A highly immunosuppressive tumor microenvironment(TME)and the presence of the blood‒brain barrier are the two major obstacles to eliciting an effective immune response in patients with high-grade glioma(HGG).Here,we tried to enhance the local innate immune response in relapsed HGG by intracranially injecting poly(I:C)to establish a robust antitumor immune response in this registered clinical trial(NCT03392545).During the follow-up,12/27(44.4%)patients who achieved tumor control concomitant with survival benefit were regarded as responders in our study.We found that the T-cell receptor(TCR)repertoire in the TME was reshaped after poly(I:C)treatment.Based on the RNA-seq analysis of tumor samples,the expression of annexin A1(ANXA1)was significantly upregulated in the tumor cells of nonresponders,which was further validated at the protein level.In vitro and in vivo experiments showed that ANXA1 could induce the production of M2-like macrophages and microglia via its surface receptor formyl peptide receptor 1(FPR1)to establish a Treg cell-driven immunosuppressive TME and suppress the antitumor immune response facilitated by poly(I:C).The ANXA1/FPR1 signaling axis can inhibit the innate immune response of glioma patients by promoting an anti-inflammatory and Treg-driven TME.Moreover,ANXA1 could serve as a reliable predictor of response to poly(I:C),with a notable predictive accuracy rate of 92.3%.In light of these notable findings,this study unveils a new perspective of immunotherapy for gliomas.

Electrocatalytically converting nitrate waste to value-added ammonia(NO3−RR)is beneficial to environmental restoration and an attractive candidate for green ammonia production[1].However,developing NO_(3)^(-)RR in an energy-saving way remains a significant challenge due to the slow kinetics inherent in multi-step electron and proton transfer,making previous investigations for NO_(3)^(-)RR generally operate at a large overpotential(<-0.2 V versus reversible hydrogen electrode(RHE))to achieve optimal efficiency and industrial current for NH3 synthesis[2].

Incompatible numerical manifold method for fracture problems

The incompatible numerical manifold method （INMM） is based on the finite cover approximation theory, which provides a unified framework for problems dealing with continuum and discontinuities. The incompatible numerical manifold method employs two cover systems as follows. The mathematical cover system provides the nodes for forming finite covers of the solution domain and the weighted functions, and the physical cover system describes geometry of the domain and the discontinuous surfaces therein. In INMM, the mathematical finite cover approximation theory is used to model cracks that lead to interior discontinuities in the process of displacement. Therefore, the discontinuity is treated mathematically instead of empirically by the existing methods. However, one cover of a node is divided into two irregular sub-covers when the INMM is used to model the discontinuity. As a result, the method sometimes causes numerical errors at the tip of a crack. To improve the precision of the INMM, the analytical solution is used at the tip of a crack, and thus the cover displacement functions are extended with higher precision and computational efficiency. Some numerical examples are given.

Prognostic implications of epidermal growth factor receptor variant Ⅲ expression and nuclear translocation in Chinese human gliomas

Objective: To determine the prognostic implications and clinical significance of epidermal growth factor receptor variant Ⅲ(EGFRvⅢ) expression and EGFRvⅢ nuclear translocation in Chinese human gliomas.Methods: We retrospectively examined EGFRvⅢ expression and EGFRvⅢ nuclear translocation using immunohistochemistry in specimens of 240 Chinese patients with glioma, including 84 World Health Organization(WHO) II gliomas, 84 WHO Ⅲ gliomas and 72 glioblastomas(WHO IV). Factors that correlated with EGFRvⅢ and EGFRvⅢ nuclear translocation expression were analyzed by the Chi-square test. Kaplan-Meier methodology and Cox regression were used for the survival analysis.Results: Log-rank tests showed that patient age, Karnofsky performance scale(KPS) score, tumor grade,EGFRvⅢ expression, EGFRvⅢ nuclear translocation, 1 p/19 q codeletion, isocitrate dehydrogenase(IDH)mutation, Ki-67 labeling index and O6-methylguanine-DNA methyltransferase(MGMT) status(P<0.05) were significantly correlated with overall survival(OS) time. Multivariate Cox regression analysis revealed that patient age, tumor grade, EGFRvⅢ nuclear translocation, 1 p/19 q codeletion, and IDH mutation(P<0.05) were significantly correlated with OS. Patients with a high level of EGFRvⅢ nuclear translocation(≥7%) had both significantly shorter OS [hazard ratio(HR): 1.920, 95% confidence interval(95% CI): 1.228-3.003, P=0.004] and progression-free survival(PFS) times(HR: 1.661, 95% CI: 1.116-2.471, P=0.012) than those with a low level of EGFRvⅢ nuclear translocation(<7%).Conclusions: A high level of EGFRvⅢ nuclear translocation in glioma is an independent factor indicating a poor prognosis, but EGFRvⅢ expression is not an independent clinical prognostic factor. The level of EGFRvⅢ nuclear translocation maybe a novel and crucial prognostic biomarker in glioma.

