Background: Insufficiency of vitamin D is one of the utmost general health issues all over the globe, including Pakistan;incompatible data are present on the possible relationship among serum vitamin D values and quality of semen. Objective: Lack of Vitamin D in body is related with a higher risk of various health problems including infertility. In our setup no such type of study had been conducted in the past on the relationship among serum vitamin D levels and infertility. This study was aimed to investigate male subjects with infertility and serum levels of vitamin D. Methods: This study was cross-sectional and performed on 243 male subjects who attended the clinic for evaluation of infertility from January 2016 to December 2017. Mean age of patients was 31.94 years with standard deviation of 7.52 years. The mean age of controls was 32.2 years with SD 8.42 years, age ranged between 20 years as minimum to 46 years as maximum. The minimum vitamin D level was 7.00 ng/dl while maximum 130 ng/dl, mean was 26.20 and standard deviation was 22.06 ng/ml. Levels of vitamin D were significantly low in (67%) male subjects with infertility. The mean vitamin D level of controls was 49.40 with SD of 35.98 ng/ml. 28% of controls had also vitamin D levels below the normal range, which indicates the prevalence of vitamin D deficiency in general population here in our setup. The participants were divided according to WHO (World Health Organization) 2010 criteria along with corresponding healthy control subjects. A questionnaire for evaluation of infertility in male subjects was filled by all the contestants. Results: Levels of vitamin D were significantly low in (67%) male subjects with infertility. While 28% of controls had also vitamin D levels below the normal range. Infertile male subjects with low or no sperm count were found insufficient for vitamin D in comparison to healthy control subjects. SOBIR1在植物基础免疫中具有重要作用,并介导了番茄Ve1对1号大丽轮枝菌生理小种的抗性。但其在棉花抗黄萎病中的作用尚不明确。本研究发现1个编码类受体蛋白激酶的基因的表达在ssn组成型免疫激活棉花突变体中与野生型相比具有明显差异。克隆了该基因的全长序列,发现其编码的蛋白质多肽与拟南芥SOBIR1具有60%的同源性,并将该基因命名为GbSOBIR1。GbSOBIR1包含1个信号肽区域,5个亮氨酸富集重复(Leucine-rich repeat,LRR)区域,1个跨膜区域和1个膜内激酶区域。表达分析显示GbSOBIR1在棉花的各个组织中均有表达,并且受到黄萎病菌侵染的诱导上调表达。利用病毒介导的基因沉默(Virus induced gene silencing,VIGS)抑制GbSOBIR1表达后,棉花对大丽轮枝菌株"V991"的抗性减弱,并且抗病相关激素合成基因ICS1、LOX2和AOS以及病程相关蛋白PR1、PR5的表达量均有所下降。同时,在拟南芥中异源超量表达GbSOBIR1后,转基因植株对黄萎病的抗性得到了增强。这表明GbSOBIR1参与了植物抗黄萎病过程,并对抗病性具有正向调节功能。通过GbSOBIR1胞内蛋白激酶结构域进行酵母双杂筛选到多个互作蛋白,其中包括1个basic helix-loop-helix(bHLH)类的转录因子GbbHLH171。通过构建干涉和超量表达GbbHLH1 71的转基因株系,证明其对于棉花抗病反应具有重要作用。利用Co-IP等试验证明GbSOBIR1可以在体外和体内条件下磷酸化GbbHLH171,磷酸化后的GbbHLH171对其下游抗病相关基因的表达具有明显增强的转录活性。通过质谱分析和点突变分析进一步明确了GbSOBIR1对GbbHLH171的磷酸化位点位于Ser-413,基于这个位点的磷酸化影响了GbbHLH171的转录活性。

Wheat(Triticum aestivum L.)exhibits a greater capacity for cadmium(Cd)absorption compared to other cereal crops,leading to elevated daily Cd intake,and posing a signiﬁcant threat to public *** the mitigation of Cd stress in sustainable and environmentally friendly way,a pot study was designed by using exogenous application of various biostimulants,i.e.,Nigella sativa and Ocimum sanctum extracts:0%,10%,and 20%in combination with the chelating agent ethylenediaminetetraacetic acid(EDTA)using 0 and 5 mg kg^(-1) under various levels of Cd stress(i.e.,0,5,10,and 15 mg kg^(-1) soil).Results revealed that Cd stress signiﬁcantly reduced the seed emergence,growth,root,and allometric characters and yield-related parameters of wheat *** most observable reduc-tion was documented in wheat plants exposed to a higher Cd concentration(15 mg kg^(-1)),followed by the lower Cd level(control).The combined application of bio-stimulants and EDTA minimized the negative impacts of Cd *** highest increase in seedling emergence(5.44%),leaf area(50.60%),number of tillers(31.02%),grain yield per plant(24.28%),biological yield(13.97%),and decrease in Cd levels in grains(40%)was noticed where 20%foliar application of *** and 10%of *** biostimulants were done using 5 mg kg^(-1) of soil-applied *** intervention demonstrated a notable reduction in Cd-induced negative effects,highlighting the potential of these substances in promoting sustainable wheat cultivation in contaminated ***,it is an eco-friendly and approachable method at theﬁeld level able to ensure food safety.

