Epidermal stem cells and skin tissue engineering in hair follicle regeneration

The reconstitution of a fully organized and functional hair follicle from dissociated cells propagated under defined tissue culture conditions is a challenge stillpending in tissue engineering. The loss of hair follicles caused by injuries or pathologies such as alopecia not only affects the patients' psychological well-being, but also endangers certain inherent functions of the skin. It is then of great interest to find different strategies aiming to regenerate or neogenerate the hair follicle under conditions proper of an adult individual. Based upon current knowledge on the epithelial and dermal cells and their interactions during the embryonic hair generation and adult hair cycling, many researchers have tried to obtain mature hair follicles using different strategies and approaches depending on the causes of hair loss. This review summarizes current advances in the different experimental strategies to regenerate or neogenerate hair follicles, with emphasis on those involving neogenesis of hair follicles in adult individuals using isolated cells and tissue engineering. Most of these experiments were performed using rodent cells, particularly from embryonic or newborn origin. However, no successful strategy to generate human hair follicles from adult cells has yet been reported. This review identifies several issues that should be considered to achieve this objective. Perhaps the most important challenge is to provide threedimensional culture conditions mimicking the structure of living tissue. Improving culture conditions that allow the expansion of specific cells while protecting their inductive properties, as well as methods for selecting populations of epithelial stem cells, should give us the necessary tools to overcome the difficulties that constrain human hair follicle neogenesis. Duplex stainless steels are being used on applications that require,especially,high corrosion resistance and overall good mechanical properties,such as the naval and oil-gas exploration *** components employed in these industries are usually obtained by machining,however,these alloys have low machinability when compared to conventional stainless *** this work,a study of the wear developed when milling duplex stainless-steel,LDX 2101,is going to be presented and evaluated,employing four types of milling tools with different geometries and coatings,while studying the influence of feed rate and cutting length in the wear of these *** used have been provided with two and four flutes,as well as three different coatings,namely:TiAlN,TiAlsiN and *** cutting behavior of these tools was analyzed;data relative to the cutting forces developed during the process were obtained;and roughness measurements of the machined surfaces were *** tools were then submitted to scanning electron microscope(sEM)analysis,enabling the identification of the wear mechanisms that tools were subjected to when machining this material,furthermore,the early stages of these mechanisms were also *** this work was done with the goal of relating the machining parameters and cutting force values obtained,identifying,and discussing the wear patterns that were observed in the coating and tools after the milling tests,providing further information on the machining of these alloys.

First report of L1014F kdr mutation in Culex quinquefasciatus in Mexico

The L1014F mutation in the voltage-sodium channel gene has been associated with resistance to DDT and pyrethroids in various arthropod species including mosquitoes. We determined the frequency of the L1014F kdr mutation in 16 field populations of Culex quinquefasciatus from Northeastern Mexico collected between 2008 and 2013. The L1014F was present in all populations analyzed with the lowest frequency （3.33%） corresponding to the population from Monclova collected in 2012, and the highest frequency （63.63%） from the Monterrey population collected in 2012. The presence of a kdr mutation in populations of Cx. quinquefasciatus from northeastern Mexico provides evidence ofpyrethroid resistance. This requires a special attention, considering that pyrethroid-based insecticides are commonly used in vector-control campaigns, especially against Aedes aegypti （L.）.

Involvement of ethylene receptors in the salt tolerance response of Cucurbita pepo

