The characterization and functions of transmembrane protein 59-like(TMEM59L),a type I transmembrane protein,are not clearly understood until now.Some TMEM59L and fluorescent fusion proteins constructs were transfected in cell lines and liposomes,and their localization was observed.The effects of protein constructs were studied by fluorescence microscopy and western blotting.This study reports a novel function of human TMEM59L(hTMEM59L)related to the expression and location of some proteins.In addition,we report two novel splice variants of human TMEM59L(hTMEM59L).The localization of mutants of this protein,lacking a middle region,and a Cterminal deletion,markedly differed from that of full-length hTMEM59L.Intracellular movement assessment in living cells showed the localization of TMEM59L to vesicular structures in the Golgi bodies,and the cell membrane was observed in living cells.Overexpression of TMEM59L markedly increased the level of amyloid precursor protein because of TMEM59L-mediated inhibition of cell membrane transport but did not affect the expression of betasecretase 2.TMEM59L overexpression also significantly increased the levels of Rab GDP dissociation inhibitor alpha and Rab GDP dissociation inhibitor beta proteins.These results suggest that TMEM59L is involved in the packaging of acidic vesicles and thereby functions in the trafficking and processing of intracellular proteins.

The flowability of granular material is commonly determined from shear cell tests,but in the real world,the granular flow is quite different in many cases.For this paper,the flowabilities in a narrow channel of five different granular materials were investigated.The flow parameters of the granular flows were determined to classify the flowabil让ies.Granular flow surface velocities and flow heights and thicknesses were measured,and it was found that the mean shear rates in the linear portions of the velocity profiles in crease with both flow rates and incli nation.A universal parameter,the ratio of the sine of the inclin ation to the shear rate in the linear part of the velocity profile,is proposed as a constant.This constant is independent of particle properties.In addition,a flowability parameter,the ratio of the mean shear rate to the cosine of the inclination,is proposed to characterize the friction between the particles in the flowing granular material.

Recommender system is an effective tool to solve the problems of information overload.The traditional recommender systems,especially the collaborative filtering ones,only consider the two factors of users and items.While social networks contain abundant social information,such as tags,places and times.Researches show that the social information has a great impact on recommendation results.Tags not only describe the characteristics of items,but also reflect the interests and characteristics of users.Since the traditional recommender systems cannot parse multi-dimensional information,in this paper,a tensor decomposition model based on tag regularization is proposed which incorporates social information to benefit recommender systems.The original Singular Value Decomposition(SVD)model is optimized by mining the co-occurrence and mutual exclusion of tags,and their features are constrained by the relationship between tags.Experiments on real dataset show that the proposed algorithm achieves superior performance to existing algorithms.

Flower development is one of the most vital pathways in plant development, during which the epigenetic regulation of gene expression is essential. DNA methylation, the most conserved epigenetic modification, participates in gene expression regulation and transposable element silencing. Honeysuckle(Lonicera japonica) is an important medicinal plant renowned for its colorful and fragrant flowers. Honeysuckle flowers change color from white to gold as a result of carotenoid accumulation during development. However, the role of DNA methylation in flower color changes is not well understood in L. japonica. Here, we performed whole-genome bisulfite sequencing and transcriptome sequencing during flowering development in honeysuckle. The results showed that a decrease in the levels of genome-wide average DNA methylation during flower development and changes in DNA methylation were associated with the expression of demethylase genes. Moreover, many genes involved in carotenoid biosynthesis and degradation, such as Lj PSY1, LjPDS1, LjLCYE, and LjCCD4, have altered expression levels because of hypomethylation, indicating that DNA methylation plays an important role in flower color changes in honeysuckle. Taken together, our data provide epigenetic insights into flower development and color change in honeysuckles.

High-quality development of ethnic minority areas is a key issue in China’s economic and social transformation.In order to explore the development trend and optimization path of high-quality development in ethnic minority areas,this study constructed a systematic evaluation index system of economic development,social development and government governance.The coupled coordination model was adopted to quantitatively analyze the spatio-temporal evolution characteristics of economic development,social development and government governance in Xinjiang from 2013 to 2020.The influencing factors of coupling coordination development of the three systems were identified by grey correlation analysis.The spatial-temporal pattern analysis results show that the coupling coordination degree of Xinjiang increased steadily,but the regional differences gradually expanded.The coupling coordination degree shows a stable structure with high center and low periphery,and a spatial pattern of low in southern Xinjiang and high in northern Xinjiang.The results of grey correlation analysis show that the grey correlation degree among economic development,social development and government governance in Xinjiang is above 0.78.The three systems of economic development,social development and government governance interact with each other,forming a complex system of high-quality development in ethnic minority areas.To promote highquality development in Xinjiang,it should take social stability and long-term stability as the overall goal,improve the quality of the production and living environment on the basis of economic development,enhance regional economic competitiveness,and take government governance as a means to optimize the environment for regional development.

A nonlinear mathematical model for hydro turbine governing system with saturation nonlinearity in small perturbation has been proposed with all the essential components,i.e. turbine,PID type governor with saturation part and generator included in the model. Existence,stability and direction of Hopf bifurcation of an example HTGS are investigated in detail and presented in forms of bifurcation diagrams and time waveforms. The analysis show that a supercritical Hopf bifurcation may exist in hydraulic turbine systems in some certain conditions. Moreover,the dynamic behavior of system with different parameters such as Tw,Tab,Tyand K are studied extensively. An example with numerical simulations is presented to illustrate the theoretical results. The researches provide a reasonable explanation for the Hopf phenomenon happened in operation of hydroelectric generating unit.

A model predictive control algorithm of closed-loop supply chain networks dynamic system is studied.Based on the established system prediction model,the calculation formula of state estimation and prediction are given;the multi-stage optimal prediction model expressions are deduced;the objective function and the feedback correction method are given;the constrained optimization algorithm based on quadratic optimization is derived;and the general steps of this algorithm is given by a numerical example.The essence of this algorithm is that through the rolling optimization and continuous implementations,adjust the order quantity,production control variables,the system has good dynamic performances in the premise of stability,the bullwhip effect is restrained.In order to get the stability and coordination control method of the complex nonlinear closed-loop supply chain networks,it provides a new method.

ZK60B(Mg-6%Zn-0.6%Zr)alloy joints fabricated by bobbin tool friction stir welding(BTFSW)with various traverse speeds were investigated.The sound joint fabricated by the BTFSW was possible under the appropriate welding parameters.The severe plastic deformation during BTFSW resulted in dispersion and segregation of the Zr-rich particles within the stirred zone(SZ)followed by evolution of a bimodal grain structure with distributed bands of 0.8-1.7μm ultrafine grains and 4.1-7.1μm equiaxed grains.Micro-hardness of SZ is substantially reduced in contrast to that of parent metal(PM)in spite of the finer grain size owing to dissolution of Mg-Zn based precipitates having hardening effects on alpha-Mg matrix.With the decrease in traverse speed,randomization degree of the plasticized metal flow increases,which is evidenced by the randomized arc line pattern at the low traverse speed.Among all defect-free joints,the 200 mm/min joint exhibits the weakest isotropy of texture within SZ and the best tensile properties,which has reduced ultimate tensile strength and yield strength by 5.4% and by 22.2%,respectively,as compared to the PM.The randomized texture hinders the joint fracturing within SZ at low elongation.Therefore,a relatively high elongation of 10.8% was achieved,which corresponded to 72% of the PM value.
