Heterozygous lipoprotein lipase knockout mice exhibit impaired hematopoietic stem/progenitor cell compartment

Background : Hematopoietic stem cells (HSC) maintain the hematopoietic system homeostasis through self- renewal and multilineage differentiation potential. HSC are regulated by the microenvironment, cytokine signaling, and transcription factors. Recent results have shown that lipid pathways play a key role in the regulation of HSC quiescence, proliferation, and division. However, the mechanism by which lipid metabolism regulates HSC proliferation and differentiation remains to be clarified. Lipoprotein lipase (LPL) is an essential enzyme in the anabolism and catabolism of very low- density lipoprotein, chylomicrons, and triglyceride- rich lipoproteins. Methods : The percentage of hematopoietic stem/progenitor cells and immune cells were determined by fluorescence- activated cell sorting (FACS). The function and the mechanism of HSCs were analyzed by cell colony forming assay and qPCR analysis. The changes in LPL^(+/−) HSC microenvironment were detected by transplantation as- says using red fluorescent protein (RFP) transgenic mice. Results : To explore the function of LPL in HSC regulation, heterozygous LPL- knockout mice (LPL^(+/−)) were established and analyzed by FACS. LPL^(+/−) mice displayed decreased hematopoietic stem/progenitor cell compartments. In vitro single- cell clono- genic assays and cell-cycle assays using FACS promoted the cell cycle and increased proliferation ability. qPCR analysis showed the expression of p57^(KIP2) and p21^(WAF1)/ ^(CIP1) in LPL^(+/−) mice was upregulated. Conclusions : LPL^(+/−) mice exhibited HSC compartment impairment due to promotion of HSC proliferation, without any effects on the bone marrow (BM) microenvironment.

Effectiveness and mechanisms of adiposederived stem cell therapy in animal models of Parkinson’s disease:a systematic review and meta-analysis

Animal models provide an opportunity to assess the optimal treatment way and the underlying mechanisms of direct clinical application of adipose-derived stem cells(ADSCs).Previous studies have evaluated the effects of primitive and induced ADSCs in animal models of Parkinson’s disease(PD).Here,eight databases were systematically searched for studies on the effects and in vivo changes caused by ADSC intervention.Quality assessment was conducted using a 10-item risk of bias tool.For the subsequent meta-analysis,study characteristics were extracted and effect sizes were computed.Ten out of 2324 published articles(n=169 animals)were selected for further meta-analysis.After ADSC therapy,the rotation behavior(10 experiments,n=156 animals)and rotarod performance(3 experiments,n=54 animals)were improved(P<0.00001 and P=0.0003,respectively).The rotation behavior test reflected functional recovery,which may be due to the neurogenesis from neuronally differentiated ADSCs,resulting in a higher pooled effect size of standard mean difference(SMD)(−2.59;95%CI,−3.57 to−1.61)when compared to that of primitive cells(−2.18;95%CI,−3.29 to−1.07).Stratified analyses by different time intervals indicated that ADSC intervention exhibited a long-term effect.Following the transplantation of ADSCs,tyrosine hydroxylase-positive neurons recovered in the lesion area with pooled SMD of 13.36[6.85,19.86].Transplantation of ADSCs is a therapeutic option that shows long-lasting effects in animal models of PD.The potential mechanisms of ADSCs involve neurogenesis and neuroprotective effects.The standardized induction of neural form of transplanted ADSCs can lead to a future application in clinical practice.

Background Alzheimer’s disease is a neurodegenerative disorder.Therapeutically,a transplantation of bone marrow mesenchymal stem cells(BMMSCs)can play a beneficial role in animal models of Alzheimer’s disease.However,the relevant mechanism remains to be fully elucidated.Main body Subsequent to the transplantation of BMMSCs,memory loss and cognitive impairment were significantly improved in animal models with Alzheimer’s disease(AD).Potential mechanisms involved neurogenesis,apoptosis,angiogenesis,inflammation,immunomodulation,etc.The above mechanisms might play different roles at certain stages.It was revealed that the transplantation of BMMSCs could alter some gene levels.Moreover,the differential expression of representative genes was responsible for neuropathological phenotypes in Alzheimer’s disease,which could be used to construct gene-specific patterns.Conclusions Multiple signal pathways involve therapeutic mechanisms by which the transplantation of BMMSCs improves cognitive and behavioral deficits in AD models.Gene expression profile can be utilized to establish statistical regression model for the evaluation of therapeutic effect.The transplantation of autologous BMMSCs maybe a prospective therapy for patients with Alzheimer’s disease.

Pennisetum giganteum z. x. lin selected by the National Juncao Center is a high-yielding and high-quality Juncao variety. It is a high-quality horticultural variety that grows upright, and has tall plants with high yield and high value. Planting should be carried out in the right time to ensure the quality of planting and promote the seedlings to be tidy and strong, and according to the row spacing of 80 cm, plant spacing of 40 cm, 2 plants per hole, about 2 000 holes per 667 m2, with the need for furrowing. At the seedling stage, the soil should be kept moist, and watered in time when it is dry and lacking water. P. giganteum is fertilizer-tolerant and sensitive to fertilizers, and requires a reasonable and economical amount of fertilizer.

