目的:神经干细胞及其支持因子均与中枢神经损伤后的神经再生有关.但目前有关神经干细胞及其衍生支持因子在体内和脊髓损伤后是否有变化的证据较少。观察神经干细胞及其衍生的神经干细胞支持因子在大鼠脊髓损伤后的变化规律.探讨它们之间的相互关系。方法:实验于2005-09/2006-03在南通大学神经科学研究所实验室完成,①实验材料:清洁级SD大鼠27只由南通大学实验动物中心提供,将12只大鼠分为假手术组和脊髓损伤组用于免疫组织化学.另15只用于RT-PCR检测.分为假手术组和脊髓损伤组.实验过程中对动物处置符台动物伦理学标准。②实验方法:按Allen重物打击法制备大鼠脊髓损伤模型,假手术组仪切除椎板,未行脊髓打击。用免疫组织化学方法鉴定脊髓组织中神经干细胞标志物nestin、胶质细胞标志物GFAP和神经干细胞衍生的神经干细胞支持因子阳性细胞在脊髓损伤后8.16 d的数量和形态变化;采用半定量RT-PCR检测神经干细胞衍生的神经干细胞支持因子mRNA在脊髓中的表达及损伤后4、8、12、16 d的变化。结果:①假手术组脊髓中央管附近出现巢蛋白阳性细胞脊髓损伤后8 d.巢蛋白阳性细胞增多,细胞变大.向外周迁移,并发出突起,以着力侧为著,胶质纤维酸性蛋白阳性细胞出现增多,并出现表达神经干细胞衍生的神经干细胞支持因子阳性类似神经元形态的细胞。脊髓损伤后16 d,nestin阳性细胞数量明显减少,以GFAP阳性细胞增生为主.神经干细胞衍生的神经干细胞支持因子阳性细胞少见。②RT-PCR检测神经干细胞支持因子mRNA在正常大鼠脊髓中有表达,脊髓损伤后4 d神经干细胞支持因子的mRNA表达上升.损伤8 d为高峰,16 d恢复到近正常水平。结论:①在正常大鼠脊髓中央管附近存在神经干细胞.在脊髓损伤后出现增殖,随后可能分化为神经胶质细胞。②在正常脊髓有神经干细胞支持因子表达.脊髓损伤后神经干细胞支持因子的mRNA表达随神经干细胞增殖和分化而增高和下降.两者在脊髓损伤修复中关系密切。

In this study,we synthesized an organic material with near-infrared emission capabilities:4-(2-(4-(9-(4-(diphenylamino)phenyl)naphtho[2,3-c][1,2,5]thiadiazol-4-yl)phenyl)-1H-phenol-1-ylidene)malononitrile(TPA).Furthermore,TPA-PEG2000 fluorescent nanoparticles were prepared via coating the shells with ***-PEG2000 exhibited strong near-infrared emission near 700 nm,with a photoluminescence quantum yield of 15.09%,indicating a high emission *** biology experiments have confirmed its low toxicity and excellent *** cholesterol and phospholipid levels in liver cancer cell membranes with low sensitivity or high drug resistance lead to increased rigidity,reduced membrane fluidity,reduced endocytic efficiency,and reduced ***,the uptake of TPA-PEG2000 nanoparticles into cells and the near-infrared fluorescence intensity can be used to evaluate the sensitivity of systemic liver cancer treatment in a simple and efficient manner.

目的:比较肌肉注射镇痛、硬膜外镇痛、静脉镇痛、皮下镇痛4种镇痛方法对开胸术后患者镇痛和镇静效果。方法:①选择2004-01/2006-01石河子大学医学院第一附属医院120例行择期胸科手术的患者,男78例,女42例;年龄32～67岁。均符合美国麻醉医师协会标准Ⅰ～Ⅱ级,且对镇痛方法知情同意。按随机数字表法将患者分为4组:肌肉注射镇痛组、硬膜外镇痛组、静脉镇痛组、皮下镇痛组,每组30例。硬膜外镇痛组:于末次硬膜外麻醉后追加负荷量内含吗啡1mg+2.5g/L布比卡因+生理盐水10mL后接镇痛泵行硬膜外镇痛,镇痛药物配方为每100mL中含吗啡6mg+布比卡因125mg+生理盐水至100mL。静脉镇痛组:静脉内推注吗啡负荷量5mg后在静脉穿刺套管针处联接镇痛装置,配方为吗啡50mg+生理盐水至100mL。皮下镇痛组:皮下注射吗啡负荷量5mg后在患者前臂置入静脉穿刺针置于皮下,连接镇痛泵,镇痛药配方为吗啡25mg+生理盐水至100mL。肌肉注射镇痛组:不用镇痛输泵,按0.1mg/kg剂量肌注哌替啶。②于术后4,8,12,24,48h采用Kecle口述描绘评分法评估术后镇痛效果,将疼痛程度(0～9分)分为:0分为无痛,7～9分为重度痛。③于术后48h用Ramsay评分法评估镇静效果(0～6分):1分为不安静烦躁,6分为深睡状态呼唤不醒。④记录术后4,8,12,24及48h患者因镇痛泵持续给药后仍然感觉疼痛而按自控按键单次给镇痛药的次数。⑤观察术后心肺系统主要并发症及不良反应。结果:行胸外科手术患者120例均进入结果分析。①镇痛效果:肌肉注射镇痛组术后4,8,12,24及48hKecle口述描绘评分法评分明显高于其他3组(P<0.01);皮下镇痛组术后12和48h明显高于硬膜外镇痛组(P<0.05),静脉镇痛组术后48h明显高于硬膜外镇痛组(P<0.05)。②镇静效果:肌肉注射镇痛组术后Ramsay评分为1分的比例为27%(8/30),明显高于其他3组(均为0,P<0.01),术后Ramsay评分为2分的比例为67%(20/30),明显低于硬膜外镇痛组[100%(30/30),P<0.01]。静脉镇痛组术后48hRamsay评分为4分的比例明显高于其他3组(均为0,P<0.01)。③患者自己按压给药次数的比较:硬膜外镇痛组患者术后48h内自己按压给药的次数明显少于静脉镇痛组和皮下镇痛组(P<0.01),皮下镇痛组患者术后4,8,24,48h自己按压给药的次数明显多于静脉镇痛组(P<0.05～0.01)。④不良事件和副反应发生情况:静脉镇痛组不良事件和副反应总发生率为40%(12/30),明显高于肌肉注射镇痛组、硬膜外镇痛组、皮下镇痛组[3%(1/30),20%(6/30),13%(4/30),P<0.01]。结论:①静脉镇痛、硬膜外镇痛、皮下镇痛均能达到良好的镇痛效果,镇痛效果优于肌肉注射镇痛。②静脉自控镇痛不良反应最多,镇静作用也最强。

