Sexual dimorphism in immune response： Testing the hypothesis in an insect species with two male morphs

It has been proposed that given that males should invest in sexual traits at the expense of their investment in immune response, females are better immunocompetent than males. Typically, this idea has been tested in monomorphic species, but rarely has been evaluated in polymorphic male species. We used Paraphlebia zoe, a damselfly with two male morphs： the black-winged morph （Black-W） develop black spots as sexual traits and the hyaline-winged morph （Hyaline-W） resembles a female in size and wings color. We predicted that Black-W should have a lower immune response than Hyaline-W, but that the latter males should not differ from females in this respect. Nitric oxide （NO） and phenoloxidase （PO） production, as well as hemolymph protein content, were used as immune markers. Body size （wing length） was used as an indicator of the male condition. The results show that, as we predicted, females and Hyaline-W had higher values of NO than Black-W, corresponding to differences in size. However, the opposite was found in relation to PO production. Females had the highest levels of hemolymph protein content, whereas no differences were found between Black-W and Hyaline-W. These results partially support the sexual selection hypothesis and are discussed in the context of the life history of this species. Black-W, Hyaline-W, and females could express the immune markers that are prioritized by their particular condition, and probably neither of them could express all immune markers in an elevated manner, as this would result in an excessive accumulation of free radicals.

Background: The protective role of (poly)phenols against metabolic disorders has been extensively studied in adults but not in ***: To assess associations of dietary (poly)phenols and their subclasses with cardiometabolic health parameters in adolescents. Methods: A cross-sectional study was conducted in 944 individuals aged 11 to 14 years enrolled in the SI! Program for Secondary Schools trial (NCT03504059). (Poly)phenol intake was assessed using semi-quantitative food frequency questionnaires and the Phenol-Explorer database. The measured cardiometabolic parameters were waist circumference (WC) age-sex z-score， blood pressure (BP) age-sex z-score， blood glucose (BG)， triglycerides (TG)， and high-density lipoprotein cholesterol (HDL-c). Multilevel mixed-effect linear regression models were applied to examine the association between (poly)phenol quintiles and cardiometabolic health ***: Compared to the lowest quintile， adolescents in the highest quintile of total (poly)phenol intake had lower WC z-scores (β=-0.09， 95%CI=-0.09;-0.08， P-for trend=0.002)， mean arterial pressure z-scores (β =-0.11， 95%CI=-0.12;-0.09， P-for trend=0.206)， and HDL-c (β=-1.52， 95%CI=-2.90;-0.14， P-for trend=0.108)， after multivariable adjustment. The WC z-scores (β=-0.11， 95%CI=-0.19;-0.03， P-for trend=0.013) and HDL-c (β=-2.16， 95%CI=-3.78;-0.53， P-for trend=0.046) were lower in the highest quintile of flavonoid intake compared to the lowest quintile. The highest quintile of phenolic acid intake was associated with a lower WC z-score (β=-0.14， 95%CI=-0.26;-0.02， P-for trend=0.011) and TG levels (β=-8.83， 95%CI=-12.37;-5.30， P-for trend <0.001)， and the highest quintile of stilbene intake with lower BG (β=-1.83， 95%CI= -3.103;-0.64， P-for trend <0.001) and TG (β=-3.22， 95%CI= -4.95;-1.49， P-for trend=0.034)， and with higher HDL-c (β=2.328， 95%CI= 1.40;3.17， P-for trend=0.466) compared to the lowest quintile. Molecular and in vitro Characterization of Field Isolates of Bovine Herpesvirus-1

Bovine Herpesvirus-1 (BoHV-1) is distributed worldwide and is a major pathogen in cattle, being the causal agent of a variety of clinical syndromes. The aim of this study was to isolate and to characterize (molecular and biological characterization) BoHV-1 from 29 immunosuppressed animals. It was possible to obtain 18 isolates, each from a different animal, such as from the respiratory and reproductive tracts. In some cases the cytopathic effect was visible 12 hours post-inoculation, and became characteristic after 36-48 hours. Biological characteristics were evaluated and compared with Iowa and Colorado-1 reference strains, and differences were found in plaque size, virus titer measured by TCID50 and PFU/mL, and one step virus curves. These results showed that some isolates had a highly virulent-like behavior in vitro, compared to the reference strains, with shorter eclipse periods, faster release of virus into the supernatants, and higher burst size and viral titer. There were no differences in glycoprotein expression of BoHV-1 isolates, measured by Western blot on monolayers. Moreover, using restriction endonucleases analysis, most of the viruses were confirmed as BoHV-1.1 and just one of them was confirmed as BoHV-1.2a subtype. These findings suggest that some wild-type BoHV-1 isolates could be useful as seeds to develop new monovalent vaccines.