Research progress on the substrate for metal-organic framework(MOF) membrane growth for separation

During the last decade, metal-organic frameworks(MOFs) have been applied in various fields due to their unique chemical and functional advantages. One of the widespread research hotspots is MOF-based membranes for separations, specifically continuous defect-free MOF membranes, which are usually grown on porous substrates. The substrate not only serves as the MOF layer support but also has a great influence on the membrane fabrication process and the final separation performance of the resultant membrane. In this review, we mainly introduce the progress focused on the substrates for MOF membranes fabrication. The substrate modifications and seeding methods aimed at synthesizing highquality MOF membranes are also summarized systematically.

Plant-derived squalene supplementation improves growth performance and alleviates acute oxidative stress-induced growth retardation and intestinal damage in piglets

Piglets are particularly susceptible to oxidative stress,which causes inferior growth performance and intestinal damage.Squalene(SQ),a natural bioactive substance enriched in shark liver oil,shows excellent antioxidant properties and can currently be obtained at a low cost from deodorizer distillate during the production of plant oil.This study aimed to evaluate the effects of plant-derived SQ supplementation on the growth performance of piglets and explore the beneficial roles of SQ against oxidative stress and intestinal injury in diquat-challenged piglets.Forty piglets were randomly divided into five groups and fed a basal diet supplemented with SQ at 0,500,1000,or 2000 mg/kg for 5 weeks.Acute oxidative stress was induced in the piglets with diquat(10 mg/kg BW)at the fourth week of the experiment,followed by a 7-d recovery period.Results showed that before the diquat challenge,SQ supplementation significantly improved growth performance(average daily gain and feed conversion ratio)and serum antioxidant status,and after the diquat challenge,SQ supplementation significantly mitigated diquat-induced growth arrest,intestinal villous atrophy,intestinal epithelial cell apoptosis,intestinal hyperpermeability,and deficiency of intestinal epithelial tight junction proteins(zonula occludens-1,occludin,and claudin-3).Under oxidative stress induced by diquat,SQ supplementation consistently improved the antioxidant status of the small intestine,liver,and muscle.In vitro,SQ was shown to alleviate hydrogen peroxide(H2O2)-induced increase of the levels of intracellular reactive oxygen species and apoptosis of porcine intestinal epithelial cells.Taken together,SQ supplementation improves growth performance and effectively alleviates acute oxidative stress-induced growth retardation and intestinal injury via improving antioxidant capacity in piglets.Our findings may provide an efficient strategy for alleviating oxidative stress-induced inferior growth performance and intestinal damage in piglets.

