Lymphatic filariasis (LF) remains a public health concern as it can cause permanent morbidity and disability to those infected. While the global elimination of LF in these endemic areas is ongoing through mass drug administration, there is the need to develop diagnostic tools that would be utilized to track the progress of total global eradication as well as perform surveillance for the recurrence of lymphatic filariasis transmission. Currently, approved LF diagnosis tools are faced with lack of specificity, low sensitivity, and periodicity dependence. Recombinant filarial antigen-based assays can address these drawbacks and offer practical instruments for LF diagnosis and surveillance. This present study, evaluated rWb-SXP-1 and rWb-123 antigens as potential diagnostic biomarker tools for Wuchereria banchrofti in human sera using microspheres-based multiplex serological assay. Based on statistical analysis using XLSTAT 2019 (Addinsoft) on data generated from multiplex technology assay, generated ROC curves for both rWb-SXP-1 and rWb-123 demonstrated 87.1% sensitivity to Wuchereria banchrofti human sera with rWb-SXP-1 antigens having the highest specificity of 96%. Indication that rWb-SXP-1 and rWb-123 antigens are capable of detecting immunoglobulin G4 (IgG4) antibodies in human sera synthesized specifically against W. banchrofti infections. Therefore, rWb-SXP-1 and rWb-123 antigens can be utilized to detect W. banchrofti infections by antibody profiling with excellent diagnostic sensitivity and specificity using microsphere-based multiplex serological tests. This method can be particularly practical for screening a large number of sera samples and/or for quick, extensive field-testing due to the high-throughput and quick formats applied.

Perceptions of Beach Stewards and Information Needs for Shorebird Stopover Sites Vary as a Function of Visitor Interests: Improving Stewardship Programs in Coastal Landscapes

Many studies have demonstrated that some of the activities of people can disturb nesting and foraging birds, particularly along coasts and estuaries. Some managers respond to human disturbance of sensitive species by closing beaches, but most have stewards monitor and restrict access to beaches or portions of beaches. Yet little is known about the type of visitor or their perceptions of beach stewards. This paper explores the general acceptance of beach stewards and their role in conservation of red knots (Calidris canutus rufa), other shorebirds, and horseshoe crabs (Limulus polyphemus) along the Delaware Bay (New Jersey) beaches by interviewing 279 visitors during the stopover period. While most people came to see the shorebird and horseshoe crab spectacle (60%), many came for the scenery, tranquility, and recreation. Just over half of the people interviewed said that the stewards were helpful (58%), although most of those that answered this question later said that they provided conservation value (86%), but only 40% said they relied on the stewards for information. On a Likert rating scale of 1 to 5 (the highest value), the mean rating for whether it was important for stewards to be present was 3.87 ± 0.09, and the rating for the presence of stewards who can provide information was 3.71 ± 0.11. Gastric peritoneal carcinomatosis-a retrospective review

AIM To characterize patients with gastric peritoneal carcinomatosis(PC) and their typical clinical and treatment course with palliative systemic chemotherapy as the current standard of *** We performed a retrospective electronic chart review of all patients with gastric adenocarcinoma with PC diagnosed at initial metastatic presentation between January 2010 and December 2014 in a single tertiary referral *** We studied a total of 271 patients with a median age of 63.8 years and median follow-up duration of 5.1 mo. The majority(n = 217, 80.1%) had the peritoneum as the only site of metastasis at initial presentation. Palliative systemic chemotherapy was eventually planned for 175(64.6%) of our patients at initial presentation, of which 171 were initiated on it. Choice of first-line regime was in accordance with the National Comprehensive Cancer Network Guidelines for Gastric Cancer Treatment. Thesepatients underwent a median of one line of chemotherapy, completing a median of six cycles in total. Chemotherapy disruption due to unplanned hospitalizations occurred in 114(66.7%), while cessation of chemotherapy occurred in 157(91.8%), with 42 cessations primarily attributable to PC-related complications. Patients who had initiation of systemic chemotherapy had a significantly better median overall survival than those who did not(10.9 mo vs 1.6 mo, P < 0.001). Of patients who had initiation of systemic chemotherapy, those who experienced any disruptions to chemotherapy due to unplanned hospitalizations had a significantly worse median overall survival compared to those who did not(8.7 mo vs 14.6 mo, P < 0.001).CONCLUSION Gastric PC carries a grim prognosis with a clinical course fraught with disease-related complications which may attenuate any survival benefit which palliative systemic chemotherapy may have to offer. As such, investigational use of regional therapies is warranted and required validation in patients with isolated PC to maximize their

