Infiltration by monocytes of the central nervous system and its role in multiple sclerosis: reflections on therapeutic strategies

Mononuclear macrophage infiltration in the central nervous system is a prominent feature of neuroinflammation. Recent studies on the pathogenesis and progression of multiple sclerosis have highlighted the multiple roles of mononuclear macrophages in the neuroinflammatory process. Monocytes play a significant role in neuroinflammation, and managing neuroinflammation by manipulating peripheral monocytes stands out as an effective strategy for the treatment of multiple sclerosis, leading to improved patient outcomes. This review outlines the steps involved in the entry of myeloid monocytes into the central nervous system that are targets for effective intervention: the activation of bone marrow hematopoiesis, migration of monocytes in the blood, and penetration of the blood–brain barrier by monocytes. Finally, we summarize the different monocyte subpopulations and their effects on the central nervous system based on phenotypic differences. As activated microglia resemble monocyte-derived macrophages, it is important to accurately identify the role of monocyte-derived macrophages in disease. Depending on the roles played by monocyte-derived macrophages at different stages of the disease, several of these processes can be interrupted to limit neuroinflammation and improve patient prognosis. Here, we discuss possible strategies to target monocytes in neurological diseases, focusing on three key aspects of monocyte infiltration into the central nervous system, to provide new ideas for the treatment of neurodegenerative diseases.

The impact of temperature on the evolution and self-deflection of spatial optical solitons in photovoltaic photorefractive media

The temperature effects on the evolution and self-deflection of bright spatial optical solitons in photo-voltaic photorefractive media were investigated by taking into account diffusion effects. The numerical results show that the evolution of the bright solitary beam depends strongly on the crystal temperature. It is also found that the bending distance of the bright solitary beam centre increases and reaches its maximum value at a characteristic temperature, and then decreases as temperature rises and ap-proaches zero at low and high temperatures. Both the maximum value and characteristic temperature increase with the input power density. The self-deflection of bright solitary beam is further studied by a perturbation technique, and the results are found to be in good agreement with that obtained by the numerical method. The diffusion process and the dark irradiance dominate the temperature depend-ence of bending distance in most values of temperature besides at the characteristic temperature and in the higher temperature regime. The diffusion process will mainly dominate the temperature dependence at the characteristic temperature and the dark irradiance will dominate in the higher tem-perature range.

Artificial biomaterials with dynamic mechano-responsive behaviors similar to those of biological tissues have been drawing great *** this study,we report a TiO_(2)-based nanowire(TiO_(2)NWs)scaffolds,which exhibit dynamic mechano-responsive behaviors varying with the number and amplitude of nano-deformation *** is found that the elastic and adhesive forces in the TiO_(2)NWs scaffolds can increase significantly after multiple cycles of *** nanofriction experiments show the triboelectric effect of increasing elastic and adhesive forces during the nano-deformation cycles of TiO_(2)NWs *** properties allow the TiO_(2)NW scaffolds to be designed and applied as intelligent artificial biomaterials to simulate biological tissues in the future.

Temperature effects of dark solitons on the self-deflection of bright solitons in a separate bright-dark soliton pair

Based on the theory of one-dimensional separate soliton pairs formed in a serial photorefractive crystal circuit, we investigated the temperature effects of the dark soliton on the self-deflection of the bright soliton in a bright-dark soliton pair. The numerical results obtained by solving the nonlinear propaga-tion equation showed that the bright soliton moves on a parabolic trajectory in the crystal and its spa-tial shift changed with the temperature of the dark soliton. The higher the temperature of the dark soli-ton was, the smaller the spatial shift of the bright soliton was. The self-bending process was further studied by the perturbation technique, and the results were found to be in good agreement with that obtained by the numerical method.

Analysis of 100 consecutive cases of resectable pancreatic neuroendocrine neoplasms： clinicopathological characteristics and long-term outcomes

The incidence rate of pancreatic neuroendocrine neoplasms （pNENs） has increased rapidly in recent years. However, the clinicopathological characteristics of pNENs are poorly understood. Medical records of patients who underwent surgery and were confirmed as pNENs by pathological examination from January 2003 to February 2015 in Qilu Hospital were reviewed retrospectively. A total of 100 patients, 36 males and 64 females, were included with a mean operation age of 46.26 ± 13.41 years. Among the 100 cases, 76 had insulinomas and 24 had non-functional pNENs. Tumor size ranged from 0.5 cm to 9 cm, and the mean size was 2.20 ± 1.40 cm. The percentages of TNM stages I, II, III, and IV tumors were 89.0%, 8.0%, 0.0%, and 3.0%, respectively. Based on the WHO classification, pNENs were classified into three grades： G1, G2, and G3. G1, G2, and G3 tumors were confirmed in 72.9%, 23.7%, and 3.4% patients, respectively. The positive rates of CgA and Syn immunohisto- chemical staining were 94.5% （69/73） and 100% （74/74）, respectively. Compared with insulinoma, non-functional pNENs have larger tumor sizes, more advanced TNM staging, a higher Ki-67 index, and a higher rate of liver metastasis （P 〈 0.05）. In conclusion, pNENs are heterogeneous tumors with varying clinical manifestations, diverse tumor biological characteristics, and different prognoses. Non-functional pNENs present a more aggressive behavioral model and have poorer prognosis than functional pNENs.

