Recent interest in recycling and reuse of waste plastic has resulted in a dramatic increase in the evaluation of waste plastic as an extender and/or modifier of bituminous binder for asphalt production. Although significant field trials of waste plastic modified asphalt have been reported in Australia since 2017, no laboratory evaluation of the effects of these products on the engineering properties of binder and asphalt has been published. In this research, two commercially available recycled waste plastic products were evaluated in the laboratory. One product is intended to be plastomeric while the other is intended to be elastomeric in nature. Compared to unmodified viscosity grade bitumen and acid modified multigrade, the recycled plastic products increased the viscosity and softening temperature of the binder and introduced significant elastic recovery. Following dry-mixing into asphalt, the recycled plastic products were associated with improved mixture deformation resistance and increased mixture stiffness. However, the mixtures containing recycled plastic were also associated with an increase in moisture susceptibility and their fatigue lives were not significantly different to those of the control mixtures. Further research is recommended to better understand the modest reduction in moisture damage resistance associated with recycled plastic, as well as the digestion of recycled plastic via the dry-mixing process.

Barren ground caribou are one of the most important natural resources for northern aboriginal peoples in Canada, and their responsible management has been identified as a top priority by northern communities and governments. This study is aimed to assess the impacts of summer range forage availability and quality on Bathurst caribou’s productivity and abundance. Despite well documented effects of habitat nutrition on individual animal, few studies have been able to link nutrition and population demographics in a quantitative fashion, probably because caribou productivity and abundance could be potentially affected by many factors (e.g., habitat, harvest, predators, diseases/parasites, extreme weather, climate change, industrial development, and pollution), and yet long-term data for many of these factors are not available. By determining the upper envelope curve between summer range indicators and caribou productivity, this study made such assessment possible. Our results indicate that summer range indicators derived from long-term remote sensing time series and climate records can explain 59% of the variation in late-winter calf:cow ratio during 1985 and 2012. As a measure of caribounet productvitiy, the late-winter calf:cow ratio, together with the mortality rate, in turn determined population dynamics.

This study explores how dust from the Ekati Diamond Mine potentially affects the availability and quality of forage on the seasonal range of the Bathurst caribou herd. Understanding the effects of dust as a source of disturbance is important because the Bathurst caribou population has declined by 93% since the middle 1980s and there are reports that caribou in general may avoid mining projects. There are several challenges for quantifying dust impacts: 1) Natural variations (e.g., topography, natural disturbance, and soil pH) may also impact forage availability and quality for caribou. To minimize their masking effect, we stratified survey sites into seven land cover classes and selected the most populous class (i.e., the dwarf shrub) for assessing the impact. 2) Within class variation (e.g., the proportion of area covered by rocks where vascular plants and lichen do not grow) can further skew the analysis. We eliminated this problem by examining only the area not covered by rocks. 3) Coarse and fine suspended particulates have different spatial coverages, chemical compositions, and pH values. Consequently, their impacts on caribou forage can be different. To distinguish their impacts, we sampled two areas: transects from the Misery Haul Road that has been in active use vs. those from a rarely used spur road outside the Misery Camp. We sampled percent vegetation cover, soil pH, and dust on leaves along these transects during the summers of 2015 and 2016. Our results indicated that the amount of dust on leaves in a zone of ~1000 m from the Misery Haul Road was 3 - 9 times than that of background sites. The zone of reduced lichen percent cover was also about 1000 m. In contrast, these road dust-induced changes in caribou forage were not observed for the dust-free transect from the spur road.

目的探讨聚乙二醇水凝胶膜(PHFs)是否能作为角膜上皮细胞(CECs)体外扩增的载体以及其能否用于角膜缘干细胞缺乏症(LSCD)的治疗。方法利用癸二酰氯、聚氧乙基甘油醚、聚已酸内酯二醇人工合成PHFs,检测其厚度、透光度和机械拉伸性能。取清洁级新西兰大白兔4只,培养原代角膜缘上皮细胞,荧光显微镜下观察培养细胞中角蛋白标志物AE1/AE3和干细胞标志物p63的表达;将细胞分为阴性对照组、阳性对照组和PHFs+CECs组,其中将阴性对照组细胞直接种植在培养皿底,加入普通细胞培养液,将阳性对照组细胞种植在培养皿后加入含有100μmol/L H_(2)O_(2)的普通细胞培养液,将PHF+CECs组细胞种植在铺有PHFs的培养皿中用普通细胞培养液培养,各组培养24 h。采用MTT和TUNEL染色法分别观察3个组细胞增生和细胞凋亡情况。采用随机数字表法将清洁级新西兰大白兔15只分为对照组、PHFs组和PHFs+CECs组,每组5只。其中对照组造模后不给予任何处置;PHFs组将空PHFs膜置于LSCD实验兔角膜表面;PHFs+CECs组为将CECs传代后种植于PHFs上构建组织工程移植片后将移植片置于LSCD兔模型的角膜表面。采用荧光素染色法检测各组实验兔角膜缺损面积大小并评分;采用苏木精-伊红染色法检测各组实验兔角膜上皮组织学特点。结果PHFs厚度不超过150μm,能承受约6 MPa的拉力,在400~700 nm可见光范围内透过率>99%。角膜缘原代培养的大多数细胞AE1/AE3染色和p63染色阳性。MTT实验结果显示,PHFs+CECs组、阴性对照组和阳性对照组A_(490)值分别为0.59±0.01、0.65±0.07和0.06±0.04,总体比较差异有统计学意义(F=12.25,P<0.05),其中PHFs+CECs组和阴性对照组A_(490)值明显大于阳性对照组,差异有统计学意义(均P<0.05)。TUNEL检测结果显示,PHFs+CECs组、阴性对照组和阳性对照组TUNEL阳性细胞率总体比较差异有统计学意义(F=13.45,P<0.05),其中PHFs+CECs组和阴性对照组TUNEL阳性细胞率明显低于阳性对照组,差异均有统计学意义(均P<0.05)。荧光素染色结果显示,随着术后时间的延长,各组角膜荧光素钠染色评分均降低,对照组、PHFs组和PHFs+CECs组角膜荧光素钠染色评分依次降低。苏木精-伊红染色结果显示,对照组存在较少形状不规则的角膜上皮细胞,PHFs组一些区域出现单层稀疏的角膜上皮细胞;PHFs+CECs组角膜上皮覆盖范围最大,细胞层数增加到3~5层,形态较规则,排列较紧密。结论PHFs韧性强、透光度高且能够体外扩增角膜上皮,适合做角膜上皮移植片,并能促进LSCD兔模型角膜上皮的修复。
