体育外交研究的纵深化趋势呼唤跨学科研究方法以推动理论的破壁。运用国际政治信号理论对冷战时期美苏围绕田径对抗赛的外交互动进行实证分析发现,美苏田径对抗赛是冷战时期两国关键的政治协商渠道,通过多轮次体育外交信号的编码、传递、解码与反馈,促成了美苏首份文化交流协定并推动两国全面合作,构建冷战中难能可贵的良性竞合关系。基于国际政治信号理论提出:体育外交促进国家间关系呈现特定效果范围,具体指向外交信息的传递、战略诚意的表达和安全困境的化解。体育外交作用机理能够提供意义共有的外交信号编码方式、构建安全可靠的外交信号交换空间和赋予生动有效外交信号转换能力。

六君子汤加味灌肠对肥胖大鼠肠道菌群影响实验研究

目的探讨六君子汤加味灌肠法对肥胖大鼠肠道菌群的影响。方法将40只雄性SD大鼠按体质量随机分为4组,分别为正常组、模型组、菌液灌肠组、六君子汤加味灌肠组,每组10只。除正常组外,其他组采用高脂饲料喂养复制肥胖大鼠模型,造模成功后,给予菌液灌肠组粪菌液2 mL灌肠,六君子汤加味灌肠组给予六君子汤加味药液2 mL灌肠,正常组与模型组给予0.9%氯化钠溶液2 mL灌肠。灌肠操作1周3次,隔天1次,连续3周。实验结束后,称重检测大鼠体质量,生物化学法检测大鼠血清中甘油三酯含量,并收集各组大鼠粪便进行16SrRNA高通量测序。结果与正常组比较,模型组大鼠体质量显著升高(P<0.01);与模型组比较,菌液灌肠组与六君子汤加味灌肠组体质量均有明显下降(P<0.01)。与正常组相比,模型组和六君子汤加味灌肠组大鼠血清甘油三酯水平升高(P<0.01);与模型组比较,菌液灌肠组大鼠血清甘油三酯水平下降(P<0.05);六君子汤加味灌肠组较菌液灌肠组大鼠血清甘油三酯水平升高(P<0.05)。高脂饮食诱导的肥胖大鼠存在肠道菌群失调,其肠道菌群物种丰富度与多样性均出现上升。六君子汤加味灌肠可上调肥胖大鼠的迷踪菌门、迷踪菌纲、氨基酸球菌科、产碱杆菌科、红螺菌科、肠杆菌科、迷踪菌科、琥珀酸弧菌科、未分类的脱硫弧菌科、蛭弧菌科、毛螺菌科、脱硫弧菌属、粪芽胞菌属、变形杆菌属、考拉杆菌属、寡源杆菌属、未分类的红螺菌属、镰刀菌属、丁酸球菌属的丰度;下调肥胖大鼠的ε变形菌纲、巴斯德氏菌科、消化链球菌科、螺杆菌科、未分类的根瘤菌科、梭菌科1、链球菌科、罗姆布茨菌属、幽门螺杆菌属、狭义梭菌属、罗氏菌属、链球菌属、苏黎世杆菌属、韦荣氏球菌属的丰度。结论菌液灌肠与六君子汤加味灌肠均能在一定程度上减缓高脂饮食诱导的肥胖大鼠体质量增加。肥胖大鼠出现肠道菌群失调,六君子汤加味灌肠可帮助肥胖大鼠重构体内肠道菌群组成,上调有益菌,下调致病菌丰度以达到新的平衡从而减缓肥胖发展进程。提示六君子汤加味灌肠可以调节肥胖大鼠肠道菌群紊乱。

调相机外冷水系统结垢分析及改进研究

某±800kV换流站300Mvar调相机组年度检修时,发现外冷水系统出现严重结垢、冷却器堵塞、冷却水池藻类滋生等现象,影响设备安全运行。通过对外冷水、补充水(城市自来水)的水质进行检测,并对外冷水所用的阻垢缓蚀剂性能进行评价,发现外冷水和补充水的硬度和碱度偏高,阻垢缓蚀剂与外冷水水质不匹配,抑制结垢效果不佳,存在明显结垢倾向,同时杀菌灭藻剂的加药方式不合理造成微生物产生抗药性,导致藻类微生物滋生严重。通过外冷水阻垢缓蚀剂筛选和性能评价,筛选出高效阻垢缓蚀剂,并应用于外冷水的处理,同时改变杀菌灭藻剂的加药方式后,外冷水系统水质大幅提升,结垢、腐蚀和藻类滋生情况大幅改善。进而提出外冷水系统的深度处理措施,进一步提高外冷水水质,减少药剂的使用量。

