Growth rate is a widely studied parameter for various cell-based biological *** rates of cell populations can be monitored in chemostats and micro-chemostats,where nutrients are continuously ***,we present an integrated microfluidic platform that enables long-term culturing of non-adherent cells as well as parallel and mutually independent continuous monitoring of(i)growth rates of cells by means of impedance measurements and of(ii)specific other cellular events by means of high-resolution optical or fluorescence *** colonies were grown in a monolayer under culturing pads,which enabled high-resolution microscopy,as all cells were in the same focal *** cell growth and division,cells leaving the culturing area passed over a pair of electrodes and were counted through impedance *** impedance data could then be used to directly determine the growth rates of the cells in the culturing *** integration of multiple culturing chambers with sensing electrodes enabled multiplexed long-term monitoring of growth rates of different yeast strains in *** a demonstration,we modulated the growth rates of engineered yeast strains using *** results indicated that impedance measurements provide a label-free readout method to continuously monitor the changes in the growth rates of the cells without compromising high-resolution optical imaging of single cells.

Ultrafast supercontinuum generation in gas-filled waveguides is an enabling technology for many intriguing applications ranging from attosecond metrology towards biophotonics,with the amount of spectral broadening crucially depending on the pulse dispersion of the propagating *** this study,we show that structural resonances in a gas-filled antiresonant hollow core optical fiber provide an additional degree of freedom in dispersion engineering,which enables the generation of more than three octaves of broadband light that ranges from deep UV wavelengths to near *** observation relies on the introduction of a geometric-induced resonance in the spectral vicinity of the ultrafast pump laser,outperforming gas dispersion and yielding a unique dispersion profile independent of core size,which is highly relevant for scaling input *** a krypton-filled fiber,we observe spectral broadening from 200 nm to 1.7μm at an output energy of B 23μJ within a single optical mode across the entire spectral *** show that the frequency generation results from an accelerated fission process of solitonlike waveforms in a non-adiabatic dispersion regime associated with the emission of multiple phase-matched Cherenkov radiations on both sides of the *** effect,along with the dispersion tuning and scaling capabilities of the fiber geometry,enables coherent ultra-broadband and high-energy sources,which range from the UV to the mid‐infrared spectral range.

AIM: To compare anxiety and depression levels in adult patients with celiac disease (CD) on a gluten-free diet (GFD) with ***: The levels of anxiety, depression and of a probable anxiety or depressive disorder were assessed by the Hospital Anxiety and Depression Scale in 441 adult patients with CD recruited by the German Celiac Society, in 235 age-and sex-matched patients with inflammatory bowel disease (IBD) in remission or with slight disease activity, and in 441 adult persons of a representative German general population sample (GP). Potential demographic (age, sex, social class, family status) and disease-related (latency to diagnosis, duration of GFD, compliance with GFD, thyroid disease) predictors of anxiety and depression in CD were tested for by regression analyses. RESULTS: The level of anxiety in CD patients was predicted (R 2 = 0.07) by female gender (P = 0.01). Female sex (OR = 3.6, 95% CI: 1.3-9.4, P = 0.01) was associated with a probable anxiety disorder. Living alone (OR = 0.5, 95% CI: 0.2-0.9, P = 0.05) was associated with a reduced risk of an anxiety disorder. The level of depression and a probable depressive disorder were not predicted by any of the demographic and medical variables tested for. The levels of anxiety in patients with CD (6.6 ± 3.4) and with IBD (6.9 ± 3.7) were higher than those of persons in the GP (4.6 ± 3.3) (both P with CD (4.2 ± 3.4), IBD (4.6 ± 3.4) and of the GP (4.2 ± 3.8) did not differ (P = 0.3). The prevalence of a probable anxiety disorder in persons with CD (16.8%) and IBD (14.0%) was higher than that of the GP (5.7%) (P ween the three groups (P = 0.1). CONCLUSION: Anxiety in adult German female celiacs on a GFD is higher than in persons of the GP. Female celiacs on a GFD should be screened for anxiety.

AIM： To investigate the effects of catalytically superior gene-directed enzyme prodrug therapy systems on a rat hepatoma model. METHODS： To increase hepatoma cell chemosensitivity for the prodrug 5-fluorocytosine （5-FC）, we generated a chimeric bifunctional SuperCD suicide gene, a fusion of the yeast cytosine deaminase （YCD） and the yeast uracil phosphoribosyltransferase （YUPRT） gene. RESULTS： In vitro stably transduced Morris rat hepatoma cells （MH） expressing the bifunctional SuperCD suicide gene （MH SuperCD） showed a clearly marked enhancement in cell killing when incubated with 5-FC as compared with MH cells stably expressing YCD solely （MH YCD） or the cytosine deaminase gene of bacterial origin （MH BCD）, respectively. In vivo, MH SuperCD tumors implanted both subcutaneously as well as orthotopically into the livers of syngeneic ACI rats demonstrated significant tumor regressions （P〈0.01） under both high dose as well as low dose systemic 5-FC application, whereas MH tumors without transgene expression （MH naive） showed rapid progression. For the first time, an order of in vivo suicide gene effectiveness （SuperCD〉〉 YCD〉〉BCD〉〉〉negative control） was defined as a result of a direct in vivo comparison of all three suicide genes. CONCLUSION： Bifunctional SuperCD suicide gene expression is highly effective in a rat hepatoma model, thereby significantly improving both the therapeutic index and the efficacy of hepatocellular carcinoma killing by fluorocytosine.

