Stability study on an anti-cancer drug 4-(3,5-bis(2-chlorobenzylidene)-4-oxo-piperidine-1-yl)-4-oxo-2-butenoic acid (CLEFMA) using a stabilityindicating HPLC method

CLEFMA, 4-(3,5-bis(2-chlorobenzylidene)-4-oxo-piperidine-1-yl)-4-oxo-2-butenoic acid, is a new chemical entity with anti-cancer and anti-inflammatory activities. Here, we report its stability in solution against stress conditions of exposure to acid/base, light, oxidant, high temperature, and plasma. The identity of the degradation products was ascertained by mass and proton nuclear magnetic resonance spectroscopy. To facilitate this study, we developed and validated a reverse phase high performance liquid chromatography method for detection of CLEFMA and its degradation. The method was linear over a range of 1–100 μg/m L; the accuracy and precision were within acceptable limits; it was stability-indicating as it successfully separated cis-/trans-isomers of CLEFMA as well as its degradation product. The major degradation product was produced from amide hydrolysis at maleic acid functionality caused by an acidic buffer, oxidant(3% hydrogen peroxide),or temperature stress(40–60 °C). The log k-p H profile showed that CLEFMA was most stable at neutral p H. In accelerated stability study we found that the shelf-life(T_(90%)) of CLEFMA at 25 °C and 4 °C was 45 days and220 days, respectively. Upon exposure to UV-light(365 nm), the normally prevalent trans-CLEFMA attained cis-configuration. This isomerization also involved the maleic acid moiety. CLEFMA was stable in plasma from which it could be efficiently extracted by an acetonitrile precipitation method. These results indicate that CLEFMA is sensitive to hydrolytic cleavage at its maleic acid moiety, and it is recommended that its samples should be stored under refrigerated and light-free conditions, and under inert environment.

This paper introduces a video application-aware cross-layer framework for joint performance-energy optimization,considering the scenario of multiple users upstreaming real-time Motion JPEG2000 video streams to the access point of a WiFi wireless local area network and extends the PHY-MAC run-time cross-layer scheduling strategy that we introduced in (Mangharam et al., 2005; Pollin et al., 2005) to also consider congested network situations where video packets have to be dropped. We show that an optimal solution at PHY-MAC level can be highly suboptimal at application level, and then show that making the cross-layer framework application-aware through a prioritized dropping policy capitalizing on the inherent scalability of Motion JPEG2000 video streams leads to drastic average video quality improvements and inter-user quality variation reductions of as much as 10 dB PSNR, without affecting the overall energy consumption requirements.

Trends in oxycodone and oxycodone-containing analgesics administration for back pain in emergency departments in the USA(2007–2018)

RNA-based therapies in animal models of Leber congenital amaurosis causing blindness

To design microstructure and microhardness in the additive manufacturing(AM)of nickel(Ni)-based superalloys,the present work develops a novel data-driven approach that combines physics-based models,experimental measurements,and a data-mining *** simulation is based on a computational thermal-fluid dynamics(CtFD)model,which can obtain thermal behavior,solidification parameters such as cooling rate,and the dilution of solidified *** on the computed thermal information,dendrite arm spacing and microhardness are estimated using well-tested mechanistic *** microstructure and microhardness are determined and compared with the simulated values for *** visualize process-structure-properties(PSPs)linkages,the simulation and experimental datasets are input to a data-mining model-a self-organizing map(SOM).The design windows of the process parameters under multiple objectives can be obtained from the visualized *** proposed approaches can be utilized in AM and other data-intensive ***-driven linkages between process,structure,and properties have the potential to benefit online process monitoring control in order to derive an ideal microstructure and mechanical properties.

GrapevineXL reliably predicts multi-annual dynamics of vine water status, berry growth, and sugar accumulation in vineyards

Climate and water availability greatly affect each season’s grape yield and *** models to accurately predict environment impacts on fruit productivity and quality is a huge *** calibrated and validated the functional-structural model,GrapevineXL,with a data set including grapevine seasonal midday stem water potential(�xylem),berry dry weight(DW),fresh weight(FW),and sugar concentration per volume([Sugar])for a wine grape cultivar(Vitis vinifera *** Franc)in field conditions over 13 years in Bordeaux,*** results showed that the model could make a fair prediction of seasonal�xylem and good-to-excellent predictions of berry DW,FW,[Sugar]and leaf gas exchange responses to predawn and midday leaf water potentials under diverse environmental conditions with 14 key *** running virtual experiments to mimic climate change,an advanced veraison(*** onset of ripening)of 14 and 28 days led to significant decreases of berry FW by 2.70%and 3.22%,clear increases of berry[Sugar]by 2.90%and 4.29%,and shortened ripening duration in 8 out of 13 simulated years,***,the impact of the advanced veraison varied with seasonal patterns of climate and soil water ***,the results showed that the GrapevineXL model can predict plant water use and berry growth in field conditions and could serve as a valuable tool for designing sustainable vineyard management strategies to cope with climate change.

China and the United States are the two most significant nations in the contemporary global food and agricultural *** addition,they are two of the most important innovators with respect to the development of new crop varieties,agro-technologies,farm products,markets and consumer issues,such as consumer resistance to genetically modified foods,among *** the face of an ever-complex web of interactions,technologies and products among producers and consumers in both nations,there are far more structural similarities than differences in the food and agriculture sectors of these two *** essay,adopting some of the themes of the Sino-American Symposium on Future Issues Affecting Quality of Life,presents a limited but representative comparative assessment of three of the most important shared challenges impacting the agricultural sectors of China and the United States for the period from 2000 to the present including 1)environmental challenges related to agricultural water supply,2)declines in farm labor and rural population,and 3)growing food-related concerns and *** both nations,excessive and unsustainable groundwater consumption has lowered water tables and limited crop *** populations and farm workforces in both nations are also declining,leading to labor challenges in both ***,concerns regarding food safety are also very similar with major challenges to the farm sector associated with consumer resistance to genetically modified food crops and sanitation issues linked to lengthening supply *** of these issues threaten the development of sustainable agricultural production systems.
