BACKGROUND Herba Patriniae and Coix seed(HC)constitute a widely utilized drug combination in the clinical management of colorectal cancer(CRC)that is known for its diuretic,anti-inflammatory,and swelling-reducing *** its efficacy has been demonstrated in a clinical setting,the active compounds and their mechanisms of action in CRC treatment remain to be fully *** To identify the active,CRC-targeting components of HC and to elucidate the mechanisms of action *** Active HC components were identified and screened using *** for each component were ***-related targets were obtained from human gene *** targets between HC and CRC were identified.A“drug-ingredient-target”network was created to identify the core components and targets *** Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses were conducted to elucidate the key pathways *** docking between core targets and key components was *** vitro experiments validated core *** Nineteen active components of HC were identified,with acacetin as the primary active *** predictive analysis identified 454 targets of the active compounds in *** mapping with 2685 CRC-related targets yielded 171 intervention targets,including 30 core *** and KEGG analyses indicated that HC may influence the phosphoinositide 3-kinase(PI3K)/Akt signaling *** docking showed that acacetin exhibited an optimal interaction with AKT1,identifying PI3K,AKT,and P53 as key genes likely targeted by HC during CRC *** inhibited HT-29 cell proliferation and migration,as well as promoted apoptosis,in *** blotting analysis revealed increased p53 and cleaved caspase-3 expression and decreased levels of p-PI3K,p-Akt,and survivin,which likely contributed to CRC *** Acacetin,the principal active compound in the HC pair,inhibited the proliferati

基于激光点云数据进行人工林调查时,由于激光扫描时树木的遮挡与自遮挡、树木被砍伐等原因造成扫描的点云数据有缺失,遗漏树木的位置判断不准确,森林调查结果误差大,解决这一问题的关键是实现缺失树木的填补。定义了离散点集共线度的概念,构建了一个基于点集共线度最大化模型并结合直线检测进行缺失数据填补的方法。模拟数据实验结果:该方法的平均准确率为97.28%;人工林数据实验结果:该方法检测到9棵缺失树的位置,共线度由0.2193增大为0.2705。实验结果表明,该方法不仅可以实现缺失位置的最优推断,加强填补后数据的共线关系,也可应用于人工林的缺失树木计数。

Elucidating the Role of Probiotics in Modulating Depression Pathophysiology: Insights from Animal Models and Clinical Trials-A Narrative Review

With the ongoing COVID-19， a significant increase in global depression rates has placed a considerable strain on healthcare systems. Conventional pharmaceutical methods have shown limitations in treating this rise in depression. Therefore， identifying new potential treatments for depression is crucial. Maintaining a balanced gut microbiota is vital for human health and immune system function due to its complex bidirectional relationship with the brain. Numerous studies have established a robust correlation between anxiety and depression disorders， as well as the composition of microbial communities in the gastrointestinal tract， providing a fresh perspective. Adjusting the balance of gut microbiota may help alleviate depression symptoms. Probiotics， live microorganisms in the human gut， can improve the microbial community， help maintain homeostasis and stress resilience to remission in patients with depression. It’s essential to explore how probiotics affect gut microbiota since their impact can differ between animal experiments and clinical trials. The variation in results may be attributed to factors like the probiotic type， administered dosage， and treatment duration. The purpose of this review is to summarize the potential mechanisms by which probiotics affect gut microbiota， drawing from recent animal and clinical studies and observations on gut microbial ecology shifts. Additionally， this review will spotlight effective probiotic species for depression relief and discuss treatments centered on restoring gut microbial balance. Furthermore， it will also discuss the potential of probiotics on host microbiota in other organs to improve depressive symptoms and demonstrate the transient nature of the need for a more nuanced understanding of the safety and potential adverse effects of probiotics.

Arecoline causes cognitive impairment and potential addiction via disturbing Kenyon cells in bumblebee brain

Areca nut is the basic ingredient of betel quid， which is chewed by hundreds of millions of people in south-eastern Asia. Chewing of areca nut has been associated with oral cancers potentially due to its specific alkaloids， among which arecoline constitutes about 90% of total fraction. Being the world’s fourth most commonly used psychoactive substance， arecoline evokes stimulation， addiction， and other direct neurological effects， while its misuse correlates to neurotoxic effects. However， what might underlie its neurotoxic mechanisms has been poorly documented. The brain is encoded by a complex network of neuronal and glial cell types， and neurotoxicity of hazardous compounds present transcriptional heterogeneity. Recently， the eusocial bumblebee has been used as a model for studying brain effects， with sophisticated cognitive capability and precisely measured brain architecture. Here， we decipher cell-type-specific mental risks to arecoline using bumblebees. Arecoline induced locomotor hyperactivity and cognitive impairment. Single nucleus RNA sequencing (snRNA-seq) unearthed arecoline-induced cell-specific responses， primarily targeted on Kenyon cells (KC). Moreover， high-dose arecoline induced distinctive cell responses among KC subtypes， particularly class I large Kenyon cell (lKC)， leading to DNA damage， excitatory/inhibitory (E/I) imbalance， and calcium dyshomeostasis， which potentially resulted in cognitive impairment. Given arecoline’s popularity and growing exposure risks to humans， neurological health risks of areca nut warrant serious consideration.