Human CD4 ＋ effector T lymphocytes generated upon TCR engagement with self-peptides respond defectively to IL-7 in their transition to memory cells

The peripheral repertoire of CD4＋ T lymphocytes contains autoreactive cells that remain tolerant through several mechanisms. However, nonspecific CD4＋ T cells can be activated in physiological conditions as in the course of an ongoing immune response, and their outcome is not yet fully understood. Here, we investigate the fate of human naive CD4＋ lymphocytes activated by dendritic cells （DCs） presenting endogenous self-peptides in comparison with lymphocytes involved in alloresponses. We generated memory cells （Tmem） from primary effectors activated with mature autologous DCs plus interleukin （IL）-2 （Tmauto）, simulating the circumstances of an active immune response, or allogeneic DCs （Tmallo）. Tmem were generated from effector cells that were rested in the absence of antigenic stimuli, with or without IL-7. Tmem were less activated than effectors （demonstrated by CD25 downregulation） particularly with IL-7, suggesting that this cytokine may favour the transition to quiescence. Tmauto and TmaHo showed an effector memory phenotype, and responded similarly to polyclonal and antigen-specific stimuli. Biochemically, IL-7-treated Tma^o were closely related to conventional memory lymphocytes based on Erk-l/2 activation, whereas Tmauto were more similar to effectors. Autologous effectors exhibited lower responses to IL-7 than allogeneic cells, which were reflected in their reduced proliferation and higher cell death. This was not related to IL-7 receptor expression but rather to signalling deficiencies, according to STAT5 activation These results suggest that ineffective responses to IL-7 could impair the transition to memory cells of naive CD4＋ T lymphocytes recognizing self-peptides in the setting of strong costimulation.

The effect of water deficit was determined on both in vitro and soil seedling as well as in cells in suspension of Agave americana *** order to do the establishment of cells,the formation of callus was induced;for it two auxins were evaluated:2,4-dichlorophenoxyacetic acid(2,4-D)and 4-mino-3,5,6-trichloropicolinic acid(picloram)at three concentrations(0.25,0.5 and 0.75 mg L−1)in three explants(leaf,root and meristems)cultured in MS semisolid *** callogenesis response was related to the type and section of the explant,as well as the regulator used,and a cell suspension was established using 0.5 mg L−1 naphthaleneacetic acid(NAA)+0.5 mg L−1 Benzylaminopurine(BAP).Seedlings were exposed to polyethyleneglycol(15%and 30%w/v)with a water potential of−0.87 and−2.67 MPa,respectively,under soil *** stress was applied through restricted *** weight,root system growth,and chlorophyll concentration were some of the parameters that were affected by the effect of water deficit on *** *** concentration values were significantly decreased by 15 at 30%PEG(19.6 SPAD units)compared to the control *** in vitro plants,the highest concentration of proline was found in the roots,being the treatment with 30%polyethylene glycol where the highest concentration of this osmoregulator was obtained(62.5 mg g−1 DW).Under restricted irrigation conditions,an increase in proline concentration was observed both in the aerial part(2.2μg 100 g−1 DW)and in the root system(1.8μg 100 g−1 DW).However,the concentrations found were approximately ten times greater,less than those found under in vitro ***,the accumulation of proline can be considered an indicator of stress in Agave Americana *** in vitro.

Photoacoustic Studies of Colloidal Silica Particles after MeV Ion-Induced Shape Deformation

Ordered arrays of colloidal submicrometer-sized silica particles deposited onto silicon wafers were irradiated with MeV Si ions. The spherical silica particles turned into oblate particles as a result of the increase of the particle dimension perpendicular to the ion beam direction and the decrease in the parallel direction. Pulsed laser photoacoustic spectroscopy was used to study the structural changes of the silica particles after the ion-induced shape deformation. Our purpose is to correlate the mechanical vibrations generated by the pulsed laser as a function of the Si irradiation parameters: ion energy and fluence. Fast Fourier transform analysis of the photoacoustic signal was carried out in order to obtain the normal vibration modes of the system. The size, size distribution and shape of the silica particles were determined by scanning electron microscopy. Our results revealed significant structural differences between the spherical and the deformed silica particles.

Comparison of Secnidazole and Fenbendazole for the Treatment of Asymptomatic Giardia Infection in Dogs

The objetive of this study was to compare a single dose of secnidazole versus multiple doses of fenbendazole for the treatment of dogs with asymptomatic Giardia *** and methods:Twenty-four asymptomatic dogs with a positive test result for Giardia spp were randomized in two equal groups to receive a single dose of secnidazole at 30 mg/kg PO,or fenbendazole at 50 mg/kg PO q24h for 3 *** parameters were evaluated before and 8 days after treatment,and feces were re-examined at days 8,15,and 30 post-treatment by fecal flotation and antigen ***:The number of positive dogs in the fenbendazole group was:1(day 8)and 3(days 15 and 30).In the secnidazole group,the number of positive cases were:4(day 8),3(day 15),and 1(day 30).Conclusion:Treatment with secnidazole or fenbendazole,were effective between 75%and 92%to eliminate the excretion of Giardia cysts in canines together with hygienic measures to control,like disinfection with quaternary ammonium of patients and their *** studies that include more animals and multiple fecal exams on consecutive days would be necessary to confirm its efficacy in dogs.

AIM： To analyze the polygraphic sleep patterns during cirrhosis progression in a rat model by repeated CCh administration. METHODS： Male Wistar rats received three weekly injections of CCl4 for 11 wk, and were analyzed before and during the induction of cirrhosis. Rats were im- planted with electrodes to record their sleep patterns. Polygraph recordings were made weekly over 11 wk for 8 h, during the light period. After a basal recording, rats received three weekly injections of CCl4. Histological confirmation of cirrhosis was performed after 11 wk. RESULTS： The results showed a progressive decrease in total wake time that reached statistical significance from the second week of treatment. In addition, there was an increase in total time of slow wave sleep （SWS）Ⅱ and rapid eye movement sleep （REM sleep） in most of the 11 wk. SWS I showed no significant variations. During the final weeks, a significant increase in REM sleep frequency was also observed. Histological analyses of the livers showed unequivocal signs of cirrhosis. CONCLUSION： These data suggest that hepatic failure produced by CCh administration is capable of modifying the sleep pattern even after only a few doses.
