Background： Bacillus spp. seem to be an alternative to antimicrobial growth promoters for improving animals＇ health and performance. However, there is little information on the effect of Bacillus spp. in combination with different dietary crude protein（CP） levels on the ileal digestibility and microbiota composition. Therefore, the objective of this study was to determine the effect of Bacillus spp. supplementation to low-（LP） and high-protein diets（HP） on ileal CP and amino acid（AA） digestibility and intestinal microbiota ***： Eight ileally cannulated pigs with an initial body weight of 28.5 kg were randomly allocated to a rowcolumn design with 8 pigs and 3 periods of 16 d each. The assay diets were based on wheat-barley-soybean meal with two protein levels： LP（14% CP, as-fed） and HP diet（18% CP, as-fed）. The LP and HP diets were supplemented with or without Bacil us spp. at a level of 0.04%（as-fed）. The apparent ileal digestibility（AID） and standardized ileal digestibility（SID） of CP and AA was determined. Bacterial community composition from ileal digesta was analyzed by Illumina amplicon sequencing and quantitative real-time PCR. Data were analyzed as a 2 × 2 factorial design using the GLIMMIX procedures of ***： The supplementation with Bacillus spp. did not affect both AID and SID of CP and AA in growing ***, there was no difference in AID of CP and AA between HP and LP diets, but SID of cystine, glutamic acid,glycine, and proline was lower（P 〈 0.05） in pigs fed the HP diets. Background Tree regeneration is a key component of resilience because it promotes post-disturbance recovery of forests. Northwestern Patagonia from Argentina is occupied by Nothofagus alpina（Na）, N. obliqua（No）, and N. dombeyi（Nd） forests that grow along an intense precipitation gradient, managed throughout shelterwood silvicultural system by technicians of the Lanin Natural Reserve. The objective of this study was to evaluate the influence of seeding cuttings over the dynamics of Nothofagus mixed forests across landscape（precipitation gradient） based mainly on the composition and abundance of tree regeneration, permanent sampling plots and generalized linear mixed models. In particular, we analysed:（i） the structure of sexual and asexual regeneration during 20 years after harvest（the dynamics of managed forests）, and（ii） the structure of sexual regeneration in primary and managed forests after > 20 years of harvest（the effect of silviculture）.Results Nd was the most abundant species in the regeneration of managed forests during all periods in both sites despite its lower contribution to the adult cohort. During the 10–20 years period after harvest, the humid site exhibited higher regeneration density than the mesic site(120,000 and 6000 ind ha-1, respectively), and it decreased afterwards. The number of established regeneration（> 2 m height） was lower for Na in the mesic site and for No in the humid site(0 and 57 ind ha-1, respectively). However, in comparison to No, Na showed a higher number of sprouted stumps and sprouts per stump, and a higher sprout height in the mesic site. No exhibited higher sprout mortality in the humid site. 