purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RpE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from polycaprolactone (pCL), and different concentrations of gelatin (5%, 15% and 30%). ARpE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARpE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARpE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARpE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARpE-19 (RpE65 and ZO-1) qRT-pCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of p 0.05 was considered to be statistically significant. Results ARpE-19 cells grew on pCL/Gelatin membranes mainly in pCL/30% gelatin combination, which had not cytotoxic effect. RpE65/cytokeratin-18/ and actin-positive ARpE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARpE-19 cells fused among, suggesting the presence of tight junctions. The expression of RpE65 and ZO-1 was unchanged. Conclusions: pCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RpE-cells and exhibited RpE-like morphology. Quality evaluation of Nothofagus pumilio seeds linked to forest management and climatic events

Background Forest ecosystems undergo significant transformations due to harvesting and climate fluctuations, emphasizing the critical role of seeding in natural regeneration and long-term structural preservation. Climate change further amplifies these dynamics, affecting phenology across species and regions. In Tierra del Fuego(Argentina), Nothofagus pumilio(lenga) forests represent the most important timber resource, and it is managed through different silvicultural strategies. This species demonstrates notable post-disturbance regeneration, yet seed fall exhibits signifi-cant variability, leading to variations in seed quality(e.g., viability). This study aims to assess fluctuations in N. pumilio seed quality, determine how it varies concerning forest management strategies, annual productivity, and the co-occurrence of climatic phenomena including El Ni?o-Southern Oscillation(ENSO) and the Southern Annular Mode(SAM).Results Viable seeds represented 18.4% of the total, notably higher in unharvested than in managed areas. Con-versely, empty seeds were more prevalent in harvested areas(> 75%). Seed quality exhibited significant differences across silvicultural treatments, except for insect-predated seeds, which had similar proportions across all areas, though dispersed retention showed higher predation. When considering years with varying production levels, high-production years favoured full and viable seeds, particularly in unharvested forests and aggregated retention, while low-production years saw reduced viability across all treatments. Quadratic models revealed that viability increased with seed production, where unharvested forests achieved the highest values. Climate variability influenced seed proportions, where ENSO+/SAM+ promoting more full and viable seeds, while ENSO–/SAM+ favoured non-predated seeds, especially in unharvested *** Seed quality varies among treatments and years with different levels of seeding. Influence of steel jacket thickness on the RC bridges' seismic vulnerability

The seismic demands of bridges subjected to strong earthquakes are mostly concentrated on piers. The main objective of this study is to assess the seismic vulnerability of retrofitted bridges with steel jackets. It analyzes the influence of the steel jacket thickness on the seismic capacity and demand of bridges. The bridges in study have two simple supported spans with superstructure composed by AASHTO type girders and a substructure formed with one circular pier over a stiff foundation. They are located on the pacific coast of Mexico that is a zone of high seismicity. The bridges seismic capacity are evaluated through nonlinear static analyses and the shear force and displacement demands were determined using capacity spectra with a group of 10 real accelerograms from a subduction source. The response spectra were scaled to several peak ground accelerations(pGA) with the aim to estimate four damage scenarios linked each one to a probability of exceedance in a specific period, for return periods in the range of 72-3500 years that represent service level and maximum considered earthquake. Results show that the confining effect provided by the steel jackets substantially improves the seismic performance of the bridges piers. The increase of jacket thickness guides to important increments on columns base shear capacity, and the influence of the steel jacket thickness depends on the limit state and bridge slenderness ratio.

Evaluation of the Impact of Driver Behavior on Back of Queues Events in Work Zones Using the SHRp2 Naturalistic Driving Study Data

The SHRp2 Naturalistic Driving Study was used to evaluate the impact of various work zone and driver characteristics on back of queue safety critical events (crash, near-crash, or conflicts) The model included 43 SCE and 209 “normal” events which were used as controls. The traces included representing 209 unique drivers. A Mixed-Effects Logistic Regression model was developed with probability of a SCE as the response variable and driver and work zone characteristics as predictor variables. The final model indicated glances over 1 second away from the driving task and following closely increased risk of an SCE by 3.8 times and 2.9 times, respectively. Average speed was negatively correlated to crash risk. This is counterintuitive since in most cases, it is expected that higher speeds are related to back of queue crashes. However, most queues form under congested conditions. As a result, vehicles encountering a back of queue would be more likely to be traveling at lower speeds.
