Cross-Talk between Extracellular S1P/S1P2 and P42/44 MAPK in bcr/abl Positive Chronic Myeloid Leukemia Cells

Objective： BCR/ABL oncoprotein-expression is associated with uncontrolled cell growth. Sphingosine kinase 1 （SPK1） regulates the production of sphingosine 1-phosphate （S1P）, a key lipid signal molecular in cell proliferation and survival. The objective of this study was to elucidate the roles of S1P and its receptors in bcr/abl positive chronic myeloid leukemia （CML） cells. Methods： The expressions of SIP receptors： S1P1, S1P2 and S1P3 in CML cells were detected by RT-PCR. SPK1 expression, activity and extracellular S1P were determined in ECV304 and HL-60 cells which were transfected with bcr/abl gene. To elucidate the relationship between the BCR/ABL, ERK/MAPK （extracellular signal-regulated kinase/mitogen-activated protein kinase）, SPK/S 1P and S 1P/S 1 P2 signal pathways, bcr/abl positive CML cell line K562 was treated with STI571, PD98059, N,N-dimethyl sphingosine （DMS） and JTE-013. Results： Retrovirus-mediated overexpression of bcr/abl gene in ECV304 and HL-60 cells resulted in upregulation of the expression, activity of SPK1 and increase of the secretion of SIP, whereas treatment of STI571 and PD98059 decreased the BCR/ABL-induced S1P secretion. Treatment of DMS reduced S1P secretion and P42/44MAPK phosphorylation. S1P2-selective antagonist JTE-013 could also decrease P42/44MAPK phosphorylation. Conclusion： These results suggest that BCR/ABL up-regulates extracellular sphingosine 1-phosphate through sphingosine kinase 1 and there is cross-talk between SPK1/S1P/S1P2 and P42/44MAPK in bcr/abl positive CML cells.

Short-term memory of danger signals or environmental stimuli in mesenchymal stem cells： implications for therapeutic potential

Mesenchymal stem/stromal cells （MSCs） possess some characteristics of immune cells, including a pro-inflammatory phenotype, an immunosuppressive phenotype, antibacterial properties and the expression of Toll-like receptor proteins. Here we show that, similar to immune cells, MSCs retain information from danger signals or environmental stimuli for a period of time. When treated with the pro-inflammatory factors lipopolysaccharide （LPS） or tumor necrosis factor-a （TNF-a）, MSCs display increased expression of IL-6, IL-8 and MCP-1. Following re-plating and several rounds of cell division in the absence of stimulating factors, the expression of IL-6, IL-8 and MCP-1 remained higher than in untreated cells for over 7 days. A spike in cytokine secretion occurred when cells were exposed to a second round of stimulation. We primed MSCs with LPS and LPS-primed MSCs had better therapeutic efficacy at promoting skin flap survival in a diabetic rat model than did unprimed MSCs. Finally, we found that several microRNAs, including miR146a, miR150 and miR155, along with the modification of DNA by 5-hydroxymethylcytosine （5hmC）, mediate the MSC response to LPS and TNF-e stimulation. Collectively, our data suggest that MSCs have a short-term memory of environmental signals, which may impact their therapeutic potential.

Intervention of Peiyuan Huayu Decoction on the neuron damage in model rats with acute subdural hematoma

Objective:To study the intervention effect of Peiyuan Huayu Decoction on the neuron damage in model rats with acute subdural hematoma (ASDH).Methods: 160 SD rats were randomly divided into four groups, and the ASDH model rats were made by stereotactic autoblood injection, and sham operation group received craniotomy without blood injection. Sham operation group and model group were normally bred after model establishment, and 6 h after model establishment, the treatment group received intragastric administration of Peiyuan Huayu Decoction, and control group received intragastric administration of Piracetam Tablets, 1 time a day. On the 1d, 3d, 5d and 7d after model establishment, the general conditions of rats (activity, food intake and mental state) were observed, blood was collected via auricula dextra, ELISA method was used to determine peripheral plasma NSE and S100β protein contents, routine HE staining was conducted after perfusion fixation, the neurons in blood injection side of brain tissue were counted, and the neuron damage was observed.Results: 26 rats were dead in the experiment. The general conditions of sham operation group were significantly better than those of other groups, treatment group was significantly better than model group and control group on the 5d group (P0.05);neuron count of sham operation group was basically stable, treatment group was not different from model group and control group on the 1d (P>0.05), treatment group was better than model group (P0.05) on the 3d, and treatment group was better than model group and control group on the 5d and 7d (P0.05), S100β protein and NSE contents decreased significantly on the 3d, and treatment group was significantly different fro

