The field of biomaterials has advanced significantly in the past *** the growing need for high-throughput manufacturing and screening,the need for modular materials that enable streamlined fabrication and analysis of tissue engineering and drug delivery schema has *** are a powerful platform that have demonstrated promise in enabling these technologies without the need to modify a bulk *** building block paradigm of using microparticles within larger scaffolds to control cell ratios,growth factors and drug release holds *** microparticles(GmPs)are a well-established platform for cell,drug and growth factor *** of the challenges in using GmPs though is the limited ability to modify the gelatin *** the present work,we hypothesized that by thiolating gelatin before microparticle formation,a versatile platform would be created that preserves the cytocompatibility of gelatin,while enabling post-fabrication *** thiols were not found to significantly impact the physicochemical properties of the ***,the thiolated GmPs were demonstrated to be a biocompatible and robust platform for mesenchymal stem cell ***,the thiolated particles were able to be covalently modified with a maleimide-bearing fluorescent dye and a peptide,demonstrating their promise as a modular platform for tissue engineering and drug delivery applications.

Screening gene function in vivo is a powerful approach to discover novel drug targets. We present high-throughput screening （HTS） data for 3 762 distinct global gene knockout （KO） mouse lines with viable adult homozygous mice generated using either gene-trap or homologous recombination technologies. Bone mass was determined from DEXA scans of male and female mice at 14 weeks of age and by microCT analyses of bones from male mice at 16 weeks of age. Wild-type （WT） cagemates/littermates were examined for each gene KO. Lethality was observed in an additional 850 KO lines. Since primary HTS are susceptible to false positive findings, additional cohorts of mice from KO lines with intriguing HTS bone data were examined. Aging, ovariectomy, histomorphometry and bone strength studies were performed and possible non-skeletal phenotypes were explored. Together, these screens identified multiple genes affecting bone mass： 23 previously reported genes （Calcr, Cebpb, Crtap, Dcstamp, Dkkl, Duoxa2, Enppl, Fgf23, Kissl/Kisslr, Kl （Klotho）, Lrp5, mstn, Neol, Npr2, Ostml, Postn, Sfrp4, S1c30a5, Sic39a13, Sost, Sumf1, Src, Wnt10b）, five novel genes extensively characterized （Cldn18, Fam20c, Lrrkl, Sgpll, Wnt16）, five novel genes with preliminary characterization （Agpat2, RassfS, Slc10a7, Stc26a7, Slc30a10） and three novel undisclosed genes coding for potential osteoporosis drug targets.

This paper explores and extends understanding of the role and significance of whole-of-society resilience pro-grammes that support cities when dealing with complex crises,like the COVID-19 *** the complexity of whole-of-society resilience as different actors locally shape it,we ask the question:How can collab-oration between formal and informal resilience practices help to enhance resilience across the‘whole-of-society’?We answer this question by reviewing the importance of whole-of-society resilience and its complexity in a city’s governance of the COVID-19 *** argue that the necessity of renewing approaches to building lo-cal resilience capabilities across the whole-of-society requires synchronisation across and between formal and informal approaches-that is,“bottom-up”and governmental initiatives-to meet the diverse needs of ***,we detail two recent practice-orientated initiatives that have taken a renewal approach to building resilience through the involvement of whole-of-society in planning recovery from COVID-19 using in-ternational standard ISO/TS 22393;and a new initiative called the National Consortium for Societal Resilience[UK+].Finally,we signpost a set of critical questions for whole-of-society resilience practice.

A 67-year-old woman underwent an orthotopic liver transplantation for end stage liver disease secondary to chronic autoimmune hepatitis. She developed sudden massive hematochezia on post-operative day 23 with hemodynamic compromise. The source of hemorrhage was found at colonoscopy after careful irrigation and inspection to be a dieulafoy lesion situated just proximal to the anorectal junction. Hemostasis was achieved with epinephrine injection and thermal coagulation.

