Protective effects of Wuwei Xiaodu Drink against chronic osteomyelitis through Foxp3^(+)CD25^(+)CD4^(+) Treg cells via the IL-2/STAT5 signaling pathway

To explore the effectiveness and safety of a Chinese medicinal decoction Wuwei Xiaodu Drink(WWXDD)in inhibiting chronic osteomyelitis via regulatory T cells signaling.The effective constitutes of WWXDD and osteomyelitis related genes were screened.Target proteins were cross-validated using the Venny database.GO function and KEGG pathway analysis were performed for target proteins,while pharmacological network was constructed.The bone properties were analyzed by HE staining and the concentrations of immune factors were measured by ELISA.The expression of CTLA-4 and Foxp3 mRNA and STAT5,p-STAT5,CTLA-4 and Foxp3 protein were detected using Real-time PCR and Western blot,respectively.FACS was used to analyze the percentages of cells.A total of 117 genes overlapped between 785 target genes of the active compounds of WWXDD and 912 osteomyelitis related genes.Inflammation-related genes,including IL-6,TNFα,IL-1βand IL-2 showed high connection degree in the drug-compound-disease-target network.GO function and KEGG pathway analysis revealed that 117 intersection genes mainly enriched in virus infection related pathways,immune related pathways and chemokine signaling pathway.Furthermore,the development of chronic osteomyelitis was suppressed in model rats after treatment with WWXDD.Meanwhile,the concentrations of IL-2 and CD4^(+)CD25^(+)Foxp3 Treg perr centages together with the levels of p-STAT5,CTLA-4 and Foxp3 were also down-regulated.Furthermore,IL-2 and WWXDD drug-containing serum exhibited opposite effects on regulating IL-2,IL-10,TGF-β1,Foxp3,CTLA4 and STAT5.In addition,a STAT5 phosphorylation inhibitor suppressed the expression of Foxp3 and CTLA-4.WWXDD can treat chronic osteomyelitis through suppressing the main regulating factors of Tregs and interfere its immunodepression.Our results bring a new solution for chronic osteomyelitis.

Evolution of gas kick and overflow in wellbore and formation pressure inversion method under the condition of failure in well shut-in during a blowout

With ongoing development of oil exploration and techniques,there is a significant need for improved well control strategies and formation pressure prediction methods.In this paper,a gas-liquid transient drift flow model was established according to the gas-liquid two-phase flow characteristics during the gas kick.A Roe scheme was used for numerical calculation based on the finite volume method.The changes of bottom-hole pressure,casing pressure,the development law of cross-sectional gas holdup,and gas velocity,along with the vertical well depth,were analyzed through simulation examples.The time-series characteristics of mud pit gain were obtained by adjusting the formation parameter.The complex nonlinear mapping relationship between the formation parameters and the mud pit gain was established.The long short-term memory network(LSTM)of deep learning was used to obtain a formation pressure inversion when the blowout is out of control and the well cannot be shut-in.Experimental data from a well were used to verify the gas-liquid two-phase transient drift flow model based on the finite volume method,demonstrating that this method is reliable,with greatly improved prediction accuracy.This approach provides theoretical support for the early monitoring of gas kick during drilling,and for well-killing design and construction after uncontrolled blowout.

Breast cancer is the most common malignant tumor in women, and the incidence of this disease has increased in recent years because of changes in diet, living environment, gestational age, and other unknown factors. Previous studies focused on cancer cells, but an increasing number of recent studies have analyzed the contribution of cancer microenvironment to the initiation and progression of breast cancer. Cancer-associated libroblasts （CAFs）, the most abundant cells in tumor stroma, secrete various active biomolecules, including extraceHular matrix components, growth factors, cytokines, proteases, and hormones. CAFs not only facilitate the initiation, growth, angiogenesis, invasion, and metastasis of cancer but also serve as biomarkers in the clinical diagnosis, therapy, and prognosis of breast cancer. In this article, we reviewed the literature and summarized the research findings on CAFs in breast cancer.

Manganese oxides are regarded as one of the most promising cathode materials in rechargeable aqueous Zn-ion batteries(ZIBs)because of the low price and high security.However,the practical application of Mn2O3 in ZIBs is still plagued by the low specific capacity and poor rate capability.Herein,highly crystalline Mn2O3 materials with interconnected mesostructures and controllable pore sizes are obtained via a ligand-assisted self-assembly process and used as high-performance electrode materials for reversible aqueous ZIBs.The coordination degree between Mn2+and citric acid ligand plays a crucial role in the formation of the mesostructure,and the pore sizes can be easily tuned from 3.2 to 7.3 nm.Ascribed to the unique feature of nanoporous architectures,excellent zinc-storage performance can be achieved in ZIBs during charge/discharge processes.The Mn2O3 electrode exhibits high reversible capacity(233 mAh g−1 at 0.3 A g−1),superior rate capability(162 mAh g−1 retains at 3.08 A g−1)and remarkable cycling durability over 3000 cycles at a high current rate of 3.08 A g−1.Moreover,the corresponding electrode reaction mechanism is studied in depth according to a series of analytical methods.These results suggest that rational design of the nanoporous architecture for electrode materials can effectively improve the battery performance.

