OBJECTIVE: To evaluate the killing effects of the cytosine deaminase (CD) gene mediated by adenovirus vector on human pancreatic cancer cell lines in vitro. METHODS: The CD gene was cloned into pAdTrack-CMV-CD, and pAdTrack-CMV-CD and pAdEasy-l were recombinated in bacteria. The newly recombinated Ad-CD containing green fluoreseent protein (GFP) was propagated in 293 cells and purified by cesium chloride gradient centrifugation. Human pancreatic cancer cell lines Patu8988 and SW1990 were infected with this virus, then 5-FC was added. XTT assay was used to estimate relative numbers of viable cells. RESULTS: The positive clones were selected by using endonuclease to digest the combinatants and the concentration of viral liquids containing the CD gene was 2×1O^(11) pfu/ml. It was found that significant cytotoxic activities were possesscd by 5-FC for the CD gene transduced pancreatic cell lines, but little effects exerted on the nontransduced pancreatic carcinoma cells. CONCLUSIONS: The CD gene mediated by adenovirus with a high infectivity is efficient for gene therapy of pancreatic carcinoma cell lines. These data demonstrate the therapeutic efficacy of an enzyme prodrug strategy in experimental pancreatic cancer.

The mixing of scalar mesons is an open problem since their structure is unclear and controversial. By introducing the ideal mixing of scalar mesons, we dynamically investigate the hyperon-nucleon interaction in the ehiral SU （ 3 ） quark model by solving the resonating group method （RGM） equation. The results show that when the ideal mixing of scalar mesons is considered and the mass of a k meson is reduced to near 780 MeV, then the hyperonnucleon scattering data can be reasonably described in the ehiral SU（3） quark model. Hence we find that the experimental mass of the n meson is around 780 MeV, and fo（600） and fo（980） mesons are the ideal mixing of scalar singlet and octet mesons.

Degenerative cervical myelopathy is a common cause of spinal cord injury,with longer symptom duration and higher myelopathy severity indicating a worse *** numerous studies have investigated serological biomarkers for acute spinal cord injury,few studies have explored such biomarkers for diagnosing degenerative cervical *** study involved 30 patients with degenerative cervical myelopathy(51.3±7.3 years old,12 women and 18 men),seven healthy controls(25.7±1.7 years old,one woman and six men),and nine patients with cervical spondylotic radiculopathy(51.9±8.6 years old,three women and six men).Analysis of blood samples from the three groups showed clear differences in transcriptomic *** analysis identified 128 differentially expressed genes that were enriched in patients with neurological *** least absolute shrinkage and selection operator analysis,we constructed a five-gene model(TBCD,TPM2,PNKD,EIF4G2,and AP5Z1)to diagnose degenerative cervical myelopathy with an accuracy of 93.5%.One-gene models(TCAP and SDHA)identified mild and severe degenerative cervical myelopathy with accuracies of 83.3%and 76.7%,*** of two immune cell types(memory B cells and memory-activated CD4^(+)T cells)predicted levels of lesions in degenerative cervical myelopathy with 80%*** results suggest that peripheral blood RNA biomarkers could be used to predict lesion severity in degenerative cervical myelopathy.

NUMERICAL STUDY ON THE PURIFICATION PERFORMANCE OF RIVERBANK

During the rain time, the runoff infiltrates into the riverbank through the collecting gutter and slope surface. The city runoff is generally polluted by organic, oil, heavy metal particulates, etc. The pollutants moving with the water through the riverbank experience advection, dispersion, diffusion, adsorption, biochemical reaction and plant uptaking processes. In this article, a mathematical model was developed to simulate the performance of pollutant removal of the riverbank. The model took those main mechanisms into account. The modified Richards equation was used in simulating flow field. The mass balance law was employed in deriving the equation for pollutant transport, where the diffusion and dispersion were described with the Fick-type law, the adsorption was macroscopically expressed as form isotherm, and the bio-chemical degradation process was assumed to follow the Monod kinetics. The NH3-N and TP were considered in the present model. The mathematical model was descritized with a finite element numerical model, which was applied to two types of model riverbanks. In the model test, the hydraulic loading was assumed to have the intermittent pattern simulating the storm runoff of certain return period, and the values of the rainfall runoff and concentrations of the NH3-N and TP were taken from the model test. The computed overall removal rates for the NH3-N and TP in 6 cases are in the range of 88%-98%, 87%-97%, respectively. The differences between the computed and tested overall removal rates for the NH3-N and TP are less than 5%. The time-varying oscillation pattern of the concentrations of the NH3-N and TP were rationally simulated, which shows that the model presented in this article can be used to assess the purification performance of the riverbank constructed with sand or soil.

Lost circulation of drilling fluid is one of the most common engineering problems in the drilling process of fractured *** this study,an oil-absorbing polymer gel synthesized using compound monomers with rigid and flexible chains was applied to control the oil-based drilling fluid loss while *** microstructure,oil-absorbing performance,and plugging performance the gel was investigated.A large number of dense pores on the surface of the gel were observed,which allowed the oil molecules to enter the internal space of the *** initial oil absorption capacity of the gel was fast,and it increased with the increase in the temperature and decrease in the particle size,reaching 20.93 g/g at140℃.At a high temperature of 140℃,the bearing pressure capacity of the gel formula containing particles of different particle sizes reached 7.6 MPa for a fracture of a width of 3 mm,showing that the oil-absorbing gel have excellent plugging performance at high *** mechanism of the gel was investigated through visualized fracture plugging *** show that the dynamic migratio n,particle-swelling,particle-bridging,particle-aggregation,deformation-filling,and compaction-plugging contribute to the whole lost circulation control process,reflecting that the plugging performance can be effectively enhanced by improving the aggregation and filling degrees of the gel with different particle sizes.

Locomotion ability, efficiency and reliability are key targets for a good robot. The linkage mechanism for robot locomotion is a discontinuous-constraint metamorphic mechanism. Here we set up equations to present the discontinuous-constraint, point out that driving and controlling are the key points to improve the performance and efficiency of the linkage mechanism. Inspired by controlling strategy of the motor nervous system in peripheral vertebrae to the locomotion, we draw off motor control and drive strategy.
