Objective Toxoplasma gondii causes one of the most common protozoal diseases of humans and animals *** the aim of designing an effective vaccine against *** infection,we have examined the immunogenicity of a DNA vaccine expressing heat shock protein 40(HSP40) against challenge with ***(type I RH and type II Pru) strains in Kunming *** The plasmid p VAX1-HSP40 was constructed and used to immunize mice by intramuscular injection for three sequential immunizations with two-week intervals,and the immune responses including specific antibodies,lymphocyte proliferation,cytokines and T lymphocyte subclasses,the survival times of RH-infected mice and the number of cysts in brains of Pru-infected ones were *** This immunization regimen significantly reduced parasite cyst burden in p VAX1-HSP40-immunized mice(1871.9 ± 142.3) compared with control mouse groups immunized with p VAX1(3479.2 ± 204.4),phosphate buffered saline(3024.4 ± 212.8),or left untreated(3275.0 ± 179.8) as healthy controls(P mmunization failed to protect mice against challenge with the virulent RH *** was significant increase in T lymphocyte subclasses(CD3 e+CD4+ T and CD3 e+CD8 a+ T lymphocytes) in splenic tissues in immunized mice compared with controls(P el of antibodies,lymphocyte proliferation and concentration of cytokines(IFN-γ,IL-2,IL-4,IL-10 and IL-12 p70) were not significantly different between immunized and control mouse groups(P mmune responses and achieved a significant reduction in the number of brain cysts in Pru-infected mice,and thus can be tested in future immunization studies along with plasmids containing other immunogenic proteins as a cocktail vaccine to fully abolish chronic toxoplasmosis.

Dense granule protein 12(GRA12) is implicated in a range of processes related to the establishment of Toxoplasma gondii infection,such as the formation of the intravacuolar network(IVN) produced within the parasitophorous vacuole(PV).This protein is thought to be important for ***-host interaction,pathogenesis,and immune evasion,but their exact role remains *** this study,the contributions of GRA12 to the molecular pathogenesis of *** infection were examined in vitro and in *** of GRA12 in type I RH and type II Pru *** strains although did not affect parasite growth and replication in vitro,it caused a significant reduction in the parasite virulence and tissue cyst burden in *** △gra12 mutants were more vulnerable to be eliminated by host immunity,without the accumulation of immunity-related GTPase a6(Igra6) onto the PV *** ultrastructure of intravacuolar network(IVN) in △gra12 mutants appeared normal,suggesting that GRA12 is not required for the IVN *** deletion of GRA12 and ROP18 induced more severe attenuation of virulence compared to single △gra12 or △rop18 mutant strains,suggesting that GRA12 may act synergistically with ROP18 in *** *** these findings indicate that although GRA12 is not essential for parasite growth and replication in vitro,it contributes to the virulence and growth of *** in mice.

This paper presents a novel fault-tolerant networked control system architecture consisting of two cells working in-line. This architecture is fault-tolerant at the level of the controllers as well as the sensors. Each cell is based on the sensor-to-actuator approach and has an additional supervisor node. It is proven, via analysis as well as OmNeT++ simulations that the production line succeeds in meeting all control system requirements with no dropped or over-delayed packets. A reliability analysis is then undertaken to quantitatively estimate the increase in reliability due to the introduction of fault-tolerance.

The liver fluke Fasciola gigantica produces a number of proteins during progression of infection inside the *** of these proteins can modulate the host immune ***,we investigated the effect of a recombinant tegumental calcium-binding EF-hand protein 4 from ***(rFg-CaBP4) on the ability of goat monocytes to produce *** isopropyl-β-dthiogalactopyranoside-induced rFg-CaBP4 protein was recognized by rat sera containing polyclonal antibodies raised against rFg-CaBP4,and its binding to the surface of monocytes was confirmed by immunofluorescence *** analysis localized native CaBP4 to the oral sucker,pharynx,genital pore and acetabulum as well as the tegument of adult *** ***-incubation of rFg-CaBP4 with concanavalin A-stimulated monocytes increased the production of interleukin 2(IL-2),IL-4,interferon-y,and transforming growth factor-β and decreased the level of ***,no change in the concentration of tumor necrosis factor-α was ***4 significantly induced the mHC-Ⅱ expression,but reduced the expression of mHC-Ⅰexpression on monocytes,in a dose-dependent *** findings support the role of CaBP4 as a modulator of the host immune response and implicate rFg-CaBP4 as a potential therapeutic target for the development of intervention to prevent or treat fascioliasis.

