A new hemostatic agent composed of Zn^(2+)-enriched Ca^(2+)alginate activates vascular endothelial cells in vitro and promotes tissue repair in vivo

To control capillary bleeding, surgeons may use absorbable hemostatic agents, such as Surgicel® and TachoSil®. Due to their slow resorption, their persistence in situ can have a negative impact on tissue repair in the resected organ. To avoid complications and obtain a hemostatic agent that promotes tissue repair, a zinc-supplemented calcium alginate compress was developed: HEMO-IONIC®. This compress is non-absorbable and is therefore removed once hemostasis has been achieved. After demonstrating the hemostatic efficacy and stability of the blood clot obtained with HEMO-IONIC, the impact of Surgicel, TachoSil, and HEMO-IONIC on cell activation and tissue repair were compared (i) in vitro on endothelial cells, which are essential to tissue repair, and (ii) in vivo in a mouse skin excision model. In vitro, only HEMO-IONIC maintained the phenotypic and functional properties of endothelial cells and induced their migration. In comparison, Surgicel was found to be highly cytotoxic, and TachoSil inhibited endothelial cell migration. In vivo, only HEMO-IONIC increased angiogenesis, the recruitment of cells essential to tissue repair (macrophages, fibroblasts, and epithelial cells), and accelerated maturation of the extracellular matrix. These results demonstrate that a zinc-supplemented calcium alginate, HEMO-IONIC, applied for 10 min at the end of surgery and then removed has a long-term positive effect on all phases of tissue repair.

Bone-marrow-derived mesenchymal stem cells and endothelial progenitor cells have some interesting biological properties that make them unique for cell therapy of degenerative and cardiovascular disorders.Although both cell populations have been already studied and used for their regenerative potentials,recently their special immunoregulatory features have brought much more attention.Mesenchymal stem cells and endothelial progenitor cells have both proangiogenic functions and have been shown to suppress the immune response,particularly T cell proliferation,activation,and cytokine production.This makes them suitable choices for allogeneic stem cell transplantation.Nevertheless,these two cells do not have equal immunoregulatory activities.Many elements including their extraction sources,age/passage,expression of different markers,secretion of bioactive mediators,and some others could change the efficiency of their immunosuppressive function.However,to our knowledge,no publication has yet compared mesenchymal stem cells and endothelial progenitor cells for their immunological interaction with T cells.This review aims to specifically compare the immunoregulatory effect of these two populations including their T cell suppression,deactivation,cytokine production,and regulatory T cells induction capacities.Moreover,it evaluates the implications of the tumor necrosis factor alpha-tumor necrosis factor receptor 2 axis as an emerging immune checkpoint signaling pathway controlling most of their immunological properties.

Celastrol targets IRAKs to block Toll-like receptor 4-mediated nuclear factor-κB activation

OBJECTIVE： Celastrol has been established as a nuclear factor-κB（NF-κB） activation inhibitor; however, the exact mechanism behind this action is still unknown. Using text-mining technology, the authors predicted that int erleukin-1 receptor-associated kinases（IRA Ks） are potential celastrol targets, and hypothesized that targeting IRAKs might be one way that celastrol inhibits NF-κB. This is because IRAKs are key molecules for some crucial pathways to activate NF-κB（e.g., the inter leukin-1 receptor（IL-1R）/Toll- like receptor（TLR） superfamily）.METHODS： The human hepatocellular cell line（Hep G2） treated with palmitic acid（PA） was used as a model for stimulating TLR4/NF-κB activation, in order to observe the potential effects of celastrol in IRAK regulation and NF-κB inhibition. The transfection of small interfering RNA was used for down-regulating TLR4, IRAK1 and IRAK4, and the Western blot method was used to detect changes in the protein expressions.RESULTS： The results showed that celastrol could effectively inhibit PA-caused TLR4-dependent NF-κB activation in the Hep G 2 cells; PA also activated IRAKs, which were inhibited by celastrol. Knocking down IRAKs abolished PA-caused NF-κB activation.CONCLUSION： The results for the first time show that targeting IRAKs is one way in which celastrol inhibits NF-κB activation.

