Late Permian to Triassic intraplate orogeny of the southern Tianshan and adjacent regions, NW China

The South Tianshan Orogen and adjacent regions of Central Asia are located in the southwestern part of the Central Asian Orogenic Belt. The formation of South Tianshan Orogen was a diachronous, scissors-like process, which took place during the Palaeozoic, and its western segment was accepted as a site of the final collision between the Tarim Craton and the North Asian continent, which occurred in the late Palaeozoic. However, the post-collisional tectonic evolution of the South Tianshan Orogen and adjacent regions remains debatable. Based on previous studies and recent geochronogical data, we suggest that the final collision between the Tarim Craton and the North Asian continent occurred during the late Carboniferous. Therefore, the Permian was a period of intracontinental environment in the southern Tianshan and adjacent regions. We propose that an earlier, small-scale intraplate orogenic stage occurred in late Permian to Triassic time, which was the first intraplate process in the South Tianshan Orogen and adjacent regions. The later large- scale and well-known Neogene to Quaternary intraplate orogeny was induced by the collision between the India subcontinent and the Eurasian plate. The paper presents a new evolutionary model for the South Tianshan Orogen and adjacent regions, which includes seven stages：（I） late Ordovician-early Silurian opening of the South Tianshan Ocean; （11） middle Silurian-middle Devonian subduction of the South Tianshan Ocean beneath an active margin of the North Asian continent; （111） late Devonian-late Carboniferous closure of the South Tianshan Ocean and collision between the Kazakhstan-Yili and Tarirn continental blocks; （IV） early Permian post-collisional magmatism and rifting; （V） late Permian-Triassic the first intraplate orogeny; （Vt） Jurassic-Palaeogene tectonic stagnation and （VII） Neocene-Quaternary intraplate orogeny.

QoE-Driven Social Aware Caching Placement for Terrestrial-Satellite Networks

In remote terrestrial-satellite networks, caching is a very promising technique to alleviate the burden of space cloudlet(e.g., cache-enabled satellite user terminal) and to improve subscribers' quality of experience(Qo E) in terms of buffering delay and achievable video streaming rate. In this paper, we studied a Qo E-driven caching placement optimization problem for video streaming that takes into account the required video streaming rate and the social relationship among users. Social ties between users are used to designate a set of helpers with caching capability, which can cache popular files proactively when the cloudlet is idle. We model the utility function of Qo E as a logarithmic function. Then, the caching placement problem is formulated as an optimization problem to maximize the user's average Qo E subject to the storage capacity constraints of the helpers and the cloudlets. Furthermore, we reformulate the problem into a monotone submodular optimization problem with a partition matroid constraint, and an efficient greedy algorithm with 1-1 e approximation ratio is proposed to solve it. Simulation results show that the proposed caching placement approach significantly outperforms the traditional approaches in terms of Qo E, while yields about the same delay and hit ratio performance compare to the delay-minimized scheme.

Highly efficient CRISPR-SaKKH tools for plant multiplex cytosine base editing

Base editing, as an expanded clustered regularly interspaced short palindromic repeats(CRISPR)-Cas genome editing strategy, permits precise and irreversible nucleotide conversion. SaKKH, an efficient variant of a Cas9 ortholog from Staphylococcus aureus(SaCas9), is important in genome editing because it can edit sites with HHHAAT protospacer adjacent motif(PAM) that the canonical Streptococcus pyogenes Cas9(SpCas9) or its variants(e.g. xCas9, Cas9-NG) cannot. However, several technical parameters of SaKKH involved base editors have not been well defined and this uncertainty limits their application. We developed an effective multiplex cytosine base editor(SaKKHn-pBE) and showed that it recognized NNARRT, NNCRRT, NNGRGT, and NNTRGT PAMs. Based on 27 targets tested, we defined technical parameters of SaKKHn-pBE including the editing window, the preferred sequence context, and the mutation type. The editing efficiency was further improved by modification of the SaKKH sgRNA. These advances can be applied in future research and molecular breeding in rice and other plants.

目的运用改良序贯法测定肥胖产妇分娩镇痛中罗哌卡因的半数有效浓度(EC_(50))。方法26例拟行分娩镇痛的产妇,BMI均≥30kg/m^(2)。罗哌卡因初始浓度为0.1%,若镇痛有效,则下一例降低罗哌卡因浓度;反之则增加浓度。罗哌卡因浓度梯度为0.01%,直至出现镇痛有效和无效交替波形成7个拐点终止研究。分娩镇痛有效标准:给予罗哌卡因首次剂量30min,产妇VAS疼痛评分≤3分。运用Probit回归分析计算肥胖产妇分娩镇痛中罗哌卡因的EC_(50)和95%有效浓度(EC_(95))及其对应95%CI。记录分娩镇痛前、镇痛后10min和30min时的VAS疼痛评分,记录皮肤瘙痒、产间发热、恶心呕吐和尿潴留等不良反应发生情况。结果肥胖产妇分娩镇痛中罗哌卡因的EC_(50)及其95%CI为0.064%(0.057%~0.070%),EC_(95)及其95%CI为0.076%(0.070%~0.106%)。1例产妇出现产间发热,所有产妇未出现皮肤瘙痒、恶心呕吐及尿潴留等不良反应。结论肥胖产妇单纯罗哌卡因分娩镇痛的EC_(50)为0.064%。

