Green tea polyphenols inhibit testosterone production in rat Leydig cells

This study investigated the acute effects of green tea extract （GTE） and its polyphenol constituents, （-）-epigallocatechin-3-gallate （EGCG） and （-）-epicatechin （EC）, on basal and stimulated testosterone production by rat Leydig cells in vitro. Leydig cells purified in a Percoll gradient were incubated for 3 h with GTE, EGCG or EC and the testosterone precursor androstenedione, in the presence or absence of either protein kinase A （PKA） or protein kinase C （PKC） activators. The reversibility of the effect was studied by pretreating cells for 15 min with GTE or EGCG, allowing them to recover for 1 h and challenging them for 2 h with human chorionic gonadotropin （hCG）, luteinizing hormone releasing hormone （LhRh）, 22（R）-hydroxycholesterol or androstenedione. GTE and EGCG, but not EC, inhibited both basal and kinase-stimulated testosterone production. Under the pretreatment conditions, the inhibitory effect of the higher concentration of GTE/EGCG on hCG/LhRh-stimulated or 22（R）- hydroxycholesterol-induced testosterone production was maintained, whereas androstenedione-supported testosterone production returned to control levels. At the lower concentration of GTE/EGCG, the inhibitory effect of these polyphenols on 22（R）-hydroxycholesterol-supported testosterone production was reversed. The inhibitory effects of GTE may be explained by the action of its principal component, EGCG, and the presence of a gallate group in its structure seems important for its high efficacy in inhibiting testosterone production. The mechanisms underlying the effects of GTE and EGCG involve the inhibition of the PKA/PKC signalling pathways, as well as the inhibition of P450 side-chain cleavage enzyme and 17β-hydroxysteroid dehydrogenase function.

The continuous assessment of child health services generates knowledge about the real needs of health services and actions in order to achieve better results and indicators. Thus, this study proposes to analyze child health indicators, based on the information systems of health services. This is a descriptive, ecological epidemiological study carried out using secondary data from the following information systems: Information System on Live Births and Mortality Information System, in a historical series of ten years (2005-2014) of the state of Rio Grande do Norte and its health micro regions. It was observed a decrease in infant, neonatal, preterm, late neonatal and postnatal mortality coefficients when compared to the first and last year of the observed period, as well as the concentration of these in micro-regions of health. There was also a positive association between the early neonatal component and infant death. In this perspective, it is important to evaluate the quality of prenatal care, childbirth, puerperium, the first week of integral health and follow-up of the child’s growth and development, as well as to relate the socio-demographic conditions that may be associated with infant morbimortality. Therefore, the importance of situational health assessment studies for the effectiveness of care provided is highlighted, identifying weaknesses and monitoring the effectiveness of strategies.

Models for the study of computational fluid dynamics in vehicles to determine aerodynamic loads usually take into account only the geometry of the body. Several constructive elements such as the wheel geometry or suspension components are disregarded in the computational models. This work presents the study of the aerodynamics of a one-fourth model passenger vehicle, which contains the wheelhouse interior elements. The goal is to identify the aerodynamic loads produced by these components and their effect on the flow dynamics. Wheel and tire set, brake components, suspension and drive shaft are contemplated. Computer simulations were performed to the vehicle speed varying from 0 to 120 km/h and included the rotation of the tire and wheel assembly, considering the tire geometry in dynamic conditions. The computational model is solved by the finite volume method, wherein the computational domain is divided into tetrahedral and hexahedral elements. The turbulence model used is the standard k −ε.

Purification and Biochemical Characterization of Native and Pegylated Form of L-Asparaginase from Aspergillus terreus and Evaluation of Its Antiproliferative Activity

L-asparaginase is a chemotherapeutic drug used in the treatment of lymphoblastic leukemia. In the present study, the extracellular L-asparaginase produced by strain (PC-1.7A) of Aspergillus terreus was purified, characterized, and modified with polyethylene glycol. Moreover, its antiproliferative activity was evaluated. The apparent molecular weight of the enzyme was found to be 136 kDa. The optimal ph and temperature for the enzyme were 9.0℃ and 40℃, respectively. The enzyme retained 100% of the activity at 40℃ for 120 min. Pegylated L-asparaginase was more thermostable and more resistant to trypsin than native enzyme. Native L-asparaginase against human normal cells did not show cytotoxicity. however, in the leukemia cell lines RS4;11 and hL60 the antiproliferative effects of native L-asparaginase were observed after 96 and 72 h of incubation, respectively. For the first time, an L-asparaginase from fungus was evaluated as an antitumor agent in human cells lines and further investigations should be conducted to improve the knowledge about this enzyme.

