Bone loss from Wnt inhibition mitigated by concurrent alendronate therapy

dysregulated Wnt signaling is associated with the pathogenesis of cancers, fibrosis, and vascular diseases. Inhibition of Wnt signaling has shown efficacy in various pre-clinical models of these disorders. One of the key challenges in developing targeted anti-cancer drugs is to balance efficacy with on-target toxicity. Given the crucial role Wnts play in the differentiation of osteoblasts and osteoclasts, acute inhibition of Wnt signaling is likely to affect bone homeostasis. In this study, we evaluated the skeletal effect of small molecule inhibitor of an o-acyl transferase porcupine（PORCN） that prevents Wnt signaling by blocking the secretion of all Wnts. Micro-computed tomography and histomorphometric evaluation revealed that the bones of mice treated with two structurally distinct PORCN inhibitors LGK974 and ETC-1922159（ETC-159） had loss-of-bone volume and density within 4 weeks of exposure. This decreased bone mass was associated with a significant increase in adipocytes within the bone marrow. Notably,simultaneous administration of a clinically approved anti-resorptive, alendronate, a member of the bisphosphonate family,mitigated loss-of-bone mass seen upon ETC-159 treatment by regulating activity of osteoclasts and blocking accumulation of bone marrow adipocytes. Our results support the addition of bone protective agents when treating patients with PORCN *** of bone toxicity can extend the therapeutic utility of Wnt pathway inhibitors.

An Expertise Management ontology (EMont) is presented with which human expertise is captured. EMont aims at describing and collectively finding improvements for complex situations in which people interact with often conflicting worldviews in order to achieve goals, shared or not. EMont is rooted in systems thinking, in particular Soft Systems Methodology, to be able to accommodate stakeholders’ worldviews and activities in complex situations. Its purpose is to model human activity systems, which is illustrated with the help of a complex situation. EMont forms the heart of the Expertise Management Methodology (EMM). This methodology can be regarded as a framework to build bodies of knowledge for particular domains systematically. EMM favors a research process of abduction. By studying one or more situations, generalized models are developed that explain the observed phenomenon best. The models are then taken as a starting point to conduct additional studies. This cyclic process is repeated thereby strengthening the models. EMont has been applied in several, diverse domains such as flood protection, health care and community resilience, over a period of more than four years. In our experience, EMont is a stable ontology, well-suited to capture human activities and identify widely supported solutions for complex situations, and has proven its value in practice. EMont has been implemented in Semantic Media Wiki to publish bodies of knowledge on the web.

The persistent challenges in attracting and retaining a diverse healthcare workforce, with a specific focus on nurses, have become increasingly pronounced in recent years. These hurdles have been exacerbated by a growing difficulty in retaining young nurses, thereby exacerbating labor shortages driven by demographic shifts and the retirement of experienced nursing professionals. While most research efforts have concentrated on the broader issue of nurse retention, our study is centered on a specific demographic—young nurses. Our research endeavors to shed light on the unique challenges faced by young nurses through a qualitative survey involving nursing students who are simultaneously employed. We seek to discern the multifaceted obstacles they encounter in both their academic environment and the healthcare organizations where they work. While certain challenges are linked to course organization, examinations, and the time required for studying, our respondents overwhelmingly emphasize the pivotal role of the work environment in facilitating the harmonization of work, family, and educational commitments. This reconciliation is achieved through measures such as flexible working arrangements and the efficient organization of nursing duties. The primary objective of our research is to provide insights into how these diverse challenges can be effectively addressed and how a range of measures can significantly contribute to the attraction and retention of nursing students, as well as the long-term retention of nurses within the healthcare system. Our recommendations are intended to be of practical use to a wide array of stakeholders, including academic institutions, particularly colleges and universities offering nursing programs, as well as hospitals, clinics, and other healthcare institutions that hire nurses. Volumetric muscle loss(VML)injuries characterized by critical loss of skeletal muscle tissues result in severe functional *** treatments involving use of muscle grafts are limited by tissue availability and donor site *** this study,we designed and synthesized an implantable glycosaminoglycan-based hydrogel system consisting of thiolated hyaluronic acid(HA)and thiolated chondroitin sulfate(CS)cross-linked with poly(ethylene glycol)diacrylate to promote skeletal muscle regeneration of VML injuries in *** HA-CS hydrogels were optimized with suitable biophysical properties by fine-tuning degree of thiol group substitution to support C2C12 myoblast proliferation,myogenic differentiation and expression of myogenic markers Myod,MyoG and ***,in vivo studies using a murine quadriceps VML model demonstrated that the HA-CS hydrogels supported integration of implants with the surrounding host tissue and facilitated migration of Pax7+satellite cells,de novo myofiber formation,angiogenesis,and innervation with minimized scar tissue formation during 4-week *** hydrogel-treated and autograft-treated mice showed similar functional improvements in treadmill performance as early as 1-week post-implantation compared to the untreated *** together,our results demonstrate the promise of HA-CS hydrogels as regenerative engineering matrices to accelerate healing of skeletal muscle injuries.

In a previous article, a model for the coronavirus pandemic was developed. This model was based on simple, uninhibited population growth with rate of infection assumed to be proportional to the existing infected population. Validity of this model was verified by testing it against the infection case data published by the Center for disease Control and the World Health Organization for the United States and the world, respectively. discrepancies between infection case data and model predictions can be accounted for by implementation of infection prevention measures enforced during the pandemic. The goal of this article is to explore which prevention measures were most effective in reducing the spread of coronavirus in the United States. It turns out that among various prevention measures implemented, lockdown is by far the most effective one.

Structural and Electronic Properties of Impurities on Boron Nitride Nanotube

The structural and electronic properties of molybdenum and magnesium substitution doping in (10,0) boron nitride nanotube (BNNT), are investigated through first-principle calculations. The electronic band structures results indicate that the molybdenum doped systems behave as n-type impurity. However, the magnesium doped systems behave as p-type impurity when magnesium replaces boron, and as a n-type impurity when the magnesium replaces nitrogen. The analysis of the energies formation shows that the molybdenum replacing a boron and nitrogen atoms are more favorable than the magnesium substitution in boron and nitrogen.
