Insight into the spoilage heterogeneity of meat-borne bacteria isolates with high-producing collagenase

Chilled chicken is inevitably contaminated by microorganisms during slaughtering and processing,resulting in spoilage.Cutting parts of chilled chicken,especially wings,feet,and other skin-on products,are abundant in collagen,which may be the primary target for degradation by spoilage microorganisms.In this work,a total of 17 isolates of spoilage bacteria that could secrete both collagenase and lipase were determined by raw-chicken juice agar(RJA)method,and the results showed that 7 strains of Serratia,Aeromonas,and Pseudomonas could significantly decompose the collagen ingredients.The gelatin zymography showed that Serratia liquefaciens(F5)and(G7)had apparent degradation bands around 50 kDa,and Aeromonas veronii(G8)and Aeromonas salmonicida(H8)had a band around.65 and 95 kDa,respectively.The lipase and collagenase activities were detected isolate-by-isolate,with F5 showing the highest collagenase activity.For spoilage ability on meat in situ,F5 performed strongest in spoilage ability,indicated by the total viable counts,total volatile basic nitrogen content,sensory scores,lipase,and collagenase activity.This study provides a theoretical basis for spoilage heterogeneity of strains with high-producing collagenase in meat.

The micaceous weathered granitic soil(WGS)is frequently encountered in civil engineering worldwide,unfortunately little information is available regarding how mica affects the physico-mechanical behaviors of WGS.This study prepares reconstituted WGS with different mica contents by removing natural mica in theWGS,and then mixes it with commercial mica powders.The geotechnical behavior as well as the microstructures of the mixtures are characterized.The addition of mica enables the physical indices of WGS to be specific combinations of coarser gradation and high permeability but high Atterberg limits.However,high mica content in WGS was found to be associated with undesirable mechanical properties,including increased compressibility,disintegration,and swelling potential,as well as poor compactability and low effective frictional angle.Microstructural analysis indicates that the influence of mica on the responses of mixtures originates from the intrinsic nature of mica as well as the particle packing being formed withinWGS.Mica exists in the mixture as stacks of plates that form a spongy structure with high compressibility and swelling potential.Pores among the plates give the soil high water retention and high Atterberg limits.Large pores are also generated by soil particles with bridging packing,which enhances the permeability and water-soil interactions upon immersion.This study provides a microlevel understanding of how mica dominates the behavior of WGS and provides new insights into the effective stabilization and improvement of micaceous soils.

Bromocriptine protects perilesional spinal cord neurons from lipotoxicity after spinal cord injury

Recent studies have revealed that lipid droplets accumulate in neurons after brain injury and evoke lipotoxicity,damaging the neurons.However,how lipids are metabolized by spinal cord neurons after spinal cord injury remains unclear.Herein,we investigated lipid metabolism by spinal cord neurons after spinal cord injury and identified lipid-lowering compounds to treat spinal cord injury.We found that lipid droplets accumulated in perilesional spinal cord neurons after spinal cord injury in mice.Lipid droplet accumulation could be induced by myelin debris in HT22 cells.Myelin debris degradation by phospholipase led to massive free fatty acid production,which increased lipid droplet synthesis,β-oxidation,and oxidative phosphorylation.Excessive oxidative phosphorylation increased reactive oxygen species generation,which led to increased lipid peroxidation and HT22 cell apoptosis.Bromocriptine was identified as a lipid-lowering compound that inhibited phosphorylation of cytosolic phospholipase A2 by reducing the phosphorylation of extracellular signal-regulated kinases 1/2 in the mitogen-activated protein kinase pathway,thereby inhibiting myelin debris degradation by cytosolic phospholipase A2 and alleviating lipid droplet accumulation in myelin debris-treated HT22 cells.Motor function,lipid droplet accumulation in spinal cord neurons and neuronal survival were all improved in bromocriptine-treated mice after spinal cord injury.The results suggest that bromocriptine can protect neurons from lipotoxic damage after spinal cord injury via the extracellular signal-regulated kinases 1/2-cytosolic phospholipase A2 pathway.

Environmental factors,including chemical/physical pollutants,as well as lifestyle and psychological factors,contribute greatly to the pathways leading to cardiometabolic diseases with a heavy disease burden and economic loss.The concept of exposomes provides a novel paradigm for combining all exposure characteristics to evaluate disease risk.A solution-like exposome requires technological support to provide continuous data to monitor vital signs and detect abnormal fluctuations.Wearable devices allow people to conveniently monitor signals during their daily routines.These new technologies empower users to more actively prevent and manage cardiometabolic disease by reviewing risk factors of the disease,especially lifestyle factors,such as sleeping time,screen time,and mental health condition.Devices with multiple sensors can monitor electrocardiography data,oxygen saturation,intraocular pressure,respiratory rate,and heart rate to enhance the exposome study and provide precise suggestions for disease prevention and management.

