BACKGROUND Colorectal cancer(CRC)is one of the most common malignancies worldwide.Given its insidious onset,the condition often already progresses to advanced stage when symptoms occur.Thus,early diagnosis is of great significance for timely clinical intervention,efficacy enhancement,and prognostic improvement.Featuring high throughput,fastness,and rich information,next generation sequencing(NGS)can greatly shorten the detection time,which is a widely used detection technique at present.AIM To screen specific genes or gene combinations in fecal DNA that are suitable for diagnosis and prognostic prediction of CRC,and to establish a technological platform for CRC screening,diagnosis,and efficacy monitoring through fecal DNA detection.METHODS NGS was used to sequence the stool DNA of patients with CRC,which were then compared with the genetic testing results of the stool samples of normal controls and patients with benign intestinal disease,as well as the tumor tissues of CRC patients.Specific genes or gene combinations in fecal DNA suitable for diagnosis and prognostic prediction of CRC were screened,and their significances in diagnosing CRC and predicting patients'prognosis were comprehensively evaluated.RESULTS High mutation frequencies of TP53,APC,and KRAS were detected in the stools and tumor tissues of CRC patients prior to surgery.Contrastively,no pathogenic mutations of the above three genes were noted in the postoperative stools,the normal controls,or the benign intestinal disease group.This indicates that tumor-specific DNA was detectable in the preoperative stools of CRC patients.The preoperative fecal expression of tumor-associated genes can reflect the gene mutations in tumor tissues to some extent.Compared to the postoperative stools and the stools in the two control groups,the pathogenic mutation frequencies of TP53 and KRAS were significantly higher for the preoperative stools(χ^(2)=7.328,P<0.05;χ^(2)=4.219,P<0.05),suggesting that fecal TP53 and KRAS genes can be used for CRC screening,diagnosis,and

COVID-19 vaccines from multiple manufacturers are needed to cope with the problem of insufficient supply.We did two singlecenter,randomised,double-blind,placebo-controlled phase 1 and phase 2 trials to assess the safety,tolerability and immunogenicity of a recombinant COVID-19 vaccine(Sf9 cells)in healthy population aged 18 years or older in China.Eligible participants were enrolled,the ratio of candidate vaccine and placebo within each dose group was 3:1(phase 1)or 5:1(phase 2).From August 28,2020,168 participants were sequentially enrolled and randomly assigned to receive the low dose vaccine,high dose vaccine or placebo with the schedule of 0,28 days or 0,14,28 days in phase 1 trial.From November 18,2020,960 participants were randomly assigned to receive the low dose vaccine,high dose vaccine or placebo with the schedule of 0,21 days or 0;14,28 days in phase 2 trial.The most common solicited injection site adverse reaction within 7 days in both trials was pain.The most common solicited systematic adverse reactions within 7 days were fatigue,cough,sore throat,fever and headache.ELISA antibodies and neutralising antibodies increased at 14 days,and peaked at 28 days(phase 1)or 30 days(phase 2)after the last dose vaccination.The GMTs of neutralising antibody against live SARS-CoV-2 at 28 days or 30 days after the last dose vaccination were highest in the adult high dose group(0,14,28 days),with 102.9(95%Cl 61.9-171.2)and 102.6(95%Cl 75.2-140.1)in phase 1 and phase 2 trials,respectively.Specific T-cell response peaked at 14 days after the last dose vaccination in phase 1 trial.This vaccine is safe,and induced significant immune responses after three doses of vaccination.

What is already known about this topic?Addressing health disparities is a worldwide priority,with a well-established acknowledgment of the influence of childhood circumstances on these discrepancies.In China,particularly among the elderly,health inequalities are a notable concern.What is added by this report?The inequality in healthy aging has increased from 2011 to 2020,both in general and concerning childhood factors.Nevertheless,the impact of early-life healthcare access and parental health behaviors on healthy aging gaps has reduced among older adults in better health within the top segment of healthy aging.What are the implications for public health practice?Efforts towards reducing regional health disparities and improving healthcare access for children,along with promoting the health and well-being of parents,especially in economically disadvantaged households,are crucial policy considerations.