Coupled conditional nonlinear optimal perturbations and their application to ENSO ensemble forecasts

Limitations are existed in current ensemble forecasting initial perturbation methods for describing the interactions among various spheres of the Earth system. In this study, a new method is proposed, namely, the coupled conditional nonlinear optimal perturbation(C-CNOP) method, which incorporates multisphere interactions much appropriately. The El Nino-Southern Oscillation(ENSO) is a typical ocean-atmosphere “coupling”(or “interaction”) phenomenon. The C-CNOP method is applied to ensemble forecasting of ENSO. It is demonstrated that the C-CNOP method can generate coupled initial perturbations(CPs) that appropriately consider initial ocean-atmosphere coupling uncertainty for ENSO ensemble forecasts. Results reveal that the CPs effectively improve the ability of ENSO ensemble-mean forecasts in both temporal variability of Nio3.4 sea surface temperature anomalies(SSTAs) and spatial variability of ENSO mature-phase SSTAs. Notably, despite the weakest ocean-atmosphere coupling strength in the tropical Pacific occurring during the boreal spring and summer, CPs still capture the uncertainties of this weak coupling when ENSO predictions are initialized at these seasons. This performance of CPs significantly suppresses the rapid increase of ENSO prediction errors due to the high ocean-atmosphere coupling instability during these seasons, and thus effectively extends the lead time of skillful ENSO forecasting. Hence, the C-CNOP method is a suitable initial perturbation approach for ENSO ensemble forecast that can describe initial ocean-atmosphere coupling uncertainty. It is expected that the CCNOP method plays a significant role in predictions of other high-impact climate phenomena, and even future Earth system predictions.

Neuroimmunology is an interdisciplinary branch of biomedical science that emerges from the intersection of studies on the nervous system and the immune system.The complex interplay between the two systems has long been recognized.Research efforts directed at the underlying functional interface and associated pathophysiology,however,have garnered attention only in recent decades.In this narrative review,we highlight significant advances in research on neuroimmune interplay and modulation.A particular focus is on early-and middle-career neuroimmunologists in China and their achievements in frontier areas of"neuroimmune interface","neuro-endocrine-immune network and modulation","neuroimmune interactions in diseases","meningeal lymphatic and glymphatic systems in health and disease",and"tools and methodologies in neuroimmunology research".Key scientific questions and future directions for potential breakthroughs in neuroimmunology research are proposed.

A new method for identifying flow pattern of spouted fluidized bed by coupling Hilbert-Huang transform characteristics of differential pressure signals in different zones

On a cold spouted fluidized bed,this study compares the characteristic differences in intrinsic mode function(IMF)energy and Hilbert–Huang spectrum between the spout zone and annulus zone under different combinations of spouted gas and fluidized gas flow rates for five typical flow patterns.The energy distribution characteristics under different flow patterns are also analyzed.The Hilbert–Huang spectrum and IMF energy of pressure difference signals exhibit distinct variations in different zones as the flow pattern changes.Moreover,there exists a correlation between the energy in the middle-frequency range and the flow pattern.Leveraging the K-means algorithm,the middle-frequency range energy of IMFs in the spout zone and annulus zone is subjected to clustering analysis,leading to the identification of partition boundaries for each flow pattern.Based on this,a flow pattern identification method of spouted fluidized bed coupled with middle-frequency range energy in spout zone and annulus zone is proposed,which has very high identification accuracy.

