Over the past two decades, numerous non-coding RNAs(ncRNAs) have been identified in different biological systems including virology, especially in large DNA viruses such as herpesviruses. As a representative oncogenic alphaherpesvirus, Marek’s disease virus(MDV) causes an important immunosuppressive and rapid-onset neoplastic disease of poultry, namely Marek’s disease(MD). Vaccinations can efficiently prevent the onset of MD lymphomas and other clinical disease, often heralded as the first successful example of vaccination-based control of cancer. MDV infection is also an excellent model for research into virally-induced tumorigenesis. Recently, great progress has been made in understanding the functions of ncRNAs in MD ***, we give a review of the discovery and identification of MDV-encoded viral miRNAs, focusing on the genomics,expression profiles, and emerging critical roles of MDV-1 miRNAs as oncogenic miRNAs(oncomiRs) or tumor suppressor genes involved in the induction of MD lymphomas. We also described the involvements of host cellular miRNAs, lincRNAs, and circRNAs participating in MDV life cycle, pathogenesis, and/or tumorigenesis. The prospects, strategies, and new techniques such as the CRISPR/Cas9-based gene editing applicable for further investigation into the ncRNA-mediated regulatory mechanisms in MDV pathogenesis/oncogenesis were also discussed, together with the possibilities of future studies on antiviral therapy and the development of new efficient MD vaccines.

Molecular dynamics simulations of helium transport through inorganic mineral nanopores

Helium transport through nanoscale inorganic mineral pores and pore throats is essential for its overall migration. To elucidate helium's transport dynamics within nanopores, we employed equilibrium and non-equilibrium molecular dynamics simulations to investigate helium's static self-diffusion and pressure-driven flow in quartz slit-shaped nanopores. We also introduced water and various gases, including hydrogen, methane, ethane, nitrogen, and carbon dioxide, into the nanopores to assess their influence on helium transport. Our findings indicate minimal helium adsorption on quartz pore surfaces. Under conditions where the pore size is less than 5 nm and the pressure under 10 MPa, environmental factors markedly influence helium diffusion. Large pore sizes, high temperatures, and low gas pressures enhance helium desorption and facilitate faster diffusion. We observed a positive correlation between helium flow velocity and factors such as pore size, pressure gradient, and surface smoothness of the pores. Notably, the presence of pore water and carrier gases in quartz nanopores, which diffuse more slowly than helium, tends to reduce helium surface adsorption and slow its diffusion. Among the carrier gases studied, nitrogen showed similar adsorption capacity, diffusivity, and stability to helium, while carbon dioxide displayed the highest adsorption capacity and the slowest diffusion rate, markedly differing from helium. Based on the simulation results, we concluded that water and carrier gases primarily function as transport mediums in helium migration, moving together with helium. Nitrogen, which shares similar properties with helium, effectively assists in this co-migration process. Conversely, carbon dioxide, due to its high adsorption capacity and slow diffusion, tends to be lost during co-migration. We rethink the segment anything model(SAM) and propose a novel multiprompt network called COMPrompter for camouflaged object detection(COD). SAM has zero-shot generalization ability beyond other models and can provide an ideal framework for COD. Our network aims to enhance the single prompt strategy in SAM to a multiprompt strategy. To achieve this, we propose an edge gradient extraction module, which generates a mask containing gradient information regarding the boundaries of camouflaged objects. This gradient mask is then used as a novel boundary prompt, enhancing the segmentation process. Thereafter, we design a box-boundary mutual guidance module, which fosters more precise and comprehensive feature extraction via mutual guidance between a boundary prompt and a box prompt. This collaboration enhances the model's ability to accurately detect camouflaged objects. Moreover, we employ the discrete wavelet transform to extract high-frequency features from image embeddings. The high-frequency features serve as a supplementary component to the multiprompt ***, our COMPrompter guides the network to achieve enhanced segmentation results, thereby advancing the development of SAM in terms of COD. Experimental results across COD benchmarks demonstrate that COMPrompter achieves a cutting-edge performance, surpassing the current leading model by an average positive metric of 2.2% in COD10K. In the specific application of COD, the experimental results in polyp segmentation show that our model is superior to top-tier methods as well. In the last decade,numerous of miRNAs have been identified in diverse virus families,particularly in *** herpesvirus2(Ga HV2)is a representativeoncogenic alphaherpesvirus that induces rapid-onset T-cell lymphomas in its natural hosts namely Marek’s disease(MD).In Ga HV2 genome,the presence of26 mature miRNAs derived from 14 precursors assembled in three clusters,namely Meq-cluster,Mid-cluster and LAT-cluster,*** Ga HV2 miRNAs,especially for those in Meq-cluster(***-M4),have been demonstrated to be critical for MD pathogenesis and/or *** downstream Mid-cluster is regulated and transcribed by a same promoter of Meq-cluster,but the role of Mid-clustered miRNAs in Ga HV2 biology remains ***,we have generated Mid-cluster and associated individual miRNA deletion mutants in GX0101 virus,a very virulent(vv)Ga HV2 *** Ga HV2-infected chickens as animal models,we found that deletion of Mid-cluster significantly increased viral pathogenicity as *** to parental GX0101 virus,the individual deletion of miR-M1 and miR-M11separately increased the viral pathogenicity or oncogenecity while the individual deletion of miR-M31 significantly decreased the mortality and gross tumour incidence of virus infected ***,our data confirms that in Ga HV2 infection,the viral oncogene meq is targeted and down-regulated by miR-M11-5p,a miR-M11-derived mature *** the best of our knowledge,this is the first report to present that the Mid-clustered miRNAs possibly act either as potential oncogene or tumour suppressor in MD lymphomagenesis.

Single-atom catalyst has garnered widespread attention to mimic mature enzymes due to its well-defined atomic structure and coordination environments. However, since the carbon–carbon（C–C） coupling reactions require synergistic catalysis of multiple sites, single-atom catalysts suffer from insufficient active sites and unclear reaction mechanisms. Controlling the reaction intermediates in a precisely targeted pocket through careful metal–organic cage design is therefore crucial. Here, we prepare a tetrahedral [Cu6L4]-type boron–imidazolate cage integrating highly active Cu sites and optimized cavity, which exhibits enzyme like specific catalytic performance in electrochemical CO2reduction reaction（CO2RR） to enhance the selectivity of *** analyses and computational calculations suggest that the single Cu site together with neighboring boron–imidazolate ligands provides suitably synergistic effects that enable the energetically favorable formation of an *COCHO intermediate, a key step determining selectivity. As a result, the [Cu6L4]-type cage of BIC-145 achieves a Faradaic efficiency of 28% for C2H4maintaining an average current density of -3.54 mA cm-2over a 5-hour electrolysis period. This work represents the first example for studying single-metal site catalysts with ultra-low coordination numbers through the rational design of metal–organic cages.
