Genome-wide association analysis reveals regulation of at-risk loci by DNA methylation in prostate cancer

Epigenetic changes are potentially important for the ontogeny and progression of tumors but are not usually studied because of the complexity of analyzing transcript regulation resulting from epigenetic *** cancer(PCa)is characterized by variable clinical manifestations and frequently unpredictable *** performed an expression quantitative trait loci(eQTL)analysis to identify the genomic regions that regulate gene expression in PCa and identified a relationship between DNA methylation and clinical *** multi-level information published in The Cancer Genome Atlas,we performed eQTL-based analyses on DNA methylation and gene *** better interpret these data,we correlated loci and clinical indexes to identify the important loci for both PCa development and *** data demonstrated that although only a small proportion of genes are regulated via DNA methylation in PCa,these genes are enriched in important cancer-related *** addition,single nucleotide polymorphism analysis identified the locations of CpG sites and genes within at-risk loci,including the 19q13.2-q13.43 and 16q22.2-q23.1 ***,an epigenetic association study of clinical indexes detected risk loci and pyrosequencing for site *** DNA methylation-regulated genes across PCa samples are a small proportion,the associated genes play important roles in PCa carcinogenesis.

Balanced and Unbalanced Components of moist Atmospheric Flows with Phase Changes

Atmospheric variables(temperature, velocity, etc.) are often decomposed into balanced and unbalanced components that represent low-frequency and high-frequency waves, respectively. Such decompositions can be defined, for instance, in terms of eigenmodes of a linear operator. Traditionally these decompositions ignore phase changes of water since phase changes create a piecewise-linear operator that differs in different phases(cloudy versus non-cloudy). Here we investigate the following question: How can a balanced–unbalanced decomposition be performed in the presence of phase changes? A method is described here motivated by the case of small Froude and Rossby numbers,in which case the asymptotic limit yields precipitating quasi-geostrophic equations with phase changes. Facilitated by its zero-frequency eigenvalue, the balanced component can be found by potential vorticity(PV) inversion, by solving an elliptic partial differential equation(PDE), which includes Heaviside discontinuities due to phase changes. The method is also compared with two simpler methods: one which neglects phase changes, and one which simply treats the raw pressure data as a streamfunction. Tests are shown for both synthetic, idealized data and data from Weather Research and Forecasting(WRF) model simulations. In comparisons, the phase-change method and no-phase-change method produce substantial differences within cloudy regions, of approximately 5K in potential temperature, due to the presence of clouds and phase changes in the data. A theoretical justification is also derived in the form of a elliptic PDE for the differences in the two streamfunctions.

The interaction of calf thymus DNA（ctDNA） with berberine（Ber） was systematically investigated via UV-Vis,fluorescence and phosphorescence spectroscopies in aqueous solution and on solid *** very weak fluorescence intensity of Ber at 525 nm in aqueous solution can be greatly enhanced by *** UV-Vis spectrum shows that Ber could bind with DNA by *** addition,strong fluorescence of Ber alone could be observed on filter paper substrate and can be quenched by ctDNA effectively,and the increase of fluorescence polarization of Ber on the filter paper implies the intercalation *** emits phosphorescence at 619 nm in the presence of thallium（I） *** lifetime of Ber increased from 38.4 ms to 43.4 ms with the increase of *** increase of lifetime is possibly attributed to the increase of rigidity of Ber after its intercalating into the ctDNA base *** explanation is also augmented by the anionic quenching *** behaves as a typical intercalating agent into *** intrinsic binding constant between Ber and ctDNA is（1.84±0.12）×10^4 L/mol and n=0.16.

