Dear Editor,Human immunodeficiency virus(HIV)is a lentivirus that,if left untreated,can lead to acquired immunodeficiency syndrome(AIDS).The virus can be divided into two types based on genetic differences:HIV-1 and HIV-2.HIV-1 is widely distributed worldwide and is the primary strain responsible for the global AIDS epidemic due to its high replication capacity,greater likelihood of transmission,and more severe clinical symptoms(Oeschger et al.,2021).

Circular RNAs (circRNAs) are a special class of single-stranded RNA molecules with covalently closed loops widely expressed in eukaryotic organisms. CircRNAs have long been considered to play important roles in various physiological and pathological processes as non-coding RNAs. However, circRNAs have recently garnered considerable attention due to their ability to be translated into peptides/proteins via internal ribosome entry site- or N6-methyladenosine-mediated pathways or rolling translation mechanisms. Furthermore, dysregulation of translatable circRNAs and their encoded proteins has been associated with developing and progressing diseases such as cancer. This review aims to summarize the driving mechanisms of circRNA translation and the available strategies in circRNA translation research. The main focus is on the emerging biological functions of translatable circRNAs, their regulatory mechanisms, and potential clinical applications in human diseases to provide new perspectives on disease diagnosis, prognosis, and targeted therapy.

Continuumtopology optimization considering the vibration response is of great value in the engineering structure design.The aimof this studyis toaddress the topological designoptimizationof harmonic excitationstructureswith minimumlength scale control to facilitate structuralmanufacturing.Astructural topology design based on discrete variables is proposed to avoid localized vibration modes,gray regions and fuzzy boundaries in harmonic excitation topology optimization.The topological design model and sensitivity formulation are derived.The requirement of minimum size control is transformed into a geometric constraint using the discrete variables.Consequently,thin bars,small holes,and sharp corners,which are not conducive to the manufacturing process,can be eliminated from the design results.The present optimization design can efficiently achieve a 0–1 topology configuration with a significantly improved resonance frequency in a wide range of excitation frequencies.Additionally,the optimal solution for harmonic excitation topology optimization is not necessarily symmetric when the load and support are symmetric,which is a distinct difference fromthe static optimization design.Hence,one-half of the design domain cannot be selected according to the load and support symmetry.Numerical examples are presented to demonstrate the effectiveness of the discrete variable design for excitation frequency topology optimization,and to improve the design manufacturability.

A multifunctional rock testing system for rock failure analysis under different stress states: Development and application

The stress state in a rock mass is complex. Stress redistribution around underground excavation may lead to various failure modes, including compressive-shear, tensile-shear, and tensile failures. The ability to perform laboratory tests with these complex stress states is significant for establishing new strength criteria. The present paper introduces a new rock testing system with “tensile-compressive-shear”loading functions. The device includes bi-directional and double-range hydraulic cylinders, auxiliary loading equipment, and roller rows that can perform direct compressive-shear tests, direct tensile tests,and direct tensile-shear tests. The testing system provides maximum vertical and lateral loading forces of2000 k N and allows testing cubical rock specimens with dimensions of 0.5 m × 0.5 m × 0.5 m. The performance of the testing machine was evaluated by testing a rock-like material based on cement mortar under compressive-shear, tensile, and tensile-shear stress states. The failure process and deformation characteristics were monitored during loading using acoustic emission(AE) transient recorder,piezoelectric AE sensors, a high-speed camera, and a thermal infrared camera. The failure mechanism was investigated by analyzing AE counts, AE amplitude, strain, and temperature changes on the rock specimen surface. The test results confirmed that the testing system could successfully simulate the abovementioned stress path. The AE counts and amplitude responses were influenced by different failure modes. The temperature response during the compressive-shear test indicated the development of a high-temperature band on the rock specimen surface. In contrast, a negligible temperature change was observed during the tensile and tensile-shear tests. The newly developed multifunctional rock testing system allows laboratory tests under various failure modes. The monitoring results of multiple variables during rock failure tests provide valuable information on failure characteristics.

An advanced stability analysis of surrounding rock can assure safe construction in tunnels.However,in traditional analysis methods,geometric information on the rock mass discontinuities is obtained by a geological compass and treated as an infinitely expanded plane in a numerical simulation,which is inaccurate in both geometric and numerical models.To solve this problem,this paper studied a control network and an image stitch method to assess the accuracy of a geometric model and proposed a more accurate method to obtain the geometric information of rock mass discontinuities based on digital photogrammetry.Based on discrete fracture network and discrete element method(DFN-DEM),this paper treated the rock mass discontinuities obtained by digital photogrammetry as finite disk planes and simulated the excavation process of the Qianyu tunnel.According to the simulation results,this paper determined the specific location of the collapse disaster on the tunnel face,which is of great significance to tunnel support design.Comparing the deformation of the surrounding rock mass on the tunnel face in the simulation results with the actual situation,this paper verified the feasible accuracy of this method in analysing the stability of the surrounding rock mass on a tunnel face in advance.

