Triple-negative breast cancer(TNBC)is the most challenging breast cancer subtype because of its aggressive behavior and limited therapeutic targets.c-Myc is hyperactivated in the majority of TNBC tissues,however,it has been considered an“undruggable”target due to its disordered ***,we developed an ultrasound-responsive spherical nucleic acid(SNA)against c-Myc and PD-L1 in *** is a self-assembled and carrier-free system composed of a hydrophilic small-interfering RNA(si RNA)shell and a hydrophobic core made of a peptide nucleic acid(PNA)-based antisense oligonucleotide(ASO)and a *** accomplished significant enrichment in the tumor by enhanced permeability and retention(EPR)effect,the controllable release of effective elements by ultrasound activation,and the combination of targeted therapy,immunotherapy and *** study demonstrated significant anti-tumoral effects in vitro and in *** cytometry showed an invigorated tumor microenvironment(TME)characterized by a significant alteration in the composition of tumor-associated macrophages(TAM)and decreased proportion of PD-1-positive(PD-1+)T effector cells after appropriate treatment of the ultrasound-responsive SNA(USNA).Further experiments verified that tumor-conditioned macrophages residing in the TME were transformed into the anti-tumoral *** finding offers a novel therapeutic strategy against the“undruggable”c-Myc,develops a new targeted therapy for c-Myc/PD-L1 and provides a treatment option for the TNBC.

Ischemic stroke is a cerebrovascular disease associated with high mortality and disability rates. Since the inflammation and immune response play a central role in driving ischemic damage, it becomes essential to modulate excessive inflammatory reactions to promote cell survival and facilitate tissue repair around the injury site. Various cell types are involved in the inflammatory response, including microglia, astrocytes, and neutrophils, each exhibiting distinct phenotypic profiles upon stimulation. They display either proinflammatory or anti-inflammatory states, a phenomenon known as ‘cell polarization.’ There are two cell polarization therapy strategies. The first involves inducing cells into a neuroprotective phenotype in vitro, then reintroducing them autologously. The second approach utilizes small molecular substances to directly affect cells in vivo. In this review, we elucidate the polarization dynamics of the three reactive cell populations(microglia, astrocytes, and neutrophils) in the context of ischemic stroke, and provide a comprehensive summary of the molecular mechanisms involved in their phenotypic switching. By unraveling the complexity of cell polarization, we hope to offer insights for future research on neuroinflammation and novel therapeutic strategies for ischemic stroke.

DNA adducts are an important class of *** adducts either as a contact(exposure)biomarkers accurately reflect the exposure dose of chemical poison to evaluate the toxicity of chemical poisons, biological monitoring and analysis of exposure to carcinogenic chemical contaminants to provide effective means for the population, but also as effector markers, screening high-risk groups,provide impor-tant information for cancer risk assessment of the organism, the same time, can be used for the development of rational strategies for cancer prevention and treatment. the vast majority of mutations in human tissues, can be determinedto be *** DNA damage occurs more frequently than *** it is the focus of toxicology research to test DNA adductsand has important application ***, the levels of endogenous DNA adducts in the body are very low,which requires the sensitivity of our detection method to be high and limits of detection to be low and better specificity. On the basis of our laboratory study, we establish and optimize UPLC-ESI-MS/MS method which can detects etheno-DNA adducts(εd A, εd C) in human *** method have a higher sensitivity,lower LOD,good recovery and need small samples(about 20 μg).This method can be applied to quantify etheno-DNA adducts(εdA, εdC) in large human blood samples. The detection limits of εdA and εdC are 1.45 fmol and 1.27 fmol.

In this paper, a rapid and effective high performance liquid chromatography(HPLC) method, using tetraazacalix[2]arene[2]triazine-modified silica gel(NCSi) as solid-phase extraction(SPE) sorbent, was developed for the simultaneous determination of two kinds of sulfonamides antibacteric in environmental water. The homemade NCSi sorbent is a novel multi-interaction and mixed mode functionalized calixarene sorbent, which can enrich different classes of sulfonamides antibacteric. By examining the conditions of the amount of the SPE material, the p H of the solution, the volume of the solution and equilibrium time, we found the optimum SPE conditions. The recoveries of this method ranged from 81.86% to 98.2% and the limits of detection of this method which were determined with signal/noise ratios of 3 ranged from 1.30 to 3.60 ng/mL.