超声处理影响乳清蛋白结构、功能和化学特性的研究较多,但人们对超声处理影响乳清蛋白固体凝胶消化行为的认识不足。同样,大豆-乳清复合蛋白基质和大豆-乳清复合蛋白乳化脂肪颗粒对乳清乳化蛋白凝胶性能的影响研究也较缺乏。本研究拟探讨不同频率和功率水平对超声预处理乳清蛋白乳液凝胶凝胶性质、微观结构和消化行为的影响。并利用大豆蛋白部分替代乳清蛋白,进一步研究大豆-乳清复合蛋白比例的大豆-乳清复合蛋白乳化脂肪颗粒和大豆-乳清复合蛋白基质对乳清蛋白乳液凝胶的凝胶性质、微观结构和消化行为的影响。主要结果如下:首先,研究了超声频率模式(单频、双频和三频)和超声功率(0-300W)对乳清蛋白结构特性(内源荧光和巯基含量)的影响。将改性乳清蛋白制备的乳液用于制备热诱导凝胶,并研究了乳清蛋白乳液凝胶(WPEG)的性质及其消化行为。使用单频和双频超声在180–240 W下预处理的乳清蛋白制备的WPEG的质构和流变性能得到增强,而使用三频超声在180W以上预处理乳清蛋白制备的WPEG的质构和流变性能则降低。经过双频超声240W预处理的乳清蛋白制备的WPEG具有最高的硬度和储能模量,分别比对照高3.07和1.41倍。根据共焦激光扫描显微镜的结果,使用双频超声预处理的乳清蛋白制备的WPEG的微观结构显示出比对照组更均匀且密集的网络结构。与对照组相比,使用双频超声预处理的乳清蛋白制备的WPEG的微结构和性能的改变延迟了胃消化前30分钟的蛋白质分解。而与对照相比,经超声预处理改性的乳清蛋白制备的WPEG的游离氨基的释放速率在肠期增加。结果表明,使用双频超声对乳清蛋白进行改性可以更有效地改善WPEG的性能,在应用超声预处理生产蛋白凝胶时应考虑超声功率。超声预处理的乳清蛋白制备的WPEG具有良好的网络结构,硬度和储能模量更好。超声预处理诱导的蛋白质部分去折叠可能使乳清蛋白更易受消化酶的作用。其次,通过热诱导手段制备了大豆和乳清复合蛋白乳液填充乳清蛋白凝胶。研究了大豆和乳清蛋白比例(10:0、8:2、6:4、4:6、2:8和0:10,w/w)对乳液填充凝胶机械性质、微观结构和消化动力学的影响。研究结果表明大豆和乳清蛋白比例的会影响乳液填充乳清蛋白凝胶的机械和微观结构。乳液中乳清蛋白比例的增加会提高乳化填充凝胶的硬度和储能模量,并且大豆和乳清蛋白比例为0:10时乳化填充凝胶的硬度和储能模量最高。但是乳液中乳清蛋白比例的增加会降低乳液填充凝胶的溶胀能力。研究表明乳清蛋白在增强凝胶网络中起到了积极作用。大豆蛋白比例较高时,凝胶微观结构可以观察到乳液颗粒的絮凝和聚结,而乳清蛋白比例较高乳液制备的填充凝胶(大豆蛋白:乳清蛋白=0:10;2:8)中,填充颗粒显示了较为均匀的粒径分布。利用两段式模型来分别模拟胃消化和肠消化过程中酸或碱消耗量的动力学,结果表明大豆蛋白比例的增加可以促进胃蛋白酶的扩散并提高消化过程的反应速率。大豆蛋白与乳清蛋白比例为0:10的时候,混合蛋白乳液填充凝胶的反应速率最低,表明乳清蛋白增加会增加凝胶的抗消化作用。这与乳清蛋白比例较高时,制备的乳液填充凝胶结构更加致密有关。结果表明,可以通过调节混合蛋白中大豆和乳清蛋白的比例来调控混合蛋白乳液填充乳清蛋白凝胶的性质特征。最后,以凝胶硬度和储能模量为指标,考察了基质总蛋白浓度(8-14%)对凝胶性质的影响,并确定凝胶基质总蛋白浓度为13%(w/v)。利用大豆蛋白部分替代乳液填充乳清蛋白凝胶基质的乳清蛋白,研究了大豆和乳清蛋白比例(10:0、8:2、6:4、4:6、2:8和0:10,w/w)对乳液填充复合蛋白凝胶特性和消化动力学的影响。凝胶硬度随着大豆蛋白比例的增加而降低(p0.05)。在溶胀能力方面,除大豆和乳清蛋白比例为10:0时溶胀率为60%外,其他凝胶的溶胀率均小于10%。增加大豆蛋白的比例可以增加了凝胶网络中的二硫键,但是会导致氢键的减少。大豆蛋白含量的增加会增加凝胶网络中聚集体数量,并导致凝胶再微观结构上显示出松散的凝胶网络。胃蛋白酶的扩散和水解速率在大豆和乳清蛋白比例为10:0的凝胶中更快,而在大豆和乳清蛋白比例为0:10的凝胶中最小。这可能是由于大豆和乳清蛋白比例为0:10的凝胶较高的硬度、致密的微观结构和较低的膨胀能力限制了胃蛋白酶在凝胶中的扩散,并限制了胃蛋白酶对可肽段中可切割位点的可及性。研究表明,通过将植物和动物蛋白结合在一起来改变基质组成,可用于开发不同凝胶特性和消化动力学的乳液凝胶。综上所述,多频超声预处理、乳液填充颗粒性质和基质特征的调节可用于调控乳液填充蛋白凝胶的性质的结构。为实现食品工业的可持续发展,可以考虑以植物蛋白部分替代动物

Laboratory Profile of HIV-2 and Dual HIV-1/HIV-2 Associated Acquired Immunodeficiency Syndrome in Nigeria

Background: HIV-2 is comparatively less pathogenic with slow progression of infection to clinical disease and consequently there is less of information on the occurrence of HIV-2 associated disease than HIV-1. We hereby describe some laboratory profiles of individuals presenting with HIV-2 and dual HIV-1/2 related AIDS at the University College hospital in Ibadan over a period of seven years. Methodology: Blood samples from patients presenting with the AIDS defining illness at the University College Hospital, Ibadan, Nigeria were tested for antibodies to