AIM: To examine the potency of 1,2,3,4,6-penta-O-galloyl-β-D-glucose (PGG) as a hepatic heme oxygen-ase-1(HO-1) inducer and its regulation in HepG2 cells. METHODS: Expression of HO-1 and NF-E2-related factor 2 (Nrf2) and activation of mitogen-activated protein (MAP) kinases were analyzed by Western blot, immuno-fluorescence assay, and flow cytometry. Transfections of HO-1 gene, small interfering RNAs for HO-1 and Nrf2, and dominant-negative gene for MAP/extracellular signal-regulated kinase (ERK) were carried out to dissect the signaling pathways leading to HO-1 expression in HepG 2 cells. REsuLTS: PGG up-regulated HO-1 expression and this expression conferred cytoprotection against oxidative injury induced by t-butyl hydroperoxide. Moreover, PGG induced Nrf2 nuclear translocation, which was found to be an upstream step of PGG-induced HO-1 expression, and ERK activation, of which pathway was involved in PGG-induced Nrf2 nuclear translocation, HO-1 expression and cytoprotection. CONCLUSION: PGG up-regulates HO-1 expression by stimulating Nrf2 nuclear translocation in an ERK-depen-dent manner, and HO-1 expression by PGG may serve as one of the important mechanisms for its hepatoprotective effects.

Case Report:Congenital atresia of the left main coronary artery in an infant

Congenital atresia of the left main coronary artery is a rare occurrence, and surgical revascularizationbypass graft is required. We here report a rare case of congenital coronary anomaly in an infant. A lO-month-old male infant was admitted to the hospital with heart failure symptoms. Echocardiographic examinations revealed mitral valve regurgitation and ischemic changes of the anterolateral atresia of the left main coronary artery with a severe papillary muscle and chordae. Coronary angiography showed hypoplastic left anterior descending artery and a circumflex coronary artery. Unfortunately, sudden cardiac arrest occurred after catheterization and the infant did not recover despite of immediate cardiopulmonary resuscitation. Further studies are needed to find a newer diagnostic method to detect coronary anomaly in an infant, and coronary angiography, if necessary, has to be performed very carefully.

Determining whether the high-latitude Bering land bridge(BLB)was ecologically suitable for the migration of mesothermal plants is significant for Holarctic phytogeographic *** studies provide a critical source of data on the latitudinal positions of different plant lineages at different times,permitting assessment of the efficacy of the BLB for *** we report exceptionally preserved fossils of Firmiana and Tilia endochrysea from the middle Miocene of South *** represents a new reliable record of Firmiana and the first discovery of the *** lineage in the fossil record of *** occurrence of these fossils in South Korea indicates that the two lineages had a distribution that extended much farther north during the middle Miocene,but they were still geographically remote from the *** light of the broader fossil record of Asia,our study shows that,in the middle Miocene,some mesothermal plants apparently inhabited the territory adjacent to the BLB and thus they were possibly capable of utilizing the BLB as a migratory *** other mesothermal plants,such as Firmiana and the *** lineages,however,are restricted to more southern regions relative to the BLB based on current fossil *** lineages may have been ecologically unable to traverse the BLB,which raises questions about the efficacy of the BLB as a universal exchange route for mesothermal plants between Asia and North America during the middle Miocene.

This study of renaturation by dilution and size exclusion chromatogra phy (SEC) addition of urea to improve yield as well as the initial and final pro tein concentrations showed that although urea decreased the rate of lysozyme ref o lding, it could suppress protein aggregation to sustain the pathway of correct r efolding at high protein concentration; and that there existed an optimum urea c oncentration in renaturation buffer. Under the above conditions, lysozyme was su ccessfully refolded from initial concentration of up to 40 mg/mL by dilution and 100 mg/mL by SEC, with the yield of the former being more than 40% and that of the latter being 34.8%. Especially, under the condition of 30 min interval time, i.e. τ>2(t_R2 -t_R1 ), the efficiency was increased by 25% and the renaturation buffe r could be recycled for SEC refolding in continuous operation of downstream proc ess.

Objective:To investigate the effects of an ethanol extract of Kalopanax septemlobus(Thunb.)***(EEKS) on cell proliferation in human hepatocellular carcinoma cells and its mechanisms of ***:Cells were treated with EEKS and subsequently analyzed for cell proliferation and flow cytometry *** of cell cycle regulators were determined by reverse transcriptase polymerase chain reaction analysis and Western blotting,and activation of eyclin-associaled kinases studied using kinase ***:The EEKS suppressed cell proliferation in both HepG2 and Hep3 B cells,but showed a more sensitive anli-proliferative activity in HepG2 *** cytometry analysis revealed an association between the growth inhibitory effect of EEKS and with G_1 phase cell cycle arrest in HepG2 cells,along with the dephosphorylation of retinoblastoma protein(pRB) and enhanced binding of pRB with the E2 F transcription factor family *** with EEKS also increased the expression of cyclin-dependent kinase(CDK) inhibitors,such as p21WAF1/CIP1 and *** any noticeable changes in G_1 cyclins and CDKs(except for a slight decrease in CDK4).Treatment of HepG2 cells with EEKS also increased the binding of p21 and p27 with CDK4 and *** was paralleled by a marked decrease in the cyclin D- and cyclin E-associated kinase ***:Overall,our findings suggest that EEKS may be an effective treatment for liver cancer through suppression of cancer cell proliferation via G_1,cell cycle arrest Further studies arc required to identify the active compounds in EEKS.