The increase in energy consumption and its collateral damage on the environment has encouraged the development of environment-friendly ceramic materials with good energy storage *** this work,(1-x)Na_(0.5)Bi_(0.5)TiO_(3)-xCa(Mg_(1/3)Nb_(2/3))O_(3) ceramics were synthesized by the solid-state reaction *** 0.88Na_(0.5)Bi_(0.5)TiO_(3)-0.12Ca(Mg_(1/3)Nb_(2/3))O_(3) ceramic exhibited a high recoverable energy storage density of 8.1 J/cm3 and energy storage efficiency of 82.4% at 550 kV/*** introduction of Ca(Mg_(1/3)Nb_(2/3))O_(3) reduced the grain size and increased the band gap,thereby enhancing the breakdown field strength of the ceramic *** method also resulted in good temperature stability(20–140℃),frequency stability(1–200 Hz),and fatigue stability over 106 *** addition,an ultrahigh power density of 187 MW/cm^(3) and a fast charge-discharge rate(t0.9=57.2 ns)can be obtained *** element method analysis revealed that the decrease of grain size was beneficial to the increase of breakdown field ***,the 0.88Na_(0.5)Bi_(0.5)TiO_(3)-0.12Ca(Mg_(1/3)Nb_(2/3))O_(3) ceramics resulted in high energy storage properties with good stability and were promising environment-friendly materials for advanced pulsed power systems applications.

Hyperplasia and migration of fibroblast-like synoviocytes(FLSs)are the key drivers in the pathogenesis of rheumatoid arthritis(RA)and joint *** Yes-associated protein(YAP),which is a powerful transcription co-activator for proliferative genes,was observed in the nucleus of inflammatory FLSs with unknown upstream *** Gene Expression Omnibus database analysis,it was found that Salvador homolog-1(SAV1),the pivotal negative regulator of the Hippo-YAP pathway,was slightly downregulated in RA ***,SAV1 protein expression is extremely ***,it was revealed that SAV1 is phosphorylated,ubiquitinated,and degraded by interacting with an important serine-threonine kinase,G protein-coupled receptor(GPCR)kinase 2(GRK2),which was predominately upregulated by GPCR activation induced by ligands such as prostaglandin E2(PGE2)in *** process further contributes to the decreased phosphorylation,nuclear translocation,and transcriptional potency of YAP,and leads to aberrant FLSs *** depletion of GRK2 or inhibition of GRK2 by paroxetine rescued SAV1 expression and restored YAP phosphorylation and finally inhibited RA FLSs proliferation and ***,paroxetine treatment effectively reduced the abnormal proliferation of FLSs in a rat model of collagen-induced arthritis which was accompanied by a significant improvement in clinical ***,these results elucidate the significance of GRK2 regulation of Hippo-YAP signaling in FLSs proliferation and migration and the potential application of GRK2 inhibition in the treatment of FLSs-driven joint destruction in RA.

Cmtm4 deficiency exacerbates colitis by inducing gut dysbiosis and S100a8/9 expression

Digital technology provides a method of quantitative investigation and data analysis for contemporary landscape spatial analysis,and related research is moving from image recognition to digital algorithmic analysis,providing a more scientific and macroscopic way of *** key to refinement design is to refine the spatial design process and the spatial improvement strategy *** the ancient city of Zhaoyu in Qixian County,Shanxi Province as an example,(1)based on obtaining the integrated data of the ancient city through the drone tilt photography,the style and landscape of the ancient city are modeled;(2)the point cloud data with spatial information is imported into the point cloud analysis platform and the data analysis is carried out from the overall macroscopic style of the ancient city to the refinement level,which results in the formation of a more intuitive landscape design scheme,thus improving the precision and practicability of the landscape design;(3)Based on spatial big data,it starts from the spatial aggregation level,spatial distribution characteristics and other evaluation index system to achieve the refinement analysis of the *** technology and methods are used throughout the process to explore the refined design path.