All-trans retinoic acid(ATRA)can reverse the malignant behaviors of hepatocellular carcinoma(HCC)cells,thereby exerting anti-HCC effect;however,the underlying mechanism is yet to be understood.This study aimed to demonstrate that ATRA is vital to ferroptosis in HCC.Ferroptosis-related genes exhibit different expression in patients with HCC compared to that in healthy individuals.A total of 20 amino acid products were detected in HepG2 cells,the expression level of 5 was decreased after ATRA treatment.ATRA improved the levels of lipid ROS,MDA,and NAPDt/NADPH,and reduced the mt-DNA copy number and changed the structure of mitochondria,in HepG2 and Hep3B cells.We found the expression of genes positively correlated with ferroptosis to increase and those negatively correlated to decrease with ATRA treatment.Inhibition of ferroptosis by Ferrostatin-1 reversed ATRA-inhibited proliferation of HCC cells,along with cell migration and invasion.GSH synthesis was blocked by ATRA,accompanied by decreased cystine content and increased glutamate content,and downregulation of the expression of GSH synthesis-related genes.Our findings suggested that ATRA inhibited the malignancy of HCC cells by improving ferroptosis,and that inhibition of GSH synthesis contributed to ATRA-induced ferroptosis.

Quantifying the density contrasts of the Earth’s inner core boundary(ICB)is crucial to understand core-mantle coupling and the generation of the geodynamo.The PKiKP/PcP amplitude ratio is commonly used to obtain the density contrast at the ICB,but its applications are limited by scattered observed data.In this study,we selected the PKiKP and PcP phases reflected at the same region of inner-core and core-mantle boundaries beneath Northeast Asia from different earthquakes for the first time,and the observations suggested that the PKiKP/PcP amplitude ratio is widely scattered.We also compared the PKiKP and PcP amplitudes,which demonstrated that the scatter cannot be attributed only to ICB anomalies but might also arise from raypath differences and heterogeneities throughout the crust and mantle.By fitting the observed PKiKP/PcP amplitude ratio,we obtained a density contrast of approximately 0.65 g cm^(-3) and a compressional velocity contrast of approximately 0.87 km s^(-1) at the ICB beneath Northeast Asia.The larger contrast values indicate the possible occurrence of local crystallization occurring at the inner core surface.

以滴熔点为113、104、92、83、78和73℃的6种窄馏分费托蜡为研究对象,采用差示扫描量热法(DSC)研究其在不同冷却速率下的非等温结晶过程,应用Jeziorny法和莫志深法对结晶动力学参数进行理论预测,通过Kissinger方程计算非等温结晶活化能。结果表明:不同蜡样品的非等温结晶过程相似,并且结晶过程依赖于冷却速率;所采用的动力学模型与试验数据吻合良好,Avrami指数n平均值在1~2之间,说明结晶过程为二次成核,成核后生长形成棒状或纤维状晶体;蜡样品的非等温结晶活化能越小或冷却速率越快,越容易发生结晶。

三元锂电池在电动汽车领域得到了越来越广泛的应用。对电池发生热失控时的喷阀射流速度场开展研究,将有助于反推热失控过程中电池内部的临界压力及压力变化,对辅助优化电池设计、准确预测火焰传播等具有重要意义。但是,锂电池在热失控过程中往往存在两次喷射,喷射产物不仅包括电池内的化学反应产气、电解液蒸发等,还包含部分颗粒和电池内部各部件的碎片等;此外,蒸发后的电解液也会在遇冷后凝结,使整个喷阀射流过程呈现气-液-固三相混合状态,提高了速度场辨识与测试的难度。因此,为实现在高温情况下喷阀射流的可视化及速度场辨识计算,本研究搭建了主要由高速相机、激光系统和电池防爆箱组成的实验台架,采用高速相机对电池热失控射流过程进行拍摄及处理。首先对流场流态开展初步分析;然后通过自适应滤波算法,有效降低成像时噪声的影响,并增强液滴的边缘细节信息;同时利用直方图均衡化进一步提高图像的对比度;最后,采用基于亚像素精度插值的互相关算法对喷射流场的速度场进行分析计算,得到了不同时刻的二维平面下的速度场数据,并进一步对不同时刻不同区域的速度特性进行了分析,为后续电池热失控机理研究和电池安全性能提升工作提供了基础。