High oxidative stress injury and bacterial infection are the main challenges that impair wound healing in diabetic ***,a hydrogel with enhanced antimicrobial and antioxidant properties was developed for rapid healing of diabetic *** this study,chitosan methacrylate-gallic acid(CSMA-GA)polymer with antioxidant activity,antimicrobial activity,and ultraviolet(UV)-triggered gelling properties was developed as a hydrogel ***,amphiphilic Pluronic F127 molecules were used to load hydrophobic chlorhexidine drug molecules to obtain F127/chlorhexidine nanoparticle(NP)with strong antibacterial ***,F127/chlorhexidine NPs were encapsulated in CSMA-GA hydrogel to further enhance its antibacterial *** hybrid hydrogel platform(CSMA-GA/F127/chlorhexidine(CMGFC))exhibited high antibacterial efficiency(>99.9%)and strong reactive oxygen species(ROS)scavenging ability(>80.0%),which effectively protected cells from external oxidative stress(upregulated superoxide dismutase(SOD)and glutathione/oxidized glutathione disulfide(GSH/GSSG)levels and downregulated malondialdehyde(MDA)levels).Moreover,in vivo results proved that the CMGFC hydrogel significantly reduced inflammatory responses(downregulated interleukin-6(IL-6)and upregulated interleukin-10(IL-10)levels),promoted angiogenesis(upregulated vascular endothelial growth factor(VEGF)and platelet endothelial cell adhesion molecule-1(CD 31)levels),and wound healing(enhanced collagen deposition and tissue remodelling).Overall,the CMGFC hydrogel with enhanced antimicrobial and antioxidant properties demonstrated significant potential to enhance diabetic wound healing.

Enhanced diabetic wound repair remained a global ***,we reported a novel hydrogel with glucose-responsive hyperglycemia regulation and antioxidant activity for enhanced diabetic wound *** this study,gallic acid(GA)with strong antioxidant activity was grafted onto chitosan(CS)chains by one-step synthesis,and then incorporated into poly(ethylene glycol)diacrylate(PEG-DA)hydrogel matrix to obtain a novel antioxidant hybrid hydrogel(PEG-DA/CS-GA).Meanwhile,polyethyleneimine(PEI)was modified with a unique glucose-sensitive phenylboronic acid(PBA)molecule to load insulin(PEIPBA/insulin nano-particles,PEI-PBA/insulin NPs),which could be immobilized in the PEG-DA/CS-GA hybrid hydrogel by the formation of dynamic borate bond between the phenylboronic acid groups on the PEI-PBA and the polyphenol groups on the *** results indicated that the PEG-DA/PEI-PBA/insulin/CS-GA(PPIC)hydrogel platform not only had remarkable biocompatibility,but also displayed extraordinary antioxidant properties(DPPH scavenging rate>95.0%),and effectively protected cells from oxidative damage(decreased MDA levels,increased Superoxide dismutase(SOD)levels and stable GSH/GSSG levels).Meanwhile,the PPIC hydrogel also exhibited unique glucose-responsive insulin release characteristics,and effectively regulated the blood glucose *** in vitro and in vivo results demonstrated that our PPIC hydrogel could promoted angiogenesis(increased VEGF and CD 31 expression),reshaped the inflammatory microenvironment(decreased IL-6 and increased IL-10 level),and achieved wound closure within 20 *** these results strongly indicated that the PPIC hydrogel represented a tough and efficient platform for diabetic wound treatment.

Stem cells are defined by their capacity for both self-renewal and directed differentiation; thus, they represent great promise for regenerative medicine. Historically, stem cells have been categorized as either embryonic stem cells （ESCs） or adult stem cells （ASCs）. It was previously believed that only ESCs hold the ability to differentiate into any cell type, whereas ASCs have the capacity to give rise only to cells of a given germ layer. More recently, however, numerous studies demonstrated the ability of ASCs to differentiate into cell types beyond their tissue origin. The aim of this review was to summarize contemporary evidence regarding stem cell availability, differentiation, and more specifically, the potential of these cells in the diagnosis and treatment of erectile dysfunction （ED） in both animal models and human research. We performed a search on PubMed for articles related to definition, iocalisation and circulation of stem cells as well as the application of stem cells in both diagnosis and treatment of ED. Strong evidence supports the concept that stem cell therapy is potentially the next therapeutic approach for ED. To date, a large spectrum of stem cells, including bone marrow mesenchymal stem cells, adipose tissue-derived stem cells and muscle-derived stem cells, have been investigated for neural, vascular, endothelial or smooth muscle regeneration in animal models for ED. In addition, several subtypes of ASCs are localized in the penis, and circulating endogenous stem cells can be employed to predict the outcome of ED and ED-related cardiovascular diseases.

Low-intensity pulsed ultrasound stimulates proliferation of stem/progenitor cells: what we need to know to translate basic science research into clinical applications

Low-intensity pulsed ultrasound (LIPUS) is a promising therapy that has been increasingly explored in basic research and clinical applications. LIPUS is an appealing therapeutic option as it is a noninvasive treatment that has many advantages, including no risk of infection or tissue damage and no known adverse reactions. LIPUS has been shown to have many benefits including promotion of tissue healing, angiogenesis, and tissue regeneration;inhibition of inflammation and pain relief;and stimulation of cell proliferation and differentiation. The biophysical mechanisms of LIPUS remain unclear and the studies are ongoing. In recent years, more and more research has focused on the relationship between LIPUS and stem/progenitor cells. A comprehensive search of the PubMed and Embase databases to July 2020 was performed. LIPUS has many effects on stem cells. Studies show that LIPUS can stimulate stem cells in vitro;promote stem cell proliferation, differentiation, and migration;maintain stem cell activity;alleviate the problems of insufficient seed cell source, differentiation, and maturation;and circumvent the low efficiency of stem cell transplantation. The mechanisms involved in the effects of LIPUS are not fully understood, but the effects demonstrated in studies thus far have been favorable. Much additional research is needed before LIPUS can progress from basic science research to large-scale clinical dissemination and application.