The Viral Load of Epstein-Barr Virus in Blood of Children after Hematopoietic Stem Cell Transplantation

Objective To detect the Epstein-Barr virus(EBV)viral load of children after hematopoietic stem cell transplantation(HSCT)using chip digital PCR(cdPCR).Methods The sensitivity of cdPCR was determined using EBV plasmids and the EBV B95-8 strain.The specificity of EBV cdPCR was evaluated using the EBV B95-8 strain and other herpesviruses(herpes simplex virus 1,herpes simplex virus 2,varicella zoster virus,human cytomegalovirus,human herpesvirus 6,and human herpesvirus 7).From May 2019 to September 2020,64 serum samples of children following HSCT were collected.EBV infection and the viral load of serum samples were detected by cdPCR.The epidemiological characteristics of EBV infections were analyzed in HSCT patients.Results The limit of detection of EBV cdPCR was 110 copies/mL,and the limit of detection of EBV quantitative PCR was 327 copies/mL for the pUC57-BALF5 plasmid.The result of EBV cdPCR was up to 121 copies/mL in the EBV B95-8 strain,and both were more sensitive than that of quantitative PCR.Using cdPCR,the incidence of EBV infection was 18.75%in 64 children after HSCT.The minimum EBV viral load was 140 copies/mL,and the maximum viral load was 3,209 copies/mL using cdPCR.The average hospital stay of children with EBV infection(184±91 days)was longer than that of children without EBV infection(125±79 days),P=0.026.Conclusion EBV cdPCR had good sensitivity and specificity.The incidence of EBV infection was 18.75%in 64 children after HSCT from May 2019 to September 2020.EBV cdPCR could therefore be a novel method to detect EBV viral load in children after HSCT.

The new X-ray imaging and biomedical application beamline BL13HB at SSRF

A new X-ray imaging and biomedical application beamline(BL13HB)has been implemented at the Shanghai Radiation Synchrotron Facility(SSRF)as an upgrade to the old X-ray imaging and biomedical application beamline(BL13W1).This is part of the Phase II construction project of the SSRF.The BL13HB is dedicated to 2D and 3D static and dynamic X-ray imaging,with a field of view of up to 48.5 mm×5.2 mm and spatial resolution as high as 0.8μm.A super-bending magnet is used as the X-ray source in BL13HB,which has a maximum magnetic field of 2.293 T.The energy range of monochromatic X-ray photons from a double-multiplayer monochromator was 8–40 keV,and the white beam mode was provided on the beamline for dynamic X-ray imaging and dynamic X-ray micro-CT.While maintaining the previous experimental setup of BL13W1,new equipment was added to the beamline experimental station.The beamline is equipped with different sets of X-ray imaging detectors for several experimental methods such as micro-CT,dynamic micro-CT,and pair distribution function.The experimental station of BL13HB is designed specifically for various in situ dynamic experiments,and BL13HB has been open to users since June 2021.

Metallographic structure,mechanical properties,and process parameter optimization of 5A06 joints formed by ultrasonic-assisted refill friction stir spot welding

Novel hybrid refill friction stir spot welding （RFSSW） assisted with ultrasonic oscillation was introduced to 5A06 aluminum alloy joints. The metallographic structure and mechanical properties of 5A06 aluminum alloy RFSSW joints formed without ultrasonic assistance and with lateral and longitudinal ultrasonic assistance were compared, and the ultrasonic-assisted RFSSW process parameters were opti- mized. The results show that compared with lateral ultrasonic oscillation, longitudinal ultrasonic oscillation strengthens the horizontal bond- ing ligament in the joint and has a stronger effect on the joint＇s shear strength. By contrast, lateral ultrasonic oscillation strengthens the ver- tical bonding ligament and is more effective in increasing the joint＇s tensile strength. The maximum shear strength of ultrasonic-assisted RFSSW 5A06 aluminum alloy joints is as high as 8761 N, and the maximum tensile strength is 3679 N when the joints are formed at a tool rotating speed of 2000 r/rain, a welding time of 3.5 s, a penetration depth of 0.2 mm, and an axial pressure of 11 kN.

