Lenalidomide regulates the CCL21/CCR7/ERK1/2 axis to inhibit migration and proliferation in diffuse large B-cell lymphoma

Background:The prognostic significance of the chemokine receptor CCR7 in diffuse large B-cell lymphoma(DLBCL)has been reported ***,the detailed mechanisms of CCR7 in DLBCL,particularly regarding its interaction with lenalidomide treatment,are not fully ***:Our study utilized bioinformatics approaches to identify hub genes in SU-DHL-2 cell lines treated with lenalidomide compared to control *** data and clinical information from 122 patients with DLBCL were analyzed to assess the correlation of CCR7 and p-ERK1/2 expression with the prognosis of ***,in vitro and in vivo experiments were conducted to clarify the role of CCR7 in the response of DLBCL to lenalidomide ***:Our bioinformatics analysis pinpointed CCR7 as a hub gene in the context of lenalidomide treatment in ***,31.14%and 36.0%(44/122)of DLBCL cases showed positive expression for CCR7 and ERK1/2 respectively,establishing them as independent prognostic factors for adverse outcomes in DLBCL via multivariate Cox regression ***,our studies demonstrated that the external application of the protein CCL21 promoted proliferation,migration,invasion,and activation of the ERK1/2 pathway in SU-DHL-2 and OCI-LY3 cell lines with high levels of CCR7 *** effect was mitigated by CCR7 silencing through siRNA,application of ERK inhibitors,or lenalidomide *** vivo experiments reinforced the efficacy of lenalidomide,significantly reducing tumor growth rate,tumor mass,serum total LDH levels,and expression of CCR7 and p-ERK1/2 in a SUDHL-2 xenograft model in nude mice(p<0.05).Conclusion:Our study clarifies the potential role of the CCL21/CCR7/ERK1/2 axis in the therapeutic effects of lenalidomide in DLBCL treatment.

Theoretical and experimental study on high-entropy flotation of micro-fine cassiterite

Tin is a critical metal for various industries,making its recovery from low-grade cassiterite ores *** study aimed to optimize the flotation recovery of cassiterite using multi-component collector *** collectors were initially selected through micro-flotation tests,leading to the identification of optimal proportions for a four-component collector system(SHA-OHA-SPA-DBIA in a 4:3:2:1 ratio).Molecular dynamics simulations and surface tension tests were used to investigate the micellar behavior of these collectors in aqueous *** adsorption characteristics were quantified using microcalorimetry,enabling the determination of collection entropy and changes in Gibbs free *** four-component collector system showed the highest entropy change and the most favorable Gibbs free energy,leading to a cassiterite recovery of above 90%at a concentration of 8.0×10^(5)mol/*** analytical techniques were employed to systematically characterize the adsorption *** findings revealed a positive correlation between the adsorption products formed by the multicomponent collectors on the cassiterite surface and the entropy ***-scale testing of the high-entropy collector system produced a tin concentrate with an Sn grade of 6.17%and an Sn recovery of 82.43%,demonstrating its substantial potential for practical applications in cassiterite flotation.

Electrochemical conversion of hypertoxic trivalent arsenic to value-added metallic arsenic can not only contribute to pollution abatement,but also resources reutilization,therefore being widely *** reduction of trivalent arsenic as a promising way is widely ***,the high efficiency conversion is retarded by the sluggish reduction kinetics of AsO33−and fierce evolution of side products of both H_(2)and toxic AsH_(3).Herein,by using the sodium citrate as the additive,the current efficiency for metal arsenic production is increased greatly from 60%to 91%,with the accompanied evolution of hypertoxic AsH_(3)being restrained from 0.15 Nm^(3)/t_(As)to 0.022 Nm^(3)/t_(As),promising a high-efficiency and green *** electrochemical tests and electrode surface characterizations aswell as DFT calculations indicate that the added sodium citrate promotes both the diffusion of reactive AsO_(3)^(3−)towards the cathode and its subsequent adsorption on the Ti cathode,contributing to smoother reduction for generating metal arsenic,with the evolution of toxic AsH_(3)being hindered at the same *** results can provide new insights for the highefficiency and greener conversion of hypertoxic trivalent arsenic to value-added metallic arsenic.

Objective To explore the toxicological mechanism of hydroquinone in human bronchial epithelial cells and to investigate whether DNA polymerase beta is involved in protecting cells from damage caused by hydroquinone. Methods DNA polymerase beta knock-down cell line was established via RNA interference as an experimental group. Normal human bronchial epithelial cells and cells transfected with the empty vector of pEGFP-C1 were used as controls. Cells were treated with different concentrations of hydroquinone （ranged from 10 μmol/L to 120 μmol/L） for 4 hours. MTT assay and Comet assay [single-cell gel electrophoresis （SCGE）] were performed respectively to detect the toxicity of hydroquinone. Results assay showed that DNA polymerase beta knock-down cells treated with different concentrations of hydroquinone had a lower absorbance value at 490 nm than the control cells in a dose-dependant manner. Comet assay revealed that different concentrations of hydroquinone caused more severe DNA damage in DNA polymerase beta knock-down cell line than in control cells and there was no significant difference in the two control groups. Conclusions Hydroquinone has significant toxicity to human bronchial epithelial cells and causes DNA damage. DNA polymerase beta knock-down cell line appears more sensitive to hydroquinone than the control cells. The results suggest that DNA polymerase beta is involved in protecting cells from damage caused by hydroquinone.

