Sexually Transmitted Diseases at Monk.y Bay Community Hospital in Mangochi, Malawi;an Analysis of Characteristics of Common Sexually Transmitted Diseases

We present the characteristics of the commonly diagnosed sexually transmitted infections (STI) at a clinic of Monk.y Bay community hospital by a retrospective study. We conducted an audit of patients’ hospital records from a STIs clinic at Monk.y Bay community hospital in Mangochi District (Malawi) covering a period from January 2019 to June 2019 (18 months). Data was tabulated in excel and analyses were made based on sex, age, male circumcision status, pregnancy and the pattern of STIs diagnosed within the chosen study period. A total number of 659 clients presented at STI clinic of Monk.y Bay community hospital between January 2018 and June 2019. Out these clients, 409 (62%) were females and 250 (38%) were males. Most of the patients were in the age range of 25 years or above (447, 68%). Majority of the patients were HIV negative (523, 79%). Out of the 250 males, 113 (45%) were circumcised. Among female patients 56 of the 409 (14%) were pregnant. The most common presentations for all cases put together were lower abdominal pain (253, 38%), then urethral discharge (189, 29%), abnormal vaginal discharge (141, 21%), genital ulcer disease (64, 10%), in that order. Syphilis, genital warts, inguinal bubo and scrotal swelling were less common. The most common presentation in males was urethral discharge (178, 71%), while in females it was lower abdominal pain (214, 52%). Our audit showed that in Mangochi (Malawi) sexually transmitted diseases of various types are more common among young adults, females and uncircumcised men. There was no much difference in the percentage of STI’s between HIV positive and HIV negative people. These results point to an alarmingly high level of risk. sexual behaviors among sexually active age groups in this part of Malawi, a country which still has one of the highest HIV prevalence in sub-Saharan Africa. This calls for continued and better research and control for transmission of STIs in the district.

Elucidating the complex dynamic cellular organization in the hypothalamus is critical for understanding its role in coordinating fundamental body functions. Over the past decade, single-cell and spatial omics technologies have significantly evolved, overcoming initial technical challenges in capturing and analyzing individual cells. These high-throughput omics technologies now offer a remark.ble opportunity to comprehend the complex spatiotemporal patterns of transcriptional diversity and cell-type characteristics across the entire hypothalamus. Current single-cell and single-nucleus RNA sequencing methods comprehensively quantify gene expression by exploring distinct phenotypes across various subregions of the hypothalamus. However, single-cell/single-nucleus RNA sequencing requires isolating the cell/nuclei from the tissue, potentially resulting in the loss of spatial information concerning neuronal network.. Spatial transcriptomics methods, by bypassing the cell dissociation, can elucidate the intricate spatial organization of neural network. through their imaging and sequencing technologies. In this review, we highlight the applicative value of single-cell and spatial transcriptomics in exploring the complex molecular-genetic diversity of hypothalamic cell types, driven by recent high-throughput achievements.

Cloning ＆ sequence identification of Hsp27 gene and expression analysis of the protein on thermal stress in Lucilia cuprina

Hsp27, a highly conserved small molecular weight heat shock.protein, is widely k.own to be developmentally regulated and heat inducible. Its role in thermotolerance is also implicated. This study is a sequel of our earlier studies to understand the molecular organization of heat shock.genes/proteins and their role in development and thermal adaptation in a sheep pest, Lucilia cuprina （blowfly）, which exhibits unusually high adaptability to a variety of environmental stresses, including heat and chemicals. In this report our aim was to understand the evolutionary relationship of Lucilia hsp27 gene/protein with those of other species and its role in thermal adaptation. We sequence characterized the Lchsp27 gene （coding region） and analyzed its expression in various larval and adult tissues under normal as well as heat shock.conditions. The nucleotide sequence analysis of 678 bps long-coding region of Lchsp27 exhibited closest evolutionary proximity with Drosophila （90.09%）, which belongs to the same order, Diptera. Heat shock.caused significant en- hancement in the expression ofLchsp27 gene in all the larval and adult tissues examined, however, in a tissue specific manner. Significantly, in Malpighian tubules, while the heatinduced level ofhsp27 transcript （mRNA） appeared increased as compared to control, the protein level remained unaltered and nuclear localized. We infer that Lchsp27 may have significant role in the maintenance of cellular homeostasis, particularly, during summer months, when the fly remains exposed to high heat in its natural habitat.

genealogy and phylogeography of Cyprinid fish Labeo rohita （Hamilton, 1822） inferred from ATPase 6 and 8 mitochondrial DNA gene analysis