植物进化出多重防御机制来使自己更好地适应复杂多变的环境。茉莉酸(Jasmonic acid,JA)是植物抵御病原侵染的重要防御激素。由生物和非生物胁迫信号引起的JA积累,通过降解具有j asmon-zim结构域的负调控蛋白JAZs从而激活了信号传导的转录因子(如MYC2),并激活下游一系列防御反应相关基因的表达。研究发现1个编码具有JAZ结构域多肽的基因GhJAZ2在棉花JA处理和接种大丽轮枝菌时的表达均明显增强。在棉花中过量表达GhJAZ2会削弱其对JA的敏感度,降低JA响应相关基因如GhPDF1.2和GhVSP的表达水平,同时使棉花对大丽轮枝菌和食草性昆虫更敏感。酵母双杂交和双分子荧光互补(Bimolecular fluorescence complementation,BiFC)分析表明,GhJAZ2可以与更多抗病反应相关蛋白互作,如病程相关蛋白GhPR10、类过敏源蛋白GhD2、NBS-LRR类蛋白质GhR1,以及具有螺旋-环-螺旋的转录因子bHLH171。与MYC2不同的是,在棉花中过量表达bHLH171激活了JA合成和信号通路,并提高了植物对大丽轮枝菌的抗性。分子和遗传证据表明,GhJAZ2可以与bHLH171互作并抑制其转录活性,从而抑制了由JA介导的防御反应。这表明JA信号路径在棉花抗黄萎病中具有重要作用,同时棉花JA信号路径与拟南芥JA调控存在差异。

Background: Diabetes Mellitus is a widespread metabolic abnormality that constitutes a most important physical dilemma in the world. Hypomagnesaemia accelerates the rate of diabetic complications. Objective: To analyze the association of magnesium levels in DM (Type 2 Diabetes Mellitus) with and without complication of Hypertension. Design: It was a comparative analytical study. Setting: This research was carried out in medicine department, Peoples Medical College Hospital, Nawabshah from March 2016 to February 2017. Samples: 245 patients with Type 2 DM after fulfilling the selection criteria were included, out of them 123 with hypertension and 122 without hypertension. Methods: After a short-lived consultation, all the subjects were categorized for variable analyses like age, gender, Type 2 DM with and without hypertension, duration of diabetes and presence of hypomagnesaemia. Joint National Committee (JNC 8) classification of Hypertension was applied to collect data. Blood samples were drawn for research purpose in fasting state for serum magnesium level analyses on the basis of hypertension and without hypertension. Results: In 245 diagnosed Type 2 diabetic patients, 160 were males and 85 were females. Out of them, 123 were hypertensive while 122 were non hypertensive. Normal magnesium was present in 120 (48.97%) and low magnesium was present in 125 (51.02%) patients overall. A decreased serum level of magnesium was observed in 52.04% hypertensive subjects with diabetes and 50% non-hypertensive subjects with diabetes. Conclusion: Frequency of decreased serum magnesium is widespread in T2DM with and without hypertension.

Viral hepatitis C is a type of illness, which is transmitted to patients by different methods in world. Here we will identify the different common risk factors for transmission of viral hepatitis C in our setup. Aim of study was to determine frequency of various risk factors in HCV positive patients. This study was cross sectional and conducted Department of Medicine PMC Hospital at Nawabshah. Duration of this study was one year from April 2016 to March 2017. After taking informed written consent, 243 diseased persons with positive anti Hepatitis C Antibodies were incorporated in this research. Frequency of variables i.e. Hepatitis C virus risk factors and different other demographic results was collected on preformed proforma. In total of 243 subjects there were 165 (67.9%) male and 78 (32.1%) females respectively. Most of the subjects have more than one risk factor. 19 (7.8%) had history of blood and blood products transfusion. IV drug abuse was detected in 08 (3.3%). Homosexuality and heterosexuality were observed in 14 (5.8%) and 12 (4.9%) subjects respectively. History of dental procedure was seen in 31 (12.8%). 228 (93.8%) had history of needle pricking in different ways. History of different surgical procedures was observed in 33 (13.8%). Calculated Mean and Standard Deviation for age was 39.2 ± 10.3 years. It was concluded that proper implementation of precautionary measures should be carried out for every human being to reduce burden of HCV illness in far near future.

