BMPRⅡ^(+)neural precursor cells isolated and characterized from organotypic neurospheres:an in vitro model of human fetal spinal cord development

Roof plate secretion of bone morphogenetic proteins(BMPs)directs the cellular fate of sensory neurons during spinal cord development,including the formation of the ascending sensory columns,though their biology is not well ***-ⅡBMP receptor(BMPRⅡ),the cognate receptor,is expressed by neural precursor cells during embryogenesis;however,an in vitro method of enriching BMPRⅡ^(+)human neural precursor cells(hNPCs)from the fetal spinal cord is *** was undertaken on intact second-trimester human fetal spinal cord using antibodies to BMPRⅡand leukemia inhibitory factor(LIF).Regions of highest BMPRⅡ^(+)immunofluorescence localized to sensory *** and meningeal-associated BMPRⅡ^(+)vascular cells were identified in both intact fetal spinal cord and cortex by co-positivity with vascular lineage markers,Cd34/*** immunostaining identified a population of somas concentrated in dorsal and ventral horn interneurons,mirroring the expression of LIF receptor/Cd118.A combination of LIF supplementation and high-density culture maintained culture growth beyond 10 passages,while synergistically increasing the proportion of neurospheres with a stratified,*** neurospheres were characterized by BMPRⅡ^(+)/MAP2ab^(+/–)/βⅢ-tubulin^(+)/nestin^(–)/vimentin^(–)/GFAP^(–)/NeuN^(–)surface hNPCs surrounding a heterogeneous core ofβⅢ-tubulin^(+)/nestin^(+)/vimentin^(+)/GFAP^(+)/MAP2ab^(–)/NeuN^(–)multipotent *** cultures from tripotential neurospheres contained neuronal(βⅢ-tubulin^(+)),astrocytic(GFAP+),and oligodendrocytic(O4+)lineage ***-activated cell sorting-sorted BMPRⅡ^(+)hNPCs were MAP2ab^(+/–)/βⅢ-tubulin^(+)/GFAP^(–)/O4^(–)in *** is the first isolation of BMPRⅡ^(+)hNPCs identified and characterized in human fetal spinal *** data show that LIF combines synergistically with high-density reaggregate cultures to support the organotypic reorganization of neurospheres,characterize

Using the polymerase chain reaction coupled with denaturing gradient gel electrophoresis to investigate the association between bacterial translocation and systemic inflammatory response syndrome in predicted acute severe pancreatitis

AIM: To investigate the use of PCR and dGGE to investigate the association between bacterial translocation and systemic inflammatory response syndrome in predicted severe ***: Patients with biochemical and clinical evidence of acute pancreatitis and an APACHE Ⅱ score ≥8 were enrolled. PCR and dGGE were employed to detect bacterial translocation in blood samples collected on d1,3, and 8 after the admission. Standard microbial blood cultures were taken when there was clinical evidence of sepsis or when felt to be clinically indicated by the supervising ***: Six patients were included. Of all the patients investigated, only one developed septic complications;the others had uneventful illness. Bacteria were detected using PCR in 4 of the 17 collected blood samples. The patient with sepsis was PCR-positive in two samples (taken on d 1 and 3), despite three negative blood cultures. Using dGGE and specific primers, the bacteria in all blood specimens which tested positive for the presence of bacterial dNA were identified as E ***: Our study confirmed thatunlike traditional microbiological techniques, PCR can detect the presence of bacteria in the blood of patients with severe AP. Therefore, this latter method in conjunction with dGGE is potentially an extremely useful tool in predicting septic morbidity and evaluating patients with the disease. Further research using increased numbers of patients, in particular those patients with necrosis and sepsis, is required to assess the reliability of PCR and dGGE in the rapid diagnosis of infection in AP.

运用扎根理论方法,探究低年级直博生学术热情消减的原因,指出学术热情消减是在个体入学前学习基础、动机与准备,教育情境中的重要他者和发展平台,以及就业前景等因素的作用下引发的行动回应,在不同个体身上体现为“主动放弃”或“被动抽离”两类维度,进而对个体后续发展趋向产生差异化的影响。据此提出了建议:贯通“后推免时期”与博士生教育阶段,提升本博衔接水平;因人施教,设定合理角色期望与支持策略;适应学生多元的发展需求,建设开放包容的博士生培养体系。

Magnetostrictive optomechanical cavities provide a new optical readout approach to room-temperature *** we report ultrasensitive and ultrahigh bandwidth cavity optomechanical magnetometers constructed by embedding a grain of the magnetostrictive material Terfenol-d within a high quality(Q)optical microcavity on a silicon *** engineering their physical structure,we achieve a peak sensitivity of26 p T∕Hz1/2 p comparable to the best cryogenic microscale magnetometers,along with a 3 d B bandwidth as high as 11.3 *** classes of magnetic response are observed,which we postulate arise from the crystallinity of the *** allows single crystalline and polycrystalline grains to be distinguished at the level of a single *** results may enable applications such as lab-on-chip nuclear magnetic spectroscopy and magnetic navigation.

青藏高原及其周边地区冰湖灾害频发,目前对其灾害特征及导致的破坏缺乏系统的分析.本研究旨在揭示冰湖灾害过去和未来的变化趋势及其对基础设施造成的影响,以指导区域及跨境流域冰湖灾害防控技术和策略的构建.研究梳理了1900-2022年间发生的298例冰湖灾害事件,发现其呈增长的趋势,尤其是1964年以来的部分冰湖灾害严重损坏了下游基础设施,不同类型冰湖灾害在空间分布、爆发和成灾特征等方面存在明显差异.例如,西斯帕冰川阻塞湖在2019-2022年连续爆发了5次溃决洪水,最近一次冲毁了下游中巴公路的桥梁和路基,影响中巴经贸.高山区冰川冰湖变化、交通和水电等基础设施及人口增长,致使冰湖灾害风险上升.为此,重大基础设施的规划应充分考虑相关风险,建立防控措施以保障人员和基础设施安全.

Single institution experience with the Ladd's procedure in patients with heterotaxy and stage Ⅰ palliated single-ventricle

AIM:To investigate and describe our current institutional management protocol for single-ventricle patients who must undergo a Ladd's ***:We retrospectively reviewed the charts of all patients from January 2005 to March 2014 who were diagnosed with heterotaxy syndrome and an associated intestinal rotation anomaly who carried a cardiac diagnosis of functional single ventricle and were status post stage I palliation.A total of 8 patients with a history of stage I single-ventricle palliation underwent Ladd's procedure during this time *** reviewed each patients chart to determine if significant intraoperative or post-operative morbidity or mortality *** also described our protocolized management of these patients in the cardiac intensive care unit,which included pre-operative labs,echocardiography,milrinone infusion,as well as protocolized fluid administration and anticoagulation *** also reviewed the literature to determine the reported morbidity and mortality associated with the Ladd's procedure in this particular cardiac physiology and if other institutions have reported protocolized care of these ***:A total of 8 patients were identified to have heterotaxy with an intestinal rotation anomaly and single-ventricle heart disease that was status post single ventricle *** of these patients were palliated with a Blaylock-Taussig shunt,one of whom underwent a Norwood *** two other patients were palliated with a stent,which was placed in the ductus *** eight patients all underwent elective Ladd's procedure at the time of gastrostomy tube *** our protocol,all patients remained on aspirin prior to surgery and had no period where they were without *** patients remained on milrinone during and after the procedure and received fluid administration upon arrival to the cardiac intensive care unit to account for *** 8 patients experienced no intraoperative or post-o