Evaluation of the shear stability of aerobic granular sludge from a pilot-scale membrane bioreactor:Establishment of a quantitative method

This work established a quantitative method to access the shear stability of aerobic granular sludge(AGS)and validated its feasibility by using the mature AGS from a pilot-scale(50 tons/day)membrane bioreactor(MBR)for treating real municipal *** results showed that the changing rate( S)of the peak area(S)of granule size distribution(GSD)exhibited an exponential relationship(R^(2)≥0.76)with the shear time(y=a−b·cx),which was a suitable indicative index to reflect the shear stability of different AGS *** limiting granule size(LGS)was defined and proposed to characterize the equilibrium size for AGS after being sheared for a period of time,whose value in terms of Dv50 showed high correlation(R^(2)=0.92)with the parameter *** free Ca^(2+)(28.44-34.21 mg/L)in the influent specifically interacted with polysaccharides(PS)in the granule’s extracellular polymeric substance(EPS)as a nucleation site,thereby inducing the formation of Ca precipitation to enhance its Young’s modulus,while Ca2+primarily interacted with PS in soluble metabolic product(SMP)during the initial granulation ***,the Young’s modulus significantly affected the parameter a related to shear stability(R^(2)=0.99).Since the parameter a was more closely related(R^(2)=1.00)to S than that of the parameter b or c,the excellent correlation(R^(2)=0.99)between the parameter a and the wet density further verified the feasibility of this method.

huanglian-Renshen-Decoction Maintains Isletβ-Cell Identity in T2DM Mice through Regulating GLP-1 and GLP-1R in Both Islet and Intestine

Objective: To elucidate the effect of huanglian-Renshen-Decoction(HRD) on ameliorating type 2 diabetes mellitus by maintaining islet β-cell identity through regulating paracrine and endocrine glucagon-like peptide-1(GLP-1)/GLP-1 receptor(GLP-1R) in both islet and intestine. Methods: The db/db mice were divided into the model(distilled water), low-dose HRD(LHRD, 3 g/kg), high-dose HRD(HHRD, 6 g/kg), and liraglutide(400 μg/kg) groups using a random number table, 8 mice in each group. The db/m mice were used as the control group(n=8, distilled water). The entire treatment of mice lasted for 6 weeks. Blood insulin, glucose, and GLP-1levels were quantified using enzyme-linked immunosorbent assay kits. The proliferation and apoptosis factors of islet cells were determined by immunohistochemistry(IHC) and immunofluorescence(IF) staining. Then,GLP-1, GLP-1R, prohormone convertase 1/3(PC1/3), PC2, v-maf musculoaponeurotic fibrosarcoma oncogene homologue A(MafA), and pancreatic and duodenal homeobox 1(PDX1) were detected by Western blot, IHC,IF, and real-time quantitative polymerase chain reaction, respectively. Results: HRD reduced the weight and blood glucose of the db/db mice, and improved insulin sensitivity at the same time(P<0.05 or P<0.01). HRD also promoted mice to secrete more insulin and less glucagon(P<0.05 or P<0.01). Moreover, it also increased the number of islet β cell and decreased islet α cell mass(P<0.01). After HRD treatment, the levels of GLP-1,GLP-1R, PC1/3, PC2, MafA, and PDX1 in the pancreas and intestine significantly increased(P<0.05 or P<0.01).Conclusion: HRD can maintain the normal function and identity of islet β cell, and the underlying mechanism is related to promoting the paracrine and endocrine activation of GLP-1 in pancreas and intestine.

The potential mechanism and clinical application value of remote ischemic conditioning in stroke

Some studies have confirmed the neuroprotective effect of remote ischemic conditioning against stroke. Although numerous animal researches have shown that the neuroprotective effect of remote ischemic conditioning may be related to neuroinflammation, cellular immunity, apoptosis, and autophagy, the exact underlying molecular mechanisms are unclear. This review summarizes the current status of different types of remote ischemic conditioning methods in animal and clinical studies and analyzes their commonalities and differences in neuroprotective mechanisms and signaling pathways. Remote ischemic conditioning has emerged as a potential therapeutic approach for improving stroke-induced brain injury owing to its simplicity, non-invasiveness, safety, and patient tolerability. Different forms of remote ischemic conditioning exhibit distinct intervention patterns, timing, and application range. Mechanistically, remote ischemic conditioning can exert neuroprotective effects by activating the Notch1/phosphatidylinositol 3-kinase/Akt signaling pathway, improving cerebral perfusion, suppressing neuroinflammation, inhibiting cell apoptosis, activating autophagy, and promoting neural regeneration. While remote ischemic conditioning has shown potential in improving stroke outcomes, its full clinical translation has not yet been achieved.