郑成功复台战争是大航海时代中西冲突规模最大的一场战争,从之发生迄今,古今国人对侵华的荷兰东印度公司(VOC)属下的海军船炮技术夸张甚多。今利用中荷一些原始资料以及调研,发现荷夷船队规模一直从4-15艘不等,此与明郑水师五六百艘御侮战船数量相比差距悬殊。荷夷船只大部分载重量100吨左右,仅有4艘达到了五六百吨。在该战争中,因为荷夷人数少,船队规模小,故最终败给了人多势众、船队规模庞大的郑成功明郑船队。

Background:The regulatory genes related to lipid metabolism affect the intramuscular fat(IMF)content and improve meat quality traits.Mediator Complex Subunit 4(MED4),a vitamin D receptor protein,affects vitamin D,livestock growth,carcass traits,and triglyceride deposition.However,the physiological function of the MED4 gene on bovine adipocyte differentiation remains unknown.Methods:This study explored the function of the MED4 gene in preadipocyte differentiation in Chinese Red Steppe cattle.The overexpression plasmid and the interference sequences of the MED4 gene were constructed to detect the effects of the MED4 gene on adipogenesis and biomakers using quantitative polymerase chain reaction and western blotting.Results:The MED4 gene had significantly high expression during preadipocyte differentiation(p<0.05).Overexpression of the MED4 gene increased the expression of the PPARγgene,a preadipocyte differentiation biomarker,improved cellular lipid droplets and triglycerides accumulation,and positively accelerated adipocyte maturation(p<0.05).Interference of the MED4 gene can negatively regulate preadipocyte differentiation.Conclusion:This study showed that the MED4 gene may affect the preadipocyte differentiation and adipogenesis in Chinese Red Steppe cattle by regulating the PPARγgene.

We hypothesized that the P2X7 receptor may be the target of isoflurane, so we investigated the roles of the P2X7 receptor and inositol triphosphate receptor in calcium overload and neuronal apoptosis induced by isoflurane in cultured embryonic rat hippocampal neurons. Results showed that isoflurane induced widespread neuronal apoptosis and significantly increased cytoplasmic Ca^2＋ Blockade of P2X7 receptors or removal of extracellular Ca^2＋ combined with blockade of inositol triphosphate receptors completely inhibited apoptosis or increase in cytoplasmic Ca^2＋. Removal of extracellular Ca^2＋ or blockade of inositol triphosphate receptor alone could partly inhibit these effects of isoflurane. Isoflurane could directly activate P2X7-gated channels and induce inward currents, but did not affect the expression of P2X7 receptor protein in neurons. These findings indicate that the mechanism by which isoflurane induced neuronal apoptosis in rat developing brain was mediated by intracellular calcium overload, which was caused by P2X7 receptor mediated calcium influx and inositol triphosphate receptor mediated calcium release.

Astrocytes can release increased levels of brain-derived neurotrophic factor during cerebral ischemia, but it is unclear whether brain-derived neurotrophic factor affects y-aminobutyric acid type A receptor function in normal neurons. Results from this study demonstrated that y-aminobutyric acid at 100 pmol/L concentration raised the intracellular calcium level in neurons treated with medium from cultured hypoxic astrocytes, and the rise in calcium level could be inhibited by y-aminobutyric acid type A receptor antagonist bicuculline or brain-derived neurotrophic factor receptor antagonist k252a, y-aminobutyric acid type A-gated current induced by 100 IJmol/L y-aminobutyric acid was in an inward direction in physiological conditions, but shifted to the outward direction in neurons when treated with the medium from cultured hypoxic astrocytes, and this effect could be inhibited by k252a. The reverse potential was shifted leftward to -93 mV, which could be inhibited by k252a and Na＋-K＋-CI cotransporter inhibitor bumetanide. Brain-derived neurotrophic factor was released from hypoxic astrocytes at a high level. It shifted the reverse potential of y-aminobutyric acid type A-gated currents leftward in normal neurons by enhancing the function of Na＋-K＋-CI- cotransporter, and caused y-aminobutyric acid to exert an excitatory effect by activating y-aminobutyric acid type A receptor.