BACKGROUND Bradycardia-induced cardiomyopathy(BIC),which is a disease resulting from bradycardia,is characterized by cardiac chamber enlargement and diminished cardiac function.The correction of bradycardia can allow for significant improvements in both cardiac function and structure;however,this disease has been infrequently documented.In this case,we conducted a longitudinal followup of a patient who had been enduring BIC for more than 40 years to heighten awareness and prompt timely diagnosis and rational intervention.CASE SUMMARY A woman who presented with postactivity fatigue and dyspnea was diagnosed with bradycardia at the age of 7.Since she had no obvious symptoms,she did not receive any treatment to improve her bradycardia during the 42-year follow-up,except for the implantation of a temporary pacemaker during labor induction surgery.As time progressed,the patient's heart gradually expanded due to her low ventricular rate,and she was diagnosed with BIC.In 2014,the patient developed atrial fibrillation,her ventricular rate gradually increased,and her heart shape gradually returned to normal.This report describes the cardiac morphological changes caused by the heart rate changes in BIC patients older than 40 years,introduces another possible outcome of BIC,and emphasizes the importance of early intervention in treating BIC.CONCLUSION BIC can induce atrial fibrillation,causing an increased ventricular rate and leading to positive cardiac remodeling.

Neurophysiological, histological, and behavioral characterization of animal models of distraction spinal cord injury: a systematic review

Distraction spinal cord injury is caused by some degree of distraction or longitudinal tension on the spinal cord and commonly occurs in patients who undergo corrective operation for severe spinal deformity.With the increased degree and duration of distraction,spinal cord injuries become more serious in terms of their neurophysiology,histology,and behavior.Very few studies have been published on the specific characteristics of distraction spinal cord injury.In this study,we systematically review 22 related studies involving animal models of distraction spinal cord injury,focusing particularly on the neurophysiological,histological,and behavioral characteristics of this disease.In addition,we summarize the mechanisms underlying primary and secondary injuries caused by distraction spinal cord injury and clarify the effects of different degrees and durations of distraction on the primary injuries associated with spinal cord injury.We provide new concepts for the establishment of a model of distraction spinal cord injury and related basic research,and provide reference guidelines for the clinical diagnosis and treatment of this disease.

A Novel Multi-Stream Fusion Network for Underwater Image Enhancement

Due to the selective absorption of light and the existence of a large number of floating media in sea water, underwater images often suffer from color casts and detail blurs. It is therefore necessary to perform color correction and detail restoration. However,the existing enhancement algorithms cannot achieve the desired results. In order to solve the above problems, this paper proposes a multi-stream feature fusion network. First, an underwater image is preprocessed to obtain potential information from the illumination stream, color stream and structure stream by histogram equalization with contrast limitation, gamma correction and white balance, respectively. Next, these three streams and the original raw stream are sent to the residual blocks to extract the features. The features will be subsequently fused. It can enhance feature representation in underwater images. In the meantime, a composite loss function including three terms is used to ensure the quality of the enhanced image from the three aspects of color balance, structure preservation and image smoothness. Therefore, the enhanced image is more in line with human visual perception.Finally, the effectiveness of the proposed method is verified by comparison experiments with many stateof-the-art underwater image enhancement algorithms. Experimental results show that the proposed method provides superior results over them in terms of MSE,PSNR, SSIM, UIQM and UCIQE, and the enhanced images are more similar to their ground truth images.

Parkinson’s disease(PD)is a complicated neurodegenerative disease,characterized by the accumulation ofα-synuclein(α-syn)in Lewy bodies and neurites,and massive loss of midbrain dopamine neurons.Increasing evidence suggests that gut microbiota and microbial metabolites are involved in the development of PD.Among these,short-chain fatty acids(SCFAs),the most abundant microbial metabolites,have been proven to play a key role in brain-gut communication.In this review,we analyze the role of SCFAs in the pathology of PD from multiple dimensions and summarize the alterations of SCFAs in PD patients as well as their correlation with motor and non-motor symptoms.Future research should focus on further elucidating the role of SCFAs in neuroinflammation,as well as developing novel strategies employing SCFAs and their derivatives to treat PD.
