NCAD v1.0: a database for non-coding variant annotation and interpretation

The application of whole genome sequencing is expanding in clinical diagnostics across various genetic disorders, and the significance of non-coding variants in penetrant diseases is increasingly being demonstrated. Therefore, it is urgent to improve the diagnostic yield by exploring the pathogenic mecha.isms of variants in non-coding regions. However, the interpretation of non-coding variants remains a significant cha.lenge, due to the complex functional regulatory mecha.isms of non-coding regions and the current limitations of available databases and tools. Hence, we develop the non-coding variant annotation database (NCAD, http://***/), encompassing comprehensive insights into 665,679,194 variants, regulatory elements, and element interaction details. Integrating data from 96 sources, spanning both GRCh37 and GRCh38 versions, NCAD v1.0 provides vital information to support the genetic diagnosis of non-coding variants, including allele frequencies of 12 diverse populations, with a particular focus on the population frequency information for 230,235,698 variants in 20,964 Chinese individuals. Moreover, it offers prediction scores for variant functionality, five categories of regulatory elements, and four types of non-coding RNAs. With its rich data and comprehensive coverage, NCAD serves as a valuable platform, empowering researchers and clinicians with profound insights into non-coding regulatory mecha.isms while facilitating the interpretation of non-coding variants.

BACKGROUND:The molecular mecha.ism of sepsis-associated acute kidney injury(SA-AKI)is *** analyzed co-differentially expressed genes(co-DEGs)to elucidate the underlying mecha.ism and intervention targets of ***:The microarray datasets GSE65682,GSE30718,and GSE174220 were downloaded from the Gene Expression Omnibus(GEO)*** identified the co-DEGs and constructed a gene co-expression network to screen the hub *** analyzed immune correlations and disease correlations and performed functional annotation of the hub *** also performed single-cell and microenvironment analyses and investigated the enrichment pathways and the main transcription ***,we conducted a correlation analysis to evaluate the role of the hub ***:Interleukin 32(IL32)was identified as the hub gene in SA-AKI,and the main enriched signaling pathways were associated with hemopoiesis,cellular response to cytokine stimulus,inflammatory response,and regulation of kidney ***,IL32 was significantly associated with mortality in SA-AKI ***,macropha.es,T cells,and NK cells were closely related to IL32 and were involved in the immune microenvironment in SA-AKI ***32 expression increased significantly in the kidney of septic ***-like receptor 2(TLR2)was significantly and negatively correlated with ***:IL32 is the key gene involved in SA-AKI and is significantly associated with ***2 and relevant immune cells are closely related to key genes.

Local air pollution is strongly affected by synoptic weather systems,such as fronts,troughs,low-altitude vortices,or high-altitude ***,few studies ha.e analyzed the meteorological properties of cold or warm air masses associated to these systems and their impact on local air *** this study,hourly observations of fine particulate matter(diameter of up to 2.5µm,i.e.,PM_(2.5)),wind(V),temperature(T),pressure(P),and precipitation(R),acquired in ha.gzhou in 2014-2020,were *** this analysis,weather patterns were categorized into 27 types;89 and 94 cases illustrating the passage of warm and cold air masses over ha.gzhou were identified,respectively;the influence of air mass temperature,wind speed,and wind direction on PM_(2.5) concentrations and local accumulation or removal was *** main results are as follows.(1)Pollution events occurred more frequently for cold tha. for warm air masses,but average pollutant concentration was lower for cold air masses;(2)48%of the cold air mass cases corresponded to PM_(2.5) decreases and 52%to PM_(2.5) increases,with strong cold air masses(ΔT_(24h)>4°C;∣V∣_(average)>4 m s^(−1))markedly reducing local pollution,but weak cold air masses(ΔT24h4°C)reduced the PM_(2.5) concentration whereas weaker winds(∣V∣_(average)ha.ply within 4 h after the passage of strong cold air masses,but more gradually within 14 h after the passage of strong warm air *** results considerably improve the current understanding of the influence of cold and warm air masses on local pollution patterns.

A method to promote aluminum hydroxide crystal growth through pickling Al(OH)_(3)as seed in the ammonia system was proposed to overcome these *** experimental results show tha.,under the conditions of pickling time of 15 min,the acid concentration of 10%,the addition of 70 g/L pickling-Al(OH)_(3)seed,and the coarse granular Al(OH)_(3)products(d0.5=85.667)can be *** cha.acterization results show tha. the pha.e of the product is gibbsite,consistent with the ***,the steps and ledges can be formed on pickling Al(OH)_(3)seed surface under the ammonia system,effectively promoting crystal *** crystal growth,the roughness of the crystal surface was first increased and then decreased,and the lamellar structure was deposited on the crystal seed *** final particles are approximately round,the surface is compact and *** growth of the product is surface reaction *** addition,the content of the AlO_(6)unit is increased and contributed to Al(OH)_(3)crystal growth.

High-power O-mode radio waves can excite artificial instabilities in the F region,according to experiments conducted at the European Incoherent Scatter Science Association(EISCAT)heating *** main instabilities include the parametric decay instability(PDI),oscillating two-stream instability(OTSI),and thermal parametric instability(TPI).The PDI and OTSI not only compete with each other,but also compete with the TPI,leading to a two-stage overshoot phenomenon:a miniovershoot occurs on a millisecond time scale after pump-on,followed by the main *** gain insight into the miniovershoot via a generalized Zakha.ov model,whereas the main overshoot can be observed as an enha.ced plasma line overshoot phenomenon in incoherent scatter radar *** can also observe tha. the zero-frequency ion line exists only in the initial heating period after a cold start and tha. the upshifted and downshifted ion lines beha.e irregularly in the *** simulation results show tha. competition between the PDI and OTSI leads to an initial peak,which we named the *** following processes,namely ion density caviton generation,and collapse and cascade in the development of the PDI,contribute to the miniovershoot phenomenon.