AIM:To understand the demographic characteristics of patients in Southwestern Ontario,Canada with ulcerative colitis(UC)in order to predict disease severity. METHODS:Records from 1996 to 2001 were exam- ined to create a database of UC patients seen in the London Health Sciences Centre South Street Hospital Inflammatory Bowel Disease *** be included, patients'charts were required to have information of their disease presentation and a minimum of 5 years of *** were reviewed using standardized data collection *** severity was generated during the chart review process,and non-endoscopic Mayo Score criteria were collected into a composite. RESULTS:One hundred and two consecutive patients'data were entered into the *** analyses revealed that 51%of the patients were male, the mean age at diagnosis was 39 years,13.7%had a first degree relative with inflammatory bowel disease (IBD),61.8%were nonsmokers and 24.5%were *** 22.5%of patients the disease was limited to the rectum,in 21.6%disease was limited to the sigmoid colon,in 22.5%disease was limited to the left colon,and 32.4%of patients had pancolitis. Standard multiple regression analysis which regressed a composite of physician global assessment of disease severity,average number of bowel movements,and average amount of blood in bowel movements on year of diagnosis and age at time of diagnosis was significant,R 2=0.306,F(7,74)=4.66,Pwith younger age at diagnosis and year of diagnosis in a longitudinal cohort of UC patients,and may identify prognostic UC indicators.

利用飞秒光电子影像技术研究了碘甲烷分子在飞秒强激光场作用下的多光子电离动力学,在实验上实现了运用飞秒强场多光子电离技术对多原子分子离子的振动量子态进行光学操控.提高了飞秒激光的强度,从1.6×1013w/cm2提高到2.5×1013w/cm2.在增大的激光强度范围内,发现了新的能量组分,并对此进行归属.通过采集光电离的光电子影像可以得到强场电离后光电子的动能分布和角度分布两方面的信息,前期的研究主要侧重于讨论光电子动能随光强的变化,重点讨论了光电子角度分布随光强的变化,通过观察光电子角度分布的变化趋势对振动量子态调控机理进一步认识.

Multimodal nonlinear endomicroscopic imaging probe using a double-core double-clad fiber and focus-combining micro-optical concept

Eudialytes are a group of complex Na-Ca zirconosilicate minerals that generally occur in peralkaline agpaitic *** that eudialytes are easily altered and commonly contain inclusions of earlier-crystallized minerals,in situ laser ablation is considered as the most suitable method to determine their U-Pb ages,and Sr,Nd and Hf isotopic *** microprobe and LA-ICP-MS analysis of eudialytes from nepheline syenites from Ilimaussaq(Greenland),Khibiny and Lovozero(Russia),Saima(China),Tamazeght (Morocco),Kipawa and Mont Saint Hilaire(Canada),Pocos de Caldas(Brazil)and Langesund Fjord (Norway),indicate that this mineral typically has high contents of U,Pb,Nb,Ta,Zr,Hf and rare earth elements(REE). Analysis of an in-house standard eudialyte by both solution and laser ablation methods demonstrates that precise and accurate U-Pb ages can be obtained from eudialyte after correction for the common Pb content. The high Sr,Nd and Hf contents in eudialyte,coupled with the generally low Rb/Sr and Lu/Hf ratios,also permit the precise determination of in situ Sr,Nd and Hf isotopic ratios by LA-(MC)-ICP-MS methods. However,some eudialytes with Rb/Sr ratio of>0.02 and Yb/Sr>0.1,and Lu/Hf>0.08(or Yb/Hf>0.4-0.5) cannot be used for in situ Sr and Hf isotopic *** is the only mineral investigated to date for which it is possible to determine simultaneously U-Pb ages and Sr,Nd and Hf isotopic compositions.

Multimodal non-linear microscopy combining coherent anti-Stokes Raman scattering,second harmonic generation,and two-photon excited fluorescence has proved to be a versatile and powerful tool enabling the label-free investigation of tissue structure,molecular composition,and correlation with function and disease *** a routine medical application,the implementation of this approach into an in vivo imaging endoscope is ***,this is a difficult task due to the requirements of a multicolour ultrashort laser delivery from a compact and robust laser source through a fiber with low losses and temporal synchronization,the efficient signal collection in epi-direction,the need for small-diameter but highly corrected endomicroobjectives of high numerical aperture and compact ***,we introduce an ultra-compact fiber-scanning endoscope platform for multimodal non-linear endomicroscopy in combination with a compact four-wave mixing based fiber *** heart of this fiber-scanning endoscope is an in-house custom-designed,single mode,double clad,double core pure silica fiber in combination with a 2.4 mm diameter NIR-dual-waveband corrected endomicroscopic objective of 0.55 numerical aperture and 180µm field of view for non-linear imaging,allowing a background free,low-loss,high peak power laser delivery,and an efficient signal collection in backward direction.A linear diffractive optical grating overlays pump and Stokes laser foci across the full field of view,such that diffraction-limited performance is demonstrated for tissue imaging at one frame per second with sub-micron spatial resolution and at a high transmission of 65%from the laser to the specimen using a distal resonant fiber scanner.