tsRNAs(tRNA-derived small RNAs),as products of the stress response,exert considerable influence on stress response and injury regulation.However,it remains largely unclear whether tsRNAs can ameliorate liver injury.Here,we demonstrate the roles of tsRNAs in alleviating liver injury by utilizing the loss of NSun2(NOP2/Sun domain family,member 2)as a tsRNAs-generating model.Mechanistically,the loss of NSun2 reduces methyluridine-U5(m^(5)U)and cytosine-C5(m^(5)C)of tRNAs,followed by the production of various tsRNAs,especially Class I tsRNAs(tRF-1s).Through further screening,we show that tRF-Gln-CTG-026(tG026),the optimal tRF-1,ameliorates liver injury by repressing global protein synthesis through the weakened association between TSR1(pre-rRNA-processing protein TSR1 homolog)and pre-40S ribosome.This study indicates the potential of tsRNA-reduced global protein synthesis in liver injury and repair,suggesting a potential therapeutic strategy for liver injury.

Epigenetic alterations and metabolic dysfunction are two hallmarks of aging.However,the mechanism of how their interaction regulates aging,particularly in mammals,remains largely unknown.Here we show ELOVL fatty acid elongase 2(Elovl2),a gene whose epigenetic alterations are most highly correlated with age prediction,contributes to aging by regulating lipid metabolism.We applied artificial intelligence to predict the protein structure of ELOVL2 and the interaction with its substrate.Impaired Elovl2 function disturbs lipid synthesis with increased endoplasmic reticulum stress and mitochondrial dysfunction,leading to key aging phenotypes at both cellular and physiological level.Furthermore,restoration of mitochondrial activity can rescue age-related macular degeneration(AMD)phenotypes induced by Elovl2 deficiency in human retinal pigmental epithelial(RPE)cells;this indicates a conservative mechanism in both human and mouse.Taken together,we revealed an epigenetic-metabolism axis contributing to aging and illustrate the power of an AI-based approach in structure-function studies.

目的：建立牵牛子配方颗粒中多指标成分的含量测定方法。方法：采用HPLC结合一测多评法同时测定绿原酸等酚酸类成分的含量。色谱条件为采用Agilent ZORBAX SB-C18(4.6 mm×250 mm,5.0μm)，流动相为乙腈-0.1%磷酸，检测波长为326 nm，流速为1.0 mL·min-1，柱温为30℃，进行梯度洗脱。建立一测多评法，以咖啡酸为内参物测定牵牛子配方颗粒中酚酸类成分的含量。同时采用外标法验证本研究建立的测定方法测定结果的准确性。结果：在各自线性范围内，新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C的相对校正因子分别为1.841、1.718、1.885、1.485、1.268、1.429；一测多评法与外标法测定结果差异不显著。结论：建立的一测多评法可准确地检测牵牛子配方颗粒酚酸类多指标成分含量，作用于牵牛子配方颗粒的质量控制。

Blastocyst complementation by pluripotent stem cell(PSC)injection is believed to be the most promising method to generate xenogeneic organs.However,ethical issues prevent the study of human chimeras in the late embryonic stage of development.Primate embryonic stem cells(ESCs),which have similar pluripotency to human ESCs,are a good model for studying interspecies chimerism and organ generation.However,whether primate ESCs can be used in xenogenous grafts remains unclear.In this study,we evaluated the chimeric ability of cynomolgus monkey(Macaca fascicularis)ESCs(cmESCs)in pigs,which are excellent hosts because of their many similarities to humans.We report an optimized culture medium that enhanced the anti-apoptotic ability of cmESCs and improved the development of chimeric embryos,in which domesticated cmESCs(D-ESCs)injected into pig blastocysts differentiated into cells of all three germ layers.In addition,we obtained two neonatal interspecies chimeras,in which we observed tissue-specific D-ESC differentiation.Taken together,the results demonstrate the capability of D-ESCs to integrate and differentiate into functional cells in a porcine model,with a chimeric ratio of 0.001-0.0001 in different neonate tissues.We believe this work will facilitate future developments in xenogeneic organogenesis,bringing us one step closer to producing tissue-specific functional cells and organs in a large animal model through interspecies blastocyst complementation.
