Long-term nonunion of bone defects has always been a major problem in orthopedic *** bone graft materials such as Poly(lactic-co-glycolic acid)/β-tricalcium phosphate(PLGA/β-TCP)scaffolds are expected to solve this problem due to their suitable degradation rate and good ***,insufficient mechanical properties,lack of osteoinductivity and infections after implanted limit its large-scale clinical ***,we proposed a novel bone repair bioscaffold by adding zinc submicron particles to PLGA/β-TCP using low temperature rapid prototyping 3D printing *** first screened the scaffolds with 1 wt%Zn that had good biocompatibility and could stably release a safe dose of zinc ions within 16 weeks to ensure long-term *** designed,the scaffold had a multi-level porous structure of biomimetic cancellous bone,and the Young’s modulus(63.41±1.89 MPa)and compressive strength(2.887±0.025 MPa)of the scaffold were close to those of cancellous *** addition,after a series of in vitro and in vivo experiments,the scaffolds proved to have no adverse effects on the viability of BMSCs and promoted their adhesion and osteogenic differentiation,as well as exhibiting higher osteogenic and anti-inflammatory properties than PLGA/β-TCP scaffold without zinc *** also found that this osteogenic and anti-inflammatory effect might be related to Wnt/β-catenin,P38 MAPK and NFkB *** study lay a foundation for the follow-up study of bone regeneration mechanism of Zn-containing *** envision that this scaffold may become a new strategy for clinical treatment of bone defects.

The Brodmann area(BA)-based map is one of the most widely used cortical maps for studies of human brain functions and in clinical practice;however,the molecular architecture of BAs remains *** present study provided a global multiregional proteomic map of the human cerebral cortex by analyzing 29 *** 29 BAs were grouped into 6 clusters based on similarities in proteomic patterns:the motor and sensory cluster,vision cluster,auditory and Broca’s area cluster,Wernicke’s area cluster,cingulate cortex cluster,and heterogeneous function *** identified 474 cluster-specific and 134 BA-specific signature proteins whose functions are closely associated with specialized functions and disease vulnerability of the corresponding cluster or *** findings of the present study could provide explanations for the functional connections between the anterior cingulate cortex and sensorimotor cortex and for anxiety-related function in the sensorimotor *** brain transcriptome and proteome comparison indicates that they both could reflect the function of cerebral cortex,but show different *** proteomic data are publicly available at the Human Brain Proteome Atlas(***).Our results may enhance our understanding of the molecular basis of brain functions and provide an important resource to support human brain research.

目的探究20种常见氨基酸在结直肠癌(CRC)患者的尿液特征;寻找CRC诊断的生物学标志物。方法纳入100名健康对照者和113名CRC患者(Ⅰ期27例,Ⅱ期28例,Ⅲ期34例,Ⅳ期24例),基于超高效液相色谱串联质谱法对尿液中的20种常见氨基酸应用多反应监测技术进行靶向定量分析,应用统计学方法分析CRC患者与健康对照氨基酸含量差异,并构建CRC诊断模型。结果CRC患者尿液中16种氨基酸含量与健康对照差异有统计学意义,Ⅰ,Ⅱ,Ⅲ和Ⅳ期各自有9种,15种,14种,17种氨基酸含量与健康对照差异有统计学意义。由异亮氨酸,天冬酰胺,脯氨酸,组氨酸,酪氨酸,苯丙氨酸6种氨基酸构建的生物标志物组合用于CRC诊断的AUC值在发现组/验证组分别为0.857/0.851,在Ⅰ,Ⅱ,Ⅲ和Ⅳ期CRC的AUC值分别为0.847,0.843,0.897,0.896。结论尿液中20种氨基酸含量变化可以用于CRC诊断,为CRC的早期诊断提供了新的研究思路。

Proteomic survey towards the tissue-specific proteins of mouse mitochondria

Mitochondrion plays the key functions in mammalian cells. It is believed that mitochondrion exerts the common biologic functions in many tissues, but also performs some specific functions correspondent with tissues where it is localized. To identify the tissue-specific mitochondrial proteins, we carried out a systematic survey towards mitochondrial proteins in the tissues of C57BL/6J mouse, such as liver, kidney and heart. The mitochondrial proteins were separated by 2DE and identified by MALDI-TOF/TOF MS. Total of 87 unique proteins were identified as the tissue-specific ones, and some representatives were further verified through ICPL quantification and Western blot. Because these issue-specific proteins are coded from nuclear genes, real-time PCR was employed to examine the mRNA status of six typical genes found in the *** combining of the expression data and the co-localization images obtained from confocal microscope, we came to the conclusion that the tissue-specifically mitochondrial proteins were widely distributed among the mouse tissues. Our investigation, therefore, indeed provides a solid base to further explore the biological significance of the mitochondrial proteins with tissue-orientation.

