Autophagy has been shown to play an important role in Parkinson’s disease.We hypothesized that skin-derived precursor cells exhibit neuroprotective effects in Parkinson’s disease through affecting autophagy.In this study,6-hydroxydopamine-damaged SH-SY5Y cells were pretreated with a culture medium containing skin-derived precursors differentiated into Schwann cells(SKP-SCs).The results showed that the SKP-SC culture medium remarkably enhanced the activity of SH-SY5Y cells damaged by 6-hydroxydopamine,reduced excessive autophagy,increased tyrosine hydroxylase expression,reducedα-synuclein expression,reduced the autophagosome number,and activated the PI3K/AKT/mTOR pathway.Autophagy activator rapamycin inhibited the effects of SKP-SCs,and autophagy inhibitor 3-methyladenine had the opposite effect.These findings confirm that SKP-SCs modulate the PI3K/AKT/mTOR pathway to inhibit autophagy,thereby exhibiting a neuroprotective effect in a cellular model of Parkinson’s disease.This study was approved by the Animal Ethics Committee of Laboratory Animal Center of Nantong University(approval No.S20181009-205)on October 9,2018.

Carbon-ion radiotherapy(CIRT)offers unique physical and biological advantages over photon radiotherapy.However,some materials and devices in the CIRT treatment room become radioactive under bombardment by therapeutic carbon-ion beams due to nuclear reactions,thereby leading to possible radiation hazards to medical staff and additional and unwanted doses to patients.This study assessed the level of induced radioactivity in the treatment room of the Heavy-Ion Medical Machine(HIMM)in Wuwei.Monte Carlo simulations using PHITS were performed for a conservative case under the conditions of maximum beam energy and intensity provided by the HIMM facility.The geometry and configuration of Treatment Room 2 of the HIMM facility in Wuwei were adopted.We evaluated the activation of air,the phantom,and the components of the beamline,such as the primary collimator(PC),ridge filter(RF),and multileaf collimator(MLC).For air activation,we calculated the medical staff immersion external exposure and inhalation internal exposure caused by the corresponding radionuclides.For phantom activation,we estimated the additional dose to the patient’s family members owing to secondary photons after treatment.In addition,the exemption or non-exemption of the component material activation was assessed.The results showed that external radiation caused by air activation was the main source of the annual effective dose at approximately 0.5 mSv/y.The induced radioactivity exposure to family members of a patient after CIRT was approximately 40μSv,sufficiently lower than the public dose limit of 1 mSv/a.The induced radioactivity of the PC,RF,and MLC was all above the exempt levels after the devices were retired,whereas the induced radioactivity of the RS and compensator could reach the exempt levels after one patient session.Our study indicated that medical staff engaged in CIRT should stay away from the high-dose-rate area of induced radioactivity along the beam direction,shorten the residence time in the treatment room as much as possible,and store

BACKGROUND Melioidosis,an infectious disease caused by Burkholderia pseudomallei(B.pseudomallei),occurs endemically in Southeast Asia and Northern Australia and is a serious opportunistic infection associated with a high mortality rate.CASE SUMMARY A 58-year-old woman presented with scattered erythema on the skin of her limbs,followed by fever and seizures.B.pseudomallei was isolated successively from the patient’s urine,blood,and pus.Magnetic resonance imaging showed abscess formation involving the right forehead and the right frontal region.Subsequently,abscess resection and drainage were performed.The patient showed no signs of relapse after 4 months of follow-up visits post-treatment.CONCLUSION We present here a unique case of multi-systemic melioidosis that occurs in nonendemic regions in a patient who had no recent travel history.Hence,it is critical to enhance awareness of melioidosis in non-endemic regions.

The use of carbon-ion radiotherapy(CIRT)is gradually increasing.Owing to the generation of high-energy secondary neutrons during CIRT,its use presents new challenges in radiation protection.Thus,secondary neutron dose distributions must be explored and evaluated under clinical scenarios based on different treatment configurations.However,neutron dose and energy spectrum measurements are often difficult.This can be primarily attributed to the inherent limitations of most neutron detectors,such as their unsuitability for spectral measurements and inaccurate responses to neutrons with energies above 20 MeV.Numerical calculation methods based on probabilistic statistical theory are fast and convenient for neutron dose evaluation.In this study,external secondary neutron doses at the heavy ion medical machine in Wuwei,which is equipped with a passive beam delivery system,were calculated using the Monte Carlo method.The dependence of neutron doses on various treatment parameters(incident carbon-ion beam energy,spatial location,field size,and spread-out Bragg peak(SOBP)width)was investigated.Furthermore,the feasibility of applying an analytical model to predict the ambient dose equivalent was verified.For the combination involving an energy of 400 MeV=u and SOBP width of 6 cm,the ambient dose equivalent per therapeutic dose(H=D)at the isocenter was 79.87 mSv=Gy:The H=D value decreased rapidly with increasing spatial distance and slightly with increasing aperture size and SOBP width.The H=D values derived from the Monte Carlo simulations were in good agreement with the results reported in the literature.The analytical model could be used to quickly predict the H=D value along the incidence direction of the beam with an error of less than 20%.Thus,our study contributes to the understanding of the relationship between neutron radiation and treatment configuration parameters,which establishes a basis for predicting non-therapeutic radiation doses in CIRT.

