6%)in individuals with high cardiovascular risk are important.

The rapid growth in software demand incentivizes software development organizations to develop exclusive software for their customers worldwide.This problem is addressed by the software development industry by software product line(SPL)practices that employ feature models.However,optimal feature selection based on user requirements is a challenging task.Thus,there is a requirement to resolve the challenges of software development,to increase satisfaction and maintain high product quality,for massive customer needs within limited resources.In this work,we propose a recommender system for the development team and clients to increase productivity and quality by utilizing historical information and prior experiences of similar developers and clients.The proposed system recommends features with their estimated cost concerning new software requirements,from all over the globe according to similar developers’and clients’needs and preferences.The system guides and facilitates the development team by suggesting a list of features,code snippets,libraries,cheat sheets of programming languages,and coding references from a cloud-based knowledge management repository.Similarly,a list of features is suggested to the client according to their needs and preferences.The experimental results revealed that the proposed recommender system is feasible and effective,providing better recommendations to developers and clients.It provides proper and reasonably well-estimated costs to perform development tasks effectively as well as increase the client’s satisfaction level.The results indicate that there is an increase in productivity,performance,and quality of products and a reduction in effort,complexity,and system failure.Therefore,our proposed system facilitates developers and clients during development by providing better recommendations in terms of solutions and anticipated costs.Thus,the increase in productivity and satisfaction level maximizes the benefits and usability of SPL in the modern era of technology.

Effects of histamine HlR-H4R-agonist on the airway epithelium of rabbits

Objective:To explore the exact role of histamine receptors in respiratory system.Methods: The cohort comprised of six groups(group I control and groupⅡ—Ⅳtreated) containing five rabbits in each group.Control-group received vehicle(sterile distilled water) and treated groups received subcutaneous histamine(100μg/kg,b.i.d.),and H1R-agonist(HTMT),H2R-agonist (amthamine),H3R-agonist(R-(-)-α-methylhistamine) and H4R-agonist(clobenpropit) each in a dose of 10μg/kg,b.***.d.for 30 subsequent days.After completion of treatment,animals were euthanized and perfused with 10% buffered formalin.Small tissue blocks of trachea and lungs were processed for paraffin embedding.Observations were recorded in sample photomicrographs taken from 10μm thick.Haematoxylin and eosin stained sections.Results:It was observed that trachea and bronchi from histamine,H1R and H4R groups had only patches of hyperplastic and hypertrophied epithelium and in general,cells in the affected region were taller heaped up. The bronchiolar epithelia from all treated groups showed hypertrophy and hyperplasia throughout with most of the cells having rounded profile and appeared to bud out from the basal cells. Conclusions:It is concluded that histamine receptors on induction via its specific agonist can induce hypertrophy,hyperplasia of respiratory tract epithelia suggesting its role akin to growth stimulating factor and warranting further long-term study.

The Effectiveness of Human Patient Simulator on Knowledge, Motivation, and Clinical Competence of the Student’s Nursing about Diabetic Ketoacidosis Management

Purpose: This study aimed to evaluate the effects of human patient simulator to improve the knowledge, motivation, and clinical competence of the students of Nursing about Diabetic Ketoacidosis (DKA) Management. Methods: The population of this research is the Students of Diploma III Keperawatan Poltekkes Bandung, Indonesia, who was attending Medical Surgical Nursing subject, one of which learning focus is about Diabetic Ketoacidosis. The research subjects were divided into 3 groups, Full SBL (a group of SBL learning with HPS), Semi SBL (a group of SBL model with Partial Task Trainer Simulation), and Conventional model. The overall sample of the research for the three groups is 90 people. Results: The analysis shows significant result with the Kruskal Wallis test. Followed by the Post Hoc test Mann Whitney test, the result shows that the treatment of Full SBL is better than Semi-SBL and Conventional model for knowledge (40.00 vs. 27.00 vs. 35.00), motivation (74.67 vs. 73.33 vs. 73.00), and clinical competence (79.00 vs. 56.50 vs. 76.00). Conclusion: The result of the research shows the effectiveness of the Human Patient Simulator towards the improvement of Knowledge, Motivation, and Clinical competence of the Students of Nursing about DKA management. In addition, the research found that Full SBL improves Knowledge, Motivation and Clinical competence better than Semi SBL and Conventional model.