Dosimetric evaluation of a beam matching procedure

Objective: The dosimetric characteristics for linear accelerators with the same model, and nominal energy are known to be very similar, as long as the machines are unaltered from the manufacturer's original specifications. In this preliminary study, a quantitative investigation of the similarity in the basic photon and electron dosimetry data from the Siemens Oncor linear accelerators at our hospital(Children's Cancer Hospital, Cairo, Egypt) was reported. methods: The output factor(OF), wedge factors(WF), percentage depth dose(PDD), and beam profile for the 6 and 10 mV photon beams were measured. Results: The measured output factors varied by less than about 1% for each field size. The difference between the maximum and minimum PDD values at each depth was less than about 1%. The difference between the beam flattnes and symetry was no more than 1% at all off-axis distances. For electron the results showed that the PDD, OF, and the beam profiles were matched within 1% differences. Conclusion: These results strongly suggest that it is feasible to establish one reference photon and electron dosimetry data set for the two machines and nominal energies.

In this study,we generated a tkl1 deletion mutant in the Toxoplasma gondii type 1 RH(RHΔtk1 l) strain and tested the protective efficacies of vaccination using RHΔtkl1 tachyzoites against acute,chronic,and congenital *** infections in Kunming *** vaccinated with RHΔtkl1 mounted a strong humoral and cellular response as shown by elevated levels of ***-specific IgG,IL-2,IL-12,IFN-γ,and *** RHΔtkl1-vaccinated mice survived a lethal challenge with 1×103 tachyzoites of type 1 RH or ToxoDB#9(PYS or TgC7) strain as well as 100 cysts or oocysts of Prugniuad *** mock-vaccinated plus infected mice have *** also protected against cyst-or oocyst-caused chronic infection,reduced vertical transmission caused by oocysts,increased litter size,and maintained body weight of pups brn to dams challenged with10 oocysts on day 5 of *** contrast,all mock-vaccinated plus oocysts-infected dams had aborted,and no fetus has *** dams remained healthy post infection,and their brain cyst burden was significantly reduced compared with mock-vaccinated dams infected with *** vivo depletion of CD4+T cells,CD8+T cells,and B cells revealed that CD8+T cells are involved in the protection of mice against *** ***,adoptive transfer of CD8+T cells from RHΔtkl1-vaccinated mice significantly enhanced the survival of naive mice infected with the pathogenic ***,these data reaffirm the importance of CD8+T cell responses in future vaccine design for toxoplasmosis and present *** tkl1 gene as a promising vaccine candidate.

The apicomplexan parasite Toxoplasma gondii can cause severe disease in pregnant women who acquire primary infection during pregnancy and in immune-suppressed *** the asexual stages,*** can transform between the rapidly dividing tachyzoites and the relatively quiescent,slow-dividing bradyzoites,forming the latent tissue *** effective treatments exist to eliminate the form of tissue cysts due to the poor understanding of the biology of chronic *** tissue cysts are characterized by the presence of amylopectin ***,the accurate roles and regulatory mechanisms of amylopectin accumulation in this parasite are not fully *** genome encodes a series of glycogenes and thus this parasite can utilize host sugars for energy and to generate glycoconjugates that are important to its growth,virulence,and ***,we investigated the basic functions of amylo-alpha-1,6-glucosidase(Aa16 GL) in the tachyzoite and bradyzoite stages of ***,using transgenic parasites that lack Aal6 GL mutants in type ⅠRH and type Ⅱ Prugniaud(Pru) *** addition,the subcellular localization of Aa16 GL was characterized by inserting a 3×HA at the 3’end of the Aal6 GL gene endogenous *** results showed that Aal6 GL localized predominantly to several small puncta within the *** characterization of△Aa16 GL mutants by plaque assay,egress assay and intracellular replication assay showed that parasites lacking Aa16 GL exhibit a slight growth defect and remain virulent in ***,although Pru△Aa16 GL tachyzoites has the ability to differentiate into bradyzoites in vitro,they displayed modest reduction of cyst formation in *** together,our results showed that Aa16 GL played minor roles in the *** growth,but likely affect the cyst formation in vivo.