HIV-1/2 using rapid test devices or ELISA. Initially reactive samples were further tested by immunoblotting for differentiation into HIV-1 or HIV-2 or HIV-1/2 dual infection. Blood samples from individuals with confirmed infections were further analyzed for CD4 cell lymphocyte number, plasma HIV-1 RNA concentration, hematological and blood chemistry parameters. The data analysis was done using descriptive statistics and Levene-S test for equality of variance. Results: Thirty five patients, 18 and 17 with HIV-2 and dual HIV-1/2 infections respectively were identified during the period covered by this study (2005-2012). The median age of the patients was 48 years old (Range: 42 - 70 years old) and mean CD4 cell count of HIV-2 patients at enrollment was 324 (Range: 16 - 696) and 350 (Range 54 - 863) per microlitre of blood for patients with dual HIV-1/2 infection. HIV-1 RNA was not detected in the plasma of the 18 patients with serological HIV-2 infection but 2 (11.8%) of the 17 patients with dual HIV-1/2 serological profile had detectable HIV-1 RNA (1,287,275 copies/ml and 1,816,491 copies/ml). Conclusion: The results emphasize the need to consider HIV-2 infection in the investigation of patients presenting with the AIDS related illness but with negative HIV-1serology. AIM To investigate the effects of orally gavaged aqueous rhubarb extract(RE) on 5-fluorouracil(5-FU)-induced intestinal mucositis in rats. METHODS Female Dark Agouti rats(n = 8/group) were gavaged daily(1 mL) with water, high-dose RE(HDR; 200 mg/kg) or low-dose RE(LDR; 20mg/kg) for eight days. Intestinal mucositis was induced(day 5) with 5-FU(150 mg/kg) via intraperitoneal injection. Intestinal tissue samples were collected for myeloperoxidase(MPO) activity and histological examination. Xenopus oocytes expressing aquaporin 4 water channels were prepared to examine the effect of aqueous RE on cell volume, indicating a potential mechanism responsible for modulating net fluid absorption and secretion in the gastrointestinal tract. Statistical significance was assumed at P < 0.05 by one-way ANOVA. RESULTS B o d y w e i g h t w a s s i g n i f i c a n t l y r e d u c e d i n r a t s administered 5-FU compared to healthy controls(P < 0.01). Rats administered 5-FU significantly increased intestinal MPO levels(≥ 307%; P < 0.001), compared to healthy controls. However, LDR attenuated this effect in 5-FU treated rats, significantly decreasing ileal MPO activity(by 45%; P < 0.05), as compared to 5-FU controls. 5-FU significantly reduced intestinal mucosal thickness(by ≥ 29% P < 0.001) as compared to healthy controls. LDR significantly increased ileal mucosal thickness in 5-FU treated rats(19%; P < 0.05) relative to 5-FU controls. In xenopus oocytes expressing AQP4 water channels, RE selectively blocked water influx into the cell, induced by a decrease in external osmotic pressure.