ATPase 6/8 gene （842 bp） of mitochondrial DNA was sequenced in Labeo rohita samples （n = 253） collected from nine rivers belonging to four river basins; Indus, Ganges, Brahmaputra and Mahanadi. Analysis revealed 44 haplotypes with high haplotype diversity （Hd） 0.694 and low nucleotide diversity （π） 0.001. The within population variation was larger （83.44%） than among population differences （16.56%）. The mean Fsv value （0.166; P 〈 0.05） for overall populations revealed moderate level of genetic structuring in the wild L. rohita populations. The haplotype network.presented a single clade for wild L. rohita population, from different rivers. Negative values for Fu＇s index （Fs）, mismatch distribution analysis indicated period of expansion in L. rohita population. The time after recent expansion was estimated for each population, between 0.042 to 0.167 mya. The pattern of Isolation by Distance （IBD） was not significant （r = -0.113, P 〈 0.287）, when all the sampling locations were compared （Mantel test）, however, when an outlier （Indus, Brahmaputra and Mahanadi） was removed from the whole population set, a clear positive correlation between pairwise Fsv and geographic distance （k.） was seen. The analysis of data demonstrated that ATPase6/8 gene polymorphism is a potential mark.r to understand genetic population structure of wild L. rohita existing in different rivers. The study identified population substructure in wild L. rohita with common ancestral origin [Current Zoology 60 （4）： 460--471, 2014].

Identification of ExpIdentification of Expressed Resistance gene Analogs from Peanut (Arachis hypogaea L.) Expressed Sequence Tags

Low genetic diversity mak.s peanut （Arachis hypogaea L.） very vulnerable to plant pathogens, causing severe yield loss and reduced seed quality. Several hundred partial genomic DNA sequences as nucleotide-binding-site leucine-rich repeat （NBS-LRR） resistance genes （R） have been identified, but a small portion with expressed transcripts has been found. We aimed to identify resistance gene analogs （RGAs） from peanut expressed sequence tags （ESTs） and to develop polymorphic mark.rs. The protein sequences of 54 k.own R genes were used to identify homologs from peanut ESTs from public databases. A total of 1,053 ESTs corresponding to six different classes of k.own R genes were recovered, and assembled 156 contigs and 229 singletons as peanut-expressed RGAs. There were 69 that encoded for NBS-LRR proteins, 191 that encoded for protein k.nases, 82 that encoded for LRR-Pk.transmembrane proteins, 28 that encoded for Toxin reductases, 11 that encoded for LRR-domain containing proteins and four that encoded for TM-domain containing proteins. Twenty-eight simple sequence repeats （SSRs） were identified from 25 peanut expressed RGAs. One SSR polymorphic mark.r （RGA121） was identified. Two polymerase chain reaction-based mark.rs （Ahsw-1 and Ahsw-2） developed from RGA013 were homologous to the Tomato Spotted Wilt Virus （TSWV） resistance gene. All three mark.rs were mapped on the same link.ge group AhIV. These expressed RGAs are the source for RGA-tagged mark.r development and identification of peanut resistance genes.

Chondrogenesis of periodontal ligament stem cells by transforming growth factor-β3 and bone morphogenetic protein-6 in a normal healthy impacted third molar

The periodontal ligament-derived mesenchymal stem cell is regarded as a source of adult stem cells due to its ***, the proof of chondrogenic potential of the cells is ***,we investigated the chondrogenic differentiation capacity of periodontal ligament derived mesenchymal stem cells induced by transforming growth factor(TGF)-p3 and bone morphogenetic protein(BMP)-*** isolation of periodontal ligament stem cells(PDLSCs) from human periodontal ligament,the cells were cultured in Dulbecco’s modified Eagle’s medium(DMEM) with 20%fetal bovine serum(FBS).A mechanical force initiated chondrogenic differentiation of the *** chondrogenic differentiation,10μg·LTGF-β3 or 100μg·LBMP-6 and the combination treating group for synergistic effect of the growth *** analyzed the PDLSCs by fluorescence-activated cell sorting and chondrogenesis were evaluated by glycosaminoglycans assay,histology,immunohistochemistry and genetic *** showed mesenchymal stem cell properties proved by FACS *** contents were increased 217%by TGF-β3 and 220%by BMP-6. The synergetic effect of TGF-β3 and BMP-6 were shown up to 281%compared to *** combination treatment increased Sox9, aggrecan and collagen II expression compared with not only controls,but also TGF-P3 or BMP-6 single treatment *** histological analysis also indicated the chondrogenic differentiation of PDLSCs in our *** results of the present study demonstrate the potential of the dental stem cell as a valuable cell source for chondrogenesis,which may be applicable for regeneration of cartilage and bone fracture in the field of cell therapy.