A canonical correlation analysis is a generic parametric model used in the statistical analysis of data involving interrelated or interdependent input and output *** is especially useful in data analytics as a dimensional reduction strategy that simplifies a complex,multidimensional parameter space by identifying a relatively few combinations of variables that are maximally *** shortcoming of the canonical correlation analysis,however,is that it provides only a linear combination of variables that maximizes these *** this in mind,we describe here a versatile,monte-Carlo based methodology that is useful in identifying non-linear functions of the variables that lead to strong input/output *** demonstrate that our approach leads to a substantial enhancement of correlations,as illustrated by two experimental applications of substantial interest to the materials science community,namely:(1)determining the interdependence of processing and microstructural variables associated with doped polycrystalline aluminas,and(2)relating microstructural decriptors to the electrical and optoelectronic properties of thin-film solar cells based on CuInSe_(2) ***,we describe how this approach facilitates experimental planning and process control.

Influence of climatic conditions,topography and soil attributes on the spatial distribution of site productivity index of the species rich forests of Jalisco,mexico

This paper presents an approach based on field data to model the spatial distribution of the site productivity index （SPI） of the diverse forest types in Jalisco, mexico and the response in SPI to site and cli-matic conditions. A linear regression model was constructed to test the hypothesis that site and climate variables can be used to predict the SPI of the major forest types in Jalisco. SPI varied significantly with topog-raphy （elevation, aspect and slope）, soil attributes （pH, sand and silt）, climate （temperature and precipitation zones） and forest type. The most important variable in the model was forest type, which accounted for 35% of the variability in SPI. Temperature and precipitation accounted for 8 to 9% of the variability in SPI while the soil attributes accounted for less than 4% of the variability observed in SPI. No significant differences were detected between the observed and predicted SPI for the individual forest types. The linear regression model was used to develop maps of the spatial variability in predicted SPI for the individual forest types in the state. The spatial site productivity models developed in this study provides a basis for understanding the complex relationship that exists between forest productivity and site and climatic conditions in the state. Findings of this study will assist resource managers in making cost-effective decisions about the management of individual forest types in the state of Jalisco, mexico.

Clones identification of Sequoia sempervirens (D. Don) Endl. in Chile by using PCR-RAPDs technique

A protocol of polymerase chain reaction-random amplified polymorphic DNAs (PCR-RAPDs) was established to analyse the gene diversity and genotype identification for clones of Sequoia sempervirens (D. Don) Endl. in Chile. Ten (out of 34) clones from introduction trial located in Voipir-Villarrica, Chile, were studied. The PCR-RAPDs technique and a modified hexadecyltrimethylammonium bromide (CTAB) protocol were used for genomic DNA extraction. The PCR tests were carried out employing 10-mer random primers. The amplification products were detected by electrophoresis in agarose gels. Forty nine polymorphic bands were obtained with the selected primers (BG04, BF07, BF12, BF13, and BF14) and were ordered according to their molecular size. The genetic similarity between samples was calculated by the Jaccard index and a dendrogram was con- structed using a cluster analysis of unweighted pair group method using arithmetic averages (UPGmA). Of the primers tested, 5 (out of 60) RAPD primers were selected for their reproducibility and high polymorphism. A total of 49 polymorphic RAPD bands were detected out of 252 bands. The genetic similarity analysis demonstrates an extensive genetic variability between the tested clones and the dendrogram depicts the genetic relationships among the clones, suggesting a geographic relationship. The results indicate that the RAPD markers permitted the identification of the assayed clones, although they are derived from the same geo- graphic origin.