Neuroprotective Effects of Tongmai Yizhi Decoction(通脉益智汤)against Alzheimer's Disease through Attenuating Cyclin-Dependent Kinase-5 Expression

Objectives： To explore the protective effects of Tongmai Yizhi Decoction（通脉益智汤, TYD）, a Chinese herb complex prescription against the impairment of cognitive functions and memory loss in amyloid beta 1–40（Aβ1-40） peptide and ibotenic（IBO）-induced Alzheimer＇s disease（AD） model rats. Methods： The in vivo model was established by injecting Aβ1-40 and IBO into left hippocampal CA1 area of Sprague-Dawley（SD） rat to mimic AD. Totally 32 SD rats were divided into 4 groups, including sham operation group, AD model group, TYD group [AD rats treated with TYD at the dosage of 19.44 g/（kg·d） for 4 weeks] and huperzine A group [AD rats treated with huperzine A at the dosage of 40.5 μg/（kg·d） for 4 weeks]. Spatial learning and memory level was detected by Morris Water Maze test. Histological morphology in the hippocampus was tested by hematoxylin-eosin（HE） staining. Cyclin-dependent kinase-5（Cdk5） protein and gene expression level were investigated by Western blot analysis and real-time quantitative polymerase chain reaction（RT-q PCR）, respectively. Results： Aβ1-40 and IBO treatment induced longer escape latency of rats, compared with sham operation group from day 25（P〈0.01）. However, TYD and huperzine A obviously shortened the escape latency from day 26（P〈0.01）. Moreover, the effect of TYD was similar to huperzine A（P〉0.05）. Furthermore, HE staining also showed that TYD and huperzine A reversed the neuropathological changes in the hippocampus triggered by Aβ1-40 and IBO. TYD and huperzine A effectively reduced the expression levels of Cdk5 protein and gene located in rat hippocampus, compared with the AD model group（P〈0.01）. Conclusion： TYD could be a promising neuroprotective agent for protecting neuron from AD injury through inhibiting Cdk5 expression.

High-throughput genotyping of single-nucleotide polymorphisms in ace-1 gene of mosquitoes using MALDI-TOF mass spectrometry

Acetylcholinesterase （ACHE） plays a vital role in the nervous system of insects and other animal species and serves as the target for many chemical agents such as organophosphate and carbamate insecticides. The mosquito, Culex pipiens complex, a vector of human disease, has evolved to be resistant to insecticides by a limited number of amino acid substitutions in ACHE1, which is encoded by the ace-1 gene. The aims of this study are to identify single nucleotide polymorphism （SNP） sites in the ace-1 gene of the C. pipiens complex and explore an economical high-throughput method to differentiate the genotypes of these sites in mosquitoes collected in the field. We identified 22 SNP sites in exon regions of the ace-1 gene. Four of them led to non-synonymous mutations, that is, Y163C, G247S, C677S and T682A. We used matrix-assisted laser desorption ionization - time-of-flight mass spectrometry for genotyping at these four sites and another site F416V, which was relevant to insecticide resistance, in 150 mosquitoes collected from 15 field populations. We were able to synchronize analysis of the five SNP sites in each well of a 384-well plate for each individual mosquito, thus decreasing the cost to one-fifth of the routine analysis. Heterozygous genotypes at Y163C and G247S sites were observed in one mosquito. The possible influence of the five SNP sites on the activity or function of the enzyme is discussed based on the predicted tertiary structure of the enzyme.

Many diseases and health conditions are closely related to various microbes,which participate in complex interactions with diverse drugs;nonetheless,the detailed targets of such drugs remain to be elucidated.Many existing studies have reported causal associations among drugs,gut microbes,or diseases,calling for a workflow to reveal their intricate interactions.In this study,we developed a systematic workflow comprising three modules to construct a Quorum Sensing-based Drug-Microbe-Disease(QSDMD)database(http://***/),which includes diverse interactions for more than 8,000 drugs,163 microbes,and 42 common diseases.Potential interactions between microbes and more than 8,000 drugs have been systematically studied by targeting microbial QS receptors combined with a docking-based virtual screening technique and in vitro experimental validations.Furthermore,we have constructed a QS-based drug-receptor interaction network,proposed a systematic framework including various drug-receptor-microbe-disease connections,and mapped a paradigmatic circular interaction network based on the QS-DMD,which can provide the underlying QS-based mechanisms for the reported causal associations.The QS-DMD will promote an understanding of personalized medicine and the development of potential therapies for diverse diseases.This work contributes to a paradigm for the construction of a molecule-receptor-microbe-disease interaction network for human health that may form one of the key knowledge maps of precision medicine in the future.