The Receptor-Lik. Cytoplasmic k.nase （OsRLCk. gene Family in Rice： Organization, Phylogenetic Relationship, and Expression during Development and Stress

Receptor-lik. cytoplasmic k.nases （RLCk.） in plants belong to the super family of receptor-lik. k.nases （RLk.）. These proteins show homology to RLk. in k.nase domain but lack.the transmembrane domain. Some of the functionally characterized RLCk. from plants have been shown to play roles in development and stress responses. Previously, 149 and 187 RLCk.encoding genes were identified from Arabidopsis and rice, respectively. By using HMM-based domain structure and phylogenetic relationships, we have identified 379 OsRLCk. from rice. OsRLCk. are distributed on all 12 chromosomes of rice and some members are located on duplicated chromosomal segments. Several OsRLCk. probably also undergo alternative splicing, some having evidence only in the form of gene models. To understand their possible functions, expression patterns during landmark.stages of vegetative and reproductive development as well as abiotic and biotic stress using microarray and MPSS-based data were analyzed. Real-time PCR-based expression profiling for a selected few genes confirmed the outcome of microarray analysis. Differential expression patterns observed for majority of OsRLCk. during development and stress suggest their involvement in diverse functions in rice. Majority of the stress-responsive OsRLCk. were also found to be localized within mapped regions of abiotic stress QTLs. Outcome of this study would help in selecting organ/development stage specific OsRLCk.genes/targets for functional validation studies.

Phylogeny of the Insect Homeobox gene (Hox) Cluster

The homeobox （Hox） genes form an evolutionarily conserved family encoding transcription factors that play major roles in segmental identity and organ specification across species. The canonical grouping of Hox genes present in the HOM-C cluster of Drosophila or related clusters in other organisms includes eight “typical” genes, which are localized in the order labial （lab）, proboscipedia （pb ）, Deformed （ Dfd）, Sex combs reduced （Scr）, Antennapedia （Antp）, Ultrabithorux （Ubx）, abdominalA （abdA）, and AbdominalB （AbdB）. The members of Hox cluster are expressed in a distinct anterior to posterior order in the embryo. Analysis of the relatedness of different members of the Hox gene cluster to each other in four evolutionarily diverse insect taxa revealed that the loci pb/Dfd and AbdB, which are farthest apart in link.ge, had a high degree of evolutionary relatedness, indicating that pb/Dfd type anterior genes and AbdB are closest to the ancestral anterior and posterior Hox genes, respectively. The greater relatedness of other posterior genes Ubx and abdA to the more anterior genes such as Antp and Scr suggested that they arose by gene duplications in the more anterior members rather than the posterior AbdB.

Mining Frequent generalized Itemsets and generalized Association Rules Without Redundancy

This paper presents some new algorithms to efficiently mine max frequent generalized itemsets （g-itemsets） and essential generalized association rules （g-rules）. These are compact and general representations for all frequent patterns and all strong association rules in the generalized environment. Our results fill an important gap among algorithms for frequent patterns and association rules by combining two concepts. First, generalized itemsets employ a taxonomy of items, rather than a flat list of items. This produces more natural frequent itemsets and associations such as （meat, milk. instead of （beef, milk., （chick.n, milk., etc. Second, compact representations of frequent itemsets and strong rules, whose result size is exponentially smaller, can solve a standard dilemma in mining patterns： with small threshold values for support and confidence, the user is overwhelmed by the extraordinary number of identified patterns and associations; but with large threshold values, some interesting patterns and associations fail to be identified. Our algorithms can also expand those max frequent g-itemsets and essential g-rules into the much larger set of ordinary frequent g-itemsets and strong g-rules. While that expansion is not recommended in most practical cases, we do so in order to present a comparison with existing algorithms that only handle ordinary frequent g-itemsets. In this case, the new algorithm is shown to be thousands, and in some cases millions, of the time faster than previous algorithms. Further, the new algorithm succeeds in analyzing deeper taxonomies, with the depths of seven or more. Experimental results for previous algorithms limited themselves to taxonomies with depth at most three or four. In each of the two problems, a straightforward lattice-based approach is briefly discussed and then a classificationbased algorithm is developed.