A hybrid machine learning optimization algorithm for multivariable pore pressure prediction

Pore pressure is essential data in drilling design,and its accurate prediction is necessary to ensure drilling safety and improve drilling efficiency.Traditional methods for predicting pore pressure are limited when forming particular structures and lithology.In this paper,a machine learning algorithm and effective stress theorem are used to establish the transformation model between rock physical parameters and pore pressure.This study collects data from three wells.Well 1 had 881 data sets for model training,and Wells 2 and 3 had 538 and 464 data sets for model testing.In this paper,support vector machine(SVM),random forest(RF),extreme gradient boosting(XGB),and multilayer perceptron(MLP)are selected as the machine learning algorithms for pore pressure modeling.In addition,this paper uses the grey wolf optimization(GWO)algorithm,particle swarm optimization(PSO)algorithm,sparrow search algorithm(SSA),and bat algorithm(BA)to establish a hybrid machine learning optimization algorithm,and proposes an improved grey wolf optimization(IGWO)algorithm.The IGWO-MLP model obtained the minimum root mean square error(RMSE)by using the 5-fold cross-validation method for the training data.For the pore pressure data in Well 2 and Well 3,the coefficients of determination(R^(2))of SVM,RF,XGB,and MLP are 0.9930 and 0.9446,0.9943 and 0.9472,0.9945 and 0.9488,0.9949 and 0.9574.MLP achieves optimal performance on both training and test data,and the MLP model shows a high degree of generalization.It indicates that the IGWO-MLP is an excellent predictor of pore pressure and can be used to predict pore pressure.

A rare missense PAX6 mutation causes atypical aniridia in a three-generation Chinese family

●AIM:To investigate the molecular diagnosis of a threegeneration Chinese family affected with aniridia,and further to identify clinically a PAX6 missense mutation in members with atypical aniridia.●METHODS:Eleven family members with and without atypical aniridia were recruited.All family members underwent comprehensive ophthalmic examinations.A combination of whole exome sequencing(WES)and direct Sanger sequencing were performed to uncover the causative mutation.●RESULTS:Among the 11 family members,8 were clinically diagnosed with congenital aniridia(atypical aniridia phenotype).A rare heterozygous mutation c.622C>T(p.Arg208Trp)in exon 8 of PAX6 was identified in all affected family members but not in the unaffected members or in healthy control subjects.●CONCLUSION:A rare missense mutation in the PAX6 gene is found in members of a three-generation Chinese family with congenital atypical aniridia.This result contributes to an increase in the phenotypic spectrum caused by PAX6 missense heterozygous variants and provides useful information for the clinical diagnosis of atypical aniridia,which may also contribute to genetic counselling and family planning.

Transparent micropatterned conductive films based on highlyordered nanowire network

Transparent conductive films that are based on nanowire networks are essential to construct flexible,wearable,and even stretchable electronics.However,large-scale precise micropatterning,especially with regard to the controllability of the organizing orientation of nanowires,is a critical challenge.Herein,we proposed a liquid film rupture self-assembly approach for manufacturing transparent conductive films with microstructure arrays based on a highly ordered nanowire network.The large-scale microstructure conductive films were fabricated through air-liquid interface self-assembly and liquid film rupture self-assembly.Six typical micropattern morphologies,including square,hexagon,circle,serpentine,etc.,were prepared to reveal the universal applicability of the proposed approach.The homogeneity and controllability of this approach were verified for multiple assemblies.With the assembly cycles increasing,the optical transmittance decreases slightly.In addition,theoretical model analysis is carried out,and the analytical formula of the speed of the film moving with the surface tension and the density of the liquid film is presented.Finally,the feasibility of this approach for piezoresistive strain sensors is verified.This fabrication approach demonstrated a cost-effective and efficient method for precisely arranging nanowires,which is useful in transparent and wearable applications.