在江苏—白鹤滩±800 kV特高压混合直流输电工程中,采用等离子体喷射触发型气体间隙开关可以对可控避雷器进行旁路控制,具有响应速度快,绝缘恢复能力强,通流容量大等优势,应用前景广泛,但触发间隙气体氛围的优化选择尚有所缺失,间隙触发特性和寿命的提升仍有待进一步研究。搭建了等离子体喷射触发实验平台,研究了等离子体喷射触发气体间隙的触发性能劣化规律,分别在SF_(6)和SF_(6)/N_(2)混合气体中探究了间隙触发的临界电路参数,在确定的电路参数下进行触发寿命实验同时探究性能劣化规律。结果表明:相同绝缘强度下,当间隙电压为10 kV时,0.2 MPa SF_(6)氛围下间隙最低可触发电压为3.5 k V,而在0.266 MPa SF_(6)/N_(2)氛围下降低为2.6 kV;在最佳触发电路参数条件和相同绝缘强度下,实验初期,SF_(6)中间隙导通时延为~270μs,等离子体最大喷射高度为4.8 cm,最大喷射速度为1296 m/s,而在SF_(6)/N_(2)中间隙导通时延为~100μs,等离子体最大喷射高度为7.2 cm,最大喷射速度为1525 m/s,而随着放电次数的增加,间隙导通时延增加,等离子体最大喷射高度降低,最大喷射速度减小,直到间隙第一次触发失败达到间隙寿命,SF_(6)中间隙的触发寿命仅有695次,而SF_(6)/N_(2)混合气体中间隙的触发寿命达到了1540次。对可控避雷器用气体间隙触发开关的性能和寿命提升提供了理论指导,同时有助于其在工程领域推广应用。

【目的】研究葡萄渣对肉母牛外周血淋巴细胞亚群、淋巴细胞转化率与凋亡率及抗氧化性的影响。【方法】选取胎次、体重相近的产后健康肉母牛20头,随机分为4组,每组5头。对照组饲喂基础饲粮,试验Ⅰ、Ⅱ、Ⅲ组每头在基础饲粮中分别添加240、300、360 g/d的葡萄渣。试验预试期15 d,正试期40 d。采集尾静脉血,测定其外周血淋巴细胞亚群、淋巴细胞的转化率与凋亡率及其抗氧化水平。【结果】①试验第20天,3个试验组肉母牛外周血CD4+T细胞比例较对照组均显著上升(P<0.05);试验第40天,试验Ⅱ、Ⅲ组CD4+T细胞比例较对照组显著升高(P<0.05),CD8+T细胞比例较对照组显著降低(P<0.05),3个试验组CD4+/CD8+比值较对照组均显著升高(P<0.05)。②试验第20、40天,试验Ⅱ、Ⅲ组母牛外周血淋巴细胞转化率均极显著高于对照组(P<0.01)。试验第40天,随着葡萄渣添加量升高,外周血淋巴细胞凋亡率呈线性下降(P<0.01)。③试验第40天,3个试验组肉母牛血清MDA含量较对照组均显著降低(P<0.05),试验Ⅰ、Ⅲ组肉母牛血清T-AOC含量均极显著高于对照组(P<0.01)。【结论】肉母牛饲粮添加不同比例葡萄渣可以改善肉牛的免疫功能,本试验条件下,葡萄渣的适宜添加水平为(300~360)g/d。

The roles and regulation of Yes-associated protein 1 in stem cells

Yes-associated protein 1(YAP1)is a downstream effector of the Hippo signaling pathway,and it is involved in tumorigenesis,tissue repair,growth,and development.In this review,the biological roles and the mechanisms of YAP1 in mediating stem cell fate decisions are discussed,including cell proliferation,differentiation,and apoptosis.In general,YAP1 promotes the proliferation and differentiation of stem cells,including embryonic stem cells and adult stem cells.It inhibits apoptosis by binding to the transcription factors,e.g.,transcriptional enhanced associate domain(TEAD),Smad,runt-related transcription factor 1/2,p73,p63,and Erb84,to maintain tissue homeostasis.The translocalization of YAP1 in cellular nuclei and the phosphorylation in the cytoplasm work as important and unusual events for the activation of YAP1.Moreover,YAP1 serves as the crosstalk for the Hippo pathway and other signaling pathways,including the Wnt and Notch pathways.It is highlighted in this review that YAP1 is an essential regulator for stem cells that have significant applications in regenerative medicine and reproductive medicine.
