Infections by the fungus Monilinia laxa,the main cause of brown rot in Europe,result in considerable losses of stone ***,we present a comprehensive transcriptomic approach to unravel strategies deployed by nectarine fruit and *** during their *** used ***-inoculated immature and mature fruit,which was resistant and susceptible to brown rot,respectively,to perform a dual RNA-Seq *** immature fruit,host responses,pathogen biomass,and pathogen transcriptional activity peaked at 14–24 h post inoculation(hpi),at which point *** appeared to switch its transcriptional response to either quiescence or *** fruit experienced an exponential increase in host and pathogen activity beginning at 6 *** analyses in both host and pathogen highlighted differences in stage-dependent *** example,in immature fruit,*** unsuccessfully employed carbohydrate-active enzymes(CAZymes)for penetration,which the fruit was able to combat with tightly regulated hormone responses and an oxidative burst that challenged the pathogen’s survival at later time *** contrast,in mature fruit,*** was more dependent on proteolytic effectors than CAZymes,and was able to invest in filamentous growth early during the *** analyses of mature fruit infected with *** indicated that,while jasmonic acid activity was likely useful for defense,high ethylene activity may have promoted susceptibility through the induction of ripening ***,we identified *** genes that were highly induced in both quiescent and active infections and may serve as targets for control of brown rot.

Gut Lymphangiopathy: Adding Fuel to the Fire in Chronic Liver Disease

The splanchnic inflammation inchronic liver disease increases intestinal angiogenesis. In the current study our aim was demonstrating that the small bowel lymphangiogenesis is associated with angiogenesis in chronic cholestasis in the rat. A stereological study of the lymphatic microcirculation in the small intestine was performed in cholestatic rats. Portal enteropathy in cholestasis increases lymphatic microvessels in the mucosa and submucosa layers. Thus, the lymphatic microvessel volume fraction was superior (p < 0.001) in the mucosa (0.16 ± 0.01) and submucosa (0.16 ± 0.01), in regard to the muscle layer 0.015 ± 0.01. The lymphatic microvessel length density was higher in the mucosa (76.89 ± 2.86 mm-2;p -2;p < 0.01), in relationship to the muscle layer (5.04 ± 2.92 mm-2). These alterations predominate in the duodenum (volume fraction: 0.10 ± 0.01 and length density: 33.55 ± 5.98 mm-2) and ileum (volume fraction: 0.16 ± 0.01 and length density: 38.62 ± 6.07 mm-2). This study demonstrates the predominance of an increased lymphangiogenic response in both end sides of the small bowel associated with chronic liver disease. Since the porto-systemic venous collateral circulation in the chronic liver insufficiency is developed in the ends of the gastrointestinal tract, the excessive duodeno-ileal lymphangiogenesis could suggest the development of amesenteric-systemic lymphatic bypass in the chronic portal hypertension.

Proteomics Analysis Reveals Post-Translational Mechanisms for Cold-Induced Metabolic Changes in Arabidopsis

Cold-induced changes of gene expression and metabolism are critical for plants to survive freezing. Largely by changing gene expression, exposure to a period of non-freezing low temperatures increases plant tolerance to freezing--a phenomenon known as cold acclimation. Cold also induces rapid metabolic changes, which provide instant protection before temperature drops below freezing point. The molecular mechanisms for such rapid metabolic responses to cold remain largely unknown. Here, we use two-dimensional difference gel electrophoresis （2-D DIGE） analysis of sub-cellular fractions ofArabidopsis thaliana proteome coupled with spot identification by tandem mass spectrometry to identify early cold-responsive proteins in Arabidopsis. These proteins include four enzymes involved in starch degradation, three HSP100 proteins, several proteins in the tricarboxylic acid cycle, and sucrose metabolism. Upon cold treatment, the Disproportio- nating Enzyme 2 （DPE2）, a cytosolic transglucosidase metabolizing maltose to glucose, increased rapidly in the centrifugation pellet fraction and decreased in the soluble fraction. Consistent with cold-induced inactivation of DPE2 enzymatic activity, the dpe2 mutant showed increased freezing tolerance without affecting the C-repeat binding transcription factor （CBF） transcriptional pathway. These results support a model that cold-induced inactivation of DPE2 leads to rapid accumulation of maltose, which is a cold-induced compatible solute that protects cells from freezing damage. This study provides evidence for a key role of rapid post-translational regulation of carbohydrate metabolic enzymes in plant protection against sudden temperature drop.

