ZNF554 Inhibits Endometrial Cancer Progression via Regulating RBM5 and Inactivating WNT/β-Catenin Signaling Pathway

BACKGROUND Thiopurine-induced leucopenia significantly hinders the wide application of *** optimization guided by nudix hydrolase 15(NUDT15)has significantly reduced the early leucopenia rate,but there are no definitive biomarkers for late risk leucopenia *** To determine the predictive value of early monitoring of DNA-thioguanine(DNATG)or 6-thioguanine nucleotides(6TGN)for late leucopenia under a NUDT15-guided thiopurine dosing strategy in patients with Crohn’s disease(CD).METHODS Blood samples were collected within two months after thiopurine initiation for detection of metabolite *** leucopenia was defined as a leukocyte count<3.5×10^(9)/L over two *** Of 148 patients studied,late leucopenia was observed in 15.6%(17/109)of NUDT15/thiopurine methyltransferase(TPMT)normal and 64.1%(25/39)of intermediate *** patients suffering late leucopenia,early DNATG levels were significantly higher than in those who did not develop late leucopenia(P=4.9×10^(-13)).The DNATG threshold of 319.43 fmol/μg DNA could predict late leucopenia in the entire sample with an area under the curve(AUC)of 0.855(sensitivity 83%,specificity 81%),and in NUDT15/TPMT normal metabolizers,the predictive performance of a threshold of 315.72 fmol/μg DNA was much more remarkable with an AUC of 0.902(sensitivity 88%,specificity 85%).6TGN had a relatively poor correlation with late leucopenia whether in the entire sample(P=0.021)or NUDT15/TPMT normal or intermediate metabolizers(P=0.018,P=0.55,respectively).CONCLUSION Proactive therapeutic drug monitoring of DNATG could be an effective strategy to prevent late leucopenia in both NUDT15/TPMT normal and intermediate metabolizers with CD,especially the former.

Regulating the key performance parameters for Hg-based IR NLO chalcogenides via bandgap engineering strategy

Recently, non-centrosymmetric（NCS） Hg-based chalcogenides have garnered significant interest due to their strong second-harmonic-generation intensities(deff), making them attractive candidates for infrared nonlinear optical（IR-NLO） application. However, achieving both wide band gaps（Eg） and large phasematched deffsimultaneously in these materials remains a challenge due to their inherent constraints on each other. In this research, we have successfully obtained two quaternary NCS Hg-based chalcogenides, Rb2HgGe3S8and Cs2HgGe3S8, by implementing a bandgap engineering strategy that involves alkali metal introduction and Hg/Ge ratio regulation. Both compounds consist of 2D [Hg Ge3S8]2–anionic layers made of 1D [HgGeS6]6–chains and dimeric [Ge2S6]4–polyhedra arranged alternately, and the charge-balanced Rb+/Cs+cations located between these layers. Remarkably, Rb2HgGe3S8and Cs2HgGe3S8exhibit overall properties required for promising IR-NLO materials, including sufficient PM deff（0.55–0.70 × AgGaS2@2050 nm）, large Eg（3.27–3.41 e V）, giant laser-induced damage thresholds（17.4–19.7 ×AgGaS2@1064 nm）, broad optical transmission intervals（0.32–17.5 μm）, and suitable theoretical birefringence（0.069–0.086@2050 nm）. Furthermore, in-depth theoretical analysis reveals that the exceptional IRNLO performance is attributed to the synergy effects of distorted [HgS4] and [GeS4] tetrahedra. Our study provides a useful strategy for enhancing the Egand advancing Hg-based IR-NLO materials, which is expected to extended and implemented in other chalcogenide systems.

