AIM To explore the kinetic changes in plasma D(-)- lactate and lipopolyssccharide(LPS)levels,and investigate whether D(-)-lactate could be used as a marker of intestinal injury in rats following gut ischemia/ reperfusion,burn,and acute necrotizing pancreatitis (ANP). METHODS Three models were developed in rats:① gut ischemia/ reperfusion obtained by one hour of superior mesenteric artery occlusion followed by reperfusion;② severe burn injury created by 30% of total body surface area(TBSA)full-thickness scald burn;and ③ ANP induced by continuous inverse infusion of sodium taurocholate and trypsin into main pancreatic duct. plasma levels of D(-)-lactate in systemic circulation and LPS in portal circulation were measured by enzymatic- spectrophotometric method and limulus amebocyte lysate (LAL)test kit,*** samples of intestine were taken for histological analysis. RESULTS One hour gut ischemia followed by reperfusion injuries resulted in a significant elevation in plasma D(-)- lactate and LPS levels,and there was a significant correlation between the plasma D(-)-lactate and LPS(r =0.719,P plasma concentrations of D(-)- lactate and LPS increased significantly at 6h postburn, and there was also a remarkable correlation between them (r = 0.877,P plasma D(-)-lactate levels in systemic blood paralleled with LPS levels in the portal vein *** measurement of plasma D(-)-lactate level may be a useful marker to assess the intestinal injury and to monitor an increase of intestinal permeability and endotoxemia following severe injuries in early stage.

AIM: To investigate the role of cytokine gene expression inorgan damage at different tissue sites during ***: Male NIH mice were subjected to cecal ligationand puncture (CLP) or sham operetion ( Sham ). Pro-inflammatory cytokine (TNFα, IL-1β and IL-6) and anti-inflammatory cytokine (IL-4) gene expression in the liverand lung tissue were assessed by RT-PCR. The permeabilityof microvascular and water content in the lungs and liverwere also ***: Significant increase in TNFα, IL-1β and IL-6 geneexpression was observed at 3 and 12 h after CLP both in theliver and lungs ( P＜ 0.01). The level of IL-4 gene expressionwas not changed after CLP in the lungs, but increased at 12h after CLP ( P ＜ 0.01) in the liver tissue. Both the liver andlungs showed a significant increase in microcirculatorypermeability at 12 h after CLP (P＜ 0.01), and the increasein the lungs was higher than that in the liver. The watermaes fractions in the liver ( P＜ 0.05) and lungs ( P＜ 0.01)were increased after CLP, and the increase in the lungshappened earlier and more severely than that in the ***: The inflammatory response in the liver andlungs wes different during sepsis. At the early stage ofsepsis, pro-inflammatory reaction dominates both in theliver and lungs. But at the later stage of sepsis, induction ofcompensatory anti-inflammatory response was seen in theliver but not in the lungs. This difference in situ activity maycontribute to the different vulnerability of organ damageduring sepsis. The strategy of systemic administration ofanti-inflammatory drugs to sepsis should be reconsidered.

There are multiple reports of autoimmune response in patients with lung cancer. To investigate whether a novel autoantibody is present in patients with lung cancer and evaluate its clinical diagnostic and prognostic value, sera from 10 patients with lung cancer and 10 normal individuals were analyzed using immunofluorescence and Western blotting. It was found that one serum sample from the patients with squamous carcinoma gave a fine speckled pattern staining in nucleus and had a high titer antinuclear autoantibody which could recognize 31 kD of nuclear protein isolated from both cancer cells and normal cells. The same patient’s serum was further used to immunoprecipitate the target antigen. The protein bands were excised from the SDS-PAGE gels and were analyzed with a Qstar Pulser I Quadrupole time-flight mass spec-trometer, and the 31 kD target antigen was identified as U1-A snRNP. To test the prevalence of anti-U1-A snRNP antibody, sera from 93 patients including 36 squmaous carcinomas (SCC), 26 adenocarcinomas (Ad), and 31 small cell carcinomas (SCLC) were screened by Western blotting. The results demonstrated that anti-U1-A snRNP antibody was present in 50% of SCC sera, 26.9% of Ad sera and 54.8% of SCLC sera. In this paper, we report for the first time that anti-U1-A snRNP antibody could be detected in the patients with lung cancer.

The chemokine CXCR4 receptor is over-expressed in a wide variety of *** this study,AMD3100,which was a prototype non-peptide antagonist of CXCR4 receptor,was labeled with;Tc.;Tc-AMD3100 was verified by thin layer radio chromatography（TLRC）.The tumor-localizing properties of;Tc-AMD3100 were evaluated and proved in mice bearing Hep-G2 tumor xenograft.;Tc-AMD3100 was a promising,novel receptor-specific radiopharmaceutical with potential application in the imaging of human tumors.