The objective of this study was to assess the validity of the step count functions in Actical accelerometers and activPAL inclinometers, compared with pedome-ter-derived step count data. Firstly, directly observed step counts over 3 treadmill speeds were compared with steps collected from 3 pedometers, accelerometers, and inclinometers in 10 adults. Secondly, step count data were derived from 22 participants who wore a pedome-ter, accelerometer, and inclinometer over 48 hours. Agreement between measurement tools was determined. All monitors appropriately measured steps in the labo-ratory conditions. In free living conditions, the mean percentage differences with pedometer-determined step counts were -7.3% and 7.0% for the Actical and ac-tivPAL monitors, respectively. With the exception of slow walking for the Actical units (ICC mill speeds, and across units during the free living condition. The 95% prediction interval ranges were wide, ranging from -68.8% to 54.2% for the Acticals, and from -39.1% to 53.2% for the activPALs. Step counts gathered from Actical and activPAL units should not be used interchangeably with pedometer-derived step count data.

To determine the role of corticotropin releasing factor receptor (CRF2) in epithelial permeability and enterocyte cell *** this purpose, we used rat Sprague Dawley and various colon carcinoma cell lines (SW620, HCT8R, HT-29 and Caco-2 cell lines). Expression of CRF2 protein was analyzed by fluorescent immunolabeling in normal rat colon and then by western blot in dissociated colonic epithelial cells and in the lysates of colon carcinoma cell lines or during the early differentiation of HT-29 cells (ten first days). To assess the impact of CRF2 signaling on colonic cell differentiation, HT-29 and Caco-2 cells were exposed to Urocortin 3 recombinant proteins (Ucn3, 100 nmol/L). In some experiments, cells were pre-exposed to the astressin 2b (A2b) a CRF2 antagonist in order to inhibit the action of Ucn3. Intestinal cell differentiation was first analyzed by functional assays: the trans-cellular permeability and the para-cellular permeability were determined by Dextran-FITC intake and measure of the transepithelial electrical resistance respectively. morphological modifications associated to epithelial dysfunction were analyzed by confocal microscopy after fluorescent labeling of actin (phaloidin-TRITC) and intercellular adhesion proteins such as E-cadherin, p120ctn, occludin and ZO-1. The establishment of mature adherens junctions (AJ) was monitored by following the distribution of AJ proteins in lipid raft fractions, after separation of cell lysates on sucrose gradients. Finally, the mRNA and the protein expression levels of characteristic markers of intestinal epithelial cell (IEC) differentiation such as the transcriptional factor krüppel-like factor 4 (KLF4) or the dipeptidyl peptidase IV (DPPIV) were performed by RT-PCR and western blot respectively. AIm:To determine the refractive accuracy of the Haigis,Barrett Universal II(Barrett),and Hill-radial basis function 2.0(Hill-RBF)intraocular lens(IOL)power calculations formulas in eyes undergoing manual cataract surgery(mCS)and refractive femtosecond laser-assisted cataract surgery(Re LACS).mETHODS:This was a REB-approved,retrospective interventional comparative case series of 158 eyes of 158 patients who had preoperative biometry completed using the IOL master 700 and underwent implantation of a Tecnis IOL following uncomplicated cataract surgery using either mCS or *** spherical equivalence(SE)was predicted using the Haigis,Barrett,and Hill-RBF *** older generation formula(Hoffer Q)was included in the *** refractive error(mE)was calculated one month *** lens factors of all formulas were retrospectively optimized to set the mE to 0 for each formula across all *** median absolute errors(medAE)and the proportion of eyes achieving an absolute error(AE)within 0.5 diopters(D)were compared between the two formulas among mCS and ReLACS eyes,***:Of the 158 eyes studied,64 eyes underwent mCS and 94 eyes underwent *** mCS eyes,the medAE did not differ between the formulas(P=0.59),however among ReLACS eyes,Barrett and Hill-RBF were more accurate(P=0.001).Barrett and Hill-RBF were both more likely to yield AEmong both groups(Pmula lead to greater refractive accuracy and likelihood of refractive success when compare to Haigis in eyes undergoing Re LACS.