Abiotic stresses have a negative effect on crop production,affecting both vegetative and reproductive *** plays a relevant role in plant response to environmental stresses,but the specific contribution of ethylene biosynthesis and signalling components in the salt stress response differs between Arabidopsis and rice,the two most studied model *** this paper,we study the effect of three gain-of-function mutations affecting the ethylene receptors CpETR1B,CpETR1A,and CpETR2B of Cucurbita pepo on salt stress response during germination,seedling establishment,and subsequent vegetative growth of *** mutations all reduced ethylene sensitivity,but enhanced salt tolerance,during both germination and vegetative growth,demonstrating that the three ethylene receptors play a positive role in salt *** salt stress,etr1b,etr1a,and etr2b germinate earlier than WT,and the root and shoot growth rates of both seedlings and plants were less affected in mutant than in *** enhanced salt tolerance response of the etr2b plants was associated with a reduced accumulation of Na^(+)in shoots and leaves,as well as with a higher accumulation of compatible solutes,including proline and total carbohydrates,and antioxidant compounds,such as *** membrane monovalent cation transporters,including Na^(+)/H^(+)and K^(+)/H^(+)exchangers(NHXs),K^(+)efflux antiporters(KEAs),high-affinity K^(+)transporters(HKTs),and K^(+)uptake transporters(KUPs)were also highly upregulated by salt in etr2b in comparison with *** aggregate,these data indicate that the enhanced salt tolerance of the mutant is led by the induction of genes that exclude Na+in photosynthetic organs,while maintaining K^(+)/Na^(+)homoeostasis and osmotic *** the salt response of etr mutants occurs via the ethylene signalling pathway,our data show that ethylene is a negative regulator of salt tolerance during germination and vegetative ***,the higher upregulation

It is known that nitrate inhibits ruminal methanogenesis,mainly through competition with hydrogenotrophic methanogens for available hydrogen(H2)and also through toxic effects on the ***,there is limited knowledge about its effects on the others members of ruminal microbiota and their *** this study,we investigated the effects of dietary nitrate inclusion on enteric methane(CH4)emission,temporal changes in ruminal microbiota,and fermentation in Holstein *** animals were maintained in individual pens for 45 *** were randomly allocated to either a control(CTR)or nitrate(NIT,containing 15 g of calcium nitrate/kg dry matter)*** emissions were estimated using the sulfur hexafluoride(sF6)tracer *** microbiota changes and ruminal fermentation were evaluated at 0,4,and 8 h *** this study,feed dry matter intake(DMI)did not differ between dietary treatments(P>0.05).Diets containing NIT reduced CH4 emissions by 27%(g/d)and yield by 21%(g/kg DMI)compared to the CTR(Ps and total volatile fatty acids(VFA)concentration did not differ between dietary treatments(P>0.05)but differed with time,and post-feeding(Pses in the concentrations of ruminal ammonia nitrogen(NH3-N)and acetate were observed,whereas propionate decreased at 4 h post-feeding with the NIT diet(Ps of total bacteria,total methanogens,Ruminococcus albus and Ruminococcus flavefaciens,and the abundance of succiniclasticum,Coprococcus,Treponema,shuttlewortia,succinivibrio,sharpea,Pseudobutyrivibrio,and selenomona(Ps,the population of total fungi,protozoa,Fibrobacter succinogenes,Atopobium and Erysipelotrichaceae L7 AE11 increased(Psion,feeding nitrate reduces enteric CH4 emissions and the methanogens population,whereas it decreases the propionate concentration and the abundance of bacteria involved in the succinate and acrylate pathways.D

Common genetic variations in CLOCK transcription factor are associated with nonalcoholic fatty liver disease

To investigate the role of gene variants and derived haplotypes of the CLOCK transcription factor in nonalcoholic fatty liver disease （NAFID） and their relation with the disease ***： A total of 136 patients with NAFLD and 64 healthy individuals were studied. Liver biopsy was performed in 91 patients. six tag sNPs showing a minor allele frequency 〉 10% （rs1554483 C/G; rs11932595 A/G; rs4580704 C/G; rs6843722 A/C; rs6850524 C/G and rs4864548 A/G） encompassing 117 kb of chromosome 4 and representing 115 polymorphic sites （P 〉 0.8） were genotyped. REsULTs： rs11932595 and rs6843722 showed significant associations with NAFLD （empiric P = 0.0449 and 0.023, respectively）. A significant association was also observed between clinical or histologic spectrum of NAFLD and rs1554483 （empiric P = 0.0399）, rs6843722 （empiric P = 0.0229） and rs6850524 （empiric P = 0.00899） and between fibrosis score and rs1554483 （empiric P = 0.02697）, rs6843722 （empiric P = 0.01898） and rs4864548 （empiric P = 0.02697）. Test of haplotypic association showed that CLOCK gene variant haplotypes frequencies in NAFLD individuals significantly differed from those in controls （empiric P = 0.0097）.CONCLUsION： Our study suggests a potential role of the CLOCK polymorphisms and their haplotypes insusceptibility to NAFLD and disease severity.

