Vitamin D receptor expression in human bone tissue and dose-dependent activation in resorbing osteoclasts

The effects of vitamin D on osteoblast mineralization are well documented. Reports of the effects of vitamin D on osteoclasts, however, are conflicting, showing both inhibition and stimulation. Finding that resorbing osteoclasts in human bone express vitamin D receptor （VDR）, we examined their response to different concentrations of 25-hydroxy vitamin D3 [25（Oh）D3] （100 or 500 nmol·L^-1） and 1,25-dihydroxy vitamin D3 [1,25（Oh）2D3] （0.1 or 0.5 nmol·L^-1） metabolites in cell cultures. Specifically, CD14＋ monocytes were cultured in charcoal-stripped serum in the presence of receptor activator of nuclear factor kappa-B ligand （RANKL） and macrophage colony-stimulating factor （M-CSF）. Tartrate-resistant acid phosphatase （TRAP） histochemical staining assays and dentine resorption analysis were used to identify the size and number of osteoclast cells, number of nuclei per cell and resorption activity. The expression of VDR was detected in human bone tissue （ex vivo） by immunohistochemistry and in vitro cell cultures by western blotting. Quantitative reverse transcription-PCR （qRT-PCR） was used to determine the level of expression of vitamin D-related genes in response to vitamin D metabolites. VDR-related genes during osteoclastogenesis, shown by qRT-PCR, was stimulated in response to 500 nmol·L^-1 of 25（Oh）D3 and 0.1-0.5 nmol·L^-1 of 1,25（Oh）2D3, upregulating cytochrome P450 family 27 subfamily B member I （CYP27B1） and cytochrome P450 family 24 subfamily A member I （CYP24A1）. Osteoclast fusion transcripts transmembrane 7 subfamily member 4 （tm7sf4） and nuclear factor of activated T-cell cytoplasmic 1 （nfatcl） where downregulated in response to vitamin D metabolites. Osteoclast number and resorption activity were also increased. Both 25（Oh）D3 and 1,25（Oh）2D3 reduced osteoclast size and number when co-treated with RANKL and M-CSF. The evidence for VDR expression in resorbing osteoclasts in vivo and low-dose effects of 1,25（Oh）2D3 on osteoclasts in vitro may therefore provide

In this paper, we are concerned with a fractional differential inequality containing a lower order fractional derivative and a polynomial source term in the right hand side. A non-existence of non-trivial global solutions result is proved in an appropriate space by means of the test-function method. The range of blow up is found to depend only on the lower order derivative. This is in line with the well-known fact for an internally weakly damped wave equation that solutions will converge to solutions of the parabolic part.

A wavelet transform method for studying the energy distribution characteristics of microseismicities associated rock failure

Microseismicity signals released during rock failure process are firstly recorded using microseismicity monitoring system.A wavelet transform scheme is then developed on the basis of the discrete wavelet transform and implemented into MATLAB to study the energy distribution characteristics of the monitored microseismicity *** wavelet transform scheme decomposes the recorded microseismicity signals into various wavelets at seven scales and eight frequency *** microseismicity energy at each frequency band is then calculated by integrating the wavelets in each *** is found that,for the microseismicity signals recorded during the uniaxial loading of the granite,the microseismicity energies are mainly distributed between the bands 7.8125-15.625 khz,15.625-31.250 khz and 31.25-62.5 khz and the percentages of the released energies at these frequency bands are 8.24%,62.72%,28.08% of the total energies,*** results reveal that the microseismicity energies at these levels are directly related to the damage mechanisms of the granite although further studies are need to identify the failure *** these monitored signals were processed using wavelet transformation to find out the frequency distribution rule and the frequency band energy varying rule of the acoustic emission(AE) signals during the different rock damage and failure *** rock failure mechanism was interpreted from the perspective of the relationship between AE signal frequency change and crack *** frequency band energy distribution histograms of the microseismicity signals at different damage stages were computed and drawn by the energy calculation method of wavelet transformation implemented into *** energy percentage of the low frequency band(d7-a7) and that of the dominated frequency band(d4-d6) and their variation rule were analyzed ***,the critical damage point is that the low frequency energy percentage is above a certain threshol

Occurrence of viable but non-culturable(VBNC)pathogenic bacteria in tap water of public places

Viable but non-culturable(VBNC)bacteria have been detected in source water and effluent of drinking water treatment processes,leading to significant underestimation of viable cell *** information exists on VBNC bacteria in tap water,particularly in public *** address this gap,a comprehensive nine-month study was conducted in a major city in south-eastern China,using culture-based and quantitative PCR with propidium monoazide(PMA)dye ***-five samples were collected from five representative public places(railway station,campus,hospital,shopping mall,and institution).The findings revealed that culturable bacteria represented only 0–17.51%of the viable 16S rRNA genes,suggesting that the majority of viable bacteria existed in an uncultured or VBNC ***,opportunistic pathogens such as Escherichia coli,Enterococcus faecalis,Pseudomonas aeruginosa,Salmonella sp.,and Shigella *** primarily detected as VBNC cells,with concentrations ranging from 1.03×10^(0)to 3.01×10^(3),1.20×10^(0)to 1.42×10^(2),1.32×10^(0)to 8.82×10^(0),1.00×10^(0)to 6.71×10^(1),and 2.07×10^(0)to 1.93×10^(2)cell equivalent/100 mL,*** *** was observed in tap water after prolonged stagnation,indicating potential risks associated with bacterial *** and temporal factors accounted for 17.1%and 26.0%,respectively,of the variation in tap water community structure during the sampling period,as revealed by 16S rRNA amplicon *** study provides quantitative insights into the occurrence of VBNC bacteria in tap water and highlights the need for more sensitive monitoring methods and microbial control techniques to enhance tap water safety in public locations.

Follicle-stimulating hormone(FSh)levels prior to prostatectomy are not related to long-term oncologic or cardiovascular outcomes for men with prostate cancer

Prior research suggests a link between circulating levels of follicle-stimulating hormone(FSh)and prostate cancer *** levels may also explain some of the observed differences in cardiovascular events among men treated with gonadotropin-releasing hormone(GnRh)antagonists compared to GnRh *** study evaluates the association between preoperative FSh and long-term cardiovascular and oncologic outcomes in a cohort of men with long follow-up after radical *** performed a cohort study utilizing an institutional biobank with annotated clinical *** levels were measured from cryopreserved plasma and compared with sex steroids previously measured from the same *** in oncologic outcomes between tertiles of FSh levels were compared using adjusted cox regression *** adverse cardiovascular events(MACE)were similarly assessed using hospital admission diagnostic codes.A total of 492 patients were included,with a median follow-up of 13.1(interquartile range:8.9–15.9)*** sulfate(DhEA-S)levels,but not other androgens,negatively correlated with FSh levels on linear regression analysis(P=0.03).There was no association between FSh tertile and outcomes of biochemical recurrence,time to castrate-resistant prostate cancer,or time to *** were identified in 50 patients(10.2%),with a mean time to first event of 8.8 *** association with FSh tertile and occurrence of MACE was *** results do not suggest that preoperative FSh levels are significantly associated with oncologic outcomes among prostate cancer patients treated with radical prostatectomy,nor do these levels appear to be predictors of long-term cardiovascular risk.