Melatonin Avoids Anatomofunctional Changes Associated to Aging in a Rat Model

Melatonin is a hormone synthesized and released primarily by the pineal gland. Its secretion fol-lows a circadian rhythm with a peak overnight. Its secretion is initiated approximately to the three months of age and continues to rise during the childhood. Previous to the puberty there is a de-crease of melatonin secretion that continues until the old age. Melatonin has effects in the body and acts through at least four mechanisms: membrane receptors, orphan nuclear receptors, calmodulin and free radicals. It has been suggested that aging can be a consequence of the oxidation of cells that eventually become vulnerable to injury and die. This work reviews the antioxidant effects of melatonin in a rodent model, on the formation of free radicals, on the MAP2 protein expression and on the electrophysiology of the hippocampus at different ages. The results indicate that melatonin maintains in a “best” state to the experimental animals compared to controls. It suggests the use of melatonin as a therapy to prevent or delay the aging effects on the cells.

目的:比较圆锥角膜患者角膜交联(CXL)、等渗(IR)和低渗核黄素(HR)的结果。方法:回顾性研究,包括29例29眼圆锥角膜患者行角膜交联术,IR组15眼,HR组14眼。参数分析包括(随访1a):最佳矫正视力(BCVA),球体和柱体,中央和较薄处角膜厚度,平均和最大角膜曲率(分别为Km和Kmax),并发症及进展。结果:观察到两组的BCVA提高:治疗前IR组为0.26±0.57,HR组为0.47±0.72。术后1a,IR组为0.13±0.79,HR组为0.29±1.52,IR组BCVA更高(P=0.018)。术后1mo,两组中央角膜厚度(μm)均减少,但在接下来的数月中增加:术前IR组为497±28μm,HR组为432±14μm;术后1a,IR组为480±31μm,HR组为424±15μm。较薄处角膜厚度也是如此,术前IR组为487±29μm,HR组为410±20μm;术后1a,IR组为468±33μm,HR组为413±13μm。两组中Km和Kmax均降低。每组6眼呈现短暂的混浊,无持续性混浊。结论:在中央角膜厚度<400μm的患者中,HR的使用似乎是一个有效的替代执行传统CXL的技术。

Novel suturing technique,based on physical principles,achieves a breaking point double that obtained by conventional techniques

BACKgrOUND Sutures have been used to repair wounds since ancient ***,the basic suture technique has not significantly *** Phase I of our project,we proposed a“double diabolo”suture design,using a theoretical physical study to show that this suture receives 50%less tension than conventional sutures,and so a correspondingly greater force must be applied to break *** To determine whether these theoretical levels of resistance were met by the new type of *** An observational study was performed to compare three types of sutures,using a device that exerted force on the suture until the breaking point was *** tension produced by this traction was *** following variables were considered:Tearing stress on entry/exit points,edge separation stress,and suture break *** study sample consisted of 30 sutures with simple interrupted stitches(group 1),30 with continuous stitches(group 2),and 30 with the“double diabolo”design(group 3).RESULTS The mean degree of force required to reach the breaking point for each of these variables(tearing,separation,and final breaking)was highest in group 3(14.56,18.28,and 21.39 kg),followed by group 1(7.36,10.38,and 12.81 kg)and group 2(5.77,7.7,and 8.71 kg).These differences were statistically significant(Pgreater force must be applied to reach the breaking point(almost twice as much as in the simple interrupted suture and more than double that required for the continuous suture).If these results are confirmed in Phase III(the clinical phase)of our study,we believe the double diabolo technique should be adopted as the standard approach,especially when the suture must withstand significant tension(e.g.,laparotomy closure,thoracotomy closure,diaphragm suture,or hernial orifice closure).

Management of pancreatic cancer in the elderly

Currently, pancreatic adenocarcinoma mainly occurs after 60 years of age, and its prognosis remains poor despite modest improvements in recent decades. The aging of the population will result in a rise in the incidence of pancreatic adenocarcinoma within the next years. Thus, the management of pancreatic cancer in the elderly population is gaining increasing relevance. Older cancer patients represent a heterogeneous group with different biological, functional and psychosocial characteristics that can modify the usual management of this disease, including pharmacokinetic and pharmacodynamic changes, polypharmacy, performance status, comorbidities and organ dysfunction. However, the biological age, not the chronological age, of the patient should be the limiting factor in determining the most appropriate treatment for these patients. Unfortunately, despite the increased incidence of this pathology in older patients, there is an underrepresentation of these patients in clinical trials, and the management of older patients is thus determined by extrapolation from the results of studies performed in younger patients. In this review, the special characteristics of the elderly, the multidisciplinary management of localized and advanced ductal adenocarcinoma of the pancreas and the most recent advances in the management of this condition will be discussed, focusing on surgery, chemotherapy, radiation and palliative care.

About an Ambiguity Appearing on the Hamiltonian Formulation of Quantum Mechanics

We study classical and quantum dynamics of the symmetric harmonic oscillator and the symmetric bouncer defined in 2-D. For these systems we get each of them for two different constants of motion, two Lagrangians and two Hamiltonians which describe the same classical dynamics. However, the quantization of these systems (using Schrödinger equation), using their two equivalents Hamiltonian, describes different quantum dynamics for each of them. This represents an ambiguity on the Hamiltonian formulation of the Quantum Mechanics.