Role of sex hormones in gastrointestinal motility in pregnant and non-pregnant rats

AIM: To correlate gastric contractility, gastrointestinal transit, and hormone levels in non-pregnant(estrous cycle) and pregnant rats using noninvasive techniques. METHODs: Female rats(n = 23) were randomly divided into(1) non-pregnant,(contractility, n =6; transit, n = 6); and(2) pregnant(contractility, n = 5; transit, n = 6). In each estrous cycle phase or at 0, 7, 14, and 20 d after the confirmation of pregnancy, gastrointestinal transit was recorded by AC biosusceptometry(ACB), and gastric contractility was recorded by ACB and electromyography. After each recording, blood samples were obtained for progesterone and estradiol determination. REsULTs: In the estrous cycle, despite fluctuations of sex hormone levels, no significant changes in gastrointestinal motility were observed. Days 7 and 14 of pregnancy were characterized by significant changes in the frequency of contractions(3.90 ± 0.42 cpm and 3.60 ± 0.36 cpm vs 4.33 ± 0.25 cpm) and gastric emptying(168 ± 17 min and 165 ± 15 min vs 113 ± 15 min) compared with day 0. On these same days, progesterone levels significantly increased compared with control(54.23 ± 15.14 ng/m L and 129.96 ± 30.52 ng/mL vs 13.25 ± 6.31 ng/mL). On day 14, we observed the highest level of progesterone and the lowest level of estradiol compared with day 0(44.3 ± 15.18 pg/mL vs 24.96 ± 5.96 pg/mL). CONCLUsION: Gastrointestinal motility was unaffected by the estrous cycle. In our data, high progesterone and low estradiol levels can be associated with decreased contraction frequency and slow gastric emptying.

Biomechanics of the Intervertebral Disc in Compression： Experimental and Numerical study

Loading history and age are factors for disc degeneration and disc biomechanics; however, their relationship is unclear. To evaluate disc biomechanics, we conducted an experimental, anatomical and numerical approach to distinguish discs with mild and severe degeneration. In the experimental procedure, 10 cadaveric lumbar discs are tested to static and dynamic compression, and the elastic and viscous moduli and the dynamic parameters are reported. The morphology of disc degeneration is gained with MRI （magnetic resonance imaging） and used to generate a nonlinear finite element model of a degenerated disc, and assisted with the experimental results in order to numerically investigate the distribution of stresses and strains within the disc. The results show a promising methodology for the study of intervertebral disc biomechanics and in general other tissues, organs and medical devices.

Aggressive blood pressure treatment of hypertensive intracerebral hemorrhage may lead to global cerebral hypoperfusion:Case report and imaging perspective

Hypoperfusion injury related to blood pressure decrease in acute hypertensive intracerebral hemorrhage continues to be a controversial topic. Aggressive treatment is provided with the intent to stop the ongoing bleeding. However, there may be additional factors, including autoregulation and increased intracranial pressure, that may limit this approach. We present here a case of acute hypertensive intracerebral hemorrhage, in which aggressive blood pressure management to levels within the normal range led to global cerebral ischemia within multiple border zones. Global cerebral ischemia may be of concern in the management of hypertensive hemorrhage in the presence of premorbid poorly controlled blood pressure and increased intracranial pressure.

Background:In January 2005,Rasulov et *** published"First experience in the use of bone marrow mesenchymal stem cells(MsCs)for the treatment of a patient with deep skin burns".Here,we present the first ever treated patient with cadaveric bone marrow mesenchymal stem cells(CMsCs)in the history of ***:A young man,who severely burned 60%of his total body surface with 30%of full-thickness burns while working with a grass trimmer that exploded,was involved in the *** were obtained from the bone marrow of a cadaver donor in a routine procurement procedure of CUCAIBA,the Province of Buenos Aires,Argentina,Ministry of Health,Transplantation Agency,cultured,expanded,and applied on the burned surfaces using a fibrin spray after early ***:so far,our preliminary experience and our early results have been very impressive showing an outstanding safety data as well as some impressive good results in the use of ***:Based on al this,we think that improvements in the use of stem cells for burns might be possible in the near future and a lot of time as well as many lives could be saved by many other research teams all over the *** will probably be a real scientific opportunity in Regenerative Medicine as well as in Transplantation.