Single atom(SA)catalysts have achieved great success on highly selective heterogeneous catalysis due to their abundant and homogeneous active *** electronic structures of these active sites,restrained by their localized coordination environments,significantly determine their catalytic performances,which are difficult to ***,we investigated the effect of localized surface plasmon resonance(LSPR)on engineering the electronic structures of single atomic ***,core–shell structures consisted of Au core and transition metal SAs loaded N-doped carbon(CN)shell were constructed,namely Au@M-SA/CN(M=Ni,Fe,and Co).It was demonstrated that plasmon-induced hot electrons originated from Au were directionally injected to the M-SAs under visible light irradiation,which significantly changed their electronic structures and meanwhile facilitated improved overall charge separation *** as-prepared Au@Ni-SA/CN exhibited highly efficient and selective photocatalytic CO_(2) reduction to CO performance,which is 20.8,17.5,and 6.9 times those of Au nanoparticles,Au@CN,and Ni-SA/CN,*** spectroscopy analysis and theoretical calculations confirmed that the plasmon enhanced Ni-SA/CN sites featured increased charge density for efficient intermediate activation,contributing to the superb photocatalytic *** work provides a new insight on plasmon and atomic site engineering for efficient and selective catalysis.

Atropine can induce autophagy independent of the M3 muscarinic acetylcholine receptor

Background: No other effects of atropine other than as an antagonist of muscarinic acetylcholine receptor (mAChR) have been found. Methods: In this study, human kidneyepithelial cells were treated with different physiological regulators. Results: Subsequently, it was found that atropine could significantly induce autophagy as demonstrated by the appearance of autophagosome-like double- or single-membrane vesicles in the cytoplasm ofhost cells and the number of GFP-LC3 dots. In addition, increased conversion of the autophagy marker protein LC3-I and LC3-II and increased p62/SQSTM1 indicatedincomplete autophagy. In addition, atropine induced autophagosome levels in a dose-dependent manner within a certain concentration range in human kidney epithelial cells. In atropine-treated mouse skeletal muscle cells containing nicotinic acetylcholinereceptors and rat cardiac muscle cells containing mAchR, atropine induced autophagy in mouse skeletal muscle cells but not in rat cardiac muscle cells. Furthermore, atropine did not induce autophagy in tissue cells containing mAchR in vivo but did in tissue cells not containing mAchR. Conclusion: This study expands the application and understanding of atropine’s action mechanism in the field of medicine.

The electrochemical oxidation of 5-hydroxymethylfurfural(HMF)to valuable chemicals is an efficient way to upgrade biomass molecules and replace traditional catalytic *** is crucial to develop efficient and low-cost earth-abundant electrocatalysts to enhance catalytic performance of HMF ***,a new type of two-dimensional(2D)hybrid arrays consisting of Ni Fe layered double hydroxides(LDH)nanosheets and bimetallic sulfide(Ni Fe S)is constructed via interface engineering for efficient electrocatalytic oxidation of HMF to 2,5-furandicarboxylic acid(FDCA).The preparation process of 2D Ni Fe LDH/NiFeS with ultrathin heterostructure involves in anchoring a Co-based metal-organic framework(Co MOF)as template onto the carbon cloth(CC)via in-situ growth,formation of NiFe LDH on the surface of Co MOF and subsequent partial *** electrocatalyst of Ni Fe LDH/Ni Fe S exhibits outstanding performance towards HMF oxidation,about 98.5%yield for FDCA and 97.2%Faraday efficiency(FE)in the alkaline electrolyte with 10 mmol/L HMF,as well as excellent stability retaining 90.1%FE for FDCA after six cycles ***,even at an HMF concentration of 100 mmol/L,the yield and FE for FDCA remain high at 83.6%and 93.6%,*** findings highlight that 2D heterostructure containing abundant interfaces between Ni Fe LDH nanosheets and Ni Fe S can enhance the intrinsic activity of LDH and thus promote the oxidation reaction ***,the synergistic effect of the bimetallic Ni Fe compounds also improved the selectivity of HMF conversion to *** present work demonstrates that constructing 2D ultrathin heterostructure of Ni Fe LDH/Ni Fe S is a facile strategy via interface engineering to enhance the intrinsic activity of LDH electrocatalysts,which would open new avenues toward low-cost and advanced 2D nanocatalysts for sustainable energy conversion and electrochemical valorization of biomass derivatives.