Several reports have promoted the root-derived Korean red ginseng （KRG; Panax ginseng） as alternative treatment for erectile dysfunction （ED）, and ginsenosides are known to be the principal active ingredients of ginseng. Recent studies showed that ginseng berries produce more ginsenosides than KRG; thus, we investigated the ability of the Korean ginseng berry extract GB0710 to relax the penile corpus cavernosum smooth muscle （CCSM） in this study. As a comparative control, the results were compared to those obtained using KRG. In addition, possible mechanisms of action for GB0710 were investigated. While KRG and GB0710 both displayed dose-dependent relaxation effects on precontracted rabbit CCSM in vitro, GB0710 was shown to be more potent than KRG. The GBO710-induced relaxation could be partially reduced by removing the endothelium. In addition, pre-treatment with several nitric oxide （NO） inhibitors significantly inhibited the relaxation of muscle strips. Furthermore, administration of GB0710 increased intracavernosal pressure （ICP） in a rat in vivo model in both a dose- and duration-dependent manner. Intracellular NO production in human microvascular endothelial cells could be induced by GB0710 and inhibited by NG-monomethyI-L-arginine. In conclusion, GB0710 had a greater relaxation effect on rabbit CCSM than did KRG extract, and increased ICP in a rat model in both a dose- and a duration-dependent manner. This relaxine effect might be mediated bv NO production.

Objective: To investigate the antibacterial activity of SHHextracted with either water or ethanol against methicillin-resistant Staphylococcus aureus(MRSA) and combinatory antimicrobial effect with ciprofloxacin(CIP) by time kill assay and checkerboard dilution test. Methods: The antibacterial activity determined by broth dilution method indicated that the antibacterial activity of Sami-Hyanglyun-Hwan(SHH) water extract(SHHW) and SHH ethanol extract(SHHE) ranged from 250 to 2000 μg/m L and 125 to 1000 μg/m L against MRSA, respectively. Results: In the checkerboard method, the combinations of SHHE with CIP had a partial synergistic or synergistic effect against MRSA. The time-kill curves showed that a combined SHHE and CIP treatment reduced the bacterial counts dramatically after 24 h. Conclusions: The present study demonstrates the therapeutic ability of SHHE against MRSA infections.

Abdominal obesity describes the accumulation of excessive fat in the abdomen. It is known that depending on its distribution, visceral obesity presents a greater danger to health than subcutaneous obesity. To properly prevent and treat visceral obesity, accurate evaluation methods are necessary, and hence quantitative VAT estimation is extremely important. CT scans are the most accurate method for estimating VAT, but it requires a great deal of time and effort, limiting its use in studying or evaluating obesity in patients. This paper proposed automatic measurement software that could quickly differentiate between and measure VAT and SAT. The method was verified using a total of 100 abdominal CT data values;this paper measured the SAT and VAT in the entire abdomen using the automatic measurement software. Additionally, through a comparative evaluation between the automated measurements and manual measurements such as BMI and waist circumference, clinical reliability and viability were validated and evaluated. Between automated measurements and manual measurements, the TAT (r = 0.995, p = 0.01), SAT (r = 0.987, p = 0.01) and VAT (r = 0.993, p = 0.01) showed high correlation. Using BMI as the main metric, the TAT for automated measurements (r = 0.674, p = 0.01) and the TAT for manual measurements (r = 0.703, p = 0.01) showed the strongest correlation. When using waist circumference, the VAT for automated measurements (r = 0.826, p = 0.01) and the VAT for manual measurements (r = 0.822, p = 0.01) showed the strongest correlation. With these results, the reliability and viability of the automatic measurement software were confirmed. The software is expected to help greatly in reducing the time and in providing objective data of VAT measurements from CT scans for clinical research.

Presurgical Mapping of Brain Tumors Using Statistical Probabilistic Anatomical Maps

Purpose: Multi-tracer neuroimaging is widely used for the diagnosis, radiotherapy, and neuro-surgery of brain gliomas. Anatomical and functional information is important to avoid having neurological deficits induced during the resection or radiation therapy of brain gliomas. The aim of this study was to investigate presurgical anatomical labeling of brain gliomas on FLT and FET using statistical probabilistic anatomic maps (SPAM), which are images of cerebral cortical, cerebellar, and subcortical volumes of interest (VOIs). Methods: FDG, FLT, and FET PET scans were acquired. FLT and FET PET images were coregistered to the FDG PET images, which were then spatially normalized onto the target brain. An inverse spatial normalization parameter was calculated and applied to SPAM. For the anatomical labeling of brain glioma regions, the volumes of brain gliomason FLT and FET images were extracted using segmentation. Probabilistic information of the glioma region was then calculated using SPAM and the segmented glioma volumes. SPM and an in-house program were used for image processing. Results: The probability of SPAM labeling a brain glioma region could be extracted using the inverse normalized SPAM and segmented glioma regions. In a sample case, the probabilistic anatomical region of the glioma included 21% of the postcentral gyrus, 12% of the superior parietal gyrus, and 6% of the angular gyrus. Conclusion: Anatomical information about brain gliomas could be extracted using SPAM. This proposed method would be optional for presurgical mapping, to avoid an additional functional mapping study that might otherwise be necessary to avoid producing neurological deficits.