The aim of this study was to evaluate morphometric and hematological parameters of juveniles Piracanjubas treated with two distinct photoperiod. Twenty nine fish with 100 g, in a factorial design were performed in 18 h light：6 h dark （18L：6D） and 6L：lSD two different photoperiod, and morphometric and hematological parameters were evaluated at 45 d and 90 d after photoperiod treatment. Benzocaine 1% was used for euthanasia. Fish were weighed and measured, so as visceral fat, liver and gonads. hematocrit, total count of white cells, lymphocytes and neutrophils were counted. Results showed that there was no significant relationship between bodyweight and standard length. Significant interaction （P = 0.00018） had been found between the fat index （FI） and gonado-somatic index （GSI） and the photoperiod and time. At 90 d, the fish kept in 18L：6D had a lower rate of fat than the fish kept in 6L：18D （P 〈 0.05）. The GSI was higher in the group 6L.＇18D compared with the 18L：6D （P 〈 0.005）. There was no interaction between photoperiod and period of treatment for total leukocyte count, however with 90 d, the total leukocyte count was decreased （P 〈 0.05）. Moreover, decrease of the hematocrit from 58.1% to 46.9% and an increase of lymphocytes from 31.5% to 43.5% had been found in fish kept in 18L：6D after 90 d （P 〈 0.05）. Fish kept in 6L：ISD presented an increase in segmented neutrophils and lymphocytes （P 〈 0.05）. It is concluded that photoperiod maintained at 6L： 18D for Piracanjubas permits a greater gonadal development and an amount of immune cells over the photoperiod maintained in 18L： 6D.

研究目的通过不同频率少林内功练习对2型糖尿病前期的进行治疗,观察和评估不同频率少林内功的治疗作用,并相互比较以明确其量效关系进行,为少林内功治疗2型糖尿病前期的临床应用提供可靠的支持。研究方法60例符合纳入标准并且不符合排除标准的社区2型糖尿病前期患者,被随机分为少林内功一组、少林内功二组、少林内功三组、少林内功四组、步行对照组及空白对照组各10例。少林内功一组、二组、三组、四组分别予1次/天、2次/天、3次/天、4次/天,每周练习五天的少林内功练习;步行对照组予中等步速(80～100步/分钟)步行30分钟,2次/天,每周步行五天;空白对照组则不干预其日常活动;共进行6个月的治疗。选用空腹血糖、餐后2小时血糖及糖化血红蛋白作为观察指标,于各组患者治疗前、治疗3个月、治疗6个月分别对患者进行评价。对治疗前后的观察指标进行组内对比,明确各组疗效;对各组间观察指标治疗前后差值进行组间对比,明确不同锻炼方法优劣及少林内功的量效关系。研究结果各组患者干预前在年龄、性别、空腹血糖、餐后2h血糖及糖化血红蛋白上的差异均无统计学意义(P>0.05),各组间基线一致,具有可比性。少林内功各组习练3个月及习练6个月后,患者各项指标与干预前相比,差异均有显著性意义(Ph血糖的组间对比,少林内功各组均显著优于空白对照组(Ph血糖及糖化血红蛋白水平,优于健康宣教;在空腹血糖、餐后2h血糖的改善上,少林内功习练3次/日优于步行训练;糖化血红蛋白改善上,少林内功习练3次/日、4次/日优于步行训练。量效关系分析揭示,少林内功习练3次/日对2型糖尿病前期有着最佳疗效。

Aim: Shigella flexneri (S. flexneri) is a gram-negative enterobacterium responsible for severe intestinal end systemic infection in humans. The bacteria can reach the liver due to degeneration of the colonic epithelium. hypoxia is present in many human diseases and can induce the expression of the transcription factor hIF-1alpha that may have a cell protective role. The influence of hypoxia and hIF-1alpha on bacterial infection, studied in this work, is unclear. hypoxia inducible factor-1alpha (hIF-1alpha) is a transcription factor that acts as a master regulator of gene expression induced by hypoxia. Methods: We compared the ability of S. flexneri to invade rat hepatocytes in primary culture both in normoxic and hypoxic conditions. We evaluated TNF-alpha released by hepatocytes, apoptosis rate and hIF-1alpha expression by confocal microscopy as well as real time PCR technique. Results: We showed that S. flexneri invaded less hepatocytes previously submitted to 24 h hypoxia (6.5% O2) than those cultivated in normoxia (21% O2). S. flexneri also induced hIF-1α expression in hepatocytes, TNF-α secretion and apoptosis. Conclusion: a) hypoxia alone was not a stimulus to TNF-α secretion, but induced cell apoptosis and hIF-1α expression;b) S. flexneri was able to invade rat hepatocytes and hypoxia apparently influenced significantly bacterial cell invasiveness;c) hIF-1α was expressed in hypoxic conditions, and it was also stimulated by S. flexneri.