The estimation of model parameters is an important subject in engineering.In this area of work,the prevailing approach is to estimate or calculate these as deterministic parameters.In this study,we consider the model parameters from the perspective of random variables and describe the general form of the parameter distribution inference problem.Under this framework,we propose an ensemble Bayesian method by introducing Bayesian inference and the Markov chain Monte Carlo(MCMC)method.Experiments on a finite cylindrical reactor and a 2D IAEA benchmark problem show that the proposed method converges quickly and can estimate parameters effectively,even for several correlated parameters simultaneously.Our experiments include cases of engineering software calls,demonstrating that the method can be applied to engineering,such as nuclear reactor engineering.

Unsteady state precipitation of M_(23)C_(6)carbides during thermal cycling in reduced activation steel manufactured by laser melting deposition

The temperature field distribution and thermal history of Fe-9Cr2WVTa reduced activation steel prepared by laser melting deposition(LMD)have been calculated with Gaussian and Ring laser beams,and the nucleation and growth behaviors of M_(23)C_(6)precipitates in the 1st,7th and 19th layers have been calculated using the modified classical nucleation theory and Svoboda Fischer Fratzl Kozeschnik model.The energy distribution shows W-shape with Ring laser beam while it shows V-shape with Gaussian laser beam,which results in the more uniform M_(23)C_(6)size in the same layer with Ring laser beam.Precipitates in the bottom(i.e.,the 1st layer)have the minimum size and the size increases with the layer number with Gaussian and Ring laser beams.The temperature history,the instantaneous nucleation rate and the size evolution of M_(23)C_(6)have been systematically discussed.The results indicate that the nucleation,growth and re-dissolution of precipitates in reduced activation steel depend on the amount of energy absorbed in the thermal cycle during LMD.The continuous accumulation of energy during the thermal cycle leads to larger M_(23)C_(6)at the top area.The unsteady state precipitation dynamics of M_(23)C_(6)carbides during thermal cycling are consistent with the simulation results.

BACKGROUND As one of the fatal diseases with high incidence,lung cancer has seriously endangered public health and safety.Elderly patients usually have poor self-care and are more likely to show a series of psychological problems.AIM To investigate the effectiveness of the initial check,information exchange,final accuracy check,reaction(IIFAR)information care model on the mental health status of elderly patients with lung cancer.METHODS This study is a single-centre study.We randomly recruited 60 elderly patients with lung cancer who attended our hospital from January 2021 to January 2022.These elderly patients with lung cancer were randomly divided into two groups,with the control group taking the conventional propaganda and education and the observation group taking the IIFAR information care model based on the conventional care protocol.The differences in psychological distress,anxiety and depression,life quality,fatigue,and the locus of control in psychology were compared between these two groups,and the causes of psychological distress were analyzed.RESULTS After the intervention,Distress Thermometer,Hospital Anxiety and Depression Scale(HADS)for anxiety and the HADS for depression,Revised Piper’s Fatigue Scale,and Chance Health Locus of Control scores were lower in the observation group compared to the pre-intervention period in the same group and were significantly lower in the observation group compared to those of the control group(P<0.05).After the intervention,Quality of Life Questionnaire Core 30(QLQ-C30),Internal Health Locus of Control,and Powerful Others Health Locus of Control scores were significantly higher in the observation and the control groups compared to the pre-intervention period in their same group,and QLQ-C30 scores were significantly higher in the observation group compared to those of the control group(P<0.05).CONCLUSION The IIFAR information care model can help elderly patients with lung cancer by reducing their anxiety and depression,psychological distress,and fatigue,improving the

Theγ-ray emitting compact symmetric objects(CSOs)PKS 1718-649,NGC 3894,and TXS 0128+554 are lobedominated in the radio emission.In order to investigate theirγ-ray radiation properties,we analyze the~14yr Fermi/LAT observation data of the three CSOs.They all show the low luminosity(1041-1043 erg s-1)and no significant variability in theγ-ray band.Theirγ-ray average spectra can be well fitted by a power-law function.These properties ofγ-rays are clearly different from theγ-ray emitting CSOs CTD 135 and PKS 1413+135,for which theγ-rays are produced by a restarted aligned jet.In the L_(γ)-Γ_(γ)plane,the three CSOs are also located at the region occupied by radio galaxies(RGs)while CTD 135 and PKS 1413+135 display a similar feature to blazars.Together with a similar radio emission property toγ-ray emitting RGs Cen A and Fornax A,we speculate that theγ-rays of the three CSOs stem from their extended mini-lobes.The broadband spectral energy distributions of the three CSOs can be well explained by the two-zone leptonic model,where theirγ-rays are produced by the inverse Compton process of the relativistic electrons in extended regions.By extrapolating the observed Fermi/LAT spectra to the very high energy band,we find that TXS 0128+554 among the three CSOs may be detected by the Cherenkov Telescope Array in the future.