A group of massive galaxies at redshifts of z■7 have been recently detected by the James Webb Space Telescope(JWST),which were unexpected to form at such an early time within the standard Big Bang *** this work,we propose that this puzzle can be explained by the presence of some primordial black holes(PBHs)with a mass of~1000M_(⊙).These PBHs act as seeds for early galaxy formation with masses of~10^(8)-10^(10)M_(⊙)at high redshift,hence accounting for the JWST *** use a hierarchical Bayesian inference framework to constrain the PBH mass distribution models,and find that the Lognormal model with the M_(c)~750M_(⊙)is preferred over other *** rapidly growing BHs are expected to have strong radiation and may appear as high-redshift compact objects,similar to those recently discovered by *** we focused on PBHs in this work,the bound on the initial mass of the seed black holes remains robust even if they were formed through astrophysical channels.

Molecular Characteristics of Cephalosporin-Resistant Neisseria gonorrhoeae Strains in China:A Retrospective Study

Objective:An increasing level of antimicrobial resistance to extended-spectrum cephalosporins has been reported,so we conducted this study to investigate the molecular characteristics of cephalosporin resistant Neisseria gonorrhoeae(NG)strains isolated from China Gonococcal Antimicrobial Resistance Surveillance Programme(China-GRSP).Methods:In total,2,038 NG isolates were collected from the China-GRSP between 2015 and *** multiantigen sequence typing(NG-MAST),multilocus sequence typing(MLST),and NG sequence typing for antimicrobial resistance(NG-STAR)were used to determine characteristics and resistant determinants of cephalosporin-resistant *** results are described as ***:Seventy-four(3.6%)cephalosporin-resistant isolates were ***_(ST7363)was the most prevalent MLST among cefixime-resistant(CFX-R)isolates,whereas MLST_(ST7365)(n=8)in was the most prevalent sequence type among ceftriaxone-resistant(CRO-R)***-R strains were mainly associated with penA-X(n=27,44.3%),penA-XIII(n=6,9.8%),penA-XVIII(n=5,8.2%),and CRO-R strains were mainly related to penA-XVIII(n=10,25.6%),penA-43(n=9,23.1%),penA-XIII(n=7,17.9%).Conclusion:The main conclusion of this study is that the predominant cephalosporin-resistant clones in China include locally emerged and imported *** CFX-R and CRO-R strains are primarily related to penA mutations,and their predominant genotypes are *** the increase in international travel,tracking high-risk-resistant clones and identifying outbreaks and their associations with epidemiological characteristics are critical to improve public health.

Flavonoids from mulberry leaves inhibit fat production and improve fatty acid distribution in adipose tissue in finishing pigs

This study evaluated the effects of flavonoids from mulberry leaves(FML)on plasma biochemical indices,serum activities of lipid metabolism-related enzymes,fat morphology,fatty acid composition,and lipid metabolism in different adipose tissues of finishing *** used 120 Chinese hybrid barrows of Berkshire and Bama mini-pigs with an average initial body weight of 45.11±4.23 *** pigs were randomly assigned to five treatment groups and fed a control diet based on corn,soybean meal,and wheat bran or a control diet supplemented with 0.02%,0.04%,0.08%,or 0.16%*** experimental group had six replicates(pens),with four pigs per *** a 7-d adaptation period,the feeding trial was conducted for 58 *** and adipose tissue samples were collected from 30 pigs(one pig per pen)at the end of the *** results showed that FML supplementation significantly decreased the feed intake to body gain ratio,the plasma concentrations of total cholesterol and free fatty acids,and the serum activity of 3-hydroxy-3-methylglutaryl coenzyme A reductase(linear or quadratic effects,P<0.05),and decreased the plasma triglyceride concentration(quadratic,P=0.07).Increasing FML supplementation increased the average daily gain and serum activities of lipoprotein lipase(linear and quadratic effects,P<0.05)and adipose triglyceride lipase(linear,P<0.05).Dietary FML supplementation decreased the adipocyte area in the dorsal subcutaneous adipose(DSA)tissue of finishing pigs(linear,P=0.05)and increased the adipocyte area in the visceral adipose tissue(quadratic,P<0.01).Increasing FML supplementation decreased the C20:1 content in DSA,abdominal subcutaneous adipose,and visceral adipose tissues of finishing pigs(P<0.05)and increased the C18:3n3 and n-3 PUFA contents(P<0.05).The lipid metabolism genes were regulated by the PPARγ-LXRa-ABCA1 signaling pathway,and their expressions differed in different adipose *** findings suggest that FML improved growth performance,regulated lipid met