Wogonin(WOG)has been demonstrated to have anti-cancer activity,but the mechanisms remain *** this study,new targets of WOG were predicted for lung cancer,bladder cancer,and colon cancer by using bioinformatics *** might primarily suppress cancers via regulating arachidonic acid,Ras,MAPK,linoleic acid,PI3K Akt,and folate biosynthesis pathways.3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium(MTS)assay showed that WOG inhibited the proliferation of A549 ***-time quantitative reverse transcription PCR(RT-qPCR)results indicated that anti-lung cancer effect of WOG was achieved by regulating the expression of 18 target genes,including AKRIBI0,AKRIC3,BDNF,CAVI,CXCL2,CYP2B6,CYP4F3,DAO,EGF,ENO3,IL6,PLA2GlB,PLA2G2F,PLA2G4A,PTGES,SLCOIBI,SLCOIB3,and *** Kaplan-Meier survival curves further confirmed that DAO,PLA2G1B,SLCO1B3 and TFAP2A were essential targets via which WOG affected lung cancer ***,BDNF,FGF2,and PTGS1 were predicted to be the targets via which WOG alleviated cancer proliferation and invasion in bladder *** for colon cancer,WOG might induce autophagy and inhibit proliferation by down-regulating NTF4 and *** study will provide clue for using wOG as a promising antineoplastic agent in basic and translational research,and bring light to the application of herbs containing WOG as food supplements.

The critical factor of spatial constraint,provided by the external confinement(e.g.,matrix),is often overlooked during photodynamic inactivation,despite playing a crucial role in determining the molecular photophysical process and subsequent antipathogen ***,as a proof-of-concept model,we employed two types of polymers with varying interaction energies with dopants to investigate the intrinsic relationship between spatial constraint and the essential excited-state behaviors of doped photosensitizer(4-(2-(5-(4-(diphenylamino)phenyl)thiophen-2-yl)ethyl)-1-methylquinolin-1-ium iodine,TPP).Through experimental investigation and theoretical calculations,we found that TPP tends to remain in the excited state for a shorter dwell time under weaker spatial constraints due to less restricted molecular motion in polyurethane(PU) ***,the singlet oxygen(^(1)O_(2)) generated from doped-TPP shows a 9.23-fold enhancement in PU than in the polyvinylchloride(PVC) *** light irradiation,the PU@TPP nanofiber can efficiently eliminate the coronavirus MHV-A59(≥99.9997%) at a 220,000-fold higher concentration than the infected *** antibacterial efficacy has also been demonstrated,with a killing rate of ≥99%.

目的:许多体内实验表明神经干细胞具有向胶质瘤细胞迁移的特性,该特性使神经干细胞有可能成为基因治疗脑胶质瘤潜在的基因携带者。实验拟观察大鼠星形胶质瘤C6细胞诱导体外培养的大鼠神经干细胞的迁移作用,并初步分离C6细胞中诱导神经干细胞迁移的蛋白。方法:实验于2005-01/2006-05在南通大学江苏省神经再生重点实验室完成。实验材料:C6细胞用含10%小牛血清的DMEM/F12培养基进行培养,使用时细胞处于对数生长期。清洁级的新生1周内的SD大鼠由南通大学实验动物中心提供,实验过程中对动物处置符合动物伦理学标准。实验方法:从新生5～7dSD大鼠大脑皮质分离培养、纯化神经干细胞。从SD新生红皮鼠大脑皮质分离、培养、纯化星形胶质细胞。采用"TranswellInserts"细胞培养室培养系统共培养36h,比较C6细胞和大鼠星形胶质细胞对体外培养的大鼠神经干细胞的迁移作用;自然系统聚丙烯酰胺凝胶蛋白电泳分离C6细胞和星形胶质细胞差异蛋白,观察各蛋白凝胶条带对神经干细胞的迁移作用。结果:体外培养C6细胞能诱导神经干细胞迁移,迁移细胞数目与星形胶质细胞相比,差异有显著意义(P<0.001)。将C6细胞中差异蛋白条带与神经干细胞共培养,与对应的星形胶质细胞蛋白条带相比,可观察到与相对分子质量约67000蛋白胶条共培养的神经细胞球在接近蛋白胶条的一侧细胞向胶条迁移生长。结论:C6细胞可以诱导体外培养的神经干细胞迁移,其总蛋白在自然系统聚丙烯酰胺凝胶电泳分离实验中获得的相对分子质量为67000的蛋白组分具有诱导神经干细胞迁移的活性。