Submillimeter interferometry has the potential to image supermassive black holes on event horizon scales,providing tests of the theory of general relativity and increasing our understanding of black hole accretion *** Event Horizon Telescope(EHT) performs these observations from the ground,and its main imaging targets are Sagittarius A~* in the Galactic Center and the black hole at the center of the m87 ***,the EHT is fundamentally limited in its performance by atmospheric effects and sparse terrestrial(u,v)-coverage(Fourier sampling of the image).The scientific interest in quantitative studies of the horizon size and shape of these black holes has motivated studies into using space interferometry which is free of these *** resolution considerations and interstellar scattering effects push the desired observing frequency to bands above 500 *** paper presents the requirements for meeting these science goals,describes the concept of interferometry from Polar or Equatorial medium Earth Orbits(PECmEO) which we dub the Event Horizon Imager(EHI),and utilizes suitable space technology *** this concept,two or three satellites orbit at slightly different orbital radii,resulting in a dense and uniform spiral-shaped(u,v)-coverage over *** local oscillator signals are shared via an inter-satellite link,and the data streams are correlated on-board before final processing on the ***-satellite metrology and satellite positioning are extensively employed to facilitate the knowledge of the instrument position vector,and its time *** European space heritage usable for both the front ends and the antenna technology of such an instrument is *** and future sensors for the required inter-satellite metrology are *** performance estimates and simulation results are given.

Dual function of a high-contrast hydrophobic-hydrophilic coating for enhanced stability of perovskite solar cells in extremely humid environments

In spite of a continuous increase in their power conversion efficiency （PCE） and an economically viable fabrication process,organic-inorganic perovskite solar cells （PSCs） pose a significant problem when used in practical applications：They show fast degradation of the PCE when exposed to very humid *** this study,the stability of PSCs under very humid conditions is greatly enhanced by coating the surface of the PSC devices with a multi-layer film consisting of ultrahydrophobic and relatively hydrophilic layers.A hydrophobic composite of poly（methyl methacrylate）（PmmA）,polyurethane （PU）,and SiO2 nanoparticles successfully retards the water molecules from very humid ***,the hydrophilic layer with moderately PmmA captures the residual moisture within the perovskite layer;subsequently,the perovskite layer *** dual function of the coating film keeps the PCE of PSCs at 17.3% for 180 min when exposed to over 95% humidity.

Ammonium metavanadate:A novel catalyst for synthesis of 2-substituted benzimidazole derivatives

Ammonium metavanadate （10 mol%） was found to be a useful catalyst for the synthesis of various 2-substituted aryl benzimidazoles. It was used as an oxidizing agent for the condensation of o-phenylenediamine with different substituted aryl aldehydes at room temperature in ethanol. The method was proved to be simple, convenient and the product was isolated with good yields （79-91%）.

Deconvolving the pre-Himalayan Indian margin-Tales of crustal growth and destruction

The metamorphic core of the Himalaya is composed of Indian cratonic rocks with two distinct crustal affinities that are defined by radiogenic isotopic geochemistry and detrital zircon age spectra. One is derived predominantly from the Paleoproterozoic and Archean rocks of the Indian cratonic interior and is either represented as metamorphosed sedimentary rocks of the Lesser Himalayan Sequence(LHS) or as slices of the distal cratonic margin. The other is the Greater Himalayan Sequence(GHS) whose provenance is less clear and has an enigmatic affinity. Here we present new detrital zircon Hf analyses from LHS and GHS samples spanning over 1000 km along the orogen that respectively show a striking similarity in age spectra and Hf isotope ratios. Within the GHS, the zircon age populations at 2800-2500 ma,1800 ma, 1000 ma and 500 ma can be ascribed to various Gondwanan source regions; however, a pervasive and dominant Tonianage population(～860-800 ma) with a variably enriched radiogenic Hf isotope signature(eHf = 10 to-20) has not been identified from Gondwana or peripheral accreted terranes. We suggest this detrital zircon age population was derived from a crustal province that was subsequently removed by tectonic erosion. Substantial geologic evidence exists from previous studies across the Himalaya supporting the Cambro-Ordovician Kurgiakh Orogeny. We propose the tectonic removal of Tonian lithosphere occurred prior to or during this Cambro-Ordovician episode of orogenesis in a similar scenario as is seen in the modern Andean and Indonesian orogenies, wherein tectonic processes have removed significant portions of the continental lithosphere in a relatively short amount of time. This model described herein of the pre-Himalayan northern margin of Greater India highlights the paucity of the geologic record associated with the growth of continental crust.