The diagnosis of prostate diseases by magnetic resonance imaging (MRI) in vivo is sometimes difficult for the lower signal to noise ratio (SNR). To increase the SNR of the prostate image, we designed a RF coil that can be inserted into rectum and was named endorectal coil. The properties of RF coil were evaluated using a network analyzer. Moreover the images and spectroscopy of a special phantom were acquired and the results were compared to those of the commercial TORSO coil (G.E. Medical Systems, USA). Our coil gave a significantly higher SNR at the region of interest (ROI). The achieved high local SNR and resulting high spatial resolution would add more anatomic and biochemical information to the diagnosis of prostate diseases.

Background and objective The value of D-dimer in the risk stratification of patients with coronary artery disease(CAD) and the relationship between D-dimer and the diseased coronary arteries remains controversial or unclear,especially in the *** study was to evaluate the usefulness of D-dimer as a biomarker in assessing the vulnerable blood in the elderly patients with coronary *** Sixty elderly (≥60 years old) male patients with suspected CAD were enrolled in this prospective study. Patients were divided into CAD group (n=41,10 with stable angina and 31 with unstable angina) and control group (n=19) according to their coronary angiography Results Clinical information including age,body mass index (BMI),smoking index,and the complications of primary hypertension or diabetes,and CAD family history was *** blood was sampled serially for the determination of total cholesterol,HDL cholesterol,LDL cholesterol,triglycerides,apoA1,apoB,glucose,uric acid,homocysteine (Hcy),hs-CRP,soluble thrombomodulin (sTM),and markers of fibrinolytic system and hypercoagulability,such as D-dimer,fibrinogen, *** extent of coronary atherosclerosis was assessed,using the Gensini scoring system on the basis of coronary angiography. Results Compared with the controls,the patients with CAD had significantly higher levels of D-dimer.D-dimer level was significantly correlated to age,hs-CRP,Hcy,and *** with D-dimer levels in the top triplicate of D-dimer level had significantly higher prevalence of unstable angina compared with patients in the lowest triplicate (OR=4.8,Z=3.28,P=0.001).In an ordinal logistic regression,the OR value of developing more serious CAD augmented 3.1-fold with each increasing triplicate of D-dimer. Patients with unstable angina had a significantly higher level of D-dimer than the controls (P=0.005),and an increasing trend compared with patients with stable angina (P=0.059),whereas there was no difference between the patients

BACKGROUND:KAI1/CD82 has been reported to attenuate the process of metastases in a variety of tumors;however,its mechanism of action in invasion has not been fully elucidated. The present study aimed to investigate the importance of KAI1 in invasion and its correlation with activation of sphingosine kinase(SPK)in human pancreatic cancer PANC1 and Miapaca-2 cell lines. METHODS:The expression of KAI1 in PANC1 and Miapaca-2 cells,which was mediated by recombinant adenovirus(Ad-KAI1), was assessed by a flow cytometer and Western *** successful infection was established,in vitro growth curve and invasive ability in Boyden Chamber assay were *** presence of KAI1 correlating with c-Met and SPK was detected by co-immunoprecipitation and[γ-32P]ATP incorporation. CONCLUSION:The results of this study for the first time demonstrated a regulatory role for KAI1 in SPK activation, which leads to decreased invasive ability in disease progression of human pancreatic cancer.

Objective To investigate the relationship between transcription factor and the change of protein expression levels in heart failure. Methods Bioinformatic method was used to analyze the data of binding-sites on the 5' flaking regions of four genes whose mRNA level changed in failing heart from three databases about nucleic acid-EMBL, transcriptional regulation factor-TRANSFAC and protein-SWISS-PORT. The expression level of selected transcription factor was determined by immunohischemical method. Results Nine transcription factors were inferred to influence the proteins' levels in occurrence and development of heart failure. Serum response factor (SRF) was selected from the nine factors and assayed. The results showed that there was a higher level of SRF in healthy group than in chronic heart failure (CHF), and the level was associated with the degree of CHF. It was also found that there was a relative higher level of SRF in the acute myocardial infarction (AMI) than that in CHF, but which was lower than the healthy. Conclusion It showed that SRF had a quantitative change in the development of heart failure, and suggested SRF might play an important regulative role in heart failure. The expression changes of proteins related to myocardial function might be regulated by the quantitative change of transcription factor(s).
