目的:研究三宝护童羹对变应性鼻炎幼鼠Th17/Treg细胞免疫失衡的影响。方法:将32只SD雌性大鼠幼鼠随机分为空白组、模型组、三宝护童羹组、地氯雷他定组,每组8只。除空白组外,其他组均以烟熏+大黄苦寒泻下法建立肺脾气虚证幼鼠模型,并运用腹腔注射卵清蛋白(OVA)和氢氧化铝Al(OH)3法进行基础致敏,再用OVA滴鼻法局部致敏,建立变应性鼻炎幼鼠模型。造模成功后,空白组、模型组、三宝护童羹组、地氯雷他定组分别予以生理盐水、生理盐水、三宝护童羹中药汤剂、地氯雷他定干混悬剂灌胃14天。观察幼鼠基础行为学改变,进行鼻部症状评分,HE染色观察鼻粘膜组织病理变化,采用ELISA法检测血清中白介素-17(IL-17)、转化生长因子β(TGF-β)水平,Western blot法检测脾脏中RORγt蛋白表达水平。结果:与空白组比较,模型组鼻黏膜上皮结构破坏,纤毛杂乱,上皮水肿,杯状细胞及嗜酸性粒细胞浸润;与模型组相比较,三宝护童羹组和地氯雷他定组给药后鼻部症状评分减少(P<0.05),鼻黏膜组织上皮纤毛较为整齐,上皮水肿减轻,杯状细胞及嗜酸性粒细胞浸润减少;与模型组相比较,三宝护童羹组和地氯雷他定组IL-17水平、脾脏RORγt蛋白表达量降低(P<0.05),TGF-β水平升高(P<0.05)。结论:三宝护童羹可改善变应性鼻炎肺脾气虚证幼鼠鼻部症状,减少炎性细胞浸润,其机制可能与调节Th17/Treg细胞免疫失衡和相关蛋白表达有关。

Cattle encephalon glycoside and ignotin(CEGI)injection is known as a multi-target neuroprotective drug that contains numerous liposoluble molecules,such as polypeptides,monosialotetrahexosyl ganglioside(GM-1),free amino acids,hypoxanthine and *** has been approved by the Chinese State Food and Drug Administration and widely used in the treatments of various diseases,such as stroke and Alzheimer's ***,the neuroprotective effects of CEGI beyond the time window of thrombolysis(within 4.5 hours)on acute ischemic stroke remain *** study constructed a rat middle cerebral artery occlusion model by suture-occluded method to simulate ischemic *** first daily dose was intraperitoneally injected at 8 hours post-surgery and the CEGI treatments continued for 14 *** of the modified five-point Bederson scale,beam balance test and rotameric test showed the neurological function of ischemic stroke rats treated with 4 m L/kg/d CEGI improved significantly,but the mortality within 14 days did not change *** MRI and 2,3,5-triphenyltetrazolium chloride staining confirmed that the infarct size in the 4 m L/kg/d CEGI-treated rats was significantly reduced compared with ischemic insult *** results of transmission electron microscopy and double immunofluorescence staining showed that the hippocampal neuronal necrosis in the ischemic penumbra decreased whereas the immunopositivity of new neuronal-specific protein doublecortin and the percentage of Ki67/doublecortin positive cells increased in CEGI-treated rats compared with untreated *** results suggest that CEGI has an effective neuroprotective effect on ischemic stroke when administered after the time window of *** study was approved by the Animal Ethics Committee of The Third Military Medical University,China.

