目的探讨丙酮酸激酶M2异构体(PKM2)在缺氧条件下心肌细胞中线粒体生物发生、线粒体动力学及能量代谢中的作用。方法给予大鼠心肌细胞系H9C2不同浓度梯度的氯化钴(CoCl_(2))刺激,模拟体内的缺氧环境,通过反转录聚合酶链反应和western blot方法检测PKM2和线粒体生物发生及能量代谢相关分子的表达量;分离H9C2心肌细胞中的胞浆和胞核蛋白,western blot检测PKM2在H9C2心肌细胞中的定位,同时免疫荧光染色检测PKM2在H9C2心肌细胞中的定位;使用三磷酸腺苷(ATP)检测试剂盒检测PKM2对缺氧条件下H9C2心肌细胞中ATP生成的影响;使用PKM2质粒和PKM2 siRNA转染H9C2心肌细胞,并给予CoCl_(2)(300μmol/L)刺激24 h,western blot检测PKM2对于H9C2心肌细胞中线粒体生物发生和线粒体融合的影响。结果PKM2在缺氧的H9C2心肌细胞中表达逐渐减少,与线粒体生物发生和功能相关分子的表达量密切相关。PKM2主要表达于心肌细胞的胞浆中,与线粒体标记分子热休克蛋白60(HSP60)共定位。PKM2过表达能促进缺氧心肌细胞中线粒体的生物发生、线粒体融合和ATP生成;反之,PKM2敲低能抑制缺氧心肌细胞中线粒体的生物发生、线粒体融合和ATP生成。结论PKM2对于缺氧条件下心肌细胞中的线粒体生物发生和线粒体动力学至关重要,从而控制心肌细胞的能量代谢。

目的构建巨核细胞/血小板特异性E1A激活基因阻遏子基因(Creg1)敲除小鼠,并诱导小鼠胚胎胎肝巨核细胞的分化,为研究Creg1在巨核细胞分化和血小板生理功能中的作用提供实验工具及方法。方法将雌性Creg1^(flox/flox)小鼠与雄性血小板因子4(Pf4)-Cre^(+)小鼠繁育,获得Creg1flox/wtPf4-Cre^(+)小鼠,进一步交配获得巨核细胞/血小板特异性Creg1敲除小鼠(Creg1^(flox/flox)Pf4-Cre^(+),简写为Creg1^(-/-))。采用聚合酶链式反应及琼脂糖凝胶电泳进行基因型鉴定。取Creg1^(flox/flox)和Creg1^(-/-)孕龄约15 d小鼠的胎肝,体外培养胎肝细胞4 d,加入血小板生成素,光镜下观察两组巨核细胞形态和大小的区别。结果本研究成功构建了Creg1^(-/-)小鼠。与Creg1^(flox/flox)小鼠比较,Creg1^(-/-)小鼠巨核细胞中Creg1 mRNA的表达量显著下降,差异有统计学意义(P<0.05)。与Creg1^(flox/flox)小鼠比较,Creg1^(-/-)小鼠胎肝巨核细胞诱导分化4 d时,光镜下观察发现,巨核细胞的直径明显变小,差异有统计学意义(P<0.05)。结论Creg1^(-/-)小鼠作为研究巨核细胞和血小板相关疾病发生机制的实验工具鼠,可能揭示尚未发现和阐明的重要病理生理学机制,为临床治疗血小板疾病提供理论基础支持。

