Fine mapping of susceptibility genes by Lewontin's linkage disequilibrium measure with application to Alzheimer's disease

Objectives To formulate an equation for fine mapping of disease loci under complex conditions and determine the marker-disease distance in a specific case using this equation. Methods Lewontin’s linkage disequilibrium (LD) measure D’ was used to formulate an equation for mapping disease genes in the presence of phenocopies, locus heterogeneity, gene-gene and gene-environment interactions, incomplete penetrance, uncertain liability and threshold, incomplete initial LD, natural selection, recurrent mutation, high disease allele frequency and unknown mode of inheritance. This equation was then used to determine the distance between a marker (ε4 within the apolipoprotein E gene, APOE) and Alzheimer’s disease (AD) loci using published *** An equation was formulated for mapping disease genes under the above conditions. If these conditions are present but ignored, then recombination fraction θ between marker and disease loci will be either overestimated or estimated with little bias. Therefore, an upper limit of θ can be obtained. AD has been found to be associated with the marker allele ε4 in Africans, Asians, and Caucasians. This suggests that the AD-ε4 allelic LD predates the divergence of peoples occurring 100?000 years ago. With the age of AD-ε4 allelic LD so estimated, the maximal distance was calculated to be 23.2 kb (mean 5.8 kb). Conclusions (1) A method is developed for LD mapping of susceptibility genes. (2) A mutation within the APOE gene itself, among others, is responsible for the susceptibility to AD, which is supported by recent evidence from studies using transgenic mice.

Higher Groundwater Selenium Exposure Is Associated with Better Memory: A Project FRONTIER Study

Background: Exposure to elements in groundwater (toxic or beneficial) is commonplace yet, outside of lead and mercury, little research has examined the impact of many commonly occurring exposures on mental abilities during the aging process. Selenium has antioxidant properties as part of the glutathione peroxidase system and may have protective effects on memory abilities. Objectives: To investigate the relationship of groundwater selenium exposure to neuropsychological status. Method: Analysis of data from 484 participants (148 men and 336 women) of Project FRONTIER, a community-based participatory research study of the epidemiology of health issues of rural-dwelling adults and elders. Results: Estimated selenium exposure (current and long-term) was specifically related to memory functioning without relation to other neurocognitive domains. The significant, positive link between selenium and memory (Immediate and Delayed) scores held regardless of APOE4 status as well as when the sample was restricted to only those without cognitive dysfunction. Current selenium was also associated with significantly reduced risk of cognitive decline prospectively. Conclusions: Higher selenium levels were associated with better memory functioning as well as reduced risk of cognitive decline among this community-based sample. Given the antioxidant properties of selenium, and the well-documented link between oxidative stress and the development of cognitive dysfunction and Alzheimer’s disease, additional research is necessary to determine the utility of groundwater selenium monitoring as a potential population-wide prevention effort against Alzheimer’s disease.

To the Editor： Pseudohypoaldosteronism Type 11 （PHAII）, also known as gordon syndrome, is a rare autosomal disease, caused by mutations in WNK1, WNK4, CUL3, or KLHL3 genes. Hitherto, about 200 individuals and families have been reported with PHAII; of these, 26 were caused by CUL3 mutation.[1,2] In pediatrics, hyperkalemia-complicated hypertension is rare, and PHAII should be considered in this condition.

本文对Neumann边界条件下的sine-gordon方程提出两类新的全离散高效保能量算法.首先考虑在两种不同空间网格上应用cosine拟谱方法去发展空间保结构格式,导出两个有限维Hamilton常微分方程系统.然后,将预估校正型的Crank-Nicolson格式和投影方法相结合,得到一类全离散保能量算法.另外,本文对sine-gordon方程引入一个补充变量,将原始模型转化成一个松弛系统,这使得保结构算法更容易被发展.本文针对等价的松弛系统仍采用cosine拟谱方法和预估校正的CrankNicolson格式进行离散,发展了另一类新的保能量算法.本文提出的数值格式不仅保持系统的原始能量,而且可以通过离散cosine变换进行高效快速求解.最后,数值实验验证了格式的数值精度、计算效率和优秀性态.

Establishment of a hyperspectral evaluation model of ocean color satellite-measured reflectance

The accuracy of the satellite-measured reflectance is a key question for data processing and oceanographic applications.A hyperspectral satellite remote sensing reflectance evaluation model(HRSREM) was developed to evaluate the accuracy of the satellite measured *** model can compute the total reflectance at the top of the atmosphere(TOA) according to observation conditions of satellites,based on a radiative transfer model with the consideration of multiple scattering effects and atmospheric absorption *** performance of the HRSREM model was examined by gordon’s algorithms,showing that the relative errors of the Rayleigh scattering reflectance and the aerosol scattering reflectance are less than 2%.The model can also compute the sky reflectance which can be validated by in-situ *** two sky reflectances match well with a spectral average error of 5.4%.The relative error of the total reflectance of the model,verified by sea-viewing wide field-of-view sensor(SeaWiFS) data,is about 3.5%.Therefore,the total reflectances at TOA,computed by the model,can be taken as reference values to evaluate the accuracy of satellite *** model was used to evaluate the accuracy of the hypersptral satellite(Hyperion) remote sensing *** Hyperion reflectance matches the total reflectance of HRSREM very well at visible and near-infrared bands with an average error of 7.3%,while the status of calibration coefficients at shortwave infrared bands are not stable with a large spectral average error of 63.5%.The reflectance evaluation of a moderate resolution imaging spectrometer(CMODIS) data indicated that relative errors are large,especially at near-infrared bands with relative errors more than 100%.The calibration coefficients of CMODIS,obtained from laboratory measurements,are not *** CMODIS data should be recalibrated for oceanographic *** performance of the HRSREM model is effective in evaluating satellite data and its

偏微分方程求解是计算流体力学等科学与工程领域中数值分析的计算核心。由于物理的多尺度特性和对离散网格质量的敏感性,传统的数值求解方法通常包含复杂的人机交互和昂贵的网格剖分开销,限制了其在许多实时模拟和优化设计问题上的应用效率。提出了一种改进的基于深度神经网络的偏微分方程求解方法TaylorPINN。该方法利用深度神经网络的万能逼近定理和泰勒公式的函数拟合能力,实现了无网格的数值求解过程。在Helmholtz、Klein-gordon和Navier-Stokes方程上的数值实验结果表明,TaylorPINN能够很好地拟合计算域内时空点坐标与待求函数值之间的映射关系,并提供了准确的数值预测结果。与常用的基于物理信息神经网络方法相比,对于不同的数值问题,TaylorPINN将预测精度提升了3~20倍。
