AIM： To assess the associations of human leukocyte antigen （HI_A） class Ⅱ DQB1＊0301 and/or DRB1＊1101 allele with spontaneous hepatitis C virus （HCV） clearance by meta-analysis of individual dataset from all studies published till date. METHODS： To clarify the impact of HLA class Ⅱ polymorphisms on viral clearance, we performed a metaanalysis of the published data from 11 studies comparing the frequencies of DQB1＊0301 and DRB1＊1101 alleles in individuals with spontaneous resolution to those with persistent infection. As we identified the heterogeneity between studies, summary statistical data were calculated based on a random-effect model. RESULTS： Meta-analyses yielded summary estimatesodds ratio （OR） of 2.36 [95%CI （1.62, 3.43）, P〈0.00001] and 2.02 [95%CI （1.56, 2.62）, P〈0.00001] for the effects of DQB1＊0301 and DRB1＊1101 alleles on spontaneous clearance of HCV, respectively. CONCLUSION： These results support the hypothesis that specific HLA class Ⅱ alleles might influence the susceptibility or resistance to persistent HCV infection. Both DQB1＊0301 and DRB1＊1101 are protective alleles and present HCV epitopes more effectively to CD4^＋T lymphocytes than others, and subjects with these two alleles are at a lower risk of developing chronic HCV infection. Large, multi-ethnic confirmatory and welldesigned studies are needed to determine the host genetic determinants of HCV infection.

CagA+ H pylori infection is associated with polarization of T helper cell immune responses in gastric carcinogenesis

AIM:To characterize the immune responses including local and systemic immunity induced by infection with H pylori,especially with CagA+ H pylori strains and the underlying immunopathogenesis. METHODS:A total of 711 patients with different gastric lesions were recruited to determine the presence of H pylori infection and cytotoxin associated protein A (CagA),the presence of T helper (Th) cells and regulatory T (Treg) cells in peripheral blood mononuclear cells (PBMCs),expression of plasma cytokines,and RNA and protein expression of IFN-γ and IL-4 in gastric biopsies and PBMCs were determined by rapid urease test,urea 14C breath test,immunoblotting test,flow cytometry ,real time RT-PCR and immunohistochemistry. RESULTS:Of the patients,629 (88.47%) were infected with H pylori ; 506 (71.16%) with CagA+ and 123 (17.30%) with CagA- strains. Among patients infected with CagA+ H pylori strains,Th1-mediated cellular immunity was associated with earlier stages of gastric carcinogenesis,while Th2-mediated humoral immunity dominated the advanced stages and was negatively associated with an abundance of Treg cells. However,there was no such tendency in Th1/Th2 polarization in patients infected with CagA- H pylori strains and those without H pylori infection. CONCLUSION:Polarization of Th cell immune responses occurs in patients with CagA+ H pyloriinfection,which is associated with the stage and severity of gastric pathology during the progression of gastric carcinogenesis. This finding provides further evidence for a causal role of CagA+ H pylori infection in the immunopathogenesis of gastric cancer.

Th1-type cytokines produced by the stimulation of Th1-type epitopes derived from definedschistosome-associated antigens are correlated with the development of resistance to the parasite *** mansoni 28 kDa glutathione-S-transferase(Sm28GST),a major detoxification enzyme,hasbeen recognized as a vaccine candidate and a phase Ⅱ clinical trial has been carried *** immunizedwith recombinant Schistosoma japonicum 28GST(Sj28GST)have shown immune protection against theparasite *** the present study,six candidate peptides(P1,P2,P3,P4,P7 and P8)from Sj28GSTwere predicted,using software,to be T cell epitopes,and peptides P5 and P6 were designed by extendingfive amino acids at the N-terminal and C-terminal of P1,*** peptide 190-211 aa in Sj28GSTcorresponding to the Th1-type epitope(190-211 aa)identified from Sm28GST was selected and named *** nine candidate peptides were synthesized or produced as the fusion protein with thioredoxin in thepET32c(+)/BL21(DE3)*** capacity to induce a Th1-type response in vitro was measured usinglymphocyte proliferation,cytokine detection experiments and flow *** results showed that P6(73-86 aa)generated the strongest stimulation effect on T cells among the nine candidate peptides,and drovethe highest level of IFN-γ and ***,P6 is a functional Th1-type T cell epitope that is differentfrom that in Sm28GST,and will be useful for the development of effective vaccines which can triggeracquired immunity against ***,our strategy of identifying the Th1-type epitope by acombination of software prediction and experimental confirmation provides a convenient and cost-savingalternative approach to previous methods.

