Drug-induced cardiomyopathy:Characterization of a rat model by[^18 F]FDG/PET and[^99m Tc]MIBI/SPECT

Background:Drug-induced cardiomyopathy is a significant medical *** diagnosis of myocardial injury is based on initial electrocardiogram,levels of circulating biomarkers,and perfusion imaging with single photon emission computed tomography(SPECT).Positron emission tomography(PET)is an alternative imaging modality that provides better resolution and sensitivity than SPECT,improves diagnostic accuracy,and allows therapeutic *** objective of this study was to assess the detection of drug-induced cardiomyopathy by PET using 2-deoxy-2-[^18F]fluoro-D-glucose(FDG)and compare it with the conventional SPECT technique with[^99m Tc]-Sestamibi(MIBI).Methods:Cardiomyopathy was induced in Sprague Dawley rats using high-dose ***[^18F]FDG/PET and[^99m Tc]MIBI/SPECT were performed before and after isoproterenol administration.[^18F]FDG(0.1 mCi,200-400μL)and[^99m Tc]MIBI(2 mCi,200-600μL)were administered via the tail vein and imaging was performed 1 hour ***-induced injury was confirmed by the plasma level of cardiac troponin and triphenyltetrazolium chloride(TTC)***:Isoproterenol administration resulted in an increase in circulating cardiac troponin I and showed histologic damage in the ***,preisoproterenol and postisoproterenol images showed alterations in cardiac accumulation of[^18F]FDG,but not of[^99m Tc]*** analysis revealed that myocardial uptake of[^18F]FDG reduced by 60%after isoproterenol treatment,whereas that of[^99m Tc]MIBI decreased by 45%.Conclusion:We conclude that[^18F]FDG is a more sensitive radiotracer than[^99m Tc]MIBI for imaging of drug-induced *** theorize that isoproterenolinduced cardiomyopathy impacts cellular metabolism more than perfusion,which results in more substantial changes in[^18F]FDG uptake than in[^99m Tc]MIBI accumulation in cardiac tissue.

Over the past few years, more and more higher education institutions have pledged to achieve carbon neutrality and designed and adopted Climate Action Plans. Although many higher institutions are adopting climate action plans, few are integrating resil-ience principles and priorities, which are essential for understanding institutions’ adaptive capacity for dealing with climate change. There is little existing research on how higher education institutions can implement climate resilience programs, behav-iors, and policies into their planning process and campus-communities. To address this gap, this case study explores Arizona State University’s process of designing and implementing a climate resilience plan and outlines best practices other higher educa-tion institutions can utilize to create their own climate resilience plan. We critically discuss the importance of climate resilience at the higher education level, outline steps necessary for designing an inclusive and holistic climate resilience plan, and provide examples of important techniques used to design the climate resilience plan.

Numerical Simulations of Heavy Rainfall over Central Korea on 21 September 2010 Using the WRF Model

On 21 September 2010, heavy rainfall with a local maximum of 259 mm d-1 occurred near Seoul, South Korea. We examined the ability of the Weather Research and Forecasting （WRF） model in reproducing this disastrous rainfall event and identified the role of two physical processes： planetary boundary layer （PBL） and microphysics （MPS） processes. The WRF model was forced by 6-hourly National Centers for Environmental Prediction （NCEP） Final analysis （FNL） data for 36 hours form 1200 UTC 20 to 0000 UTC 22 September 2010. Twenty-five experiments were performed, consisting of five different PBL schemes--Yonsei University （YSU）, Mellor-Yamada-Janjic （MYJ）, Quasi Normal Scale Elimination （QNSE）, Bougeault and Lacarrere （BouLac）, and University of Washington （UW）--and five different MPS schemes--WRF Single- Moment 6-class （WSM6）, Goddard, Thompson, Milbrandt 2-moments, and Morrison 2-moments. As expected, there was a specific combination of MPS and PBL schemes that showed good skill in forecasting the precipitation. However, there was no specific PBL or MPS scheme that outperformed the others in all aspects. The experiments with the UW PBL or Thompson MPS scheme showed a relatively small amount of precipitation. Analyses form the sensitivity experiments confirmed that the spatial distribution of the simulated precipitation was dominated by the PBL processes, whereas the MPS processes determined the amount of rainfall. It was also found that the temporal evolution of the precipitation was influenced more by the PBL processes than by the MPS processes.

Obesity has a multifactorial etiology and is known to be a state of chronic low-grade inflammation,known as *** state is associated with the development of metabolic disorders such as glucose intolerance and nonalcoholic fatty liver *** is a glycolytic metabolite and a crucial node in various metabolic ***,its role and molecular mechanism in obesity and associated complications are *** this study,we reported that pyruvate substantially inhibited adipogenic differentiation in vitro and its administration significantly prevented HFD-induced weight gain,white adipose tissue inflammation,and metabolic *** identify the target proteins of pyruvate,drug affinity responsive target stability was employed with proteomics,cellular thermal shift assay,and isothermal drug response to detect the interactions between pyruvate and its molecular ***,we identified cytosolic phospholipase A2(cPLA2)as a novel molecular target of pyruvate and demonstrated that pyruvate restrained diet-induced obesity,white adipose tissue inflammation,and hepatic steatosis in a cPLA2-dependent *** with global ablation of cPLA2 in mice showed that the protective effects of pyruvate were largely abrogated,confirming the importance of pyruvate/cPLA2 interaction in pyruvate attenuation of inflammation and ***,our study not only establishes pyruvate as an antagonist of cPLA2 signaling and a potential therapeutic option for obesity but it also sheds light on the mechanism of its ***’s prior clinical use indicates that it can be considered a safe and viable alternative for obesity,whether consumed as a dietary supplement or as part of a regular diet.

High yield reproducible rat model recapitulating human Barrett's carcinogenesis

AIM To efficiently replicate the biology and pathogenesis of human esophageal adenocarcinoma(EAC) using the modified Levrat model of end-to-side esophagojejunostomy. METHODS End-to-side esophagojejunostomy was performed on rats to induce gastroduodenoesophageal reflux to develop EAC. Animals were randomly selected and serially euthanized at 10(n = 6),17(n = 8),24(n = 9),31(n = 6),38(n = 6),and 40(n = 6) wk postoperatively. The esophagi were harvested for downstream histopathology and gene expression. Histological evaluation wascompleted to determine respective rates of carcinogenic development. Quantitative reverse transcriptionpolymerase chain reaction was performed to determine gene expression levels of MUC2,CK19,and CK20,and results were compared to determine significant differences throughout disease progression *** The overall study mortality was 15%. Causes of mortality included anastomotic leak,gastrointestinal hemorrhage,stomach ulcer perforation,respiratory infection secondary to aspiration,and obstruction due to tumor or late anastomotic stricture. 10 wk following surgery,100% of animals presented with esophagitis. Barrett's esophagus(BE) was first observed at 10 wk,and was present in 100% of animals by 17 wk. Dysplasia was confirmed in 87.5% of animals at 17 wk,and increased to 100% by 31 wk. EAC was first observed in 44.4% of animals at 24 wk and increased to 100% by 40 wk. In addition,two animals at 38-40 wk post-surgery had confirmed macro-metastases in the lung/liver and small intestine,respectively. MUC2 gene expression was progressively down-regulated from BE to dysplasia to EAC. Both CK19 and CK20 gene expression significantly increased in a stepwise manner from esophagitis to EAC. CONCLUSION Esophagojejunostomy was successfully replicated in rats with low mortality and a high tumor burden,which may facilitate broader adoption to study EAC development,progression,and therapeutics.