Adding PD-1 blockade in the neoadjuvant regimens for locally advanced rectal cancer(LARC)patients with microsatellite stable(MsS)/mismatch repair-proficient(pMMR)tumors is an attractive,but debatable *** phase 2,multicenter,prospective,single-arm study enrolled patients from 6 centers from June 2021 to November *** advanced rectal cancer(LARC,cT_(3-4a)N_(0)M_(0) and cT_(1-4a)N_(1-2)M_(0))patients aged≥18 years with the distance from distal border of tumor to anal verge≤10 cm(identified by Magnetic Resonance Imaging)were qualifed for *** patients received long-course radiotherapy(50 Gy/25 fractions,2 Gy/fraction,5 days/week)and three 21-day cycles capecitabine(850-1000 mg/m2,bid,po,day1-14)and three 21-day cycles tislelizumab(200 mg,***,day8)as *** mesorectal excision(TME)was 6-12 weeks after the end of radiotherapy to achieve radical resection.A total of 50 patients were enrolled in this *** pathological complete response rate was 40.0%[20/50,95%confidence interval(CI):27.61-53.82%],while 15(30.0%,95%CI:19.1-43.75%),9(18.0%,95%CI:9.77-30.8%),2(4.0%,95%CI:1.10-13.46%)patients respectively achieved grade 1,2,and 3 tumor ***-related adverse events(TRAEs)occurred in 28(56.0%)LARC patients,including 26(52.0%)with grade I-II and 2(4.0%)with grade II(1 with grade 3 immune-related colitis and 1 with grade 3 rash).PD-1 blockade plus long-course chemoradiotherapy(CRT)showed promising therapeutic effects according to pathological complete response rate and is well-tolerated in LARC patients.A larger randomized controlled study is desired to further validate the above findings.

Background Bariatric surgery offers a productive resolution of type 2 diabetes mellitus （T2DM）.The development of T2DM vasculopathy is due to chronic inflammation,which increases matrix metalloproteinase-9 （MMP-9） and tissue inhibitor of matrix metalloproteinase-1 （TIMP-1） *** study sought to examine MMP-9 and TIMP-1 expression in the thoracic aorta after duodenal-jejunal bypass （DJB） surgery on a T2DM rat model induced by a high-fat diet and low dose streptozotocin （STZ）.Methods Twenty-one T2DM Wistar rats induced by high-fat diet and low dose STZ were randomly divided into DJB and sham duodenal-jejunal bypass （S-DJB） *** Wistar rats were fed a normal diet as a *** of gastrointestinal function post-operation and resumption of a normal diet completed the *** weight,blood glucose,blood lipid levels,and MMP-9 and TIMP-1 expression levels in aortic endothelial cells were measured *** DJB rats showed significant weight loss 2 weeks post-operation compared with S-DJB *** surgery,DJB rats showed significant improvement and steady glycemic control with improved insulin sensitivity and glucose *** also exhibited improved lipid metabolism with a decrease in fasting free fatty acids （FFAs） and triglycerides （all P ＜0.05）.Immunohistochemistry showed decreased MMP-9 and TIMP-1 expression 12 weeks after surgery （P ＜ 0.01）.Conclusions DJB surgery on an induced T2DM rat model improves blood glucose levels and lipids,following a high-fat diet and low dose STZ *** addition,DJB decreased MMP-9 and TIMP-1 expression in vascular endothelial cells,which may play an important role in delaying the development of T2DM vascular disease.

Effects of system parameters on bright dissipative holographic photovoltaic solitons

In a photovoltaic dissipative system, evolution and stability of a bright dissipative holographic photo- voltaic (DHP) soliton are completely determined by the given dissipative system, i.e., they are impacted by the system parameters, such as the system temperature, the photovoltaic field of the crystal, the angle between two input beams, and the angle of polarized direction. Such DHP solitons can be easily amplified or absorbed by adjusting these system parameters. They have potential applications in op- tical switches or repeaters.

Growth, structure and upconversion mechanisms of Ba_2ErCl_7 crystal

The methods to grow a novel upconversion crystal Ba2ErCI7 are reported in this note. Ba2ErCI7 material was synthesized with Er2O3, BaCI2-2H2O and NH4CI by using a so-called "direct synthesis method". High quality Ba2ErCI7 single crystals with size up to 4 mm×8 mm×15 mm were grown by using the Czochralski and the Bridgman methods. Intense yellow-green luminescence was detected when the crystal was pumped by an 803 laser diode (LD). Transmittance spectrum was measured by using HITACH U-3500 spectrophotometer at room temperature for the first time, in which one can see that the transmittance is high in the range from 350 to 1 600 nm and the cut-off wavelength of the crystal was 230 nm. The upconversion mechanisms were discussed.