Fatty acid uptake is extremely important for the survival and growth of the intracellular parasite Toxoplasma *** this study,CRISPR-Cas9 gene editing technology was used to investigate the role of four lipid synthesis enzymes,namely Glycerol-3-phosphate dehydrogenase(G3 PDH),malonyl CoA-acyl carrier protein transacylase(Fabd),Acyl-ACP thiolesterase(TE)and diacylglycerol acyltransferase(DGAT),in the virulence and infectivity of Type Ⅰ RH and Type Ⅱ Prugniaud(Pru) *** analysis of the tachyzoites stage showed that Fabd protein was located in the apicoplast;however,the expression level of the other three proteins were undetectable in tachyzoites *** with wild type(WT) strains,the growth of RHΔG3 PDH,RHΔTE and RHΔDGAT in vitro and their virulence in vivo were not significantly ***,RHΔFabd exhibited a significantly reduced growth rate,compared with the WT ***,the deletion of Fabd attenuated the virulence of Type Ⅱ Pru strain and reduced the formation of cysts in *** data improved our understanding of the role of lipid synthesis enzymes in the pathogenesis of ***.

In the present study,a dense granule protein 17(gra17) and novel putative transporter(nptl) double deletion mutant of Toxoplasma gondii RH strain were *** protective efficacy of vaccination using RHΔgra17Δnpt1 tachyzoites against acute,chronic,and congenital toxoplasmosis was studied in a mouse *** using RHΔgrfa17Δnpt1 induced a strong humoral and cellular response,as indicated by the increased levels of *** specific IgG,interleukin 2(IL-2),IL-10,IL-12,and interferon-gamma(IFN-γ).Vaccinated mice were protected against a lethal challenge dose(103 tachyzoites) of wild-type homologous(RH) strain and heterologous(PYS and TgC7) strains,as well as against 100 tissue cysts or oocysts of Pru *** also conferred protection against chronic infection with 10 tissue cysts or oocysts of Pru strain,where the numbers of brain cysts in the vaccinated mice were significantly reduced compared to those detected in the control(unvaccinated+infected) *** addition,vaccination protected against congenital infection with 10 *** Pru oocysts(administered orally on day 5 of gestation) as shown by the increased litter size,survival rate and the bodyweight of pups born to vaccinated dams compared to those born to unvaccinated+infected *** brain cyst burden of vaccinated dams was significantly lower than that of unvaccinated dams infected with *** data show that *** RHΔgra17Δnpt1 mutant strain can protect mice against acute,chronic,and congenital toxoplasmosis by balancing inflammatory response with immunogenicity.

引言/目的犬弓首蛔虫是一种被忽视的人畜共患寄生虫,可以感染人类和狗等其它动物。而犬蛔虫感染终末宿主进展的分子机制大部分仍不清楚。microRNAs (miRNAs)作为一种小的非编码RNA在犬弓首蛔虫感染宿主期间发挥重要的调控作用。因此本研究通过高通量测序方法对犬弓首蛔虫感染的比格犬的肝脏进行miRNAs的检测,旨在探讨这些差异的miRNAs在犬弓首蛔虫感染宿主期间发挥的作用,为探究犬弓首蛔虫与终宿主间相互作用提供理论依据。