目的探讨做为微环境的滋养层细胞在体外大量扩增晚期内皮祖细胞(EPC)中的作用。方法预铺辐射后的滋养层细胞,而后植入分离的外周血单个核细胞,计数培养21 d后获得的晚期EPC克隆数目,并检测目的细胞免疫表型,用Matrigel鉴定其体外血管形成能力。采用动态显微镜追踪单个干细胞在滋养层细胞上的分化过程。结果培养21 d时,在预铺晚期EPC和人脐静脉内皮细胞(HUVEC)为滋养层的培养环境下,每100ml外周血单个核细胞分别获得(40.0±3.9)和(39.3±3.1)个晚期EPC克隆,均明显高于无滋养层条件下的(2.0±1.3)个(P均<0.05)。目的细胞可表达CD31、CD34、eNOS、FLt-1、P1H12、Sendo、VEcadherin和CD117,并在体外与Matrigel形成管状结构。动态追踪示单个干细胞的扩增是在有滋养层细胞参与下完成不对称分裂的。结论通过提供滋养层细胞方法可以大量扩增晚期EPC,滋养层细胞可能参与了早阶段内皮干细胞的不对称分裂扩增。

目的通过实验验证一个体外大量扩增晚期内皮祖细胞（EPC）的方法。方法先预铺经辐射后的滋养层细胞（晚期EPC或脐带脐静脉内皮细胞），而后植入分离的人外周血单个核细胞，计数并对比培养21d后获得的晚期EPC克隆数目，并检测目的细胞免疫表型，应用基质胶（matrigel）鉴定其体外管状结构形成能力。结果21d时在预铺的两种滋养层细胞培养条件下获得的晚期EPC克隆数目均比无滋养层细胞的培养方法高20倍（40．0±3．9、39．3±3．1比2．0±1．3，均P〈0．05）；且细胞克隆出现的早。目的细胞表达CD31、CD34、内皮型一氧化氮合酶（eNOS）、血管内皮细胞生长因子受体Fit-1、CDl46的2个表型（P1H12、Sendo）、血管内皮钙黏蛋白（VEcadherin）、CDll7，且具有能在体外与matrigel形成管状结构的功能。结论通过提供滋养层细胞的方法可以大量扩增晚期EPC，提示微环境在晚期EPC扩增中起重要作用。

促炎细胞因子活化内皮细胞,使之表达黏附分子(cellular adhesion molecules,CAMs),从而增加对血循环中白细胞黏附力,使白细胞流速逐渐变慢,直至静止于血管内壁,并最终穿越血管。上述过程是多种急慢性炎症的早期步骤。为探讨雷公藤红素抗炎症价值,本文以体外培养的人脐静脉内皮细胞(HUVECs)作为模型,观察该化合物对HUVECs活化表达CAMs及黏附白细胞能力的影响。MTT检测表明,红素在400 nM以上浓度,剂量依赖地减少活细胞数量(即毒性作用)。细胞周期分析表明,在400 nM以上浓度,红素呈现G/M期阻滞作用。非毒性剂量的红素与HUVECs预先共育6小时,对TNFα(10 ng/ml)刺激ICAM-1,VCAM-1和E-Selectin表达,有剂量依赖的抑制作用。红素在200μM浓度对ICAM-1和VCAM-1抑制率在90%以上(IC分别为51和52 nM),对E-Selectin抑制率达73%(IC:94 nM)。红素预育6小时,对IL-1β(5 ng/ml)或IFN-r(200 ng/ml)刺激HUVECs表达ICAM-1,VCAM-1和E-Selectin同样有显著抑制作用。与其不同,地塞米松(2μM),布洛芬(200μM),甲氨喋呤(2μM)或丙丁酚(100μM)与HUVECs预育6小时,对细胞因子刺激黏附分子表达,均无显著抑制作用。另外,先用TNFα(10 ng/ml)或IL-1β(5 ng/ml)或IFN-r(200 ng/ml)刺激HUVECs 6小时,使之处于活化状态,再加入非毒性剂量雷公藤红素18小时,红素对已经活化细胞仍有明显抑制黏附分子表达作用。TNFα(10 ng/ml)显著提高HUVECs对外周血单个核细胞(PBMCs)黏附能力,而雷公藤红素能明显逆转TNFα这一效应。红素不改变细胞表面TNFα受体(TNFRⅠ或TNFRⅡ)水平,提示红素作用靶位于细胞因子受体的下游。雷公藤使细胞总ICAM-1(细胞表面加细胞内)含量下降,提示黏附分子蛋白合成受到红素抑制。但细胞总ICAM-1减少主要是细胞膜外水平下降所导致,细胞内ICAM-1并不降低,该结果提示红素可能影响蛋白转运。总之,红素在非毒性剂量,对多种细胞因子诱导内皮细胞合成及表面表达黏附分子,有强烈抑制作用,并减少内皮细胞黏附白细胞能力。这些药理作用显示,红素作为新的抗炎症药物值得进一步研究。

雷公藤红素(tripterine)是从雷公藤中分离到的第一个单体,其药理作用研究,在上个世纪70年代,有零星开展。到80年代末期,因雷公藤在临床应用方面疗效已经肯定,进一步促进了对该植物有效成分的药理研究,红素药理研究,从这个时候也算正式启动,作者中部分人员也是从80年代末开始进入这个领域的,至今不缀,将近20年。