Characterization of CD4~+ T Cell Responses in Mice Infected with Schistosoma japonicum

To better understand the interaction between Schistosoma japonicum and its murine host,wecharacterized the immune response of CD4~+ T cells generated during an experimental *** infectionbased on different key aspects,from gene expression to cell *** oligonucleotide microarrayswere used to compare gene expression profiles of CD4~+ T cells from spleens of mice at 0,3,6 and 13weeks *** cytometry analysis was used to determine type 1 and type 2 cytokine-secretingCD4~+ T cells,to test apoptosis of CD4~+ T cells and to count CD4~+CD25~+ T cells,a kind of regulatorysubpopulation of CD4~+ T *** percentage of interleukin-4-producing CD4~+ T cells was found to bemuch higher than that of γ-interferon-producing cells,especially after stimulation with *** eggantigen,which was consistent with type 1 and type 2 cytokine gene expression in the *** also showed that *** induced the increased expression of Th2 response-related genes,whereassome transcripts related to the Th1 responsive pathway were *** cytometry analysis showeda marked increase in the apoptotic CD4~+ T cells from 6 weeks post-infection and in the ratio of CD4~+CD25~+to CD4~+ T cells in infected mice after 13 *** therefore concluded that experimental infection of micewith *** resulted in a Th2-skewed immune response,which was to a great extent monitored bythe immune regulatory network,including cytokine cross-modulation,cell apoptosis and the subpopulationof regulatory cells.

Microstructure and water-swelling mechanism of red-bed mudstone in the Xining region,Northeastern Tibetan Plateau

This paper examines the effect of the microstructure and electrical conductivity(EC)on the swelling ratio and pressure in red-bed mudstone sampled from arid areas in the Xining region in the northeastern Tibetan Plateau.A series of laboratory tests,including swelling experiments,X-ray diffraction(XRD),and scanning electron microscope(SEM),was carried out for mechanical and microstructural *** coupled influence of the EC and microstructural parameters on the expansion ratio and pressure was investigated,and the weight coefficients were discussed by the entropy weight *** results revealed an increasing exponential trend in EC,and the maximum swelling speed occurred at an EC of approximately 10 μS/*** addition,a method for predicting the expansion potential is proposed based on the microstructure,and its reliability is verified by comparing with swelling experimental *** addition,according to the image analysis results,the ranges of the change in the clay minerals content(CMC),the fractal dimension(FD),the average diameter(AD)of pores,and the plane porosity(PP)are 23.75%-53%,1.08-1.17,7.53-22.45 mm,and 0.62%-1.25%,***,mudstone swelling is negatively correlated with the plane porosity,fractal dimension and average diameter and is linearly correlated with the clay mineral ***,the weight values prove that the microstructural characteristics,including FD,AD,and PP,are the main factors influencing the expansion properties of red-bed mudstones in the Xining *** on the combination of macro and micro-analyses,a quantitative analysis of the swelling process of mudstones can provide a better reference for understanding the mechanism of expansion behavior.