Evaluation of root traits and water use efficiency of different cotton genotypes in the presence or absence of a soil-hardpan

Cotton(Gossypium spp.) is an important fiber and oil crop grown worldwide. Water and nutrient stresses are major issues affecting cotton production globally. Root traits are critical in improving water and nutrient uptake and maintaining plant productivity under optimal as well as drought ***, root traits have rarely been utilized in cotton breeding programs, a major reason being the lack of information regarding genetic variability for root traits. The objective of this research was to evaluate ten selected cotton genotypes for root traits and water use efficiency. The tested genotypes included germplasm lines(PD 1 and PD 695) and cultivars that are currently grown in the southeastern USA(PHY 499 WRF, PHY 444 WRF, PHY 430 W3 FE, DP 1646 B2 XF, DP 1538 B2 XF, DP 1851 B3 XF, ng5007 B2 XF, and ST 5020 GLT). Experiments were conducted under controlled environmental conditions in 2018 and 2019. A hardpan treatment was included in the second year to evaluate the effect of a soil hardpan on root traits and water use efficiency. Genotype PHY 499 WRF ranked at the top and ng5007 B2 XF ranked at the bottom for root morphological traits(total and fine root length, surface area,and volume) and root weight. PHY 499 WRF was also one of the best biomass producers and had high water use efficiency. PHY 444 WRF, PHY 430 W3 FE, and PD-1 were the other best genotypes in terms of root traits and water use efficiency. All genotypes had higher values for root traits and water use efficiency under hardpan conditions. This trend indicates a horizontal proliferation of root systems when they incur a stress imposed by a hardpan. The genotypic differences identified in this research for root traits and water use efficiency would be valuable for selecting genotypes for cotton breeding programs.

The SARS-CoV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)*** study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron BA.2 and compared with 338 patients infected with non-variant of concern(non-VOC)-D614G.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both BA.2 and non-VOCD614G regardless of host gender,age and presence of ***,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both BA.2 and *** presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUC¼0.91,95%CI 0.88–0.94),and sgRNA ORF7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual *** potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.

Increased tumor necrosis factor receptor 1 expression in human colorectal adenomas

AIM: To determine the expression statuses of tumor necrosis factor (TNF)-α, its receptors (TNF-R) and downstream effector molecules in human colorectal adenomas. METHODS: We measured the serum concentrations of TNF-α and its receptors in 62 colorectal adenoma patients and 34 healthy controls. The protein expression of TNF-α, TNF-R1, TNF-R2 and downstream signals of the TNF receptors, such as c-Jun N-terminal kinase (JNK), nuclear factor-κ B and caspase-3, were also investigated in human colorectal adenomas and in normal colorectal mucosal tissues by immunohistochemistry. Immunofluorescence confocal microscopy was used to investigate the consistency of expression of TNF-R1 and phospho-JNK (p-JNK). RESULTS: The serum levels of soluble TNF-R1 (sTNF-R1) in adenoma patients were significantly higher than in the control group (3.67 ± 0.86 ng/mL vs 1.57 ± 0.72 ng/mL, P ng characteristic analysis revealed the high diagnostic sensitivity of TNF-R1 measurements (AUC was 0.928) for the diagnosis of adenoma, and the best cut-off level of TNF-R1 was 2.08 ng/mL, with a sensitivity of 93.4% and a specificity of 82.4%. There were no significant differences in the serum levels of TNF-α or sTNF-R2 between the two groups. Immunohistochemistry showed high levels of TNF-R1 and p-JNK expression in the epithelial cells of adenomas. Furthermore, a high incidence of co-localization of TNF-R1 and p-JNK was identified in adenoma tissue. CONCLUSION: TNF-R1 may be a promising biomarker of colorectal adenoma, and it may also play an important role in the very early stages of colorectal carcinogenesis.

To the Editor： To date, epilepsy is one of the most common neurological disorders globally, affecting more than 50 million people worldwide. It is debilitating and represents a large health-care and economic burden, especially when treatment may often be expensive and not necessarily effective. It has been suggested that the burden of lifetime epilepsy has been significantly increasing in China in recent decades, The International League Against Epilepsy has defined epilepsy in 2014 as a neurological disorder with at least two unprovoked seizures occurring more than 24 h apart.