Layer Hall effect induced by hidden Berry curvature in antiferromagnetic insulators

The layer Hall effect describes electrons spontaneously deflected to opposite sides at different layers, which has been experimentally reported in the MnBi2Te4thin films under perpendicular electric fields. Here, we reveal a universal origin of the layer Hall effect in terms of the so-called hidden Berry curvature, as well as material design principles. Hence, it gives rise to zero Berry curvature in momentum space but non-zero layer-locked hidden Berry curvature in real space. We show that, compared to that of a trivial insulator, the layer Hall effect is significantly enhanced in antiferromagnetic topological insulators. Our universal picture provides a paradigm for revealing the hidden physics as a result of the interplay between the global and local symmetries, and can be generalized in various scenarios.

Advances in MET tyrosine kinase inhibitors in gastric cancer

Gastric cancer is among the most frequently occurring cancers and a leading cause of cancer-related deaths globally. Because gastric cancer is highly heterogenous and comprised of different subtypes with distinct molecular and clinical characteristics, the management of gastric cancer calls for better-defined, biomarker-guided, molecular-based treatment strategies. MET is a receptor tyrosine kinase mediating important physiologic processes, such as embryogenesis, tissue regeneration, and wound healing. However, mounting evidence suggests that aberrant MET pathway activation contributes to tumour proliferation and metastasis in multiple cancer types, including gastric cancer, and is associated with poor patient outcomes. As such, MET-targeting therapies are being actively developed and promising progress has been demonstrated, especially with MET tyrosine kinase inhibitors. This review aims to briefly introduce the role of MET alterations in gastric cancer and summarize in detail the current progress of MET tyrosine kinase inhibitors in this disease area with a focus on savolitinib, tepotinib, capmatinib, and crizotinib. Building on current knowledge, this review further discusses existing challenges in MET alterations testing, possible resistance mechanisms to MET inhibitors, and future directions of MET-targeting therapies.

Semi-derived Ringel-Hall algebras and Hall algebras of odd-periodic relative derived categories

Let t be a positive integer and A be a hereditary abelian category satisfying some finiteness conditions. We define the semi-derived Ringel-Hall algebra of A from the category CZ/t（A） of Z/t-graded complexes and obtain a natural basis of the semi-derived Ringel-Hall algebra. Moreover, we describe the semiderived Ringel-Hall algebra by the generators and defining relations. In particular, if t is an odd integer, we show an embedding of the derived Hall algebra of the odd-periodic relative derived category in the extended semi-derived Ringel-Hall algebra.

Distribution of per-and polyfluoroalkyl substances in blood,serum,and urine of patients with liver cancer and associations with liver function biomarkers

Studies have shown that per-and polyfluoroalkyl substances(PFASs)may be hepatotoxic in animals or humans.However,data on clinical epidemiology are very limited.In this study,21PFASs were determined in patients with liver diseases,with the highest median concentrations detected in the serum sample(26.7 ng/mL),followed by blood(10.7 ng/mL)and urine(5.02 ng/mL).Higher total PFAS concentrations were found in hepatocellular carcinoma(HCC)patients compared to non-HCC patients,with significant discrepancies in serum and blood samples.Besides,significant correlations were also found among PFAS concentrations and age,gender,body mass index(BMI),and liver function biomarkers levels.For example,PFAS concentrations are significantly higher in males than in females;Several serum PFASs concentrations increase with age and BMI,while the serum perfluorohexane sulfonic acid(PFHxS)concentrations are negatively correlated with age.In addition,multiple regression models adjusted for age,gender and BMI found that increased serum perfluorobutane sulfonic acid(PFBS),perfluoroheptane sulfonic acid(PFHpS)and perfluorohexylphosphonic acid(PFHxPA)conentrations are correlated with elevated alkaline phosphatase(ALP),aspartate aminotransferase(AST),and alpha-fetoprotein(AFP)(plinical hepatotoxicity of PFAS.