Examination of two-phase behaviors in porous media during pool boiling

The enhancement of pool boiling heat transfer using porous media has been extensively *** the two-phase distribution and evolution in porous media are crucial to the heat transfer performance,including the critical heat flux(CHF)and heat transfer coefficient(HTC),direct observation of the two-phase flow inside the media is limited owing to the blockage of the direct view from the porous *** this study,pool boiling visualization experiments were conducted on porous samples with different throat widths in deionized *** results showed that the HTC increased with the throat ***,the growth-contraction cycle of the vapor region and the formation and drying of the wall liquid film inside the porous media were *** vapor region,including the maximum and minimum areas in the boiling cycle,was quantitatively ***,the relationship between the minimum gas-phase area and HTC peak was identified.A one-dimensional transient model was developed considering solid skeleton heat conduction,liquid film evaporation,and vapor region growth to quantitatively study the influence of heat flux on the internal two-phase *** model successfully captured the maximum gas-phase area,duration of boiling cycles,and HTC trends at specific heat *** results of this quantitative study provide insights into the internal two-phase distribution and evolution induced by pool boiling.

Flotation separation of scheelite from calcite using luteolin as a novel depressant

This paper proposes luteolin(LUT)as a novel depressant for the flotation-based separation of scheelite and calcite in a sodium oleate(NaOL)*** suitability of LUT as a calcite depressant is confirmed through micro-flotation *** pH=9,with LUT concentration of 50 mg·L^(-1) and NaOL concentration of 50 mg·L^(-1),scheelite recovery reaches 80.3%.Calcite,on the other hand,exhibits a recovery rate of 17.6%,indicating a significant difference in floatability between the two ***,the surface modifica-tions of scheelite and calcite following LUT treatment are characterized using adsorption capacity testing,Zeta potential analysis,Fourier transform infrared spectroscopy(FT-IR),X-ray photoelectron spectroscopy(XPS),and atomic force microscopy(AFM).The study in-vestigates the selective depressant mechanism of LUT on *** capacity testing and Zeta potential analysis demonstrate sub-stantial absorption of LUT on the surface of calcite,impeding the further adsorption of sodium oleate,while its impact on scheelite is ***-IR and XPS analyses reveal the selective adsorption of LUT onto the surface of calcite,forming strong chemisorption bonds between the hydroxyl group and calcium ions *** directly illustrates the distinct adsorption densities of LUT on the two miner-al ***,LUT can effectively serve as a depressant for calcite,enabling the successful separation of scheelite and calcite.

Gene Transcription Profile in Mice Vaccinated with Ultraviolet-attenuated Cercariae of Schistosoma japonicum Reveals Molecules Contributing to Elevated IFN-γ Levels

Reduced magma generation and its implications for the Pulang giant porphyry Cu-polymetallic deposit in Northwest Yunnan,China

The Pulang giant porphyry Cu-Mo polymetallic deposit is located in the Zhongdian area in the center of the Sanjiang Tethys tectonic domain,which was formed by the westward subduction of the Garze-Litang oceanic slab beneath the Zhongza ***-pyrrhotite-pyritemolybdenite occurs as disseminations,veins,veinlets,and stockworks distributed in the K-silicate alteration zone in the monzonite porphyry,which is superimposed by *** chemical compositions of biotite and amphibole analyzed by electron probe microanalysis(EPMA)indicate that the ore-forming magma and exsolved fluids experienced a continuous decrease in the oxygen fugacity(fO_(2)).Primary amphibolite and biotite(type I)crystallized at relatively high temperatures(744-827°C)and low fO_(2)(log fO_(2)=−12.26 to−11.91)during the magmatic *** fluids exsolved from the magma have a relatively lower temperature(621-711°C)and fO_(2)(log fO_(2)=−14.36 to−13.32)than the original *** addition,the presence of a high abundance of pyrrhotite and an insufficiency of primary magnetite and sulfate in the ore(i.e.,anhydrite and gypsum)indicate that the deposit may be a reduced porphyry *** and fluid fO_(2)results,combined with previous research on magmatic fO_(2)at the Pulang deposit,indicate that the magma associated with the reduced Pulang ore assemblages was initially generated as a highly oxidized magma that was subsequently reduced by sedimentary rocks of the Tumugou Formation.