Bioinformatics analysis of human kallikrein 5 (KLK5) expression in metaplastic triple‐negative breast cancer

Background:Metaplastic breast carcinoma(MBC)is a rare breast cancer subtype;most cases are triple‐negative breast cancers(TNBCs)and are poorly responsive to conventional systemic therapy.Few potential diagnostic and prognostic markers for distinguishing between metaplastic TNBC and nonmetaplastic TNBC have been discovered.We performed bioinformatic analysis to explore the underlying mechanism by which metaplastic TNBC differs from nonmetaplastic TNBC and provides potential pathogenic genes of metaplastic TNBC.Methods:Differentially expressed genes(DEGs)in metaplastic tumors and nonmetaplastic tumors from TNBC patients were screened using GSE165407.The GSE76275 data set and The Cancer Genome Atlas(TCGA)database were used to screen DEGs in TNBC and non‐TNBC.Metascape and DAVID were used for the Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis and Gene Ontology(GO)analysis of DEGs.Online databases,including UALCAN,GEPIA,HPA,Breast Cancer Gene‐Expression Miner,and quantitative PCR and western blot,were used to examine KLK5 messenger RNA and protein expression in breast cancer.Analysis of KLK5‑associated genes was performed with TCGA data,and the LinkedOmics database was used to detect the genes co‐expressed with KLK5.STRING(Search Tool for the Retrieval of Interacting Genes)and Cytoscape were used to screen for hub genes.Kaplan‑Meier plotter was used for survival analysis.Results:KLK5 was identified among the DEGs in nonmetaplastic TNBC and metaplastic TNBC.The KLK5 gene was overexpressed in nonmetaplastic TNBC but downregulated in metaplastic TNBC.KEGG and GO analyses revealed that epithelial‐to‐mesenchymal transition was a pathogenic mechanism in metaplastic TNBC and an important pathway by which KLK5 and its associated genes DSG1and DSG3 influence metaplastic TNBC progression. 目的探讨基于护理程序的整体护理干预在慢性充血性心力衰竭患者中的应用效果。方法选取2016年8月至2018年8月于胶州市人民医院就诊的72例慢性充血性心力衰竭患者,采用随机数表法分为对照组和观察组,每组36例。对照组患者接受常规护理干预,观察组患者接受基于护理程序的整体护理干预。比较两组护理前后焦虑自评量表(SAS)、抑郁自评量表(SDS)、明尼苏达心力衰竭生活质量量表(MLHFQ)评分及护理工作满意度。结果护理后,两组SAS、SDS评分均低于护理前,观察组SAS、SDS评分均低于对照组,差异有统计学意义(均P<0.05)。护理后,两组患者MLHFQ评分均低于护理前,观察组MLHFQ评分低于对照组,差异有统计学意义(均P<0.05)。观察组护理工作满意度高于对照组,差异有统计学意义(P<0.05)。结论基于护理程序的整体护理干预可有效缓解慢性充血性心力衰竭患者负面情绪,改善其生活质量,提高其对护理工作的满意度。

忆阻器是一种可以通过改变电阻来储存信息或者进行计算的电路元件。但脉冲电路脉宽达到纳秒级较为困难,这限制了忆阻器改变电阻的速度。而脉冲激光器的脉宽很短,很容易达到纳秒级。Cu_(2)Se是典型的在梯度温场作用下空穴和Cu+同时参与定向迁移的热电材料,在无梯度温场时Cu^(+)迁移不可逆,并且热场或梯度温场是比电场、激光、氧化还原反应等所需能量低的能量转换形式,因此Cu_(2)Se有可能成为一种消耗能量更低的可用温差驱动的热电忆阻器材料。本工作模拟建立了用脉冲激光器照射Cu_(2)Se样品后在不同时刻厚度方向上的温度分布模型,根据其热电性能计算了不同位置的温差电势与电场强度,同时制备了Cu_(2)Se热电忆阻器并测定了其电学性质。实验结果表明,厚度为70μm、测试探针间距为1 mm的Cu_(2)Se样品从高阻态转变为低阻态时需要的重置电压为0.47 V,从低阻态转变为高阻态时需要的重置电压为0.40 V,其重置电场强度小于0.5 V/mm,证明了Cu_(2)Se热电材料不需要一个初始高电压来激发其忆阻器效应,给出了热电忆阻器的工作原理。计算结果,表明经脉冲激光作用后Cu_(2)Se样品在厚度方向上的电场强度(50~0.56 V/mm)均大于电阻转变所需要的电场强度(0.5 V/mm),在理论上证明了用激光照射Cu_(2)Se热电材料制作热电忆阻器件的可行性。

目的探究家庭参与式护理干预对慢性心功能不全患者社会支持及生活质量的影响。方法选取2016年8月至2018年8月于胶州市人民医院就诊的102例慢性心功能不全患者,采用随机数表法分为对照组和观察组,每组51例。对照组患者接受常规护理,观察组患者接受家庭参与式护理。比较两组患者护理前后社会支持评定量表(SSRS)、简明健康状况调查量表(SF-36)评分及护理满意度。结果干预后,观察组SSRS、SF-36评分均高于对照组,差异有统计学意义(均P<0.05).对照组和观察组护理满意度分别为72.55%(37/51),94.12%(48/51)。观察组护理满意度高于对照组,差异有统计学意义(P<0.05)。结论家庭参与式护理可有效改善慢性心功能不全患者社会支持度和生活质量,提高患者对护理的满意度。