目的探究胃癌及肝癌中差异表达的基因,并分析其生物学功能及其在临床应用中的潜在价值。方法在基因表达综合数据库中选取胃癌相关数据集GSE79943及肝癌相关数据集GSE84402进行分析。通过对两个数据集的基因表达数据进行差异分析确定共同差异表达的基因,进一步利用GO富集和KEGG途径富集分析揭示差异基因的主要生物学功能和通路参与情况。结果筛选出117个在胃癌和肝癌中共同差异表达的基因。采用GO富集分析软件进行117个基因的分析发现,这些基因在多种金属离子和无机物的发生及转运,如铜、锌、钙离子等,以及细胞对无机物的代谢过程中起着至关重要的作用。采用KEGG富集分析软件对117个差异基因的基因功能进行分析发现,这些基因中更多发挥着无机物吸收和代谢过程的功能。通过对GO和KEGG富集分析的结果进行分析,获得了8个与无机物代谢联系紧密的主要基因,分别为MT1G、MT2A、MT1E、MT1X、MT1F、MT1H、MT1M和MT1HL1,且在两种疾病中均有着显著的低表达。结论通过对胃癌和肝癌基因表达数据的差异分析,揭示了在这两种癌症中共同差异表达的基因群,并发现其在无机物代谢过程中的重要作用。这些差异基因可能参与调控肿瘤细胞对金属离子的吸收和代谢,从而影响肿瘤的发展,可以作为潜在的治疗靶点及胃癌和肝癌的预后评估指标。

360°videos enable viewers to watch freely from different directions but inevitably prevent them from perceiving all the helpful *** mitigate this problem,picture-in-picture(PIP)guidance was proposed using preview windows to show regions of interest(ROIs)outside the current view *** identify several drawbacks of this representation and propose a new method for 360° film watching called *** enhances traditional PIP by adaptively arranging preview windows with changeable view ranges and *** addition,AdaPIP incorporates the advantage of arrow-based guidance by presenting circular windows with arrows attached to them to help users locate the corresponding ROIs more *** also adapted AdaPIP and Outside-In to HMD-based immersive virtual reality environments to demonstrate the usability of PIP-guided approaches beyond 2D *** user experiments on 2D screens,as well as in VR environments,indicate that AdaPIP is superior to alternative methods in terms of visual experiences while maintaining a comparable degree of immersion.

The fatigue damage caused by flow-induced vibration(FIV)is one of the major concerns for multiple cylindrical structures in many engineering *** FIV suppression is of great importance for the security of many cylindrical *** active and passive control methods have been employed for the vibration suppression of an isolated cylinder undergoing vortex-induced vibrations(VIV).The FIV suppression methods are mainly extended to the multiple cylinders from the vibration control of the isolated *** to the mutual interference between the multiple cylinders,the FIV mechanism is more complex than the VIV mechanism,which makes a great challenge for the FIV *** efforts have been devoted to vibration suppression of multiple cylinder systems undergoing FIV over the past two *** control methods,such as helical strakes,splitter plates,control rods and flexible sheets,are not always effective,depending on many influence factors,such as the spacing ratio,the arrangement geometrical shape,the flow velocity and the parameters of the vibration control *** FIV response,hydrodynamic features and wake patterns of the multiple cylinders equipped with vibration control devices are reviewed and *** FIV suppression efficiency of the vibration control methods are analyzed and compared considering different influence *** research on the FIV suppression of multiple cylinders is suggested to provide insight for the development of FIV control methods and promote engineering applications of FIV control methods.

As the second of Earth's Trojan asteroids, 2020 XL_(5) is worthy of rendezvous and even sample return missions in many aspects. In this paper, a rendezvous mission to Earth's second Trojan asteroid 2020 XL_(5) is ***, due to its high inclination and large eccentricity, direct impulsive transfer requires large amounts of fuel consumption. To address this challenge, we explore the benefits of electric propulsion and multi-gravity assist techniques for interplanetary missions. These two techniques are integrated in this mission design. The design of a low-thrust gravity-assist(LTGA) trajectory in multi-body dynamics is thoroughly investigated,which is a complex process. A comprehensive framework including three steps is presented here for optimization of LTGA trajectories in multi-body dynamics. The rendezvous mission to 2020 XL_(5) is designed with this three-step approach. The most effective transfer sequence among the outcomes involves Earth–Venus–Earth–Venus-2020 XL_(5). Numerical results indicate that the combination of electric propulsion and multi-gravity assists can greatly reduce the fuel consumption, with fuel consumption of 9.03%, making it a highly favorable choice for this rendezvous mission.