建立HPLC-MS/MS法同时测定人全血中他克莫司和五酯胶囊4种主要成分五味子甲素、五味子乙素、五味子醇甲、五味子酯甲的浓度,并用于免疫抑制剂他克莫司联用保肝中药五酯胶囊临床药物浓度监测的研究中。采用了WatersAcquity UPLC■BEH C18色谱柱(50×2.1mm,1.7μm),以5mmol/L乙酸铵-0.1%甲酸水溶液为水相,乙腈为有机相,流速为0.3mL/min,柱温60℃。各成分均采用他克莫司同位素化合物(13C,2H4)作为内标(IS),全血样本首先经硫酸锌处理,再加入含内标的乙腈沉淀蛋白,最后离心后取上清液进样,质谱采用电喷雾离子源,以多反应离子监测(MRM)、正离子扫描模式检测。建立的HPLC-MS/MS法在1~100ng/m L范围内,他克莫司和五酯胶囊的4种主要成分色谱响应与浓度相关性良好,定量下限均为1ng/m L;他克莫司、五味子甲素、五味子乙素、五味子醇甲、五味子酯甲以及内标的保留时间分别为2.54、2.33、2.65、1.14、1.56、2.54min;准确度、回收率和经内标校准后的基质效应及稳定性数据均符合相关要求。方法灵敏、专属性强、准确性好,适用于人全血中他克莫司、五味子甲素、五味子乙素、五味子醇甲、五味子酯甲的浓度测定。

Objeetive To evaluate the effectiveness and to explore the mechanism of photodynamic therapy(PDT)for transplanted C6 glioma in rat brain and nude mice subeutis with different photosensitizers. Methods PDT was performed to treat transplanted C6 glioma in rat brain and nude mice subcutis using three different groups of photosensitizers as endogenous photoseusitizer of ALA,exogenous photosensitizer of HPD and both of ***-tumor-bearing group with PDT irradiation,tumor-beating group with simple light irradiation,and tumor-bearing group without irradiation were established as *** ethological changes, imaging features,and survival time of rats were *** microscope(LM),transmission electron microscope(TEM),and laser confocal scanning microscope(LCSM)were used to observe the morphological *** All the tumor-beating rats developed hemiparalysis,intermittent hyperspasmia of whole body with different extents within 3 days,and afterward the symptoms were alleviated gradually for PDT *** the PDT groups with three different photoseusitizers(ALA,HPD,ALA+HPD),tumor volumes increased transiently, then decreased gradually within 3 day,and reached 9.32±1.10mm ,7.87±1.51mm,7.20±3.26mm on 9th day after irradiation,and there was no death within 80 ***,the tumor volume of tumor-bearing group with simple light irradiation increased progressively and reached 92.05±10.10mm simultaneously,and mean survival was 38.5 days(P＜0.01).Various extents of interstitium edema or intracellular edema were developed in all experiment and control *** effect achieved peak after 3 days and recovered gradually after 5 days for non-tumor-bearing *** edema aggravated gradually for the tumor-bearing group with simple light irradiation,the vascular proliferation was active,and tumor cell growth was *** damage,rupture and hemorrhage in the tumor emerged gradually 1 day after PDT with three different photoseusitizers,as well as lysis and nec

Objective We sought to determine the safety and efficacy of using magnetoencephalography (MEG)to guide the resection of gliomas in eloquent *** From May to September of 2006,MEG was performed in 10 patients(age,5-57 years)before an attempted surgical resection of gliomas in functional *** 1.5-T MRI imagines,Functional location of MEG was used to identify and coregister eloquent cortices pertinent to motor(5 cases),speech(3 cases),motor and speech(1 cases),and sense and speech(1 cases)activation with respect to the *** with neuronavigation system,the MEG guided guided resection of gliomas in *** acquired brain-mapping or cortical stimulating were used to verify the veracity and postoperative CT and MR to assess the completeness of surgical resection in 2 *** Tumors included 9 astrocytomas,classified in WHO Ⅰ(4 cases),Ⅱ(1),Ⅲ(2),Ⅳ(2),and 1 oligogliomas. In every case,the preoperative brain activation study accurately determined the location of neurologic function. After surgery,one patient had a transient hemiparesis and another had a temporary apraxia.8 patients had radiographically complete resections,l cases got a subtotal resection,l with astrocytumas had incomplete resections because of the proximity of their tumors to functional areas(***).Four patients with an astrocytoma in the motor area received postoperative radiation *** date,radiographic tumor progression has not been seen in any patient with either a partial or a complete resection with a median follow-up of 6 weeks(range,3-12 weeks).Conclusions MEG-functional location was accurate for identifying areas of neumlogic function before surgical resection of gliomas in functional *** with complete radiographic resections or with incompletely resected can be safely after surgery.