Food safety has as a Global Concern problem, and illicit drugs in food detection and drug residual problem gradually become a hot issue. Albendazole is anthelmintic benzimidazole most widely used as veterinary medicine for the treatment of infections caused *** of albendazole has potential harm to human body, so many countries have regulations maximum residue and the method has established for detecting albendazole and its metabolites in animal tissues. However, most of detection methods had serious impurity interference, so it is necessary to establish a new method for sample preparation. In this paper, a high-performance liquid chromatography with fluorescence detection(HPLC-FLD) method, using acidic alumina and MCX solid-phase extraction(SPE) cartridges was developed for extracting and purifying the albendazole and its metabolites in animal *** limits of detection(LOD) were about 1-10 ng/m L, the limits of quantification(LOQ) were 4.0 and 20.0ng/m L for albendazole and its metabolites. The intraday and interday RSDs(n=6) of peak areas and retention times were in the range of 2.0.-15%. In addition, overall recoveries through the extraction and SPE purification ranged from 78.4% to 102.8%. Compared with others detection methods, the two cartridges(acidic alumina and MCX) in series connection was proposed SPE-HPLC–FLD method had highly effective and good purification effects for trace analysis of the albendazole and its metabolites.

Porous carbon nanofibers(CNFs) using polyacrylonitrile(PAN), Cellulose nanocrystals(CNCs) and Silica nanoparticles(Si NPs) of self-supporting were prepared by electrospining and subsequent stabilization, carbonization, activation and acidification processes. Compared with the traditional fibers,the strength and toughness of the electrospun nanofibers were significantly increased. After treatment,the resultant CNFs showed higher porosity and reduced average diameters compared to as-spun *** surface area and pore volume of 926 m/g and 0.318 cm/g were obtained when composite nanofibers under the optimal proportion were treated with air at 270℃ for 3h and follow on with N2 at 800℃ for 6h and CO for 30 min. The performance of an electrochemical capacitor can be characterized by galvanostatic and cyclic voltammetry methods. The research show that the addition of CNCs and Si NPs resulted in reduced fiber diameters, increased porosity, reduced thermal stabilities and enhanced hardness and slowed cyclization reactions of PAN in the electrospun nanofibers. A new methods is thus provided for CNCs and Si NPs as a low-cost material to prepare high surface areas electrode material and reduce the using of polymer as an industrial waste.

Magnetic solid phase extraction(MSPE) has been introduced for separation and preconcentration of organic and inorganic species from complex matrices. Compared to the conventional extraction methods such asliquid–liquid extraction(LLE) and SPE magnetic solid phasemicro extraction(MSPME)is a promising technique that pro-vides cleaner extracts with less solvent consumption, and notablyimproves analytical parameters. Moreover, it combines the extrac-tion, enrichment, and sample introduction in one single step. And a magnetic adsorbent plays a key role in the MSPE procedure. In the present paper, Tetraoxocalix[2]arence[2]triazine coated magnetic nanoparticles(FeO@SiO@TCAT) was prepared and characterized. The performance using FeO@SiO@TCAT as MSPE sorbent was investigated using five polycyclic aromatic hydrocarbons(PAHs) as *** MSPE conditions were optimized for PAHs in the environmental water. It can achieve the highest recovery when 20 mg FeO@SiO@TCAT was used for PAHs extraction from 400 m L water at p H7. The adsorption equilibration time is 10 min, and 5 mL acetonitrile was used as elution solvent,and then it was evaporated to near dryness by N2 blow, then dissolved in the 1 m L acetonitrile. The 1m L sample solution was determined by HPLC with fluorescence *** method recoveries ranged from 80.1% to 103.3%(RSDs 1.1%-7.3%, n=5) for five PAHs indicating that the developed HPLC method based on FeO@SiO@TCAT MSPE cleanup possessed high *** linear range was between 4-400 pg/m L, the limits of detection of five PHAs were between 0.13-3.60 pg/m L. The method was successfully used for PAH determination in the real environmental water samples. The method showed good extraction efficiency for PAHs due to Tetraoxocalix[2]arence[2]triazine having benzene rings, which interacted with PAHs based on π-πinteraction. The MSPE extraction is simple because of the use of the magnetic nanoparticles FeO@SiO@TCAT. This SPE material can be widely used in the sample

以烟草中含量丰富的大环二萜类化合物2,7,11-西柏三烯-4,6-二醇(CBT)为母体,其C6位羟基经酯化/醚化,合成了28个西柏三烯-4-醇-6-羧酸酯及西柏三烯-4-醇-6-烷氧类化合物.采用核磁共振波谱(NMR)和高分辨质谱(HRMS)对目标化合物进行了结构表征,并初步评价了其生物活性.细胞毒活性测试结果表明,部分化合物对肿瘤细胞的抑制作用与阳性对照顺铂相当,其中α-2,7,11-西柏三烯-4-醇-6-O-苯乙酸酯(1 i)对乳腺癌细胞株MCF-7的IC 50值为15.45μmol/L,α-2,7,11-西柏三烯-4-醇-6-O-(2-氟)苯乙酸酯(1 t)对肺癌细胞株SMMC-7721的IC 50值为11.44μmol/L.