The potential mechanism of Isodon suzhouensis against COVID-19 via EGFR/TLR4 pathways

Corona Virus Disease 2019 (COVID-19) has brought the new challenges to scientific research. Isodon suzhouensis has good anti-inflammatory and antioxidant stress effects， which is considered as a potential treatment for COVID-19. The possibility for the treatment of COVID-19 with suzhouensis and its potential mechanism of action were explored by employing molecular docking and network pharmacology. Network pharmacology and molecular docking were used to screen drug targets， and Lipopolysaccharide (LPS) induced RAW264.7 and NR8383 cells inflammation model was used for experimental verification. Collectively a total of 209 possible linkages against 18 chemical components from suzhouensis and 1194 COVID-19 related targets were selected. Among these， 164 common targets were obtained from the intersection of suzhouensis and COVID-19. GO and KEGG enriched 582 function targets and 87 target proteins pathways， respectively. The results from molecular docking studies revealed that rutin， vitexin， isoquercitrin and quercetin had significant binding ability with SARS-CoV-23CLpro and ACE2. In vitro studies showed that Isodon suzhouensis extract (ISE) may inhibit the activation of PI3K/Akt pathway and the expression level of downstream pro-inflammatory factors by inhibiting the activation of EGFR in RAW264.7 cells induced by LPS. In addition， ISE was able to inhibit the activation of TLR4/NF-κB signaling pathway in NR8383 cells exposed to LPS. Overall， the network pharmacology and in vitro studies conclude that active components from suzhouensis have strong therapeutic potential against COVID-19 through multi-target， multi-pathway dimensions and can be a promising candidate against COVID-19.

Background: Thioacetamide (TAA) has been used extensively in the development of suitable animal models of acute and chronic liver injury employing various doses, times and routes of its administration, particularly in drinking water due to its resemblance with human liver fibrosis and cirrhosis. The aim of this study was to investigate and compare hematological alteration during the acute and chronic liver inflammation. Methods: Acute Liver inflammation was induced in Wistar rats via intraperitoneal injection of thioacetamide and the animals were sacrificed 12 h after the TAA administration. Induction of chronic liver inflammation was performed by continuous administration of TAA in the drinking water (200 mg/L) during 18 weeks of experiment. After that all animals were sacrificed and Blood samples were collected for further analysis. Results: Single intra peritoneal injection of TAA (300 mg/kg B.W.) induced an acute condition with hematological changes including leukocytosis with marked neutrophilia (P = 0.0429), lymphopenia, thrombocytosis as well as increased hemoglobin concentration (P 0.05) and decline of erythrocytic count (P = 0.0009). Eighteen weeks of uninterrupted supply of TAA (200 mg/L) in drinking water lead to chronic inflammation and the hematological alterations were leucopenia (P = 0.0197) accompanied with neutropenia and thrombocytopenia. Increase in RBCs (P = 0.0073) and Hb contents was also observed with a decline of red cell indices. Conclusion: Taken together these findings we can conclude that the animals respond differently under acute and chronic inflammatory condition with TAA administration. Leukocytosis with marked neutrophilia, thrombocytosis as well as increased hemoglobin concentration and decline of erythrocytic count were observed in acute while leucopenia accompanied with neutropenia and thrombocytopenia and increase in RBCs, Hb and Hct was also observed with a decline of other red cell indices during chronic phase.

Background: A relationship among vitamin D and pulmonary tuberculosis. Objective: To conclude the occurrence of Vitamin D insufficiency in subjects suffering from pulmonary TB. Design: This was a cross sectional study. Setting: Current study was performed at department of medicine, Peoples Medical College Hospital Nawabshah starting from January 2017 to December 2017. Samples: 385 AFB positive subjects of pulmonary tuberculosis after achieving the criteria of selection were recruited. Material and Methods: After a brief discussion, the patients were characterized for variable evaluation such as age, gender, sputum AFB and duration of disease and presence of low vitamin D levels. Sputum AFB criterion was used for Data collection. Fasting blood samples were collected for analysis of vitamin D levels in all subjects of AFB positive pulmonary tuberculosis. Results: In 385 diagnosed patients with smear positive PTB, 273 (70.9%) males and 112 (29.1%) were female. Vitamin D levels were within normal or sufficient range in 134 (34.8%), below normal range in 251 (65.2%) cases (insufficient in 09.56% (24/251) and deficient in 90.43% (227/251) cases). Conclusion: Decreased levels of vitamin D were common in subjects with AFB smear Positive PTB cases.