Background:The human brain is the most complex organ in the body,and it is important to have a better understanding of how the protein composition in the brain regions contributes to the pathogenesis of associated neurological ***:In this study,a comparative analysis of the frontal and temporal cortex proteomes was conducted by isobaric tags of relative and absolute quantification(iTRAQ)labeling and two-dimensional liquid chromatographytandem mass spectrometry(2D LC-MS/MS).Brain protein was taken from relatively normal tissue that could not be avoided of damage during emergent surgery of the TBI(traumatic brain injury)patients admitted in Beijing Tiantan Hospital from 2014 to *** cases were *** frontal lobes and 4 temporal lobes proteome were analyzed and the proteins were *** Ontology(GO),Ingenuity Pathway Analysis(IPA),and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were used to analyze the biological function of identified proteins,unchanged proteins,and differentially expressed proteins(DEPs).Results:A total number of 2127 protein groups were identified in the frontal and temporal lobe proteomes.A total of 1709 proteins could be quantitated in both the frontal and temporal *** 90 DEPs,14 proteins were screened highly expressed in the temporal cortex,including MAPT,SNCG,ATP5IF1,GAP43,HSPE1,STMN1,NDUFS6,LDHB,SNCB,NDUFA7,MRPS36,EPDR1,CISD1,and *** addition,compared to proteins expressed in the frontal cortex,14 proteins including EDC4,NIT2,VWF,ASTN1,TGM2,SSB,CLU,HBA1,STOM,CRP,LRG1,SAA2,S100A4,and VTN were a low expression in the temporal *** biological process enrichment showed that unchanged proteins between the frontal and temporal cortex mainly take part in regulated exocytosis,axon guidance,and vesicle-mediated *** KEGG pathway analysis showed that unchanged proteins between the frontal and temporal cortex mainly take part in oxidative phosphorylation,carbon metabolism,H

This paper deals with numerical methods for solving one-dimensional(1D)and twodimensional(2D)initial-boundary value problems(IBVPs)of space-fractional sine-Gordon equations(SGEs)with distributed *** 1D problems,we construct a kind of oneparameter finite difference(OPFD)*** is shown that,under a suitable condition,the proposed method is convergent with second order accuracy both in time and *** implementation,the preconditioned conjugate gradient(PCG)method with the Strang circulant preconditioner is carried out to improve the computational efficiency of the OPFD *** 2D problems,we develop another kind of OPFD *** such a method,two classes of accelerated schemes are suggested,one is alternative direction implicit(ADI)scheme and the other is ADI-PCG *** particular,we prove that ADI scheme can arrive at second-order accuracy in time and *** some numerical experiments,the computational effectiveness and accuracy of the methods are further ***,for the suggested methods,a numerical comparison in computational efficiency is presented.

Dominant woody plant species recognition with a hierarchical model based on multimodal geospatial data for subtropical forests

Since the launch of the Google Earth Engine(GEE)cloud platform in 2010,it has been widely used,leading to a wealth of valuable ***,the potential of GEE for forest resource management has not been fully *** extract dominant woody plant species,GEE combined Sen-tinel-1(S1)and Sentinel-2(S2)data with the addition of the National Forest Resources Inventory(NFRI)and topographic data,resulting in a 10 m resolution multimodal geospatial dataset for subtropical forests in southeast *** and texture features,red-edge bands,and vegetation indices of S1 and S2 data were computed.A hierarchical model obtained information on forest distribution and area and the dominant woody plant *** results suggest that combining data sources from the S1 winter and S2 yearly ranges enhances accuracy in forest distribution and area extraction compared to using either data source ***,for dominant woody species recognition,using S1 winter and S2 data across all four seasons was *** terrain factors and removing spatial correlation from NFRI sample points further improved the recognition *** optimal forest extraction achieved an overall accuracy(OA)of 97.4%and a maplevel image classification efficacy(MICE)of 96.7%.OA and MICE were 83.6%and 80.7%for dominant species extraction,*** high accuracy and efficacy values indicate that the hierarchical recognition model based on multimodal remote sensing data performed extremely well for extracting information about dominant woody plant *** the results using the GEE application allows for an intuitive display of forest and species distribution,offering significant convenience for forest resource monitoring.
